The work presented in this dissertation deals with establishing efficient methods for solving some algorithmic problems, which have applications to Bioinformatics. After a short introduction in Chapter 1, an algorithm for genome mappability problem is presented in Chapter 2. Genome mappability is a measure for the approximate repeat structure of the genome with respect to substrings of specific length and a tolerance to define the number of mismatches. The similarity between reads is measured by using the Hamming distance function. Genome mappability is computed for each position in the string and has several applications in designing high-throughput short-read sequencing experiments. …

Still image emotion recognition (ER) has been receiving increasing attention in recent years due to the tremendous amount of social media content on the Web. Many works offer both categorical and dimensional methods to detect image sentiments, while others focus on extracting the true social signals, such as happiness and anger. The use of ultraviolet (UV) light and chlorine dioxide (ClO2) as an advanced oxidation process (AOP) has been investigated at the bench-scale to understand the effects of their use on disinfection by-product (DBP) formation potential (FP) in chlorinated groundwater (GW) and surface water (SW) supplies. Two GWs and two SWs of varying qualities were subject to a series of AOP treatment sequences at the bench scale: sodium hypochlorite, to serve as a baseline; ClO2-Cl2, UV-Cl2, and UV-ClO2-Cl2. Rigid bodies impacting liquid pools incite splashing and air-entraining cavities that depend on impactor shape, entry speed, surface texture, and free surface conditions. In this body of work, we investigate the influence of interfacial properties on water entry dynamics for free-falling spherical projectiles. Our investigations translate to everyday fluid-structure interactions that are complex and generally conclude beyond the temporal acuity of the naked eye. High-speed videography between 1000 -- 3200 fps is used to digitize the water entry process and measure salient splash features under different initial conditions. Humans use gestures to emphasize ideas and disseminate information. Their importance is apparent in how we continuously augment social interactions with motion—gesticulating in harmony with nearly every utterance to ensure observers understand that which we wish to communicate, and their relevance has not escaped the HCI community's attention. For almost as long as computers have been able to sample human motion at the user interface boundary, software systems have been made to understand gestures as command metaphors. Central nervous system disorders produce the undesired, approximately rhythmic movement of body parts known as pathological tremor. This undesired motion inhibits the patient's ability to perform tasks of daily living and participate in society. Typical treatments are medications and deep brain stimulation surgery, both of which include risks, side effects, and varying efficacy. Since the pathophysiology of tremor is not well understood, empirical investigation drives tremor treatment development. This study developed and analyzed the use of a live virtual constructive (LVC) framework capable of simulating ionizing radiation damage to Unmanned Aerial Vehicles (UAV) during a nuclear power plant disaster. UAV response promises greater safety to humans over helicopters as well as provides longer survivability in the presence of irradiated environments. However, electronics in unmanned systems are subject to radiation damage and over time eventual failure. An example of these technologies is Blockchain, which has appeared to overcome security problems potentially. This study examines the "Spanish" influenza pandemic of 1918-1919 in the U.S. South, using case-studies of Jacksonville, Savannah, New Orleans, and Nashville to sculpt a "Southern flu" more identical to the Global South and the developing world than the rest of the U.S. I examine poverty and political and economic paralysis in the years between the end of Reconstruction and 1918, and the poor results of political indifference on public health and disease control. I also analyze the social and institutional racism against persons of color that defined high infectious disease mortality in Southern cities.

In her 2007 essay “Slow Death (Sovereignty, Obesity, Lateral Agency),” Lauren Berlant asserts that “in the scene of slow death, a condition of being worn out by the activity of reproducing life, agency can be an activity of maintenance” (759). Alcohol Use Disorder (AUD) has multiple health and social consequences which negatively affect individuals’ lives. However, the decision to utilize treatment is influenced by a variety of social factors. The stigma associated with AUD may impact individuals’ willingness to seek treatment after drinking is viewed as an issue. A highly stigmatized disorder, perceptions of AUD may be influenced by medical/moral lay theories, responsibility attributions and implicit theories. Combat sports can be defined as sports that involve the physical domination of opponents in order to win. Previous research on combat sports as they relate to psychopathy and anti-social behavior has produced limited information regarding subtypes of trait psychopathy. This research investigates faculty socialization of college physics instructors and its effect on teaching practice. Results of two studies—a statistical analysis about professional characteristics and teaching practice and an ethnography about culture, professional roles, and teaching practice—are integrated to inform our understanding of the impact of socialization on teaching practice. These findings have the potential to improve institutional and organizational faculty development, as well as improve individual teaching practice and, by extension, student persistence and success. The purpose of this qualitative study was to explore the awareness, knowledge, and skills of institutional agents who interact with college students who are undocumented, to inform professional development and practice. Through a survey and interviews, participants revealed familiarity with students who are undocumented and discussed the support provided to them. All interview participants understood the experience of assisting students who are undocumented, as evidenced by six major themes: (a) an understanding of student financial needs, (b) recognizing students' anxiety, (c) providing consistent guidance, (d) establishing supportive collaborations, (e) gaining foundational knowledge, and (f) sharing professional development with others. The objective of this dissertation was to develop novel methods of patterning inorganic and organic materials, develop biocompatibility evaluations, and subsequently apply these methods toward developing biosensors and lab-on-a-chip devices, such as Interdigitated Electrodes (IDEs) and Microelectrode Arrays (MEAs) on non-traditional (such as nanostructured and plasmonic) polymer substrates or deploy these methods to enhance precision cellular placement on traditional (glass) MEA substrates. An investigation of 1,4-dioxane and enantiomeric ibuprofen mass transfer in a nanofiltration (NF) membrane process has been completed. Pilot-scale experiments using a 267 gallon per minute (gpm) split-feed, center-port NF process treating pH 6.5 groundwater revealed a consistent 12 percent removal of 1,4-dioxane despite the variable feed concentration (180 nanograms per liter (ng/L) to 38,400 ng/L) when the water flux and temperature were held constant. Bench-scale, flat-sheet NF membrane experiments treating pH 4.0 synthetic water displayed a 34.5 to 49.5 percent removal of racemic ibuprofen. Removal values were dependent on feedwater concentration (1 to 1,500 microgram per liter (µg/L)), pH, …

Until recently, complete understanding of the endogenous activity of pathologically relevant genes was out of reach and research was confined to in situ work, plasmid-based constructs and artificial model systems. The development and expansion of the CRISPR/Cas9 genome editing technique has enabled us to explore the molecular underpinnings of gene activation using the cell's own endogenous regulatory environment. In this work, we report on the development of a novel tool to monitor the endogenous activity of a causative gene in Parkinson's disease, a-synuclein. Globally, coastal habitats are experiencing degradation due to the increase of human population growth, development along coastlines, and a constantly changing climate. This threatens the future production of critical ecosystem services such as shoreline stabilization, water filtration, nursery grounds for marine fauna, and many more. To combat these losses, resource managers are actively restoring coastal habitat. Past research suggests restoring habitats has mixed results; numerous factors influence restoration success. This thesis is composed to two studies, each of which relies on recent advances in time-dependent density functional theory (TD-DFT) to accurately model fluorescence spectra. The first study focuses on evaluating the fluorescence spectrum of polycyclic aromatic hydrocarbons (PAHs) having a molecular weight of 302. Three isomers are analyzed including dibenzo[a,j]fluoranthene, dibenzo[a,k]fluoranthene and dibenzo[a,l]fluoranthene. These isomers differ in the placement of one of six aromatic rings and in the amount that each molecule deviates from planarity. Receptor-interacting protein 2 (RIP2) is a kinase which mediates signaling downstream of the bacterial peptidoglycan sensors Nucleotide-binding Oligomerization Domain (NOD) 1 and 2. Genetic loss or pharmaceutical inhibition of RIP2 has been shown to be beneficial in multiple inflammatory disease models with the effects largely attributed to reducing pro-inflammatory signaling downstream of peptidoglycan recognition. However, given the widespread expression of this kinase and its reported interactions with numerous other proteins, it is possible that RIP2 has many other unappreciated roles.
