Project-based instruction is an instructional methodology based in constructivism that aligns with recommendations from the American Statistical Association for teaching statistics, including incorporating real-world data and examples, collaboration, and scaffolding (GAISE College Report ASA Revision Committee, 2016; GAISE Steering Committee, 2024; Krajcik & Blumenfeld, 2005; Tishkovskaya & Lancaster, 2012). This study examined current use of project-based instruction in United States higher education courses in statistics and the supports for and challenges to implementing this type of instruction.

This autoethnography examines the impact of intimate partner violence on the educational outcomes of a PhD student. Through a qualitative study of personal narratives and media and document analysis, this research explores the challenges faced by a survivor of assault during an online graduate program, including physical and mental well-being, academic performance, the impact of social support structures, and responses from the University of Kentucky. By integrating lived experiences with existing literature, the study identifies gaps in university policies and offers recommendations for creating more supportive educational environments for online graduate students who have endured intimate partner violence. In Thomas Hardy’s Tess of the D’Urbervilles, a group of fallen women see “themselves and surrounding nature forming an organism of which all the parts harmoniously and joyously interpenetrated each other” (65). As relational, boundary-dissolving, ecological subjects, these women are empowered above society’s judgment, made “sublime as the moon and stars” (65). By analyzing such representations of ecological relationality in Victorian fiction, my dissertation reveals new forms of agency for women and racialized others. This dissertation analyzes the political capacity of feeling in American women’s autobiographical and semi-autobiographical writing from and about their work during the First World War. Women produced emotionally complex accounts that resist easy categorization, and I argue that attending to feeling clarifies not only their emotional experiences with war work but their social and political projects. This dissertation closely examines portrayals and rhetorics of feeling, considering the intersections of gender, class, race, and labor in impacting how feelings are depicted, negotiated, and conveyed. Lake Tanganyika is a large, ancient freshwater body with exceptional biodiversity that is located in the East African Rift System. Lake Tanganyika’s size and longevity make its sedimentary record useful for understanding environmental changes that cross spatial and temporal boundaries. Karst aquifers represent some of the world’s most productive yet vulnerable groundwater systems, supplying freshwater to approximately 10% of the global population across 15% of Earth’s ice-free land surface. Their distinctive hydrological characteristics— including rapid recharge, preferential flow paths, and limited natural filtration— create unique vulnerabilities to both climate variability and contamination that pose significant challenges for sustainable water resource management. This dissertation presents a comprehensive multi-scale analysis of karst aquifer vulnerability through three interconnected studies that examine climate change impacts, microbial contamination dynamics, and contaminant transport processes across diverse geographic and hydrogeologic settings.

Epigenetic clocks are algorithms that use epigenetic biomarkers of aging to calculate an organism’s ‘biological age.’ Through discursive analysis of commercially available epigenetic clock marketing materials, scientific articles, and interviews with actors in the longevity biotechnology industry, this study examines the commercialization of epigenetic clocks in the form of consumer test-kits. The study argues that longevity biotechnology companies leverage the malleable and fluid qualities of epigenetic markers of biological age to extend the domains of profitable therapies. This thesis explores how the Houston Climate Justice Museum (HCJM) functions as a site for environmental/climate justice education through art and community engagement. Drawing on interviews, participant observation, and document analysis, this research examines how the museum’s curating practices generate new understandings of environmental in/justices, fosters local partnerships, and creates a welcoming space for community dialogue. This dissertation examines how geospatial technologies are used to make slums in Mumbai legible, calculable, and hypervisible, and how these practices reshape urban land governance. It situates mapping, remote sensing, and algorithmic systems within their political-economic contexts, showing how technologies intending to see the city often function as instruments of dispossession. At the turn of the twentieth century, rapid industrialization and urban expansion transformed American cities, creating urgent challenges in governance, public health, and environmental management. In response, women across the United States extended their domestic expertise into the public sphere, spearheading initiatives to improve sanitation, waste disposal, and urban infrastructure. This dissertation examines the Louisville Women’s City Club (LWCC) as a case study of municipal housekeeping, demonstrating how middle-class women used their traditional roles to influence systemic reform in Louisville, Kentucky. “A True And Broad Sympathy:” Thomas Hughes, Medievalism, And Social Justice In The Nineteenth Century, Sarah E. Stapleton

Thomas Hughes, author of Tom Brown’s Schooldays (1857), used medievalism to help solve social problems of the Nineteenth Century. This contravenes the traditional view of medievalism, especially Anglo-Saxonism, since others have used it to justify the subjugation of people of lower social status, namely the poor in England, enslaved African Americans in the United States, and Indians under British rule.

While the use of medievalism is veiled in Hughes’ work, the subtext of Tom Brown’s Schooldays demonstrated Hughes’ Anglo-Saxonism. Chapter one estimates the effect on infant health of drinking water contamination attributable to tropical cyclones. This dissertation explores how economic uncertainty and oil price shocks influence the U.S. labor market, focusing on the roles of age distributions, manufacturing characteristics, and labor skills. Chapter One investigates how uncertainty and age distribution affect employment volatility across U.S. states using data from 2000 Q1 to 2017 Q4. The findings show that states with a larger share of prime-aged workers experience lower employment volatility in the face of economic uncertainty. Protein Arginine Methyltransferase 5 (PRMT5) is a member of PRMT family that controls both intrinsic and extrinsic oncogenic processes [1]. Protein arginine methyltransferase 5 (PRMT5) is a type II methyltransferase that exerts widespread influence over gene expression, splicing, DNA repair, and immune regulation through symmetric dimethylation of arginine residues on histone and non-histone proteins [2]. The addition of methyl groups induces gene silencing through producing dictatorial histone marks like H2AR3ne2s, H3R8me2s and H4R3mes [3], which act as epigenetic repressive marks on gene promoters. Prior studies indicated Borrelia burgdorferi SpoVG protein was regulated, SpoVG binds specifically to a number of nucleic acids, and SpoVG binds to the non-variable section of the major virulence factor vlsE. Although a direct transcriptional activator or inhibitor was not elucidated, it is shown that SpoVG is regulated based on cell culture density independent of the growth rate of B. burgdorferi, indicating that SpoVG regulation is outside the scope of the alternative sigma factor RpoS. Using a dCas9 CRISPRi construct to target SpoVG resulted in protein expression trending towards upregulation of the BosR-RpoS regulon, and a marked decrease …

Among the conditions treated is substance use disorder (SUD), which afflicted 48.5 million (16.7%) Americans aged 12 and older in 2023 (NSDUH). This dissertation explores the dynamic interplay between the immune system and the brain across stroke and aging using clinical, post-mortem, and pre-clinical models. The immune system plays critical roles in shaping brain function, responding to injury, and mediating recovery. These studies aim to enhance our understanding of neuroimmune dynamics across spatial and temporal dimensions.

Three related analyses were performed to try to understand the immediate effects of Medicaid expansion on hospital systems in Kentucky. Medicaid expansion led to a large, sudden increase in the number of individuals eligible for various health care needs. This sudden increase in demand for health services lead to initial hypotheses regarding hospitals’ ability to handle this demand. The first analysis examined hospital average length of stay (LOS) using a linear mixed-effects model. There is a specific focus on the impact that gain-of-function SHP-2 mutations may impact binding to DDX3 via conformational change. Using immunoprecipitation protocols as well as western blotting we were able to provide insights into SHP-2 molecular mechanisms and subsequent binding to DDX3.
