Stem cells off er an enormous pool of resources for the understanding of the human body. One proposed use of stem cells has been as an autologous therapy. The use of stem cells for neurodegenerative diseases has become of interest. Clinical applications of stem cells for Alzheimer disease, Parkinson disease, amyotrophic lateral sclerosis, and multiple sclerosis will increase in the coming years, and although great care will need to be taken when moving forward with prospective treatments, the application of stem cells is highly promising.

The pro-oncogenic transcription factor STAT3 is constitutively activated in a wide variety of tumours that often become addicted to its activity, but no unifying view of a core function determining this widespread STAT3-dependence has yet emerged. We show here that constitutively active STAT3 acts as a master regulator of cell metabolism, inducing aerobic glycolysis and down-regulating mitochondrial activity both in primary fibroblasts and in STAT3-dependent tumour cell lines. As a result, cells are protected from apoptosis and senescence while becoming highly sensitive to glucose deprivation. We show that enhanced glycolysis is dependent on HIF-1a up-regulation, while reduced mitochondrial activity is …

A study investigating what factors are present leading to pilots submitting voluntary anomaly reports regarding their flight performance was conducted. The study employed statistical methods, text mining, clustering, and dimensional reduction techniques in an effort to determine relationships between factors and anomalies. A review of the literature was conducted to determine what factors are contributing to these anomalous incidents, as well as what research exists on human error, its causes, and its management. The self-assembly of chiral amphiphilic molecules in aqueous solutions is of particular interest because the chirality of individual molecules is often expressed in their supermolecular structures. Self-assembled tubes made of chiral amphiphilic molecules represent useful supramolecular architectures which hold promise as controlled release vehicles for drug delivery, encapsulates for functional molecules, and nanoreactors for chemical reactions. In a distributed wireless system, multiple network nodes behave cooperatively towards a common goal. Though such assumptions on cooperation are desirable (e.g., controlling the transmit power level, reducing interference for each other, revealing private information, adhering to network policies) for analyzing and modeling, certain nodes belonging to a real-world system have often shown to deviate. This study evaluated alternative sulfide treatment processes for potable water systems that rely on groundwater supplies. Research for this study was conducted at the Imperial Lakes (IL) and Turner Road (TR) water treatment plants (WTPs) in Polk County, Florida. These WTPs are in the process of refurbishment and expansion, and will require the installation of a new groundwater well. The IL and TR WTPs both rely upon groundwater sources that contain total sulfide at concentrations ranging from 1.4 to 2.6 mg/L. This thesis presents a study on the strength properties of the different pervious pavement systems installed at the Stormwater Management Academy field laboratory at University of Central Florida (UCF), Orlando. The strength tests were performed both in the laboratory and in the field. Laboratory testing was conducted to determine the compressive strength and flexural strength of the various pavement surfaces. The general intent of my thesis is to write two novellas that show the differences in ways of life in two completely dissimilar states, and the conflicts that occurred therein. Therefore, the novellas will include a variation in gender, setting, and conflict. The main characters will be roughly the same age, but I will explore how the setting they live in has affected conflicts they have to deal with. Subspecific taxonomic designations solely based on morphological characters can often lead to erroneous assumptions about the evolutionary history of populations. This study sought to investigate evolutionary questions and conservation implications associated with morphological subspecific designations of island populations. To this end, I focused my attention on the Lower Keys of Florida, a unique chain of islands with well-known geologic history and rich in endemic, endangered subspecies. In this dissertation, I analyze the 20th century text, A Room of One's Own, by Virginia Woolf (2005), and I engage with Woolf's concept of a woman's need for a room of her own in which she can be free to think for herself, study, write, or pursue other interests away from the oppression of patriarchal societal expectations and demands. Through library-based research, I identify four screens in Woolf's work through which she viewed and critiqued culture, and I use these screens to reconceptualize "a room of one's own" in 21st Century terms. Ring opening metathesis polymerization (ROMP) is established as one of the efficient controlled living polymerization methods which have various applications in polymer science and technology fields. The research presented in this dissertation addresses several applications of multifunctional well-defined norbornene-based block copolymers synthesized by ROMP using ruthenium-based Grubbs catalysts. These novel block copolymers were applied to stabilize maghemite nanoparticles, creating the superparamagnetic polymeric nanocomposites. The Jaggregation properties of the porphyrin dyes were improved via self-assembly with a customized norbornene polymer. Applications of surface plasmon polaritons (SPPs) have thus far emphasized visible and near-infrared wavelengths. Extension into the long-wave infrared (LWIR) has numerous potential advantages for biosensors and waveguides, which are explored in this work. A surface plasmon resonance (SPR) biosensor that operates deep into the infrared (3-11 µm wavelengths) is potentially capable of biomolecule recognition based on both selective binding and characteristic vibrational modes. Interleukin-7 (IL-7) is an essential lymphocyte growth factor required for the survival and proliferation of mature T-cells. As a therapeutic agent, IL-7 has the potential to restore T-cell numbers following immune depletion and to promote immunity against cancers. While the survival function of IL-7 is well established, less is known about how it supports T-cell expansion, a critical feature of the immune response. To study the biological effects of IL-7 on T-cell growth, we developed an in vitro culture technique to expand T-cells ex vivo. Current treatment for type I diabetes includes delivery of insulin via injection or pump, which is highly invasive and expensive. The production of chloroplast-derived proinsulin should reduce cost and facilitate oral delivery. Therefore, tobacco and lettuce chloroplasts were transformed with the cholera toxin B subunit fused with human proinsulin (A, B, and C peptides) containing three furin cleavage sites (CTB-PFx3).
