Detection Of Alzheimer's Disease And Other Diseases Using An Improved Photoaffinity Labeling Method, Boyd E. Haley

An improved method for diagnosing a disease comprising detecting a disease-specific biochemical marker macromolecule within a sample of extracted cerebral spinal fluid or serum is disclosed. In particular, a radioactively labeled photoaffinity probe is used to diagnose a disease. For instance, Alzheimer's disease can be diagnosed by detecting a disease-specific protein having a molecular weight of about 42,000 daltons, i.e., glutamine synthetase. Also included is a method for detecting other disease states, such as cancer, by detecting the presence or absence of specific nucleotide binding proteins.

Antibodies against HIV-1 proteins in HIV-1-infected individuals share a cross-reactive idiotype defined by the monoclonal antiidiotypic antibody 1F7 (5). Using a computer algorithm based on the molecular recognition theory, regions of inverse hydropathy between the variable sequence of 1F7 and human monoclonal anti-HIV-1 antibodies were identified, which are assumed to be involved in idiotype-antiidiotype contacts. A peptide was designed from the proposed contact in the variable heavy chain framework 3-complementarity determining region 3 (FR3-CDR3) of human antibodies and was synthesized. This peptide is recognized by the antiidiotype 1F7 and inhibits the binding of 1F7 to human anti-HIV-1 antibodies which express …

We have generated an IgG1 murine monoclonal anti-idiotype antibody (Ab2) designated 3H1, which mimics a specific epitope on the carcinoembryonic antigen (CEA). Patients with CEA positive tumors are immunologically "tolerant" to CEA. We used 3H1 as a surrogate for CEA for vaccine therapy of 12 patients with advanced colorectal cancer. Each of the patients received a minimum of four intracutaneous injections of aluminum hydroxide precipitated 3H1 at either 1, 2, or 4 mg dosage per injection. 9 of 12 patients demonstrated anti-anti-idiotypic (Ab3) response to 3H1. All nine patients generated specific anti-CEA antibody demonstrated by reactivity with radiolabeled purified CEA; …

A kit and method of non-destructively radiolabelling a biomolecule substrate by halogenation, including the steps of: a) reacting a secondary amine with a chlorinating agent to produce an N-chloro secondary amine, (b) reacting the N-chloro secondary amine with a salt of a radiolabelled halogen to produce a radiolabelled halogenating agent, (c) reacting a solution of the biomolecule substrate with the radiolabelled halogenating agent, and (d) recovering a radiohalogenated biomolecule from the reaction of step (c). The kit and method allow for more complete iodination with a lessening of side reactions such as decomposition of the biomolecule substrate to be labelled.

Compositions containing as the active ingredient a spin-trapping reagent, preferably α-phenyl butyl nitrone (PBN) or spin-trapping derivatives thereof, in a suitable pharmaceutical carrier for administration to a patient are disclosed for treating or preventin symptoms associated with aging or other conditions associated with oxidative tissue damage. Other spin-trapping agents can also be used, such as 5,5-dimethyl pyrroline N-oxide (DMPO) or α-(4-pyridyl 1-oxide)-N-tert-butylnitrone (POBN), and other spin-trapping derivatives thereof. A method and apparatus are provided for magnetohydrodynamic acoustic-resonance, near-IR spectroscopy. The method includes a step of applying to a subject under study a magnetic field having a strength between 2,00-10,000 gauss, near-IR radiation having a wavelength between 800-3,000 nm and an acoustic wave having a frequency between 10 khz-1 Mhz. The method also includes the steps of inducing vibration of ions in the magnetic field and detecting an electric wave generated magnetohydrodynamically by the acoustic wave induced vibration of the ions. Next is the collecting of the electrical, acoustical and near-IR spectra and the analyzing of the collected spectra. …

Lipoprotein lipase (LPL) is a central enzyme in lipoprotein metabolism and is expressed predominantly in adipose tissue and muscle. In these tissues, the regulation of LPL is complex and often opposite in response to the same physiologic stimulus. In addition, much regulation of LPL occurs post-transcriptionally. The human LPL cDNA is characterized by a long 3′-untranslated region, which has two polyadenylation signals. In this report, human adipose tissue expressed two LPL mRNA species (3.2 and 3.6 kb) due to an apparent random choice of sites for mRNA polyadenylation, whereas human skeletal and heart muscle expressed predominantly the longer 3.6-kb mRNA …

A composition which is a contraceptive with potent anti-HIV activity. The composition is a high energy coprecipitate of nonoxynol-9 oligomers, polyvinylpyrrolidone (PVP) and iodine or PVP-I and shows a pronounced synergistic, anti-HIV effect between the compounds of the composition. A method of obtaining a contraceptive and anti-HIV environment in a female comprising administering to said female an effective amount of a high energy coprecipitate.

Acid mine drainage (AMD) from an abandoned underground coal mine in the Jones Branch watershed in McCreary County, KY, substantially reduced water quality in Jones Branch. Downstream from the mine seeps, the pH was routinely below 4.5 and concentrations of most heavy metals, especially iron, were elevated. A cattail wetland (1,022 m2) was constructed on Jones Branch in 1988-1989 to obviate the effects of the AMD. Chemical transport is being monitored in the root zone of three agricultural management systems at the Ohio Management Systems Evaluation Area (OMSEA). Atrazine and alachlor concentration data from soil cores taken to a depth of 0.9 m and partitioned into the increments of 0.0 to 0.15, 0.15 to 0.3, 0.45 to 0.6, and 0.75 to 0.9 m show the herbicides remained in the top 0.15 m of the profile during the 1991 and 1992 growing seasons. The slow movement of herbicides was partly due to below normal rainfall during the period. Since the herbicides have not been transported out of …

In June 1992, the United States Supreme Court decided Lucas v. South Carolina Coastal Council. The case involved a claim for compensation against the State of South Carolina by a landowner who was prohibited from placing structures on two of his beachfront lots. The Court declared that the landowners must be compensated when government regulations deprive them of all economically beneficial or productive uses of their property unless the proscribed uses were not permitted as part of their original titles.

The occurrence of Giardia lamblia cysts and Cryptosporidium parvum oocysts was determined from source and treated drinking water supplies at selected water treatment facilities in Eastern Kentucky. Giardia and Cryptosporidium are two human protozoan parasites often found in water supplies due to fecal contamination. Characteristic cysts and oocysts were identified from large volume water samples, typically 100 gallons for raw samples and 1,000 gallons for finished water, using an indirect fluorescent antibody specific for Giardia and Cryptosporidium. Source water from four different water treatment facilities utilizing a reservoir, river, and two creeks were assayed. All sites exhibited Giardia lamblia …

This review discusses recent literature on the chemical and physiological factors that influence the absorption, distribution, and excretion of aluminum in mammals, with particular regard to gastrointestinal absorption and speciation in plasma. Humans encounter aluminum, a ubiquitous yet highly insoluble element in most forms, in foods, drinking water, and pharmaceuticals. Exposure also occurs by inhalation of dust and aerosols, particularly in occupational settings. Absorption from the gut depends largely on pH and the presence of complexing ligands, particularly carboxylic acids, with which the metal can form absorbable neutral aluminum species. Use Of Riparian Vegetated Filter Strips To Reduce Nitrate And Fecal Contamination In Surface Water, Mark S. Coyne, Robert L. Blevins, Rebecca A. Gilfillen

This research assessed fecal bacteria trapping in surface runoff by grass filters and their potential to enhance NO3- removal via denitrification. Grass filter strips 9.0 m long trapped over 99% of the soil in surface runoff in 1992. Fecal coliform removal was less than 75%. In 1993, 9.0 and 4.5 m grass filter strips trapped 99 and 95% of the sediment, respectively. Fecal coliform trapping efficiency was 90% in 9.0 m grass filters and 75% in 4.5 m filters. Fecal streptococci trapping efficiency was 77% in 9.0 m grass filters and only 56% in 4.5 m filters. Fecal …

Increasing evidence suggests heterogeneity in the molecular pathogenesis of cystic fibrosis (CF). Mutations such as deletion of phenylalanine at position 508 (delta F508) within the cystic fibrosis transmembrane conductance regulator (CFTR), for example, appear to cause disease by abrogating normal biosynthetic processing, a mechanism which results in retention and degradation of the mutant protein within the endoplasmic reticulum. Other mutations, such as the relatively common glycine-->aspartic acid replacement at CFTR position 551 (G551D) appear to be normally processed, and therefore must cause disease through some other mechanism. Adding carbon-rich materials to fields, like manure, may enhance denitrification. Grass filters, which are used to trap surface runoff from these fields, may also provide a carbon-rich environment that favors water infiltration and denitrification. Nitrous oxide (N2O) may be evolved these settings. It is a radiatively important trace gas and intermediate in the denitrification pathway and several other microbial processes. We measured N2O flux, after simulated rain, using a soil cover technique in poultry-manured plots and grass filters receiving their runoff.
