A Laudable Ambition Fired Her Soul Conduct Fiction Helps Define Republican Womanhood, Female Communities, And Women's Education In The Works Of Judith Sargent Murray, Hannah Webster Foster, And Susanna Haswell Rowson, Jessica Crystal Workman Jan 2011

A Laudable Ambition Fired Her Soul Conduct Fiction Helps Define Republican Womanhood, Female Communities, And Women's Education In The Works Of Judith Sargent Murray, Hannah Webster Foster, And Susanna Haswell Rowson, Jessica Crystal Workman

Electronic Theses and Dissertations

This study examines the major works of Judith Sargent Murray, Hannah Webster Foster, and Susanna Haswell Rowson, three major writers of the 1790s whose writing responds to the ideologies of the early American Republic. I suggest that Murray, Foster, and Rowson write conduct fiction which responds to the changing attitudes toward women and education after the American Revolution. Using fiction, these authors comment on the republican woman, the need for women’s education, and the necessity for women to gather in communities for support. Despite the prevailing notion that reading too many novels would corrupt young women, Judith Sargent Murray’s novella, …

Stable Expression Of Tuberculosis Vaccine Antigen In Lettuce Chloroplasts, Priya Saikumar Lakshmi Jan 2011

Stable Expression Of Tuberculosis Vaccine Antigen In Lettuce Chloroplasts, Priya Saikumar Lakshmi

Electronic Theses and Dissertations

Tuberculosis (TB) is caused by Mycobacterium tuberculosis and is one of the leading reasons of death by an infectious bacterial pathogen. The development of TB vaccines has been recognized as a major public health priority by the World Health Organization. In this study, a potential candidate antigen, ESAT-6 (6 kDa early secretory antigenic target) was fused with cholera toxin B subunit (CTB). Transplastomic lettuce plants were generated expressing these fusion proteins. Site-specific transgene integration into the chloroplast genome was confirmed by polymerase chain reaction and Southern blot analysis. In transplastomic leaves, expression levels of fusion protein (CTB-ESAT6) varied depending upon …

Evolutionary Relationships Among Staphylococci And The Prevention Of Staphylococcus Aureus Nasal Colonization, Ryan Paul Lamers Jan 2011

Evolutionary Relationships Among Staphylococci And The Prevention Of Staphylococcus Aureus Nasal Colonization, Ryan Paul Lamers

Electronic Theses and Dissertations

Staphylococcus is a significant cause of human infection and mortality, worldwide. Currently, there are greater than 60 taxa within Staphylococcus, and nearly all are pathogenic. The collective potential for virulence among species of Staphylococcus heightens the overall clinical significance of this genus and argues for a thorough understanding of the evolutionary relationships among species. Within Staphylococcus, aureus is the most common cause of human infection, where nasal carriage of this bacterium is a known risk factor for autoinfection. The predisposition to infection by nasal carriers of S. aureus, and the ease with which strains are transferred between individuals, suggests that …

Prediction Of Survival Of Early Stages Lung Cancer Patients Based On Er Beta Cellular Expressions And Epidemiological Data, Evgeny Martinenko Jan 2011

Prediction Of Survival Of Early Stages Lung Cancer Patients Based On Er Beta Cellular Expressions And Epidemiological Data, Evgeny Martinenko

Electronic Theses and Dissertations

We attempted a mathematical model for expected prognosis of lung cancer patients based on a multivariate analysis of the values of ER-interacting proteins (ERbeta) and a membrane bound, glycosylated phosphoprotein MUC1), and patients clinical data recorded at the time of initial surgery. We demonstrate that, even with the limited sample size available to use, combination of clinical and biochemical data (in particular, associated with ERbeta and MUC1) allows to predict survival of lung cancer patients with about 80% accuracy while prediction on the basis of clinical data only gives about 70% accuracy. The present work can be viewed as a …

Indirect Estimates Of Gene Flow And Its Conservation Implications In The Striped Newt (Notophthalmus Perstriatus), Sarah Elizabeth May Jan 2011

Indirect Estimates Of Gene Flow And Its Conservation Implications In The Striped Newt (Notophthalmus Perstriatus), Sarah Elizabeth May

Electronic Theses and Dissertations

This study used indirect methods to estimate patterns of gene flow in a rare salamander species, the striped newt (Notophthalmus perstriatus). First, we used combined genetic and ecological methods to determine whether populations that appear to exist in two regions separated by 125 km, exhibited genetic and ecological distinctness such that the regions demarcate separate conservation units. Using mtDNA (cyt-b), we found that haplotypes were shared between localities within each region but none were shared between regions. Niche-based distribution modeling revealed significant differences in the ecological setting between the two regions. In combination, the absence of evidence for recent genetic …

The Effects Of Urbanization On Cypress (Taxodium Distichum) In Central Florida, Lisa A. Mccauley

Electronic Theses and Dissertations

Urbanization is accelerating in the United States and is contributing to fragmentation of natural habitats, causing changes in species composition and declines in native species. Human population growth in Orlando is typical of growth in the southeastern United States and throughout the range of cypress (Taxodium distichum). Orlando has numerous isolated cypress wetlands, called cypress domes, and many remain among the current urbanized area. This makes Orlando ideal to study the effects of urbanization on cypress domes. Specifically, I tested how urbanization and its effects on fragmentation, hydrology, and fire regime) affected (a) the numbers and spatial pattern of cypress …

Synthesis, Characterization And Antibacterial Activity Of Silver Embedded Silica Nanoparticle/Nanogel Formulation, Roseline Menezes Jan 2011

Synthesis, Characterization And Antibacterial Activity Of Silver Embedded Silica Nanoparticle/Nanogel Formulation, Roseline Menezes

Electronic Theses and Dissertations

The antibacterial property of silver (Ag) has been known since ancient time. It is reported in the literature that silver nanoparticles (AgNPs) exhibit improved antibacterial properties in comparison to silver ions of equivalent metallic Ag concentration. Such improvement in antibacterial activities is due to the high surface area to volume ratio of AgNPs (which facilitates interaction with the bacterial cells), increased release of silver ions and direct intra-cellular uptake of AgNPs leading to localized release of Ag ions. To date, over 300 consumer products containing AgNPs are available in the market and the inventory is rapidly expanding. The antibacterial efficacy …

Direct, Hands-On Or Inquiry Instruction A Study Of Instructional Sequencing And Motivation In The Science Classroom, Jamie Vander Wiede

Electronic Theses and Dissertations

Currently, a debate exists between the strengths and weaknesses of direct and inquiry instruction. Inquiry instruction is related to positive effect on learner motivation whereas supporters of direct instruction point to its ability to adequately support learners’ working memories (Hmelo-Silver, Duncan, & Chinn, 2007; Kirschner, Sweller, & Clark, 2006; Kuhn, 2007; Sweller, 1988). This study examined the possibility of combining the best features of both inquiry and direct instruction by sequencing them together. A two-part lesson on electrical circuits was presented in three separate sequences of instruction to middle school students to determine if differences in student motivation and academic …

Expression Of Trichoderma Reesei Beta]-Mannanase In Tobacco Chloroplasts And Its Utilization In Lignocellulosic Woody Biomass Hydrolysis, Pankaj M. Agrawal

Electronic Theses and Dissertations

Lignocellulosic ethanol offers a promising alternative to conventional fossil fuels. One among the major limitations in the lignocellulosic biomass hydrolysis is unavailability of efficient and environmentally biomass degrading technologies. Plantbased production of these enzymes on large scale offers a cost effective solution. Cellulases, hemicellulases including mannanases and other accessory enzymes are required for conversion of lignocellulosic biomass into fermentable sugars. β- mannanase catalyzes endo-hydrolysis of the mannan backbone, a major constituent of woody biomass. In this study, man1 gene encoding β-mannanase was isolated from Trichoderma reesei and expressed via the chloroplast genome. PCR and Southern hybridization analysis confirmed the site-specific …

The Effects Of Presentation Mode And Pace On Learning Immunology With Computer Simulation A Cognitive Evaluation Of A Multimedia Learning Resource, Radakovich Kristy Bradley

Electronic Theses and Dissertations

Multimedia learning tools have the potential to benefit instructors and learners as supplemental learning materials. However, when such tools are designed inappropriately, this can increase cognitive taxation and impede learning, rendering the tools ineffective. Guided by the theoretical underpinnings provided by cognitive load theory and the cognitive theory of multimedia learning, this study sought to empirically evaluate the effectiveness of a multimedia simulation tool aimed at teaching immunology to novices in an instructional setting. The instructional mode and pace of the tool were manipulated, the three levels of each variable yielding nine experimental groups. The effects of mode and pace …

Modeling Transport And Protein Adsorption In Microfluidic Systems, Craig Finch Jan 2011

Modeling Transport And Protein Adsorption In Microfluidic Systems, Craig Finch

Electronic Theses and Dissertations

Mass transport limitations and surface interactions are important phenomena in microfluidic devices. The flow of water is laminar at small scales and the absence of turbulent mixing can lead to transport limitations, especially for reactions that take place at surfaces. Microscale devices have a high ratio of surface area to volume, and proteins are known to adsorb preferentially at interfaces. Protein adsorption plays a significant role in biology by mediating critical processes such as the attachment of cells to surfaces, the immune response and the coagulation of blood. Simulation tools that can quantitatively predict transport and protein adsorption will enable …

In Search Of Satanists Examining The Accounts Of Deviant Religious Practitioners, Chris P. Mcdaniel

Electronic Theses and Dissertations

Despite the glut of literature focusing on numerous aspects of mainline American religious life, there is a surprising dearth of information regarding deviant religious practitioners. Importantly, there remains a lack of focus on the specificities of religious and spiritual deviance, and the accounts of those who engage in such practices. This exploratory study closely examines the members of one such religion; specifically, the Church of Satan. Despite the stigma associated with Satanism, individuals continue to willfully engage in such practices. Research uses face-toface, semi-structured interviews to better discern the rationale behind Satanic worship as understood by modern-day believers. Particular attention …

Neuronal culture systems have many applications, such as basic research into neuronal structure, function, and connectivity as well as research into diseases, conditions, and injuries affecting the brain and its components. In vitro dissociated neuronal systems have typically been derived from embryonic brain tissue, most commonly from the hippocampus of E18 rats. This practice has been motivated by difficulties in supporting regeneration, functional recovery and long-term survival of adult neurons in vitro. The overall focus of this dissertation research was to develop a dissociated neuronal culture system from human and animal adult brain tissue, one more functionally and developmentally correlative …

Discovery And Optimization Of Novel Small-Molecular Inhibitors Suppressing Stat3-Dependent Tumor Process, Xiaolei Zhang Jan 2011

Discovery And Optimization Of Novel Small-Molecular Inhibitors Suppressing Stat3-Dependent Tumor Process, Xiaolei Zhang

Electronic Theses and Dissertations

With the critical role of aberrantly active Signal Transducer and Activator of Transcription (Stat) 3 protein in many human cancers, selective small-molecule inhibitors targeting the dimerization event which is required for stat3 activation, would be valuable as therapeutic agents. And the inhibitors will be useful chemical probes to clarify the complex biological functions of Stat3. By computational and structural analyses of the interaction between Stat3 and the lead dimerization disruptor, S3I-201, we have designed a diverse set of analogs. One of the most active analogs, S3I-201.1066 is derived to contain a cyclo-hexyl benzyl moiety on the amide nitrogen, which increases …

Impact Of Land Management On House Mice And Red Imported Fire Ants, Jesse R. Abelson Jan 2011

Impact Of Land Management On House Mice And Red Imported Fire Ants, Jesse R. Abelson

Electronic Theses and Dissertations

Understanding of mechanisms that limit the abundance and distribution of species is central to ecology. The failure of mechanisms to regulate populations can result in population outbreaks. There have been two outbreaks of house mice in the past decade in central Florida. In my study, I examine the efficacy of landscape management in the form of mowing and plowed soil barriers to limit or prevent outbreaks of house mice in a former agricultural area. House mouse populations were highly variable, but were unaffected by mowing or plowed soil barriers. Red imported fire ants were ubiquitous in the study area regardless …

The Application Of Two-Photon Absorbing Probes In Lysosomal, Zinc Ion Sensing And Folate Receptor-Targeted Bioimaging, Xuhua Wang Jan 2011

The Application Of Two-Photon Absorbing Probes In Lysosomal, Zinc Ion Sensing And Folate Receptor-Targeted Bioimaging, Xuhua Wang

Electronic Theses and Dissertations

Two-photon fluorescence microscopy (2PFM) has become a powerful technique for bioimaging in non-invasive cancer diagnosis and also investigating the mechanization and original of a variety of diseases by tracking various biological processes. Because the fluorescence emission by two photon absorbing (2PA) is directly proportional to the square of the intensity of excitation light, this intrinsic property of 2PA provides 2PFM great advantages over traditional one-photon fluorescence microscopy (1PFM), including high 3D spatial localization, less photodamage and interference from biological tissue because of using longer wavelength excitation (700-1300 nm). However, most 2PA probes are hydrophobic and their photostabilities are questionable, severely …

Metal Blacks As Scattering Centers To Increase The Efficiency Of Thin Film Solar Cells, Deep R. Panjwani Jan 2011

Metal Blacks As Scattering Centers To Increase The Efficiency Of Thin Film Solar Cells, Deep R. Panjwani

Electronic Theses and Dissertations

Metal nano particles are investigated as scattering centers on front surface of thin-film solar cells to improve efficiency. The principle is that scattering, which is enhanced near the plasmon resonance frequency of the particle and depends on particle size, increases the effective optical path length of incident light, leading to more light absorption in active layer of thin film solar cell. The particular types of particles investigated here are known as "metal-black", well known as an IR absorber for bolometric infrared detectors. Gold-black was deposited on commercial thin-film solar cells using a thermal evaporator in a nitrogen ambient at pressures …

Genetically Modified Es Cells Enhance Cardiac Repair And Regeneration In The Infarcted Heart, Carley E. Glass Jan 2011

Genetically Modified Es Cells Enhance Cardiac Repair And Regeneration In The Infarcted Heart, Carley E. Glass

Electronic Theses and Dissertations

Transplanted embryonic stem (ES) cells following myocardial infarction (MI) contribute to limited cardiac repair and regeneration with improved function. Therefore novel strategies are still needed to enhance the efficacy by which ES cells differentiate into cardiac cell types and inhibit adverse remodeling in the infarcted myocardium. Our studies evaluate whether genetic manipulation of transplanted ES cells employing miR- 1, a pro-cardiac microRNA, and TIMP-1, an anti-apoptotic and anti-fibrotic protein, will enhance cardiac myocyte differentiation, inhibit native cardiac apoptosis, and reduce fibrosis in the infarcted myocardium. Furthermore, we assess levels of associated pro- (caspase-3, PTEN) and anti-(Akt) apoptotic proteins as well …

The Relative Recoverability Of Dna And Rna Profiles From Forensically Relevant Body Fluid Stains, Charly Parker Jan 2011

The Relative Recoverability Of Dna And Rna Profiles From Forensically Relevant Body Fluid Stains, Charly Parker

Electronic Theses and Dissertations

Biological material (fluids or tissues) whether from the victim or suspect is often collected as forensic evidence, and methods to obtain and analyze the DNA found in that material have been well established. The type of body fluid (i.e. blood, saliva, semen, vaginal secretions, and menstrual blood) from which the DNA originated is also of interest, and messenger RNA typing provides a specific and sensitive means of body fluid identification. In order for mRNA profiling to be utilized in routine forensic casework, RNA of sufficient quantity and quality must be obtained from biological fluid stains and the methods used for …

Modeling Carbon Allocation, Growth And Recovery In Scrub Oaks Experiencing Aboveground Disturbance, Troy J. Seiler Jan 2011

Modeling Carbon Allocation, Growth And Recovery In Scrub Oaks Experiencing Aboveground Disturbance, Troy J. Seiler

Electronic Theses and Dissertations

Allocation of assimilated carbon amongst plant metabolic processes and tissues is important to understanding ecosystem carbon cycles. Due to the range of spatio-temporal scales and complex process interactions involved, direct measurements of allocation in natural environments are logistically difficult. Modeling approaches provide tools to examine these patterns by integrating finer scale process measurements. One such method is root:shoot balance, where plant growth is limited by either shoot activity (i.e. photosynthesis) or root activity (i.e. water and nutrient uptake). This method shows promise for application on frequently disturbed systems which perturb aboveground biomass and thus create imbalances in root and shoot …

Variational Embedded Solitons, And Traveling Wavetrains Generated By Generalized Hopf Bifurcations, In Some Nlpde Systems, Todd Blanton Smith Jan 2011

Variational Embedded Solitons, And Traveling Wavetrains Generated By Generalized Hopf Bifurcations, In Some Nlpde Systems, Todd Blanton Smith

Electronic Theses and Dissertations

In this Ph.D. thesis, we study regular and embedded solitons and generalized and degenerate Hopf bifurcations. These two areas of work are seperate and independent from each other. First, variational methods are employed to generate families of both regular and embedded solitary wave solutions for a generalized Pochhammer PDE and a generalized microstructure PDE that are currently of great interest. The technique for obtaining the embedded solitons incorporates several recent generalizations of the usual variational technique and is thus topical in itself. One unusual feature of the solitary waves derived here is that we are able to obtain them in …

Translocation Of The Cholera Toxin A1 Subunit From The Endoplasmic Reticulum To The Cytosol, Michael Prentice Taylor Jan 2011

Translocation Of The Cholera Toxin A1 Subunit From The Endoplasmic Reticulum To The Cytosol, Michael Prentice Taylor

Electronic Theses and Dissertations

AB-type protein toxins such as cholera toxin (CT) consist of a catalytic A subunit and a cell-binding B subunit. CT proceeds through the secretory pathway in reverse, termed retrograde trafficking, and is delivered to the endoplasmic reticulum (ER). In order for the catalytic A1 subunit to become active it must separate from the rest of the holotoxin, and this dissociation event occurs in the ER lumen. CTA1 assumes an unfolded conformation upon dissociation from the holotoxin and is recognized by ERassociated degradation (ERAD), a quality control system that recognizes and exports misfolded proteins to the cytosol for degradation by the …
