Microbiology, Immunology, and Molecular Genetics Faculty Publications

Alcohol-induced dysregulation of microglial activity is associated with neuroinflammation, cognitive decline, heightened risk for neurodegenerative diseases, alcohol dependence, and escalation of alcohol drinking. Given the challenge of longitudinally sampling primary microglia, we optimized an in vitro method to differentiate peripheral blood mononuclear cells (PBMC) from rhesus macaque (RM) into induced microglia-like cells (RM-iMGLs). The RM-iMGLs displayed transcriptional profiles distinct from monocyte progenitors and closely resembling primary microglia. Notably, morphological features showed that differentiated RM-iMGLs derived from subjects with chronic alcohol consumption (CAC), while bigger, exhibited a bipolar-like morphology. Young Black men who have sex with men (YBMSM) in the southern United States, particularly in Fulton County, Georgia, face disproportionately high rates of HIV and unique structural barriers to pre-exposure prophylaxis (PrEP) uptake and adherence. Sustained adherence among YBMSM remains low due to intersecting structural, social, and behavioral barriers. To address these disparities, this paper presents an adapted implementation of the evidence-based M-Cubed (Mobile Messaging for Men) mHealth intervention. Prior studies of M-Cubed and similar mHealth interventions have shown promising outcomes in increasing PrEP adherence among Black MSM populations. Background: New findings indicate that environmental factors, such as air pollution, could contribute to cognitive decline among older adults. Nonetheless, extensive longitudinal studies in the U.S. investigating this connection are few and far between.

Objective: This study assessed the relationship between long-term exposure to ambient air pollutants and cognitive function in older adults in the United States.

Methods: We examined data from the Health and Retirement Study (2010-2018), which included 234,250 observations from adults aged 50 and older. Cognitive performance was evaluated based on immediate and delayed word recall, a 27-point cognitive summary score, backward from 20, and …

Maternal mortality rates rob families across the U.S. of a mother figure, and each death is preventable. The U.S. has the highest maternal mortality rate of high-earning countries. This study assesses the effects of the VIPCorps program, a course on the top 3 risk factors of maternal mortality, on improving knowledge and attitudes towards victims of IPV. Students in helping professions at the University of Kentucky (n = 223) were recruited via on campus advertising and remotely allocated to either the VIPCorps program or a mock maternal mortality training control. Background: This study addresses the gap in education and training for medical intensive care unit (MICU) nurses regarding the care of hospice patients. The traditional medical model of healthcare prioritizes illness treatment, often leading to inadequate end-of-life support and increased hospitalization during patients' final days. The rise of in-hospital hospice programs, or scatter-bed hospice models, highlights the need for nurses to enhance their skills in providing end-of-life compassionate care. The MICU at the University of Kentucky’s Chandler Hospital serves as a focal point for this initiative, given its role in caring for critically ill patients who require hospice attention.

Purpose/objective

We propose a novel lattice deployment for spatially fractionated radiotherapy (SFRT) treatments. In this approach, a larger diameter high-dose sphere is centrally placed in the bulky tumor mass and surrounded by smaller diameter high-dose spheres.

Materials/methods

Thirty SFRT patients (10 head and neck [HN], 10 abdominal/pelvis, and 10 chest/lung cases) treated with an MLC-based crossfire method were retrospectively analyzed. Eleven differential hole-size lattice patterns were benchmarked against the clinically delivered SFRT plans (1 cm diameter cylinders, 2 cm spacing) and the standard uniform lattice pattern (1.5 cm diameter spheres, 3 cm spacing). Purpose: To determine the distributions of published α/β values measured in vitro and the resulting uncertainties in the equivalent dose in 2 Gy fractions (EQD 2 ) when applied to high-dose-rate brachytherapy (HDR-BT) for cervical cancer.

Methods and Materials: An analysis of 98 published α/β values from 31 papers was conducted, spanning 23 cervical cancer cell lines. Values were further divided into squamous cell carcinoma and adenocarcinoma histologies. Probability distributions were fit to histograms using the bootstrapped Kolmogorov-Smirnov goodness-of-fit test. Both average and most probable α/β values for cervical cancer were determined. Sweden has been lifted as a prominent example of a successful feminist policy enactor, and has built credibility as a humanitarian actor both domestically and transnationally. Simultaneously, intersectionally marginalized groups have attested to patterns of social exclusion and feelings of political disenfranchisement in Swedish society, calling into question Sweden’s reputation for feminist exceptionalism. This dissertation project centers on unpacking this dynamic by examining how Swedish gender equality policymaking institutions design and implement feminist policy, and how those efforts are interpreted and contested by intersectionally marginalized groups of Afro-Swedish feminist activists. This research study examines the challenges Black women encounter when in predominantly white, male academic and professional science settings, where they experience dual discrimination because of their race and gender. These biases perpetuate the negative stereotype that Black women are less competent than white men. This mixed methods study examines how Black women navigate their race, gender, and science identities within predominantly white institutions through their doctoral programs and professional science careers. However, there is limited research on the professional competencies necessary for success in this role. This study explores the relationship between the ACPA and NASPA professional competencies RDs and their supervisors identify as most critical for RD success. Women higher education administrators encounter distinct stressors which can significantly impact wellbeing and lead to burnout. These challenges include implicit bias, difficulty achieving work-life balance, and increasing institutional expectations. This study utilizes a Mixed Methods Action Research (MMAR) framework to assess stress levels, trait mindfulness, coping mechanisms, and the effectiveness of a structured mindfulness-based intervention. While the quantitative analysis revealed only a slight increase in trait mindfulness and no notable reduction in stress levels, the qualitative findings showcased meaningful personal benefits to the participants. International school leaders worldwide have navigated their schools through a variety of crises. Their unique lived experiences offer insights into the role of trust in leadership, particularly during times of crisis.

This qualitative study employed a phenomenological approach to explore the lived experiences of 12 experienced international school leaders. Data was collected through semi-structured interviews. Using the theoretical lenses of authentic leadership, sensemaking, loose coupling, and crisis leadership, this study illuminates the central role of trust in leading schools through crises.

This dissertation examined the adaptive instructional leadership practices of instructors teaching dual credit career and technical education (CTE) classes in Connecticut, particularly in advanced manufacturing technologies (AMT). The study drew on survey responses from a broader group of course instructors and a purposeful sample of six instructors and six adult students who participated in semi-structured interviews. All instructors taught dual credit classes in partnership with Connecticut State Community College in hands-on manufacturing, engineering, or similar workshop environments designed to prepare high school aged students for careers and develop practical, job-ready skills in settings that closely replicate industry conditions. Individuals with hip-related pain exhibit altered movement patterns, and worse fear of movement (kinesiophobia), compared to asymptomatic controls. Exacerbation of hip pain during movement and the role of hip symptom duration may help further our understanding of specific movement patterns that are associated with hip-related pain. A better understanding of the potential connections between hip-related symptom duration, kinesiophobia, hip pain during movement, and lower limb mechanics in those with hip-related pain may allow for more efficient referrals to clinicians and lead to more effective rehabilitation. The purpose of this qualitative narrative study is to explore the experiences of spouses, specifically female spouses, of retired U. S. military veterans who pursued the completion of postsecondary degrees following the retirement of the military service member. The study employs narrative inquiry, guided by the Biographic Narrative Interview Method approach to interviewing. The generated data is analyzed utilizing thematic and narrative analysis methods, and a framework of two researchers’ sensitizing concepts is applied to understand and create meaning from the participants’ narratives. Purpose: To explore the potential relationship between tobacco retailer proximity/density around Kentucky elementary, middle, and high schools, and students’ perceptions, perceived norms, susceptibility to, and use of e-cigarettes. Design: A mixed-methods analysis of youth’s self-reported survey results and geographical data. This investigation examined how movement intent influences neuromuscular excitation strategies during fatiguing resistance exercise, comparing maximal intended concentric efforts versus self-selected volitional efforts during load-equated Smith machine back squat exercise to quantify reserve capacity and characterize underlying neuromuscular mechanisms. Twenty-three resistance-trained participants (12 males, 11 females) completed a repeated-measures design, with replication, across six visits, performing smith machine back squat repetitions-to-failure at 70% 1RM under maximal intent (accelerate rapidly) and volitional intent (self-selected effort) conditions. Maximal voluntary isometric contractions used rapid (instantaneous 10-second effort) and ramp (5-second build-up, 5-second maximal) protocols. Physical inactivity remains a pressing challenge in Appalachian Kentucky, where high obesity rates intersect with limited access to structured exercise facilities. This dissertation investigated the extent to which participation in outdoor recreation supports adherence to physical activity (PA) recommendations and explored community awareness of local public lands. Surveys from 52 adult participants in Knox County revealed strong engagement in outdoor activity, with participation significantly predicting compliance with national PA guidelines. Educational attainment was also associated with higher rates of PA compliance, pointing to the influence of literacy and awareness in health behaviors. Magnetic heating mediated by magnetic iron oxide nanoparticles (MION) has several clinical applications including cancer thermal therapy, tissue thawing for organ preservation, and magnetogenetics. However, current models of magnetic heating, including the widely used linear response theory, inadequately predict MION heating efficiency. An experimental study covering alternating magnetic field (AMF) strengths and frequencies ranging from 3.98 – 27.85 kA/m and 103.6 – 984.1 kHz, respectively, with a range of MION core diameters 9.5- 30.3 nm was conducted to elucidate the relationships between these AMF parameters and MION sizes and the resulting heating efficiencies. Complete Streets is a vital strategy for developing an equitable transportation network that provides access to all users and transportation options to communities. A growing interest in this concept has been noted in the United States, with over 1,700 Complete Streets-related policies implemented nationwide. The Kentucky Transportation Cabinet (KYTC) has developed the “Complete Streets, Roads, and Highways Manual” and the “Complete Streets Policy” aiming to promote a safer and more inclusive transportation system statewide through Complete Streets implementation. The growing shift of commuters towards active transportation (AT) in the U.S. has led transportation agencies to place greater emphasis on improving bicycle and pedestrian facilities. Such improvement efforts can be integrated into multi-modal highway planning and development or be implemented through stand-alone pedestrian and bicycle projects both resulting in additional AT demand. Vapor intrusion, the migration of subsurface vapors into indoor air spaces, is an important source of indoor air contamination to consider at hazardous waste sites contaminated by volatile organic compounds (VOCs). There is considerable uncertainty and variability in the factors that influence vapor intrusion processes, which poses a significant challenge for decision making, despite advancements in vapor intrusion science.
