The development of a highly sensitive ruthenium-based fluorescent staining solution isdescribed in this dissertation. The in-house synthesized ruthenium complex (RuMS)containing both sulfonated and non-sulfonated ligand has detection limit of 1 ng ofprotein that is better than colloidal coomassie, silver and ruthenium complex containingall sulfonated ligands (RuBPS). RuMS stain has 100-fold dynamic range and does notinterfere with subsequent mass spectral identification of proteins. The capability of inhousesynthesis of the staining solution makes it a viable cost-effective alternative to theexpensive commercially available fluorescent stain, Sypro Ruby. The low detection limit,broad linear dynamic range and compatibility with mass spectrometry, make thedevelopment of this …

We have developed CO2-selective membranes to purified hydrogen and nitrogenfor fuel cell processes. Hydrogen purification impacts other industries such as ammoniaproduction and flue gas purification at reduced costs.Dense chitosan membranes were used for the first time to separate CO2 from amixture of 10% CO2, 10% H2, and 80% N2 at temperatures of 20 – 150oC and feedpressures of 1.5 atm – 5 atm. At 1.5 atm and 20 – 150oC, dry chitosan membranesachieved CO2 permeabilities, CO2/N2 and CO2/H2 separation factors of 0.383 – 24.3barrers, 10.7 – 3.40, and 4.54 – 1.50, respectively. Successful forest management requires an understanding of the habitat requirements of individual species at multiple spatial scales. The cerulean warbler (Dendroica cerulea) is a neotropical migratory songbird that has recently gained widespread attention as a species of conservation and management concern. It breeds in mature, deciduous forests of eastern North America and has experienced precipitous range-wide declines over the last 40 years. Although Kentucky likely supports one of the largest breeding populations of the species, no information exists on cerulean warbler habitat selection within the state. Suspended Sediment And Pathogen Transport In Two Inner Bluegrass Karst Ground-Water Basins, Woodford County, Kentucky, Thomas M. Reed

Multiple parameters were monitored during an 18-month period in two karst groundwater basins in Woodford County, Kentucky, in order to assess the effects of land use on water quality. Blue Hole Spring drains a primarily urban area, whereas spring SP-2 drains an agricultural area. Water-quality parameters were monitored manually weekly or biweekly, as well as more frequently during storms. Discharge (Q), temperature (T), specific conductance (SC), and turbidity were continuously monitored and logged at 15-minute intervals. Phase measuring Profilometry (PMP) is an important technique used in 3D data acquisition. Many variations of the PMP technique exist in the research world. The technique involves projecting phase shifted versions of sinusoidal patterns with known frequency. The 3D information is obtained from the amount of phase deviation that the target object introduces in the captured patterns. Using patterns based on single frequency result in projecting a large number of patterns necessary to achieve minimal reconstruction errors. Taxonomic Revision Of Zelomorpha Ashmead, 1900 And Hemichoma Enderlein, 1920 (Braconidae: Agathidinae) With A Phylogenetic Analysis Of Color Patterns, Carlos Eduardo Sarmiento-Monroy

A revision and a phylogenetic analysis of the genera Zelomorpha Ashmead, 1900 and Hemichoma Enderlein, 1920 were conducted. Phylogenetic analyses used molecular and morphological data. A total of 39 sequences were obtained for COI (887 bases long) and 57 for 28S (1254 bases long). DNA sequences were aligned manually and also aligned with ClustalW (Thompson et al. 1997). Parsimony, Maximum Likelihood, and Bayesian approaches were applied to phylogenetic analyses with each gene region analyzed separately and in a combined analysis. For many, contemporary theatre is represented by the musical. The form remains, however, virtually unstudied by literary scholars. In part, this may be a result of the difficulty of accessing the texts. Reading a musical from a traditional codex is no easy matter. The integration of text and music in a musical make it inappropriate to separate the two. One can try to follow along with a cast recording. In most cases, though, this is awkward. For insects that develop inside discrete hosts both host size and host quality constrain offspring growth, influencing the evolution of body size and life history traits. This dissertation examines the effects of host size, host quality, and intraspecific competition on life history and associated traits of populations of the seed-feeding beetle S. limbatus adapted to different host plants, and quantifies population differences in phenotypic plasticity. Populations of the study correspond to divergent clades of the species phylogeography (Colombia and United States).

Subsurface solutional pathways make limestone terrains sensitive to changes in soil properties that regulate flows to the epikarst. This study examines biogeomorphic factors responsible for changed water movements and erosion in fluviokarst slopes deforested 200 years ago along the Kentucky River, Kentucky. In this project, infiltration and water content data from forest and fescue grass soil profiles were analyzed within a detailed overview of system factors regulating hillslope hydrology. Results show that grass has growth and rooting characteristics that tend to create a larger volume of lateral water movement in upper soil layers than occurs under forests. This sets up …

The initial study herein assessed mineral digestibility in situations when reducedamounts of inorganic and organic (proteinates) trace minerals (TM) were fed in finishingpigs, and their long-term effects on body mineral status. The second study was aslaughter-investigation that tested the impact of lean growth potential on phosphorusbody accretion from 30 to 110 kg.Organic TM exhibited neither improvement in digestibility nor in total retention;fecal excretion responded quantitatively to mineral intake independently of the source.Contents of copper in kidney and zinc in liver were higher for pigs fed the organic form.Phosphorus content was linearly related to live weight, empty body weight, andnitrogen content. …

The Indian Chipko movement is analyzed as a case study employing a geographically-informed political ecology approach. Political ecology as a framework for the study of environmental movements provides insight into the complex issues surrounding the structure of Indian society, with particular attention to its ecological and political dimensions. This framework, with its focus on social structure and ecology, is distinct from the more traditional approaches to the study of social movements, which tend to essentialize their purpose and membership, often by focusing on a single dimension of the movement and its context. Salamanders of the genus Ambystoma are a unique model organism system because they enable natural history and biomedical research in the laboratory or field. We developed Sal-Site to integrate new and existing ambystomatid salamander research resources in support of this model system. Variable-rate technology (VRT) adds complexity to application equipment, thereby confounding the assessment of applicator performance. The intent of this investigation was to assess the rate response of various VRT granular applicators: two spinner spreaders (A and B), and two pneumatic applicators (C and D). Variable-rate (VR) tests were conducted to quantify the rate response characteristics (delay and transition times) for the applicators. A sigmoidal function was used to model the rate response for five of the six tests. Applicator A exhibited a linear response during decreasing rate changes. The present invention provides a monoclonal anti-idiotype antibody 11D10 that elicits an immune response against a specific epitope of a high molecular weight mucin of human milk fat globule (HMFG) and a hybridoma that produces 11D10. The hybridoma that produces 11D10 was selected by specific procedures. 11D10 induces an immunological response to HMFG in nice, rabbits, monkeys and patients with advanced HMFG-associated tumors. This invention provides compositions derived from polynucleotide sequences encoding the variable light and/or variable heavy regions of monoclonal anti-idiotype antibody 11D10, as well as polypeptides encoded thereby. Diazinon [O,O-diethyl O-2-isopropyl-6-methyl(pyrimidine-4-yl) phosphorothioate] and imidacloprid [1-(1-[6-chloro-3-pyridinyl]methyl)-N-nitro-2-imidazolidinimine] are applied to lawns for insect control simultaneously with nitrogenous fertilizers such as urea, but their potential effect on urease activity and nitrogen availability in turfgrass management has not been evaluated. Urease activity in enzyme assays, washed cell assays, and soil slurries was examined as a function of insecticide concentration. Intact cores from field sites were used to assess the effect of insecticide application on urease activity in creeping bentgrass (Agrostis palustris Huds.) and bluegrass (Poa pratensis L.) sod. BACKGROUND: Unloading of skeletal muscle causes atrophy and loss of contractile function. In part, this response is believed to be mediated by the transcription factor nuclear factor-kappa B (NF-kappaB). Both curcumin, a component of the spice turmeric, and N-acetylcysteine (NAC), an antioxidant, inhibit activation of NF-kappaB by inflammatory stimuli, albeit by different mechanisms. In the present study, we tested the hypothesis that dietary curcumin or NAC supplementation would inhibit unloading-induced NF-kappaB activity in skeletal muscle and thereby protect muscles against loss of mass and function caused by prolonged unloading.

BACKGROUND: Creatine kinase (CK) links phosphocreatine, an energy storage system, to cellular ATPases. CK activity serves as a temporal and spatial buffer for ATP content, particularly in fast-twitch skeletal muscles. The extraocular muscles are notoriously fast and active, suggesting the need for efficient ATP buffering. This study tested the hypotheses that (1) CK isoform expression and activity in rat extraocular muscles would be higher, and (2) the resistance of these muscles to fatigue would depend on CK activity.

Several primary murine and human B lymphomas and cell lines were found to constitutively express high levels of the activated form of c-jun N-terminal kinase (JNK), a member of the mitogen-activated protein (MAP) kinase family. Proliferation of murine B lymphomas CH31, CH12.Lx, BKS-2, and WEHI-231 and the human B lymphomas BJAB, RAMOS, RAJI, OCI-Ly7, and OCI-Ly10 was strongly inhibited by SP600125, an anthrapyrazolone inhibitor of JNK, in a dose-dependent manner. The lymphoma cells underwent apoptosis and arrested at the G2/M phase of cell cycle. Furthermore, JNK-specific small interfering RNA (siRNA) inhibited the growth of both murine and human B lymphomas. …

BACKGROUND: Runx transcription factors play critical roles in the developmental control of cell fate and contribute variously as oncoproteins and tumor suppressors to leukemia and other cancers. To discover fundamental Runx functions in the cell biology of animal development, we have employed morpholino antisense-mediated knockdown of the sea urchin Runx protein SpRunt-1. Previously we showed that embryos depleted of SpRunt-1 arrest development at early gastrula stage and underexpress the conventional protein kinase C SpPKC1.

RESULTS: We report here that SpRunt-1 deficiency leads to ectopic cell proliferation and extensive apoptosis. Suppression of the apoptosis by pharmacological inhibition of caspase-3 prevents the …

Few studies have examined the water quality impact of manure use in no-tillage systems. BACKGROUND: HIV Associated Dementia (HAD) is a common complication of human immunodeficiency virus (HIV) infection that erodes the quality of life for patients and burdens health care providers. Intravenous drug use is a major route of HIV transmission, and drug use is associated with increased HAD. Specific proteins released as a consequence of HIV infection (e.g., gp120, the HIV envelope protein and Tat, the nuclear transactivating protein) have been implicated in the pathogenesis of HAD.
