Prostate cancer is the second leading cause of cancer death among men in the United States. The androgen receptor (AR) antagonist enzalutamide is an FDA-approved drug for treatment of patients with late-stage prostate cancer and is currently under clinical study for early-stage prostate cancer treatment. After a short positive response period to enzalutamide, tumors will develop drug resistance. In this study, we uncovered that DNA methylation was deregulated in enzalutamide-resistant cells. DNMT activity and DNMT3B expression were upregulated in resistant cell lines. Enzalutamide induced the expression of DNMT3A and DNMT3B in prostate cancer cells with a potential role of p53 …

The primary objective was to examine the role of pelvic fluid observed during transvaginal ultrasonography (TVS) in identifying ovarian malignancy. A single-institution, observational study was conducted within the University of Kentucky Ovarian Cancer Screening trial from January 1987 to September 2019. We analyzed true-positive (TP), false-positive (FP), true-negative (TN), and false-negative (FN) groups for the presence of pelvic fluid during screening encounters. Measured outcomes were the presence and duration of fluid over successive screening encounters. Of the 48,925 women surveyed, 2001 (4.1%) had pelvic fluid present during a TVS exam. Because the effects of age, menopausal status, weight and body mass index (BMI) on ovarian detectability by transvaginal ultrasound (TVS) have not been established, we determined their contributions to TVS visualization of the ovaries. A total of 29,877 women that had both ovaries visualized on their first exam were followed over 202,639 prospective TVS exams. All images were reviewed by a physician. While visualization of both ovaries decreased with age, one or both ovaries could be visualized in two of every three women over 80 years of age. The fat acceptance (FA) movement aims to counteract weight stigma and discrimination against individuals who are overweight/obese. We developed a supervised neural network model to classify sentiment toward the FA movement in tweets and identify links between FA sentiment and various Twitter user characteristics. We collected any tweet containing either “fat acceptance” or “#fatacceptance” from 2010–2019 and obtained 48,974 unique tweets. We independently labeled 2000 of them and implemented/trained an Average stochastic gradient descent Weight-Dropped Long Short-Term Memory (AWD-LSTM) neural network that incorporates transfer learning from language modeling to automatically identify each tweet’s stance toward the FA movement. Mild cognitive impairment (MCI) or dementia often leads to behavioral and psychiatric symptoms of dementia (BPSD). Sensory processing abnormalities may be associated with BPSD. The purpose of this study was to explore relationships among sensory processing, behavior, and environmental features within the homes of people with MCI or dementia. This project used mixed methods to assess participants’ sensory processing, care partner perspectives on behaviors, and in situ observations of the home environment. Altered fatty acid metabolism continues to be an attractive target for therapeutic intervention in cancer. We previously found that colorectal cancer (CRC) cells with a higher metastatic potential express a higher level of fatty acid translocase (CD36). However, the role of CD36 in CRC metastasis has not been studied. Here, we demonstrate that high expression of CD36 promotes invasion of CRC cells. Consistently, CD36 promoted lung metastasis in the tail vein model and GI metastasis in the cecum injection model. There is a growing trend in Australia and other developed countries for deintensification of animal production from grasslands and for grasslands to be used for purposes other than livestock grazing. These shifts are occurring for a variety of reasons including unsustainable land management practices, environmental pollution, loss of conservation value, poor commodity prices and declining rural infra-structure and communities. Gadolinium, and possibly praseodymium, are relatively enriched in the magnetic 24 fractions of Class F fly ashes from Central Appalachian coal sources. Approximately 10-15% of American women will be raped by a spouse in their lifetime (Bureau of Justice Statistics, 2002). Research suggests that the American public regards spousal rape as wrong but not rape (Lynch et al., 2017). Research on spousal rape has centered on heterosexual couples despite a higher likelihood that LGBTQ+ individuals will experience severe intimate partner violence (IPV) (Rollé et al., 2019). Coffee production in Vietnam has increased almost 100-fold within the past three decades, positioning it to become the second-largest coffee exporter in the world after Brazil. The U.S. is a top market for Vietnamese coffee. In this study, we estimate a conditional demand system of expenditure share equations for coffee from Vietnam, Brazil, Colombia, and an aggregate of seven other countries in the U.S. coffee market using the Linear Approximation Almost Ideal Demand System (LA/AIDS) and based on monthly time-series data from 2000 to 2020 with a total of 252 observations. The first article uses a comparative analysis tool to analyze the potential of spent hemp as an animal feed or feed ingredient based on its yield, nutritional composition and market price. The current dissertation, based on three separate and yet closely related studies, tested the longitudinal dynamic relationships of maternal and paternal parenting factors with adolescent deviance, the direct links as well as underlying mechanisms. Study 1, based on longitudinal data collected from 570 early adolescents, part of the Brno Longitudinal Study of Youth (BLSY), used latent growth modeling and tested the development of three parenting measures, including parental solicitation, knowledge, and peer approval, and deviance during early adolescence, as well as the developmental (i.e., growth to growth) links of these three parenting measures with adolescent deviance over time. Artificial intelligence (AI) and other advanced technologies pose new questions for philosophers of science regarding epistemology, science and values, and the history of science. I will address these issues across three essays in this dissertation. The first essay concerns epistemic problems that emerge with existing accounts of scientific explanation when they are applied to deep neural networks (DNNs). Causal explanations in particular, which appear at first to be well suited to the task of explaining DNNs, fail to provide any such explanation. Background: Students with complex communication needs (CCN) rely on the use of augmentative and alternative communication (AAC) to support their communication; however, many students are leaving high school without an effective form of communication. To build capacity and increase school professionals’ knowledge and self-efficacy in AAC, this thesis used an adaptation of Project ECHO (Extension for Community Healthcare Outcomes) for use with AAC.

Methods: Using a post then pre-retrospective survey, participants self-reported changes in knowledge, confidence, and overall satisfaction with ECHO sessions. We investigated if listening to songs changes breathing pattern which changes autonomic responses such as heart rate (HR) and heart rate variability (HRV) or change in breathing pattern is a byproduct of listening to songs or change in breathing pattern as well as listening to songs causes changes in autonomic responses. Seven subjects (4 males and 3 females) participated in a pilot study where they listened to two types of songs and used a custom developed biofeedback program to control their breathing pattern to match the one recorded during listening to the songs.

Appalachian counties within Kentucky have higher rates of HPV-related cancers, including cervical cancer. HPV vaccines are effective at reducing the risk of cervical cancer, but vaccination coverage in Appalachia remains low. The proposed program relies on recommended strategies to improve vaccine uptake by increasing access and demand for HPV vaccines. This will be done through school vaccine clinics and parent education on HPV, cervical cancer, and HPV vaccines.

Objectives: Cancer of the endometrium is the most common female reproductive cancer in the United States, but is understudied in relation to stage at diagnosis, which is a potential predictor of survival. BACKGROUND: Continuous glucose monitoring (CGM) improves diabetes control by reducing HbA1c and decreasing rates of hypoglycemia. Despite clinical benefits and clear guidelines from the American Diabetes Association, <20% of patients with type 1 diabetes mellitus (T1DM) use CGM. Studies report disparities in CGM among youth with T1DM by race/ethnicity, socioeconomic status, and region, but, insight into patterns of CGM utilization in adults with T1D is limited. METHODS: This project retrospectively analyzed CGM utilization among individuals with T1DM age 18-64 using the Truven MarketScan® Commercial Claims and Encounters (CCAE) database from 1/1/2016-12/31/2018. CGM utilization was determined using National Drug Code (NDC), Current Procedural Terminology (CPT), or Healthcare Common Procedure Coding system II (HCPCS II) claim identifiers. Individuals with >365 days of continuous medical and pharmacy insurance eligibility were followed from their first Type 1 diabetes-related diagnosis code until the end of the study period or >120 day gap in coverage. Using descriptive statistical analysis, CGM utilization was examined in the overall population and was stratified by patient demographics (age, sex, region), and clinical characteristics (insulin pump, diabetes complications). Black people die at the hands of police at a disproportionate rate. In the United States, Black citizens are three times more likely to be killed by police than White citizens. This, along with other factors, has caused a lack of trust, legitimacy, and relationships between police and the Black community. Examining this problem from a socio-ecological and social constructivist perspective suggests that the solution encompasses the use of community feedback and experiences to build and develop a model of community-oriented policing that addresses the issues of the community being served.
