BCR::ABL1-like B-lymphoblastic leukaemia (B-ALL) neoplasms lack the BCR::ABL1 translocation but have a gene expression profile like BCR::ABL1 positive B-ALL. This includes alterations in cytokine receptors and signalling genes, such as CRLF2, ABL1, ABL2, JAK2, PDGFRB and EPOR. Cases with CRLF2 rearrangements account for approximately 50% of cases of Philadelphia-like acute lymphoblastic leukaemia (Ph-like ALL), and the frequency of specific genomic lesions varies with ethnicity such that IGH::CRLF2 translocations are more common in Hispanics and Native Americans. We report two cases of BCR::ABL1-like ALL, with significant eosinophilia. A Hispanic man in his early 20s and a Hispanic woman in her 50s …

Osteosarcoma (OS) is a relatively rare bone malignancy that primarily affects children and young adults and is associated with significant morbidity and mortality. Cisplatin is a mainstay of treatment, but its efficacy is limited by off-target toxicities. Immunotherapy is not effective due to a poor antigenic tumor microenvironment. Here, we address these challenges by using manufactured M1 macrophage-derived vesicles (MVs) loaded with cisplatin. Human blood and mouse RAW 264.7 M1 macrophages were used to prepare empty (E-MVs) and cisplatin-loaded MVs (C-MVs). Human OS cell lines were used in vitro and in a tibia xenograft mouse model to evaluate the anti-cancer …

Characterizing Agroecosystem Effects Of Industrial Hemp On A Rotational Row Cropping System And Microbial Communities, Kent D. Pham
Characterizing Agroecosystem Effects Of Industrial Hemp On A Rotational Row Cropping System And Microbial Communities, Kent D. Pham
Theses and Dissertations--Plant and Soil Sciences
The implementation of crop rotation on a farm can have many benefits for agroecosystem health and production. This practice can potentially reduce nutrient amendments, disrupt pest and disease cycles, and increase crop yields. In Kentucky, one common rotation consists of either corn or soybean grown the first year, followed by the other crop, repeated every two years. Industrial hemp, grown for fiber or grain, is an emerging crop that has shown promise as a rotation alternative, although not much is known about its effects on soil health, agroecosystem functioning, and microbial communities. To better characterize hemp’s role within American agriculture, …

Factors Impacting Germination, Emergence, And Establishment Of Direct Seeded And Transplanted Hemp (Cannabis Sativa L.), Paul Cockson Dr.
Theses and Dissertations--Plant and Soil Sciences
Industrial hemp (IH) establishment has many challenges including: poor emergence, low stand establishment, and variable plant development in direct seeded hemp. In floral hemp production, quality transplant production is a challenge with seedlings often becoming root bound and etiolated. In both direct seeded and transplant hemp, the relationship between root vigor and vegetative growth has not been studied and may prove insightful for managing emergence and establishment challenges. The goals of this study were three-fold: 1) to measure and describe root system architecture (RSA) and root phenotypes observed in selected IH cultivars especially in regard to identifying agronomically important RSA …
Measurement Of Moisture Levels In Oils And Lubricants Using A Novel Moisture Sensor, Aaron Swartz
Measurement Of Moisture Levels In Oils And Lubricants Using A Novel Moisture Sensor, Aaron Swartz
Theses and Dissertations--Electrical and Computer Engineering
It is very challenging to measure moisture levels in oils. There is not a good method to measure it. Using the novel moisture sensor, we tried various methods to measure the moisture levels in oils. The initial trials are to immerse the sensor chip into the oils to see any sensor reading changes with the change of moisture levels in oils. It was observed that the sensor reading was unstable. The idea for immersion of the sensor chip into the oils failed. The last idea is to heat the oil to let all moisture evaporate fully. Before heating, the dew …
Computing With Photonic Phase Change Memory, David B. Pippen
Computing With Photonic Phase Change Memory, David B. Pippen
Theses and Dissertations--Electrical and Computer Engineering
A recent breakthrough in silicon photonics includes the discovery and use of phase changing materials (PCMs). These materials can be programmed to store nonvolatile values, and when a stored value in a PCM cell is read, it changes the amplitude of the read signal, imprinting the value held into the PCM cell on the amplitude of the read signal. This thesis proposes a new approach to using PCM cells not only for photonic memory but also as a substrate to perform multiplications in the photonic domain. The proposed multiplier uses PCM cells to encode amplitude-analog weight values and differing lengths …
Scalable Hypergraph Structure Learning With Diverse Smoothness Priors, Benjamin T. Brown
Scalable Hypergraph Structure Learning With Diverse Smoothness Priors, Benjamin T. Brown
Theses and Dissertations--Electrical and Computer Engineering
In graph signal processing, learning weighted connections between nodes from signals is a fundamental task when the underlying relationships are unknown. With the extension of graphs to hypergraphs, where edges can connect more than two nodes, graph learning methods have similarly been generalized to hypergraphs. However, the absence of a unified framework for calculating total variation has led to divergent definitions of smoothness and, consequently, differing approaches to hyperedge recovery. This challenge is confronted in this work through generalization of several previously proposed hypergraph total variations, allowing ease of substitution into a vector-based optimization. To this end, a novel hypergraph …
Electromagnetic Integral Equation Methods For High-Order Field Predictions, Jordon N. Blackburn
Electromagnetic Integral Equation Methods For High-Order Field Predictions, Jordon N. Blackburn
Theses and Dissertations--Electrical and Computer Engineering
Methods like the Method of Moments (MoM) or the locally-corrected Nyström (LCN) method are employed to discretize and solve electromagnetic integral equations. This process results in large, dense systems of linear equations that must be solved. In many cases, the elements of the system matrix can be computed analytically or approximated with high-order numerical methods. In this thesis, various approaches are presented to improve the accuracy and efficiency of integral equation solutions.
The second chapter derives a modified form of the low-rank matrix approximation algorithm known as the adaptive cross approximation (ACA). The original ACA has been observed to lose …
Afapbp: Aggregate Function Accelerated Parallel Bit-Pattern Computing, Charles Z. Armstrong
Afapbp: Aggregate Function Accelerated Parallel Bit-Pattern Computing, Charles Z. Armstrong
Theses and Dissertations--Electrical and Computer Engineering
Classical computing models have proven sufficient for problems of the complexity class ``P", but problems of a higher complexity class like ``NP", ``NP-Hard", etc. have been shown to be more resistant to efficient computation. Quantum Computing is an alternative computing model that specifically targets performing computations within the ``NP" complexity class in close to linear time. However, Quantum Computing has its own set of problems. Methods for dealing with quantum decoherence, error correction, and difficulty in scaling have all inhibited Quantum Computing from becoming a commonly used computational model. This thesis introduces AFAPBP (Aggregate Function Accelerated Parallel Bit Pattern), a …
Multi-Objective Design Optimization Of Power Converters For Electric Aircraft Propulsion, Ben Luckett
Multi-Objective Design Optimization Of Power Converters For Electric Aircraft Propulsion, Ben Luckett
Theses and Dissertations--Electrical and Computer Engineering
As global focus shifts to the electrification of the aviation sector, the need for high efficiency, lightweight, and reliable electric aircraft propulsion power converter systems has become apparent. These goals can be somewhat conflicting with each other, and a single multi-domain-optimized solution is not guaranteed. The search for a design which presents satisfactory merits becomes a drudge through various trade-off studies which can expend vast quantities of manpower and time. As a remedy to this, design automation allows the process to be computer-assisted. This dissertation presents the core fundamentals for a general multi-objective design optimization framework intended for the design …

Implementation Of A Pilot Post-Code Debrief Tool Initiated By The Chaplains And Unit Lead Nurses Within The Medicine Intensive Care Unit, Sarah Stigall
DNP Projects
Background: Post-code debriefs are shown to improve teamwork and communication, increase performance, and reduce burnout. According to the University of Kentucky HealthCare (UKHC) policy, teams are to perform a post-code debrief immediately after an in-hospital cardiac arrest (IHCA). However, the current policy does not provide guidance on what to include during a debrief session. At UK post-code debrief sessions are documented as occurring 60-72% of the time; however, personal observation and discussions with other bedside and rapid response nurses suggest that post-code debriefs are underperformed when compared to the documentation. Purpose: This study aimed to explore barriers to post-code debriefs …

Nurse-Led Cbt For Depression In Heart Transplant Patients: A Feasibility Study, Matthew R. Wallace
DNP Projects
Background: Heart transplant recipients are at an elevated risk of experiencing depressive symptoms compared to the general population. Those with depression post-transplant face increased complications, e.g., infection, organ rejection, prolonged hospitalization, and higher mortality rates. However, specific psychotherapeutic interventions tailored for heart transplant recipients remain limited. Nevertheless, there is substantial evidence supporting the effectiveness of various interventions for adults with heart failure, a condition frequently diagnosed in these patients. This suggests that successful strategies for managing depressive symptoms in heart failure patients could be adapted for heart transplant recipients.
Purpose: The purpose of this project was to evaluate …

Advancing Root Trait Measurements For A Deeper Understanding Of Soil Biogeochemistry, Brian Rinehart
Advancing Root Trait Measurements For A Deeper Understanding Of Soil Biogeochemistry, Brian Rinehart
Theses and Dissertations--Plant and Soil Sciences
Climate change is disrupting (agro)ecosystems, with the potential for significant losses in crop productivity and ecosystem resilience due to changes in water and biogeochemical cycling. These disruptions can also release more carbon dioxide into the atmosphere through the degradation of soil organic matter. Plant roots have emerged as a potential tool for mitigating these effects, given their roles in plant function and in driving soil biogeochemistry. While this interest is growing, our ability to implement root trait selection is limited due to constraints on characterizing roots and limited clarity on the effects of traits on plant and ecosystem function. My …

Revolutionizing Grapevine Breeding Programs: A Mobile Genome Editing System For Perennial Crops, Bridget Bolt
Revolutionizing Grapevine Breeding Programs: A Mobile Genome Editing System For Perennial Crops, Bridget Bolt
Theses and Dissertations--Plant and Soil Sciences
Grapevine (Vitis vinifera) faces unprecedented challenges from climate change, with up to 90% of current growing regions potentially becoming unsuitable for production within decades. Traditional breeding approaches require 25-30 years for variety development, creating a time constraint incompatible with the pace of environmental change. This research establishes the approach for a novel mobile RNA-based gene editing platform that addresses longstanding bottlenecks in grapevine improvement while preserving cultivar identity and reducing breeding timelines.
This research developed a three-component system for transgene-free gene editing in grapevine. First, a mobile CRISPR/Cas9 system was established using Agrobacterium rhizogenes transformation and the cut-and-dip …
The Effects Of Technology-Based Self-Monitoring Intervention To Increase On-Task Behaviors For Adolescents With Intellectual And Developmental Disabilities, Deris Cox Jr
Theses and Dissertations--Educational, School, and Counseling Psychology
This study aims to assess the effects of technology-based self-monitoring interventions and to promote independent functioning skills for adolescents with intellectual and developmental disabilities (IDD). It also seeks to replicate the findings of Rosenbloom et al. (2019). IDD typically appears in adolescents before the age of 18 and affects an individual’s ability to learn, reason, and manage daily tasks. Behavior analysis is a common approach used to teach individuals with developmental disabilities new skills, decrease challenging behaviors, and enhance quality of life.

In this work, we discovered a novel organometallic gold(III) macrocycle, Au-Mac1, that demonstrates anticancer potency in a panel of triple-negative breast cancer cells (TNBC), and based on this complex, a biotinylated-Au-Mac1 probe was designed for target identification via chemoproteomics, which uncovered the engagement of HMOX2 of the heme-energy metabolism pathway. Using orthogonal chemical biology and molecular biology approaches, including immunoblotting, flow cytometry, and cellular thermal shift assays, it was confirmed that Au-Mac1 engages HMOX2 in cells. Downstream effects of Au-Mac1 on the depletion of mitochondrial membrane proteins and bioenergetics point to the potential role of HMOX2 in cancer. Importantly, Au-Mac1 …

Gamma-aminobutyric acid type A receptors (GABAARs) are essential for maintaining the excitation–inhibition balance in the central nervous system. Genetic variations of GABAARs result in a variety of neurological disorders, such as epilepsy. A key pathogenic mechanism involves protein misfolding and defective assembly of GABAARs in the endoplasmic reticulum (ER), resulting in impaired surface expression and loss of function. Here, we investigated three trafficking-deficient variants of the GABAAR α1 subunit (GABRA1), including D219N (ClinVar Variation ID: 127232), G251D (Variation ID: 419523), and P260L. We demonstrated that selective pharmacological activation of the IRE1/XBP1s signaling …
Ion Hydration In Bulk And Nanoconfined Water: Insights From Machine Learning Force Fields, Zachary D. Baker
Ion Hydration In Bulk And Nanoconfined Water: Insights From Machine Learning Force Fields, Zachary D. Baker
Theses and Dissertations--Chemical and Materials Engineering
Understanding ionic hydration remains a central challenge in physical chemistry and materials science, as the interactions between ions and water molecules govern diverse phenomena ranging from electrolyte transport to selective ion separation. While experimental techniques have provided invaluable insights into solvation energetics and coordination numbers, they often lack atomistic resolution, particularly under nanoscale confinement where direct measurement becomes infeasible. Molecular dynamics (MD) simulations can bridge this gap; however, conventional classical force fields are limited by their simplified, fixed functional forms and empirical parameterization, whereas ab initio molecular dynamics (AIMD) achieves higher accuracy at the expense of severe computational cost and …

BACKGROUND One third of organ donors suffer catastrophic brain injury (CBI). There are no standard guidelines for the management of traumatic CBI prior to brain death, and not all trauma centers have institutional CBI guidelines. In addition, there is high variability in management between institutions with guidelines. Catastrophic brain injury guidelines vary and may include various combinations of hormone therapy, vasopressors, fluid resuscitation, and other practices. We hypothesized that centers with CBI guidelines have higher organ donation rates than those without. METHODS This prospective, observational EAST-sponsored multicenter trial included adult (18+ years old) traumatic-mechanism CBI patients at 33 level I …
Machine Learning Models For Molecular-Based Functional Organic Materials, Moses Ogbaje
Machine Learning Models For Molecular-Based Functional Organic Materials, Moses Ogbaje
Theses and Dissertations--Chemistry
Organic semiconductors (OSC) are of interest for a wide range of flexible optoelectronics applications, including transistors, solar cells, and sensors, to name a few. Despite their promise, the design and optimization of OSC pose significant challenges due to the complexity of the structures of the molecular building blocks, varied packing configurations of these building blocks in the solid state, which impacts the optical and electronic response, and sensitivity of the solid-state packing to material processing conditions. Accurately predicting the solid-state properties of OSC traditionally requires high-level quantum mechanical methods. These methods, however, can be computationally demanding, particularly for large molecules …
