Background: After anterior cruciate ligament reconstruction (ACLR), chronic changes in knee joint biomechanics during higher level tasks, such as running, may negatively impact long-term knee joint health. Among the factors that contribute to these chronic changes, the influence of quadriceps strength on knee joint biomechanics during running is not well understood. Hypothesis: Higher involved limb quadriceps strength (peak torque and rate of torque development [RTD]) and limb symmetry index (LSI) will be positively associated with greater peak knee flexion angle and peak knee extensor moment during running. Quantile regression for panel data has become a common model of interest for econometricians and statisticians. The theoretical literature has addressed important questions, including identification, estimation, and statistical inference. Although recent advances have enabled practitioners to estimate flexible models under a wide range of assumptions, practical issues have not been fully addressed. In this paper, we offer a guide to empirical practice intended to help applied researchers navigate the challenges of estimation and inference for panel quantile models. High-resolution time series recorded by soil water dielectric sensors are often affected by other processes. Dielectric soil matric potential sensors, such as the TEROS 21, exhibit temperature sensitivity, resulting in deviations of readings from the true value due to soil temperature fluctuations. However, methods for correcting this effect remain limited. The objective of this study was to create a straightforward, scale-based statistical approach to mitigate the influence of daily temperature oscillations on matric potential dielectric readings. The temperature sensitivity correction function (TSCF) identifies, quantifies, and smooths diurnal fluctuations caused by soil temperature dynamics. We provide a detailed description of the …

Apolipoprotein E4 (APOE4) is the strongest risk allele associated with the development of late onset Alzheimer’s disease (AD). Across the CNS, astrocytes are the predominant expressor of APOE while also being critical mediators of neuroinflammation and cerebral metabolism. APOE4 has been consistently linked with dysfunctional inflammation and metabolic processes, yet insights into the molecular constituents driving these responses remain unclear. Utilizing complementary approaches across humanized APOE mice and isogenic human iPSC astrocytes, we demonstrate that ApoE4 alters the astrocyte immunometabolic response to pro-inflammatory stimuli. Introduction: Participatory action research methods like Photovoice are uniquely equipped to identify structural determinants of health based on lived experiences of how they influence proximal and distal population health outcomes. This study uses Photovoice data to examine how structural determinants impact community health needs.
Methods: Qualitative thematic analysis used focus group discussion data (25 transcripts) from four Kentucky counties that embedded Photovoice into their local health department community health assessment processes. Coded excerpts were examined for ways in which participants' community health concerns reflected structural issues rather than individual health behaviors.

Objective: The rise in adolescent handgun carriage over the past decade represents a significant public health concern. Using nationally representative data, this study examines risk factors associated with handgun carriage among U.S. adolescents.
Methods: We used data on adolescents aged 12–17 years from the 2021 and 2022 National Survey on Drug Use and Health ( n = 20,869), with past-year handgun carriage defined as a binary variable. Logistic regression estimated odds ratios (ORs) and 95% confidence intervals (CIs) to determine factors associated with handgun carriage across adolescent age groups (12–13 years, 14–15 years, 16–17 years).

The utilization of extensive datasets, such as those generated by DNA or RNA sequencing, has become a central focus in drug discovery and personalized medicine (PerMed). Nonetheless, these datasets are constrained by the absence of protein-functionality testing, which affects physiological responses. In this study, we employed PamGene PamStation technology to quantify protein function by kinase activity across more than 500 signaling pathways from patients with hepatocellular carcinoma (HCC). An interplay of growth factors and signaling pathways governs the development and maintenance of lymphatic vasculature, ensuring proper fluid homeostasis and immune function. Disruption of these regulatory mechanisms can lead to congenital lymphatic disorders and contribute to various pathological conditions. However, the mechanisms underlying the molecular regulation of these processes remain elusive. Here, we reveal a critical and previously unappreciated role for the signaling scaffold protein Shoc2 in lymphangiogenesis. We demonstrate that loss of Shoc2 results in near-complete loss of lymphatic vasculature in vivo and senescence of lymphatic endothelial cells in vitro. Background/Objectives: Duchenne muscular dystrophy (DMD) is a fatal, progressive neuromuscular disorder caused by mutations in the dystrophin gene, leading to the absence of functional dystrophin protein. As the largest gene in the human genome, the DMD locus is highly susceptible to mutations, contributing to a prevalence of approximately 1 in 3800–6300 live male births worldwide. This review aims to provide a comprehensive and critical synthesis of current and emerging therapeutic strategies for DMD.

Cannabis is increasingly legalized in more areas of the USA and marketed under the misnomer “medical cannabis.” Yet, there are no official dosing guidelines or approved medical uses for plant cannabis by the Food and Drug Administration. Due to significant federal barriers, research on efficacy, safety, and adverse effects of cannabis is limited. Introduction: Pediatric cancer patients suffer mental health deficits. Patients who are vulnerable with respect to socioeconomic or other sociodemographic factors may be at heightened risk for worse mental health outcomes during cancer for many reasons including entering treatment with an existing mental health disorder. The purpose of this study was to describe the prevalence and trajectory of mental health before and after a childhood cancer diagnosis in a socio-demographically at-risk sample.

Background: Colorectal cancer (CRC) incidence and mortality rates continue to be elevated even though effective screening methods are widely available. To increase CRC screening in primary care practices, our team developed a tablet-based digital health program (mPATH) designed to identify patients needing CRC screening, provide education, and empower patients to request a screening test via the program.

Objective: Glucose tolerance (GT) is a major effector for adipose tissue (AT) remodeling in obesity, yet its molecular mechanisms remain incompletely defined. We hypothesized that the biophysical and molecular profiles of AT-derived small extracellular vesicles (sEVAT) change in response to glucose availability and differ by GT status.

This systematic review and meta-analysis synthesized the body of research on behavioral interventions for improving group engagement. Fifty studies were included (n = 10 group designs; n = 40 single case designs [SCDs]). Most research on interventions to improve group engagement was conducted in elementary and middle school general education settings. Class-Wide Function-Related Intervention Teams (n = 6), the Good Behavior Game (n = 6), Tootling (n = 8), and multicomponent approaches (n = 9) were the most commonly reported intervention types. Introduction: Discrimination is the unfair treatment of people based on their identity. Youth who experience discrimination may cope with associated distress by using tobacco products, with amplified health consequences depending on the type of product.
Methods: We used data from the 2023 National Youth Tobacco Survey (n = 22 069) and derived an overall continuous measure from the 15-item Adolescent Discrimination Distress Index, as well as three continuous discrimination distress sub-indices: educational, institutional, and peer (range for each: 0–5). Background: Poppers (alkyl nitrites) are widely used among sexually diverse men to enhance sexual pleasure. However, research has historically been dominated by risk-centered frameworks such as associations with HIV or other STIs, overlooking user motivations related to hedonic reward, intimacy, and agency. This conceptual oversight is particularly evident in the Global South. Objectives: This study examined the relationship between childhood mental health conditions and incident major depression in middle-aged and older adults.
Methods: This longitudinal panel study used 12 years of data from 27,220 adults aged 51 and older in the U.S. Health and Retirement Study. Childhood mental health conditions were assessed retrospectively with a question about having a depression, drug/alcohol, or other emotional problem before age 16. Cox regression was used to estimate hazard ratios (HRs).

In the face of increasingly dire environmental crises, proposed solutions have gravitated toward imperatives for faster, cleaner, and more efficient technologies and resources. While practical solutions are necessary, they are entirely insufficient to deal with an underlying issue driving environmental degradation: unbridled instrumental reason. In this dissertation, I suggest a theoretical posture, cultus, which attempts to balance the necessity of instrumental action with the guiding counterweight of receptivity. Furthermore, I argue that such personal receptivity is uniquely fostered in aesthetic experiences, attuning the individual to the non-identical and renewing a relationship of care.

The rapid proliferation of Internet of Things (IoT) devices and cyber–physical systems (CPS) in domains such as smart healthcare, intelligent transportation, and industrial automation has led to unprecedented volumes of heterogeneous data. While advances in deep learning and large-scale foundation models have enabled powerful data-driven decision making, their deployment in real-world distributed environments remains fundamentally constrained by limited computation, communication bandwidth, energy resources, and data privacy requirements. Electric vehicles (EVs) and rooftop solar photovoltaic (PV) systems are increasingly being integrated into residential settings, creating new opportunities for vehicle-to-grid (V2G) and vehicle-to-home (V2H) operations. In these systems, the EV battery functions as a controllable energy storage unit that can charge from the grid or PV and discharge energy to supply household load or export to the grid for a profit. By intelligently scheduling this bidirectional power exchange, households can reduce electricity costs and enhance PV utilization. Scientific simulations and instruments now produce data at rates that overwhelm the storage, memory, and network subsystems of modern high-performance computing (HPC) facilities. Error-bounded lossy compression reduces data movement costs while bounding reconstruction error, yet three barriers limit its adoption in mission-critical workflows: existing compressors cannot guarantee the accuracy of domain-specific quantities of interest (QoIs) derived from compressed data; compression-induced artifacts such as posterization, blocking, and interpolation banding erode user confidence in decompressed fields; and significant compressibility in the quantization index arrays of interpolation-based pipelines remains unexploited. Task-oriented dialog (TOD) systems enable conversational interfaces for complex tasks like flight booking and restaurant reservations. However, deploying TOD systems at scale faces three critical barriers: scalability, generalization, and evaluation. Scalability is primarily restricted by the human-annotation bottleneck, as current systems depend on vast quantities of manually labeled data for every new domain, making deployment prohibitively expensive. Generalization remains a persistent challenge, as systems optimized for known domains often suffer significant performance degradation when encountering new, unseen ones. The integration and modeling of high-dimensional, heterogeneous biological data remain central challenges in computational biology due to complex feature dependencies and pervasive missingness. This dissertation addresses these challenges by developing novel generative frameworks for multimodal data reconstruction, imputation, and interaction prediction. In generative modeling, we focus on capturing structural and causal dependencies in sparse biological systems. We first introduce CausalGeD, a causality-aware diffusion framework that leverages Granger-causal attention for biologically coherent spatial gene expression generation. For over a century, archaeological investigations in the Lower Ohio River Valley have provided extensive evidence of Indigenous Ancestors’ lifeways and practices. However, interpretations of shell-bearing sites have largely been shaped by Eurocentric and Western perspectives, often framing Indigenous actions as responses to environmental shifts rather than as expressions of dynamic social, spiritual, and ecological relationships. Within this historical context, explanations for the relatively abrupt decline of freshwater mollusk collection at the end of the Archaic Period (ca. 3000 cal yr BP) have primarily emphasized environmental change, particularly fluctuations in riverine conditions and resource availability. Vision-based monitoring of Wire Arc Additive Manufacturing (WAAM) using supervised deep learning represents the state of the art in anomaly detection, but such approaches require large labeled datasets that are costly to obtain and typically limited to laboratory conditions. To address these limitations, this work proposes a hybrid deep learning–statistical process monitoring (SPM) framework tailored to the stochastic nature of conventional arc welding processes such as GMAW-based additive manufacturing, where existing methods often overfit. Tyrosine phosphorylation has emerged as a central regulatory mechanism in innate immunity. Building on our recent studies that Syk and EGFR sequentially phosphorylate TLR9 to fully activate it, we discuss how similar mechanisms operate across other Toll-like receptors and the cytosolic DNA sensor STING. Evidence from complementary systems reveals that receptor and nonreceptor tyrosine kinases, including Src-family kinases, Syk, BTK, and EGFR, form an integrated signaling network that triggers receptor activation, trafficking, and downstream gene expression. Background: Pregnancy requires precisely timed immune adaptations to maintain foetal tolerance while enabling timely initiation of labour, a process often conceptualised as the ‘immune clock’ of pregnancy. Disruption of this immune clock contributes to adverse obstetric outcomes. While maternal opioid use disorder (OUD) is a recognised risk factor for poor maternal and neonatal health, its impact on maternal immune landscape at delivery remains poorly understood.

Ovulatory defects are the leading cause of female infertility. Widespread exposure to endocrine-disrupting chemicals, such as phthalates, may contribute to the high prevalence of failed ovulation among infertile women. Using primary ovarian granulosa cells obtained from women, we tested the hypothesis that exposure to an environmentally relevant mixture of phthalate metabolites (MPTmix) impairs essential mediators of the ovulatory process, including progesterone (P4), progesterone receptor (PGR), and prostaglandins (PGs). The composition of the MPTmix was derived from urinary phthalate levels in women. Aortic aneurysms and dissections are devastating vascular diseases with high mortality, yet no pharmacological therapy has proven effective in halting growth or preventing rupture. Surgical and endovascular repair remain the only treatment options for advanced disease. Animal models have been indispensable in defining mechanisms and testing candidate therapies, but the diversity of protocols, strain-dependent variability, and heterogeneous endpoints complicate interpretation and translation. This review provides an update focused on how to match models to specific research questions.
