Salvinorin A (SA) is a κ-opioid receptor agonist and atypical dissociative hallucinogen found in Salvia divinorum. Despite the resurgence of hallucinogen studies, the effects of κ-opioid agonists on human brain function are not well-understood. This placebo-controlled, within-subject study used functional magnetic resonance imaging for the first time to explore the effects of inhaled SA on strength, variability, and entropy of functional connectivity (static, dynamic, and entropic functional connectivity, respectively, or sFC, dFC, and eFC). SA tended to decrease within-network sFC but increase between-network sFC, with the most prominent effect being attenuation of the default mode network (DMN) during the …

A Thorough Analysis And Categorization Of Bacterial Interrupted Adenylation Domains, Including Previously Unidentified Families, Taylor A. Lundy, Shogo Mori, Sylvie Garneau-Tsodikova

Pharmaceutical Sciences Faculty Publications

Interrupted adenylation (A) domains are key to the immense structural diversity seen in the nonribosomal peptide (NRP) class of natural products (NPs). Interrupted A domains are A domains that contain within them the catalytic portion of another domain, most commonly a methylation (M) domain. It has been well documented that methylation events occur with extreme specificity on either the backbone (N-) or side chain (O- or S-) of the amino acid (or amino acid-like) building blocks of NRPs. Here, through taxonomic and phylogenetic analyses as well as multiple sequence alignments, we evaluated the similarities and …

Determination Of Steels In Kentucky Bridges, Theodore Hopwood Ii, Christopher Goff, Sudhir Palle Oct 2020

Determination Of Steels In Kentucky Bridges, Theodore Hopwood Ii, Christopher Goff, Sudhir Palle

Kentucky Transportation Center Research Report

The Kentucky Transportation Cabinet (KYTC) maintains over two hundred steel bridges constructed with types of steel which are unknown. Kentucky Transportation Center (KTC) researchers have developed a procedure for determining steel tensile properties (0.2% yield strength and ultimate tensile strength) which can be used to help identify unknown steels. The procedure involves extracting small coupons from the lower flanges of steel deck girder bridges. Coupons are then machined into sub-sized tensile testing specimens and suitable shapes for chemical analyses. Sub-sized tensile tests returned values generally comparable to those provided in mill certifications of steels purchased for laboratory trials. Several analytical …

Importation Of Macrocyclic Lactone Resistant Cyathostomins On A Us Thoroughbred Farm, Martin K. Nielsen, M. Banahan, R. M. Kaplan

Maxwell H. Gluck Equine Research Center Faculty Publications

Anthelmintic resistance in equine cyathostomins is both widespread and highly prevalent in the benzimidazole and tetrahydropyrimidine classes; however, reports of resistance to macrocyclic lactone (ML) drugs are sparse and sporadic. This study reports a case of clear ML resistance in a group of Thoroughbred yearlings imported from Ireland to the US in 2019. Fecal egg count reduction (FECR) following ivermectin administered in February 2020 demonstrated 100% reduction in the US bred yearlings, but 93.5%, 70.5%, and 74.5% reduction in three groups of the imported yearlings. The two former groups were then retreated with ivermectin, yielding FECRs of 33.8% and 23.5%, …

Biology Faculty Publications

Temperature and water potential are two important environmental factors influencing germination and subsequent seedling establishment. Seed germination requirements vary with species and with the environment in which the seeds are produced. Stipa species dominate large areas of the Eurasian zonal vegetation, but comparisons of germination requirements between Stipa species from different habitats is limited. We investigated the effects of temperature and water potential on seed germination of S. grandis, S. purpurea, and S. penicillata from habitats with low temperatures and relatively abundant rainfall (cool habitats) and S. glareosa, S. breviflora, S. gobiea, and S. bungeana …

Radiation therapy is one of the most prevalent procedures for cancer treatment, but the risks of malignancies induced by peripheral beam in healthy tissues surrounding the target is high. Therefore, being able to accurately measure the exposure dose is a critical aspect of patient care. Here a radiation detector based on an organic field‐effect transistor (RAD‐OFET) is introduced, an in vivo dosimeter that can be placed directly on a patient's skin to validate in real time the dose being delivered and ensure that for nearby regions an acceptable level of low dose is being received. This device reduces the errors …

Collecting remotely sensed spectral data under varying ambient light conditions is challenging. The objective of this study was to test the ability to classify grayscale targets observed by portable spectrometers under varying ambient light conditions. Two sets of spectrometers covering ultraviolet (UV), visible (VIS), and near−infrared (NIR) wavelengths were instrumented using an embedded computer. One set was uncalibrated and used to measure the raw intensity of light reflected from a target. The other set was calibrated and used to measure downwelling irradiance. Three ambient−light compensation methods that successively built upon each other were investigated. The default method used a variable …

Microglial-Associated Responses To Comorbid Amyloid Pathology And Hyperhomocysteinemia In An Aged Knock-In Mouse Model Of Alzheimer's Disease, David J. Braun, Edgardo R. Dimayuga, Josh M. Morganti, Linda J. Van Eldik

Sanders-Brown Center on Aging Faculty Publications

BACKGROUND: Elevated blood homocysteine levels, termed hyperhomocysteinemia (HHcy), is a prevalent risk factor for Alzheimer's disease (AD) in elderly populations. While dietary supplementation of B-vitamins is a generally effective method to lower homocysteine levels, there is little if any benefit to cognition. In the context of amyloid pathology, dietary-induced HHcy is known to enhance amyloid deposition and certain inflammatory responses. Little is known, however, about whether there is a more specific effect on microglia resulting from combined amyloid and HHcy pathologies.

METHODS: The present study used a knock-in mouse model of amyloidosis, aged to 12 months, given 8 weeks of …

Electrical and Computer Engineering Faculty Patents

Methods and systems are provided herein for measuring 3D apparatus (e.g., manual tool or tool accessory) orientation. Example implementations use an auto-nulling algorithm that incorporates a quaternion-based unscented Kalman filter. Example implementations use a miniature inertial measurement unit endowed with a tri-axis gyro and a tri-axis accelerometer. The auto-nulling algorithm serves as an in-line calibration procedure to compensate for the gyro drift, which has been verified to significantly improve the estimation accuracy in three-dimensions, especially in the heading estimation.

Nursing Faculty Publications

Background

The nurse workforce shortage, partially caused by high work turnover, is an important factor influencing the quality of patient care. Because previous studies concerning Chinese nurse work turnover were predominantly quantitative, they lacked insight into the challenges faced by nurses as they transition from university to their career. A successful transition can result in new nurses’ commitment to the career. As such, this study sought to understand how new nurses commit to the career, and focused on identifying facilitators and barriers to such commitment.

Methods

This was a qualitative study using a grounded theory design. Through purposive sampling, clinical …

Atom Identifiers Generated By A Neighborhood-Specific Graph Coloring Method Enable Compound Harmonization Across Metabolic Databases, Huan Jin, Joshua M. Mitchell, Hunter N. B. Moseley

Molecular and Cellular Biochemistry Faculty Publications

Metabolic flux analysis requires both a reliable metabolic model and reliable metabolic profiles in characterizing metabolic reprogramming. Advances in analytic methodologies enable production of high-quality metabolomics datasets capturing isotopic flux. However, useful metabolic models can be difficult to derive due to the lack of relatively complete atom-resolved metabolic networks for a variety of organisms, including human. Here, we developed a neighborhood-specific graph coloring method that creates unique identifiers for each atom in a compound facilitating construction of an atom-resolved metabolic network. What is more, this method is guaranteed to generate the same identifier for symmetric atoms, enabling automatic identification of …

Greenhouse Spatial Effects Detected In The Barley (Hordeum Vulgare L.) Epigenome Underlie Stochasticity Of Dna Methylation, Moumouni Konate, Michael J. Wilkinson, Julian Taylor, Eileen S. Scott, Bettina Berger, Carlos M. Rodriguez Lopez Sep 2020

Greenhouse Spatial Effects Detected In The Barley (Hordeum Vulgare L.) Epigenome Underlie Stochasticity Of Dna Methylation, Moumouni Konate, Michael J. Wilkinson, Julian Taylor, Eileen S. Scott, Bettina Berger, Carlos M. Rodriguez Lopez

Horticulture Faculty Publications

Environmental cues are known to alter the methylation profile of genomic DNA, and thereby change the expression of some genes. A proportion of such modifications may become adaptive by adjusting expression of stress response genes but others have been shown to be highly stochastic, even under controlled conditions. The influence of environmental flux on plants adds an additional layer of complexity that has potential to confound attempts to interpret interactions between environment, methylome, and plant form. We therefore adopt a positional and longitudinal approach to study progressive changes to barley DNA methylation patterns in response to salt exposure during development …

Suspected Malignant Hyperthermia And The Application Of A Multidisciplinary Response, Laura Ebbitt, Eric Johnson, Brooke Herndon, Kristina Karrick, Aric Johnson

Pharmacy Practice and Science Faculty Publications

Purpose: Malignant hyperthermia (MH) is a critical and potentially life-threatening emergency associated with inhaled anesthetic and depolarizing neuromuscular blocker administration. This is a single center’s response to MH. Summary: When signs of MH are observed, a page for “anesthesia STAT-MH crisis” is called, triggering a multidisciplinary response, including the deployment of a Malignant Hyperthermia Cart. The MH cart and the delegation of duties allows nurses, physicians and pharmacists to quickly understand their role in the stabilization, transition and recovery of a suspected MH patient. Conclusion: This case highlights the importance of multi-disciplinary involvement in these rare, but potentially fatal, cases.

Encoding The Odor Of Cigarette Smoke, Timothy S. Mcclintock, Naazneen Khan, Yelena Alimova, Madeline Aulisio, Dong Y. Han, Patrick Breheny

Physiology Faculty Publications

The encoding of odors is believed to begin as a combinatorial code consisting of distinct patterns of responses from odorant receptors (ORs), trace-amine associated receptors (TAARs), or both. To determine how specific response patterns arise requires detecting patterns in vivo and understanding how the components of an odor, which are nearly always mixtures of odorants, give rise to parts of the pattern. Cigarette smoke, a common and clinically relevant odor consisting of > 400 odorants, evokes responses from 144 ORs and 3 TAARs in freely behaving male and female mice, the first example of in vivo responses of both ORs and …

Misinformation More Likely To Use Non-Specific Authority References: Twitter Analysis Of Two Covid-19 Myths, Joseph Mcglynn, Maxim Baryshevtsev, Zane A. Dayton

Communication Graduate Research

This research examines the content, timing, and spread of COVID-19 misinformation and subsequent debunking efforts for two COVID-19 myths. COVID-19 misinformation tweets included more non-specific authority references (e.g., “Taiwanese experts”, “a doctor friend”), while debunking tweets included more specific and verifiable authority references (e.g., the CDC, the World Health Organization, Snopes). Findings illustrate a delayed debunking response to COVID-19 misinformation, as it took seven days for debunking tweets to match the quantity of misinformation tweets. The use of non-specific authority references in tweets was associated with decreased tweet engagement, suggesting the importance of citing specific sources when refuting health misinformation.
