Reactive Oxygen Species Mediate Tnfr1 Increase After Trpv1 Activation In Mouse Drg Neurons, Fei Ma, Liping Zhang, Karin N. Westlund
Physiology Faculty Publications
Background: Transient receptor potential vanilloid subtype 1 (TRPV1) is activated by low pH/protons and is well known to be involved in hyperalgesia during inflammation. Tumor necrosis factor α (TNF-α), a proinflammatory cytokine, is involved in nociceptive responses causing hyperalgesia through TNF receptor type 1 (TNFR1) activation. Reactive oxygen species (ROS) production is also prominently increased in inflamed tissue. The present study investigated TNFR1 receptors in primary cultured mouse dorsal root ganglion (DRG) neurons after TRPV1 activation and the involvement of ROS. C57BL/6 mice, both TRPV1 knockout and wild type, were used for immunofluorescent and live cell imaging. The L4 and …

This analysis describes the organizational and operational characteristics of local public health agencies participating in an initial cohort of five (5) public health PBRNs in the U.S. We examine variation in practice settings within and between PBRNs; compare practice settings to state and national norms; and identify opportunities for comparative research that can be conducted through PBRNs.
Externalities In The Classroom: How Children Exposed To Domestic Violence Affect Everyone’S Kids, Scott E. Carrell, Mark L. Hoekstra
Externalities In The Classroom: How Children Exposed To Domestic Violence Affect Everyone’S Kids, Scott E. Carrell, Mark L. Hoekstra
University of Kentucky Center for Poverty Research Discussion Paper Series
There is widespread perception that externalities from troubled children are significant, though measuring them is difficult due to data and methodological limitations. We estimate the negative spillovers caused by children from troubled families by exploiting a unique data set in which children’s school records are matched to domestic violence cases. We find that children from troubled families significantly decrease their peers’ reading and math test scores and increase misbehavior in the classroom. The achievement spillovers are robust to within-family differences and when controlling for school-by-year effects, providing strong evidence that neither selection nor common shocks are driving the results.

BACKGROUND: Combination therapy in managing psychiatric disorders is not uncommon. While combination therapy has been documented for depression and schizophrenia, little is known about combination therapy practices in managing attention-deficit/hyperactivity disorder (ADHD). This study seeks to quantify the combination use of ADHD medications and to understand predictors of combination therapy.
METHODS: Prescription dispensing events were drawn from a U.S. national claims database including over 80 managed-care plans. Patients studied were age 18 or over with at least 1 medical claim with a diagnosis of ADHD (International Classification of Diseases, Ninth Revision, Clinical Modification [ICD-9-CM] code 314.0), a pharmacy claim for …

Phylogenetic Analyses Reveal Monophyletic Origin Of The Ergot Alkaloid Gene Dmaw In Fungi, Miao Liu, Daniel G. Panaccione, Christopher L. Schardl
Plant Pathology Faculty Publications
Ergot alkaloids are indole-derived mycotoxins that are important in agriculture and medicine. Ergot alkaloids are produced by a few representatives of two distantly related fungal lineages, the Clavicipitaceae and the Trichocomaceae. Comparison of the ergot alkaloid gene clusters from these two lineages revealed differences in the relative positions and orientations of several genes. The question arose: is ergot alkaloid biosynthetic capability from a common origin? We used a molecular phylogenetic approach to gain insights into the evolution of ergot alkaloid biosynthesis. The 4-γ,γ-dimethylallyltryptophan synthase gene, dmaW, encodes the first step in the pathway. Amino acid sequences deduced …

The use of hormonal contraception is commonly associated with weight gain. The purpose of this retrospective study was to explore the association between the use of hormonal contraception and weight change in an outpatient adolescent patient population in Lexington, Kentucky. Data were gathered from the charts of 259 adolescent female patients seen in an outpatient adolescent clinic for up to 72 months. At the initial visit, all patients were evaluated for risk-taking behavior and received preventive counseling. Patients were categorized as either contraception users or non-users (control group). Contraception users were subdivided into Depot Medroxyprogesterone Acetate (DMPA) user group or …

The gynecologist may be the only physician that an adolescent depends on for primary and reproductive healthcare services. Because adolescent females often make personal healthcare decisions without the benefit of supportive parents/caregivers, it is imperative that the gynecologist understands the developmental issues encountered during this phase of life. The objective of this study is to identify cited complaints of adolescent females seeking reproductive care and, thus, provide gynecologists with needed information about the medical/mental health issues that may be encountered during routine reproductive care exams. The study group was adolescent females between the ages of 12 and 20 years old. …

Driver Of A Semi-Tractor Pulling Twin Trailers Fatally Injured In Single Vehicle Crash, Kentucky Injury Prevention And Research Center
Driver Of A Semi-Tractor Pulling Twin Trailers Fatally Injured In Single Vehicle Crash, Kentucky Injury Prevention And Research Center
Fatality Case Reports--Motor Vehicle
In the spring of 2008, a 40-year-old tractor-trailer driver died after his vehicle left the interstate highway, crashed, and was engulfed in flames. The tractor drifted over the fog line and rumble strip onto the right shoulder, began to skid and left the highway. The tractor slid through the grass beside the roadway, struck a concrete culvert, sideswiped a small tree and then hit a large tree head-on. The tractor and trailers then skidded back down the embankment toward the highway where the entire unit came to rest. The tractor and lead trailer overturned onto the driver’s side. The second …

The isolation and characterization of cDNAs encoding poly(ADP-ribose) glycohydrolase (PARG) enzymes and the amino acid sequences of PARGs from several species are described. PARG is involved in the cellular response to DNA damage and its proper function is associated with the body's response to neoplastic disorder inducing agents and oxidative stress. Expression vectors containing the cDNAs and cells transformed with the vectors are described. Probes and primers that hybridize with the cDNAs are described. Expression of the cDNA in E. coli results in an enzymatically active protein of about 111 kDa and an active fragment of about 59 kDa. Methods …

A Gene Expression Predictor Of Response To Egfr-Targeted Therapy Stratifies Progression-Free Survival To Cetuximab In Kras Wild-Type Metastatic Colorectal Cancer, Justin M. Balko, Esther P. Black
Pharmaceutical Sciences Faculty Publications
BACKGROUND: The anti-EGFR monoclonal antibody cetuximab is used in metastatic colorectal cancer (CRC), and predicting responsive patients garners great interest, due to the high cost of therapy. Mutations in the KRAS gene occur in ~40% of CRC and are a negative predictor of response to cetuximab. However, many KRAS-wildtype patients do not benefit from cetuximab. We previously published a gene expression predictor of sensitivity to erlotinib, an EGFR inhibitor. The purpose of this study was to determine if this predictor could identify KRAS-wildtype CRC patients who will benefit from cetuximab therapy.
METHODS: Microarray data from 80 metastatic CRC patients subsequently …

Coal Truck Driver Dies In Rollover After Tire On Trailer Fails, Kentucky Injury Prevention And Research Center
Fatality Case Reports--Motor Vehicle
In the fall of 2006, a twenty-three year old male tractor-trailer driver who was hauling coal, died when a rear outer tire on the trailer failed. After the tire failed, the trailer slipped off the right side of the road, overturned, and went over an embankment. The trailer remained attached to the tractor as both went over the embankment, flipping the entire unit upside down. A witness called emergency medical services who arrived and called the county coroner. The coroner arrived and declared the driver dead at the scene. It was determined by Kentucky State Police that the driver was …

University Of Kentucky Adult Learning Focused Institution (Alfi) Task Force: Findings And Recommendations, Randolph Hollingsworth
Undergraduate Education White Papers
This report to Provost Kumble Subbaswamy details the wide-ranging work over two years of the UK Adult Learning Focused Institution (ALFI) Task Force. The findings in sum assert that the undergraduate population at the University is primarily of a traditional age and that little explicit attention is given to adult learners in UK's stated policies or procedures. The recommendations are that by becoming more "adult friendly" the University could also provide more flexibility and promise for success to all its students.

BACKGROUND: Sporadic Parkinson's disease (PD) is a progressive neurodegenerative disorder with unknown cause, but it has been suggested that neuroinflammation may play a role in pathogenesis of the disease. Neuroinflammatory component in process of PD neurodegeneration was proposed by postmortem, epidemiological and animal model studies. However, it remains unclear how neuroinflammatory factors contribute to dopaminergic neuronal death in PD.
FINDINGS: In this study, we analyzed the relationship among inducible nitric oxide synthase (iNOS)-derived NO, mitochondrial dysfunction and dopaminergic neurodegeneration to examine the possibility that microglial neuroinflammation may induce dopaminergic neuronal loss in the substantia nigra. Unilateral injection of lipopolysaccharide (LPS) …
