Plant embryogenesis is an intriguing developmental process that is controlled by many genes. AGAMOUS Like 15 (AGL15) is a MADS-domain transcriptional regulator that accumulates preferentially during this stage. However, at the onset of this work it was unknown which genes are regulated by AGL15 or how AGL15 is regulated. This dissertation is part of the ongoing effort to understand the biological roles of AGL15. To decipher how AGL15 functions during plant development, a chromatin immunoprecipitation (ChIP) approach was adapted to obtain DNA fragments that are directly bound by AGL15 in vivo. Despite distinct dissimilarities, diverse cancers express several common pro-tumorigenic traits. We present here evidence that the pro-apoptotic protein Par-4 utilizes one such common tumorigenic trait to become selectively activated and induce apoptosis in cancer cells. Elevated PKA activity noted in cancer cells activated the apoptotic function of ectopic Par-4 or its SAC domain, which induces apoptosis selectively in cancer cells and not in normal or immortalized cells. PKA preferentially phosphorylated Par-4 at the T155 residue within the SAC domain in cancer cells. Moreover, pharmacological-, peptide- or siRNA-mediated inhibition of PKA activity in cancer cells resulted in abrogation of both T155 …

Matrix metalloproteinase-2 (MMP-2, gelatinase A, type IV collagenase) is a member of a family of zinc-dependent metalloendopeptidases that functions in the degradation of elastin, collagens, and other components of extracellular matrix (ECM). Both secretion and activation of MMP-2 are elevated in human atherosclerotic lesions and abdominal aortic aneurysms (AAA). In this dissertation project, we sought to test the hypothesis that MMP-2 plays a critical role in both atherosclerosis and AAA. We also sought to determine the detailed mechanism. A major goal of this research was to evaluate a variety of cell lines for theirpermissiveness to equine arteritis virus (EAV) infection and then identify the mechanismthat restricts EAV infection in certain cell lines. The cell lines BHK-21, RK-13, andC2C12 were found to support productive infection with EAV strain VBS53, whereasHela, Hep-2, and L-M cell lines exhibited limited susceptibility to infection with thisvirus. In the course of the study, it was found that the Hela cell line became moresusceptible to infection with EAV strain VBS53 after extended serial passage. This study investigated the interaction between polychlorinated biphenyls (PCBs) and selenium to explain the mechanism involved that could affect selenium metabolism and its anti-cancer property. PCBs congeners and mixtures were previously found to reduce hepatic Se and Se-dependent glutathione peroxidase activity. I hypothesized that certain PCB congeners affect selenium metabolism in the rat liver resulting in diminished antioxidant capacity of selenoproteins, which could alter the ability of Se to protect against PCBs induced tumor promotion. Kentucky plays an important role in the complex U.S. beef cattle industry. Thisstudy focused on the feeder cattle production sector of Kentucky's beef cattle industry.Primarily a cow-calf state with a substantial backgrounding sector, Kentucky is a largesupplier of feeder cattle to the cattle finishing sector. Price relationships within themarket for Kentucky feeder cattle were examined using historical price data fromKentucky livestock auction markets. Biodegradable polymer materials, specifically poly(lactic-co-glycolide) (PLGA) can be used as bone replacements for bone regeneration. Scaffolds can be prepared to be porous to induce bone growth into a scaffold so that it is replaced with natural tissue as the polymer degrades. However, simply using PLGA will result in formation of scar tissue rather than regeneration of natural bone. Therefore focus has turned to attaching growth factors to the PLGA molecules to elicit a specific cellular response when the implant is placed in the body. Sexual conflict occurs when individuals of one sex express traits that reduce the fitness of their mates. Males of many species harass females to gain copulations, which benefits males by increasing the number of offspring they sire but imposes energetic and opportunity costs on the females they harass. This thesis examined the fitness costs of sexual harassment to females, the energetic costs of mating to males, and the factors influencing the intensity of male competition for mates in the western mosquitofish Gambusia affinis. In this study, mitochondrial transcript information from a recent EST project wasextended to obtain complete mitochondrial genome sequence for 5 tiger salamandercomplex species (Ambystoma mexicanum, A. t. tigrinum, A. andersoni, A. californiense,and A. dumerilii). For the first time, aspects of mitochondrial transcription in arepresentative amphibian are described, and then complete mitochondrial sequencedata are used to examine salamander phylogeny at both deep and shallow levels ofevolutionary divergence. The available mitochondrial ESTs for A. mexicanum (N=2481)and A. t. tigrinum (N=1205) provided 92% and 87% coverage of the mitochondrialgenome, respectively. For economical and ecological reasons, efficiency and profitability ofswine production relies heavily on the way pigs utilize key nutrients such as P,which is considered a potential pollutant of water ecosystems. Although cerealgrains and oilseed meals contain enough P to fulfill the biological needs of pigs,most of this P is tightly bound as phytate. As pigs do not have enough phytase(PHY) to cleave P from phytate, it is excreted in the feces. To prevent adeficiency, diets have traditionally been supplemented with highly availableinorganic sources of P. Follicle stimulating hormone (FSH) comprises an ?? subunit and a ?? subunit,whereas the FSH receptor consists of two halves with distinct functions, the N-terminalextracellular exodomain and C-terminal membrane associated endodomain. FSH initiallybinds to exodomain, and the resulting FSH/exodomain complex modulates the endodomainand generates signal. The purpose of this study was to evaluate masticatory muscle (MM) and temporomandibular joint (TMJ) pain patients regarding the prevalence of Posttraumatic Stress Disorder (PTSD) symptoms, and evaluate the level of psychological dysfunction and its relationship to PTSD symptoms in these patients. This study included 445 adult patients (male = 42, female = 403). Psychological questionnaires included the Symptom Check List-90-Revised (SCL-90-R), the Multidimensional Pain Inventory, the Pittsburgh Sleep Quality Index and the PTSD Check List Civilian. A series of acute (96h) toxicity tests were conducted on six species of crayfish inthe family Cambaridae. Toxicity values fell into a sensitive group comprising Orconectesplacidus and Procambarus acutus (LC 50= 0.368mg Cd/L - 0.487mg Cd/L; LC10= 0.048mg Cd/L - 0.092 mg Cd/L) and tolerant group comprising O. juvenilis, O. virilis, P.alleni, and P. clarkii (LC 50= 2.44 mg Cd/L - 3.30 mg Cd/L; LC10= 0.386 mg Cd/L -0.947 mg Cd/L). This dissertation details three studies dealing with the manipulation of nucleicacids. In the first investigation, each of the four natural nucleobases were analyzed for theability to serve as a universal template at the ligation junction of a T4 DNA ligasereaction. This resulted in the first instance of sequence-independent ligation catalyzed byany DNA ligase. Although all of the nucleobases display universal templatingcapabilities, thymidine and guanosine provided the most effective results. In addition,lowered MgCl2 and ATP concentrations, as well as the inclusion of DMSO, also aided inthe sequence-independent ligation reported here. Cytochrome P4501A (CYP1A), a xenobiotic metabolizing enzyme found in allvertebrates, is highly induced following exposure to a number of organic contaminants.Several populations of teleosts residing in highly contaminated areas have been found toexhibit resistance to the toxic effects of contaminants, a condition characterized byreduced expression of CYP1A.Within this work I demonstrated that expression of CYP1A mRNA, protein, andactivity in caged rainbow trout (Oncorhynchus mykiss) was an effective biomarker ofpolychlorinated biphenyl (PCB) contamination. A restored elk (Cervus elaphus) population in eastern Kentucky may be affecting ecosystem processes in a landscape composed of reclaimed grassland expanses and isolated forest remnants. Elk routinely select forested ridge-tops as resting and ruminating sites. These locations are characterized by sparse or absent leaf litter, trampled and diminished vegetation, large deposits of dung, and urine-saturated soils. In fall 2003, a series of fenced ungulate exclosures were constructed; 8 on highly disturbed forested ridge-tops and 8 on reclaimed grasslands. In the southern Mexican state of Oaxaca, many indigenous communities further their struggles for greater political and cultural autonomy by working with transnational non-governmental organizations (NGOs). Communication technology (what I call comtech) is increasingly vital to these intersecting socio-spatial relations of activism and advocacy. In this dissertation, I examine how comtech offer indigenous individuals and organizations with the means for visualizing their political-cultural agendas. In 1975, the US Fish and Wildlife Service listed the grizzly bear as a threatened species under the Endangered Species Act. Following the listing, a recovery plan was drafted in which the Bitterroot Ecosystem of central Idaho and extreme western Montana was one of six proposed grizzly bear recovery areas. It was the only one of the six, however, which did not contain a resident population of grizzlies. The Fish and Wildlife Service eventually accepted a proposal submitted by a coalition of environmental and timber industry groups. Degenerative suspensory ligament desmitis (DSLD) is a debilitating disease of connective tissues seen in many breeds but has become prevalent in the Peruvian Pasohorse. DSLD is believed to be a genetic disorder caused by one primary founder and most likely has a recessive mode of inheritance although a dominant or co-dominant mode of inheritance has not been ruled out.

This dissertation discusses the implied reader in the EnCuento series illustrated childrens stories. All the stories are written by well-known Hispanic authors. This work elucidates historical, cultural, and semiotic gaps in the reading process. It explores the ways in which textual elements- such as style, focalization, and manipulation of readers expectations - affect the implied readers ability to produce or extract meaning. Unlike straight turning, the effective cutting conditions and tool geometry in contour turning operations are changing with changing workpiece profile. This causes a wide variation in machining performance such as chip flow and chip breakability during the operation. This thesis presents a new methodology for optimizing the machining performance, namely, chip breakability and surface roughness in contour finish turning operations. Tuberous sclerosis complex (TSC) 1 and TSC2 are thought to be involved in protein translational regulation and cell growth, and loss of their function is a cause of TSC and lymphangioleiomyomatosis (LAM). However, TSC1 also activates Rho and regulates cell adhesion. We found that TSC2 modulates actin dynamics and cell adhesion and the TSC1-binding domain (TSC2-HBD) is essential for this function of TSC2. Expression of TSC2 or TSC2-HBD in TSC2-/- cells promoted Rac1 activation, inhibition of Rho, stress fiber disassembly, and focal adhesion remodeling. Evaluation Of Us 119 Pine Mountain Safety Improvements: Ihsdm Analysis Of Post Construction, Will Conkin, Nikiforos Stamatiadis
Kentucky Transportation Center Research Report
The spot improvements on US 119 in Letcher County between Whitesburg and Partridge were an attempt to improve safety throughout the corridor. To achieve this goal, the roadway alignment and cross section were changed at various locations. In addition, problems related to truck traffic were considered in redefining the roadway geometries. The total length of the spot improvement locations was approximately 6.9 miles.

Methods and apparatus for analyzing the chemical composition of vulnerable plaques with an intravascular catheter having a near-infrared light source, a fiber-optic probe, a mechanism for directing the light from the light source into a blood vessel, and detectors for detecting light reflected or scattered by the tissue. The light source may be a tunable laser, and may transmit an incident beam having a wavelength ranging from 1400 to 4100 nm. A computer may be included to receive and process the spectral data in the analysis of the vulnerable plaques. A catheter system may be configured to provide near-IR spectrometric …
