The purpose of this case study is to understand how management and leadership ideas that were present in Toyota Motor Manufacturing Kentucky influenced management and leadership practices adopted by Scott County Public Schools during 2002-2011. Data for the study were collected during the summer of 2011, using individual and focus group interviews including teachers, administrators, community and Toyota Motor Manufacturing Kentucky employees; on-site participant observations, and documents.

Smoking during pregnancy remains a significant public health issue despite knowledge about the adverse maternal and fetal health effects. This research had six purposes: identifying effective smoking cessation strategies for low income pregnant women; identifying characteristics of Kentucky women who smoke during pregnancy; estimating the role of smoking on birth outcomes in Kentucky; exploring the impact of tobacco reduction on birth outcomes; identifying the characteristics of women participating in the Giving Infants and Families Tobacco-free Starts (GIFTS) pilot program; and evaluating the impact of GIFTS on smoking status and birth outcomes.

This study examines agriculture in the Inner Bluegrass Region of Central Kentucky from 1850 to 1880. It utilizes an agroecological perspective, which interprets agriculture through the lens of ecology, to highlight the complex natural and cultural factors that combined to form one of the nation's most prosperous agricultural systems during the nineteenth century. Chapter One explores the agroecosytem Bluegrass farmers created and maintained, emphasizing dynamics in crop and livestock diversity and agricultural technology. Circadian clocks are responsible for daily rhythms in gastrointestinal function which are vital for normal digestive rhythms and health. The present study examines the roles of the circadian pacemaker, the suprachiasmatic nuclei (SCN), and the sympathetic nervous system in regulation of circadian gastrointestinal rhythms in Mus musculus. Surgical ablation of the SCN abolishes circadian locomotor, feeding, and stool output rhythms when animals are presented with food ad libitum, while restricted feeding reestablishes these rhythms temporarily. In intact mice, chemical sympathectomy with 6- hydroxydopamine has no effect on feeding and locomotor rhythmicity, but attenuates stool output rhythms. Again, restricted …

Disease management along the boundaries of wildlife reserves is a growing conservation problem worldwide, as infected wildlife can migrate outside protected areas and pose a threat to livestock and human health. The bison Bison bison population in Yellowstone National Park has long been infected with Brucella abortus, the bacterium causing bovine brucellosis. Concern over migratory bison transmitting B. abortus to cattle herds on lands adjacent to Yellowstone has led to proposals for bison vaccination. Model simulations suggest that vaccination is unlikely to eradicate B. abortus from Yellowstone bison but could be an effective tool for reducing the level of …

Badges of status are supposed to have insignificant production costs, so use costs are thought to be most important in ensuring signal reliability. Use costs arise from the use of the status signal in social interactions. Social experiences that arise from the use of inappropriate signals in social interactions may drive mechanisms that result in reduced fitness for inappropriate signalers. The role of social control, probing and familiarity in producing use costs was explored. Due to the severity of foodborne pathogen illnesses, the widespread use and implementation of rapid detection methods in food testing labs is increasingly important. The liver is an essential organ for cholesterol homeostasis. If this process becomes dysregulated, cardiovascular disease (CVD) develops. Zinc-fingers and homeoboxes 2 (Zhx2) as an important hepatic gene regulator and contributes to CVD. BALB/cJ mice, with mutant Zhx2 allele, have fewer atherosclerotic plaques compared to other strains on a high fat diet. In my dissertation, I focused on the liver phenotype in BALB/cJ mice on a high-fat diet and found increased liver damage compared to wild-type Zhx2 mice. These data indicates that reduced Zhx2 in the liver leads to CVD resistance, but increases liver damage. Therefore, Zhx2 has an important …

Dietary saturated fat intake contributes to diabetes and cardiovascular disease, as shown in numerous animal and human studies. However, the hypothesis that stearic acid, a saturated fat, has beneficial effects on these conditions has not been adequately tested. Leptin receptor deficient mice (db/db) and wild-type mice were fed either chow or a high fat diet enriched in either stearic acid or oleic acid for ten weeks. The progression of diabetes was evaluated with blood glucose, insulin, and metabolic parameter measurements. Heart failure contributes to nearly 60,000 deaths per year in the USA and is often caused by hypertension and preceded by the development of left ventricular hypertrophy (LVH). LVH is usually accompanied by intensive interstitial and perivascular fibrosis which may contribute to arrhythmogenic sudden cardiac death. Emerging evidence indicates that LV dysfunction in patients and animal models of cardiac hypertrophy is closely associated with perivascular inflammation.

To assess strategies aimed at minimizing the release of trace elements and the impact of disposal of coal waste materials on the environment, two long-term leaching experiments of up to five months duration were performed using waste materials from two plants cleaning high and low sulfur bituminous coal. The organic anion transporting polypeptide 4c1 (Oatp4c1) was previously identified as a novel uptake transporter predominantly expressed at the basolateral membrane in the rat kidney proximal tubules. Its functional role was suggested to be a vectorial transport partner of an apically-expressed efflux transporter for the efficient translocation of physiological substrates into urine, some of which were suggested to be uremic toxins. In vitro studies in polarized cell lines showed that upon transfection rat Oatp4c1 localizes at the apical membrane. Nutrition Education Professionals (NEP) working for Extension Agents of Kentucky were surveyed to evaluate their use of a variety of tools in nutrition education in three areas: nutrition, food safety, and food preparation. The purpose of this research was to determine the perception and usage of video streaming/electronic media among community nutrition professionals as a means to better educate individuals in Kentucky and furthermore, to determine if demographics of NEP affect their utilization of this technology. The purpose of this study is to assess the need for menu labeling of calories based on the ability of food consumers to identify lower calorie options. By surveying consumers in Kentucky and Ohio and health professionals in Kentucky about their awareness of caloric content, basic nutrition knowledge and ability to choose lower calorie options, we can predict the potential benefit of menu labeling initiatives and the amount of education that will be necessary for consumers to effectively use menu labeling to make informed decisions on calorie intake. The body composition of female collegiate athletes was measured using the Bod Pod® device. The sample consisted of 75 student athletes, aged 18 to 22 years old. Five sports at the university level were represented, including basketball, gymnastics, soccer, swimming & diving, and soccer. Participants were measured at the preseason and postseason periods. Overall, participants in all five sports were not found to change significantly in total body mass, fat mass, fat free mass, percent body fat, or body mass index from the preseason period to the postseason period at the alpha = 0.05 level. The objective of this qualitative longitudinal study was to assess stakeholder perceptions after one year of a new school feeding program to contrast and compare to pre-program expectations. Semi-structured interviews were conducted using questions reviewed by an in-country collaborator for cultural sensitivity and appropriateness. Follow up questions were determined from participant response. The interviews were analyzed by performing content analysis using open and axial coding to find key concepts in the data. Algae can be used to treat wastewater and manure while producing a feedstock for renewable energy. Algae require nutrients to achieve their maximum growth and manure could provide those nutrients, thereby reducing the cost of algae production and the impact of manure treatment. Algae concentration during cultivation is a critical variable that is difficult to measure due to the high concentration of suspended solids present in manure. This dissertation addresses methods to measure algae concentration in the presence of manure solids.

Corn nitrogen (N) applications are still done on a field basis in Kentucky, according to previous crop, soil tillage management and soil drainage. Arginine deiminase (ADI), an arginine-metabolizing enzyme involved in cell signaling, is dysregulated in multiple inflammatory diseases and cancers. We hypothesized that pegylated ADI (ADI-PEG) provide protection against colitis.
METHODS: Dextran sodium sulfate colitis was induced in IL-10-deficient and BALB/c (WT) mice. ADI-PEG was administered i.p., and inflammatory mediators and pathology were evaluated.
RESULTS: Acute colitis in mice was manifested by increases in inflammatory biomarkers, such as serum amyloid A (SAA, P < 0.001), IL-12 p40, and disease index (3-Fold). In contrast, ADI-PEG significantly decreased clinical disease index, SAA levels, and inflammatory cytokines in blood as well as in colonic explants. Animals developed moderate (2.2 ± 0.3 WT) to severe (3.6 ± 0.5 IL-10 deficient) colonic pathology; and ADI-PEG treatment significantly improved the severity of colitis (P < 0.05). Marked infiltration of CD68+ macrophages and iNOS expression were detected in colonic submucosa in colitic animals but not detected in ADI-PEG-treated animals.
CONCLUSION: ADI-PEG attenuated inflammatory responses by suppression of macrophage infiltration and iNOS expression in colitic animals. ADI-PEG can serve as a potential therapeutic value in IBD.

Over the last three decades, neoliberal restructuring of the economy created a symbiosis of debt and discipline. New legal regimes and strategic use of monetary policy displaced Keynesian welfare, facilitated financialization of the economy, broke the power of organized labor, and expanded debt to sustain aggregate demand. Public laws and policies created afield of possibility within which financial markets extended their reach and brought ever-increasing sections of the working classes and the marginalized within the ambit of the credit economy.
