One of the pathological hallmarks of Alzheimer's disease (AD) is the amyloid plaque deposition in aging brains by aggregation of amyloid-β (Aβ) peptides. In this study, the effect of chloroplast derived myelin basic protein (MBP) fused with cholera toxin subunit B (CTB) was investigated in advanced diseased stage of human and mouse AD brains. The CTB-fusion protein in chloroplasts facilitates transmucosal delivery in the gut by the natural binding ability of CTB pentameric form with GM1 receptors on the intestinal epithelium. Further, bioencapsulation of the MBP within plant cells confers protection from enzymes and acids in the digestive system. Here, …

Malaria, the disease caused by Plasmodium sp., claims the lives of over 1 million people every year, with Plasmodium falciparum causing the highest morbidity. Rapidly acquiring drug resistance is threatening to exhaust our antimalarial drug arsenal and already requires the utilization of combination drug therapy in most cases. The global need for novel antimalarial chemical scaffolds has never been greater. Screening of natural product libraries is known to have higher hit rates than synthetic chemical libraries. Female mentoring success was investigated as an undergraduate intervention utilizing career development practices to reduce dysfunctional career thinking and STEM major retention in first year freshmen females within a living-learning community. Repeated measures MANOVAs and canonical correlations in the causal comparative research design evaluated mentoring’s influence on first year females. Male voluntary participants (n = 126) formulated the comparison group, and female voluntary participants (n = 75) filled the treatment group. The purpose of this action research study was to document 8th grade students' experiences at a residential marine learning facility as they discovered the interconnections between life on Earth and its dependency on ocean health. My goal was for students to take their new knowledge and share it with others in a quest to become educated and caring stewards of the Earth's ocean. In this research, five different coagulants were evaluated to determine their effectiveness at removing turbidity, color and dissolved organic carbon (DOC) from a surface water in Sarasota County, Florida. Bench-scale jar tests that simulated conventional coagulation, flocculation, and sedimentation processes were used. Iron-based coagulants (ferric chloride and ferric sulfate) and aluminum-based coagulants (aluminum sulfate, polyaluminum chloride (PACl) and aluminum chlorohydrate (ACH)) were used to treat a highly organic surface water supply (DOC ranging between 10 and 30 mg/L), known as the Cow Pen Slough, located within central Sarasota County, Florida. Humankind’s tremendous industrial and technological progress over the last two centuries has been driven by the natural abundance and availability of fossil fuels. As those reserves deplete, the prudent course of action would be to develop other readily available fuel sources. Some research efforts using biomolecules involve the hydrogenases and nitrogenases with the goal of evolving dihydrogen. At the nitrogenase active site, the iron-molybdenum cofactor (FeMo-co) catalyzes the reduction of dinitrogen and protons to form ammonia and dihydrogen. Human activities are a major driver of biodiversity degradation and loss, especially in tropical forest areas, where forest-fringe towns and villages depend on the forests for their livelihoods. In order to reduce threats that human activities pose to biodiversity, livelihoods support programs are employed as economic incentives for biodiversity conservation. These programs support the livelihoods activities of local communities, with the aim of triggering favorable attitudes and behaviors towards conservation, and ultimately reduce biodiversity degradation. Human land use legacies have significant and long lasting impacts across landscapes. However, investigating the impacts of ancient land use legacies ( > 400 years) remains problematic due to the difficulty in detecting ancient land uses, especially those beneath dense canopies. The city of Caracol, one of the most important Maya archaeological sites in Belize, was abandoned after the collapse of the Maya civilization (ca. A.D. 900), leaving behind numerous structures, causeways, and agricultural terraces that persist beneath the dense tropical forest of western Belize. The Ras superfamily of GTPases are important regulators of morphogenesis involved in control of cytoskeletal dynamics, intracellular trafficking, apical-basal polarity and cell migration. Mis-regulation of GTPase signaling interferes with development and is linked to pathogenesis. Traditionally, GTPase signaling has been depicted as a series of independent linear pathways. However, recently it has become apparent that multiple GTPases can interact to regulate a single cellular process, functioning in poorly understood networks of cross talk between pathways during development. This work explores a binary reagent approach to increase the specificity of covalent inhibitors. In this approach, two ligand analogs equipped with inert pre-reactive groups specifically bind a target biopolymer. The binding event brings the pre-reactive groups in proximity with each other. The two groups react, generating active chemical intermediates that covalently modify and inactivate the target. In the present study we compare the new approach with the traditional single-component reagent strategy using DNA polymerase from bacteriophage T4 as a model target biopolymer. The goal of this research was to determine whether communication apprehension impacted reading comprehension in sixth, seventh, and eighth grade students and to examine the impact of family socio-economic status. Many studies have demonstrated the negative relationship between communication apprehension and academic achievement, however, studies of elementary and middle school students have been conspicuously missing from this research. Findings of this study indicated that the levels of communication apprehension rose slightly as grade level increased. Results showed that females in the study had higher levels of communication apprehension than males. The study also found that those students receiving free and …

This study examines lived experiences of veterinarians. A common feature of being a veterinarian is curing and caring for nonhuman animals. It is the love and connection most veterinarians share for animals that ignite their journey to become an animal doctor. Data collected during semi-structured interviews with 17 veterinarians reveal that there are many more intricacies to the job than just animal medicine. These veterinarians suggest that they must treat animals as learning tools during veterinary training and once they complete school, they deal with people and business on a regular basis. Massively parallel arrays of single walled carbon nanotubes (SWNT) have attracted significant research interests because of their ability to (i) average out inhomogeneities of individual SWNTs, (ii) provide larger on currents, and (iii) reduce noise to provide higher cutoff frequency for radio frequency applications. However, the array contains both metallic and semiconducting SWNTs and the presence of metallic nanotube in an aligned array negatively affects the device properties. Functional properties of living cells depend on the thermodynamic variables such as temperature and pressure. A unique tool to investigate volume effects on structure and metabolism of the cell and biomolecules is pressure perturbation. We have developed a new setup that enables micro-spectroscopy and optical imaging of individual live cells at variable pressure from 0.1 to 400 MPa. Following characterization of the setup, pressure and temperature effects on the secondary structure of the peptide Poly-L-glutamic acid (PGA) in deuterated water buffer solution were investigated. The determination of ancestry is an important part of an individual's identification when creating a biological profile. This thesis scrutinizes postcranial variation using over 65 osteometric sorting measurements in an attempt to identify those measurements that display the most significant differences among Koreans, Africans, and Europeans. Data was collected from four American skeletal collections and one South Korean skeletal collection for a total sample population of 306 individuals: 24 of Korean ancestry, 66 of African ancestry, and 216 of European ancestry. An ambitious long-term goal for neuroevolution, which studies how artificial evolutionary processes can be driven to produce brain-like structures, is to evolve neurocontrollers with a high density of neurons and connections that can adapt and learn from past experience. Yet while neuroevolution has produced successful results in a variety of domains, the scale of natural brains remains far beyond reach. The following is a compilation of field data collected in 2011 and 2012 in Apalachicola, FL as part of a five year study assessing the ecological effects of sea level rise in the northern Gulf of Mexico. Non-protein-coding RNAs play critical regulatory roles in cellular life. Many ncRNAs fold into specific structures in order to perform their biological functions. Some of the RNAs, such as riboswitches, can even fold into alternative structural conformations in order to participate in different biological processes. In addition, these RNAs can transit dynamically between different functional structures along folding pathways on their energy landscapes. Elevated concentrations of nutrients, particularly nitrate, in groundwater and springs in Florida are a growing resource management concern. Stormwater infiltration basins, which are a common stormwater management practice in the well-drained karst terrain areas of Florida, are a potentially important source of nutrients to the groundwater system because stormwater exits the basin by only evaporation or infiltration. The research presented in this dissertation provides the results of a comprehensive assessment of the water treatment requirements for the City of Sarasota. The City’s drinking water supply originates from two sources: (1) brackish groundwater from the Downtown well field, and (2) Floridan surficial groundwater from the City’s Verna well field. At the time the study was initiated, the City treated the brackish water supply using a reverse osmosis process that relied on sulfuric acid for pH adjustment as a pretreatment method. This thesis focuses on the numerical and theoretical modeling of the interaction of charged droplets from the single and multiplexed electrospray. A modified two-element membrane pressure vessel assembly has been used to monitor process operational changes in a full-scale reverse osmosis (RO) water treatment plant (WTP). This study evaluated the effectiveness of the assembly as an on-line monitoring device intended to detect scale formation conditions when connected to an operating RO process train. This study was implemented to support the requirements of a larger University of Central Florida (UCF) research project ongoing at the city of Sarasota’s Public Works and Utilities (City) water treatment facilities located in Sarasota, Florida. Stormwater runoff from highways is a source of pollution to surface water bodies and groundwater. This project develops a bio-detention treatment and harvesting system that is incorporated into roadside swales. The bio-detention system uses Bold & Gold™, a type of biosorption activated media (BAM), to remove nutrients from simulated highway runoff and then store the water in underground vaults for infiltration, controlled discharge, and/or irrigation and other non-potable applications. In order to design a bio-detention system, media characteristics and media/water quality relationships are required. In 2008, the State of Florida established a recycling goal of 75% to be achieved by 2020. In response to the Florida goal Orange County (OC), Florida has made the development and implementation of an efficient strategy for landfill diversion of its solid waste a top priority. The Florida Department of Environmental Protection (FDEP) estimated that 23 % of municipal solid waste was generated by construction and demolition (C&D) activities in 2009, with only 30 percent of C&D debris being recycled. Stormwater runoff can be a great concern in the State of Florida due to the impact the quality of the runoff water can have on the natural water bodies. Stormwater runoff can carry pollutants and sediments which can cause both physical and biological risks in an aquatic ecosystem such as a lake, river, or pond. Polymers, namely the chitosan-based dual polymer system, can be used remove the sediment from this runoff to ensure the safety of the state’s water bodies.
