Ruminative Thought In Individuals With Borderline Personality Features, Brian Thomas Upton Jan 2011

Ruminative Thought In Individuals With Borderline Personality Features, Brian Thomas Upton

University of Kentucky Master's Theses

Borderline personality disorder (BPD) is characterized by patterns of intense negative affect, interpersonal difficulties, and maladaptive impulsive behaviors, and is associated with impairments in social and occupational functioning. Rumination is a maladaptive form of repetitive thought that maintains and intensifies emotional disturbance and is associated with behavioral dysregulation. This study tested several hypotheses about relationships between rumination and borderline personality features. This study included 117 college student participants, 88 female students and 29 male students, most of whom (84%) identified themselves as Caucasian. Participants completed a series of measures which included a writing sample to sample repetitive thought. Findings consistently …

Development And Characterization Of Novel Monoclonal Antibodies For Studying Prion Pathogenesis, Chu-Chun Weng Jan 2011

Development And Characterization Of Novel Monoclonal Antibodies For Studying Prion Pathogenesis, Chu-Chun Weng

University of Kentucky Master's Theses

Monoclonal antibodies (mAbs) recognizing different regions of PrP are potential tools in the study of prion diseases and immunotherapy. We used shuffled recombinant prion protein containing elk and mouse PrP as antigen to produce monoclonal antibodies in mice. We found that mAb 5C6 mapped to a discontinuous epitope comprised of amino acid 132 and 158 (mouse numbering). Monoclonal anibody 9E9 which maps to a unique N-terminal epitope at amino acid preferentially recognized cervid PrP. In contrast, the epitope of mAb 9H9 is located in the C-terminus and only reacted with mouse and hamster. The epitope for mAb 7H11 appears to …

“Obtuse Women”: Venereal Disease Control Policies And Maintaining A “Fit” Nation, 1920-1945, Evelyn Ashley Sorrell

University of Kentucky Master's Theses

Public health officials and social reformers grew concerned over the prevalence of gonorrhea and syphilis following World War I. The initiatives put in place by authorities to control the spread of venereal disease lacked any concern for women’s health and sought to control their newly found independence and mobility. This thesis examines public health policies related to venereal disease control from 1920-1945 and how these regulations affected women in the United States. Laws and social reform measures such as pre-marital blood tests, the Sheppard-Towner Maternity and Infancy Act, and the use of quarantining prostitutes during World War I and World …

Similarly, heavy viewers perceived medical miracles to occur less often than non-viewers. Interestingly, heavy viewers perceived medical dramas to be less credible than non-viewers. In addition, this …

Combinatorial Biosynthetic Derivatization Of The Antitumoral Agent Gilvocarcin V, Micah Douglas Shepherd Jan 2011

Combinatorial Biosynthetic Derivatization Of The Antitumoral Agent Gilvocarcin V, Micah Douglas Shepherd

University of Kentucky Doctoral Dissertations

Gilvocarcin V (GV), the principal product of Streptomyces griseoflavus Gö 3592 and other Streptomyces spp., is the most prominent member of a distinct class of antitumor antibiotics that share a polyketide derived coumarin-based aromatic core. GV and other members of this class including polycarcin V from Streptomyces polyformus, often referred to as gilvocarcin-like aryl C-glycosides, are particularly interesting because of their potent bactericidal, virucidal and antitumor activities at low concentrations while maintaining low in vivo toxicity. Although the precise molecular mechanism of GV bioactivity is unknown, gilvocarcin V has been shown to undergo a photoactivated [2+2] cycloaddition of …

T-Phylloplanin And Cis-Abienol, Two Natural Products From Tobacco Have Broad Spectrum, Anti-Fungal Activities, Brian Christopher King Jan 2011

T-Phylloplanin And Cis-Abienol, Two Natural Products From Tobacco Have Broad Spectrum, Anti-Fungal Activities, Brian Christopher King

University of Kentucky Doctoral Dissertations

Tobacco phylloplanins (T-phylloplanin) are a group of closely-related glycoproteins that are formed and disposed at the interface between the plant aerial surface (the phylloplane) and the atmosphere. They are synthesized in short procumbent trichomes and are secreted to aerial surfaces where they are thought to serve the plant as a first line of defense against fungal pathogens. Here it is shown using in vitro and in planta assays that tobacco and sunflower phylloplanins have broad-spectrum antifungal activities against spores - and also hyphae for two species - of several true fungi. Field tests show that T-phylloplanin reduces diseases caused by …

Computer Simulation Of A Hollow-Fiber Bioreactor: Heparan Regulated Growth Factors-Receptors Binding And Dissociation Analysis, Changjiang Zhang

University of Kentucky Doctoral Dissertations

This thesis demonstrates the use of numerical simulation in predicting the behavior of proteins in a flow environment.

A novel convection-diffusion-reaction computational model is first introduced to simulate fibroblast growth factor (FGF-2) binding to its receptor (FGFR) on cell surfaces and regulated by heparan sulfate proteoglycan (HSPG) under flow in a bioreactor. The model includes three parts: (1) the flow of medium using incompressible Navier-Stokes equations; (2) the mass transport of FGF-2 using convection-diffusion equations; and (3) the cell surface binding using chemical kinetics. The model consists of a set of coupled nonlinear partial differential equations (PDEs) for flow and …

Effect Of Azithromycin On Macrophage Phenotype During Pulmonary Infections And Cystic Fibrosis, Theodore James Cory Jan 2011

Effect Of Azithromycin On Macrophage Phenotype During Pulmonary Infections And Cystic Fibrosis, Theodore James Cory

University of Kentucky Doctoral Dissertations

Azithromycin improves clinical outcomes in patients with cystic fibrosis (CF), specifically in patients infected with Pseudomonas aeruginosa. Azithromycin shifts macrophage programming away from a pro-inflammatory classical (M1) phenotype, and towards an anti-inflammatory alternative (M2) phenotype; however, little is known about this mechanism, nor of its impact upon immune response to pulmonary infection. We set out to determine the mechanism by which azithromycin is able to alter macrophage phenotype, and assess the effect of azithromycin induced macrophage polarization on inflammation during pulmonary infections.

Utilizing macrophage cell culture, we found that azithromycin increased IKKβ, a signaling molecule in the NFκB pathway, which …

Bayesian Semiparametric Generalizations Of Linear Models Using Polya Trees, Angela Schoergendorfer Jan 2011

Bayesian Semiparametric Generalizations Of Linear Models Using Polya Trees, Angela Schoergendorfer

University of Kentucky Doctoral Dissertations

In a Bayesian framework, prior distributions on a space of nonparametric continuous distributions may be defined using Polya trees. This dissertation addresses statistical problems for which the Polya tree idea can be utilized to provide efficient and practical methodological solutions.

One problem considered is the estimation of risks, odds ratios, or other similar measures that are derived by specifying a threshold for an observed continuous variable. It has been previously shown that fitting a linear model to the continuous outcome under the assumption of a logistic error distribution leads to more efficient odds ratio estimates. We will show that deviations …

Should Pseudoephedrine (Pse) Be Moved From Behind The Counter To Prescription-Only? An Analysis Of Pse As An Otc Medication And A Precursor For Methamphetamine Production, Jerry Dillon

MPA/MPP/MPFM Capstone Projects

Kentucky State Senator Tom Jensen recently sponsored Senate Bill 45, which would change pseudoephedrine (PSE) from an over-the-counter to a schedule V prescription-only drug. The bill failed, but the debate continues as to whether Kentucky should make PSE, a precursor required to manufacture illicit methamphetamine, a controlled drug that is available by prescription only.

Methamphetamine abuse has been increasing in Kentucky as well as in the rest of the country, despite efforts to control the sale of PSE through federal and state legislation. The Combat Methamphetamine Epidemic Act (CMEA) of 2006 and Kentucky’s initiative to electronically monitor the sales of …

U.S. State Employee Pension Systems: An Investigation Into The Causes Of Unfunded Liabilities, Chuck Truesdell Jan 2011

U.S. State Employee Pension Systems: An Investigation Into The Causes Of Unfunded Liabilities, Chuck Truesdell

MPA/MPP/MPFM Capstone Projects

Defined benefit retirement plans for state employees have come under fire, both financially and politically, following recent market volatility and subsequent losses in pension investments. Asset losses matched with liabilities that are set years in advance translate to pressures on state policymakers and plan administrators to either find ways to improve the finances of these pension systems or transition to defined contribution plans that put the financial risk on individual employees rather than the state government. Because most states are legally and contractually obligated to pay retiree benefits regardless of the pension system’s financial condition — even to the point …

Kentucky has 1500 square miles of surface water and 1500 miles of navigable waterways. Fifty-one of the 120 county seats are located on rivers, and over 25 percent of the state’s population lives along waterways. Not surprisingly, flooding is the most common natural hazard in Kentucky, and many structures have been built over the years in floodplain areas. Although many local ordinances have been enacted across the State in recent years to limit new construction in these areas, existing structures still experience frequent flood damages. These structures are required by the federal government to maintain flood insurance, and the National …
