Fluorescence microscopy has emerged as a popular and powerful tool within biology research, owing to its exceptional signal contrast, specificity, and the versatility of the various microscope designs. Fluorescence microscopy has been used to study samples across orders of magnitude in physical scale ranging from tissues to cells, down to single-molecules, and as such has led to breakthroughs and new knowledge in a wide variety of research areas. In particular, single-molecule techniques are somewhat unique in their ability to study biomolecules in their native state, which enables the visualization of short-lived interactions and rare events which can be highly relevant …
Knowledge And Attitudes Of Nurse Practitioner's Assessment Of Adults With Signs And Symptoms Of Posttraumatic Stress Disorder, Kelli Lipscomb
Knowledge And Attitudes Of Nurse Practitioner's Assessment Of Adults With Signs And Symptoms Of Posttraumatic Stress Disorder, Kelli Lipscomb
Electronic Theses and Dissertations, 2020-2023
As the numbers of primary care physicians decline, nurse practitioners (NPs) are filling this gap as primary care providers in many settings. The number of adults presenting to their primary care provider with signs and symptoms of posttraumatic stress disorder (PTSD) each year is unknown. The devastating effects of untreated PTSD include high rates of homelessness, violence, drug abuse, suicidality and a decreased quality of life. Little is known as to the breadth of NPs knowledge of PTSD sequelae, epidemiology and screening protocols. Increased perceived self-efficacy, tied to positive attitudes about PTSD knowledge and screening, result in overall higher rates …
A Mathematical Model For Predicting Animal Population Persistence On Fragmented Landscapes, Allyson Jones
A Mathematical Model For Predicting Animal Population Persistence On Fragmented Landscapes, Allyson Jones
Electronic Theses and Dissertations, 2020-2023
The effects of roads, buildings, and cities on animal populations are widespread and, often times, disastrous. These structures fragment animals' homes, inhibiting their ability to obtain essential resources and to reproduce. The question arises then: Under what circumstances can an animal population persist in a fragmented landscape? To attempt to answer this question, we present a spatially explicit reaction-diffusion model with varying growth and diffusion rates that incorporates animal behavior at points where habitats are fragmented for four different habitats. The outcome of extinction or persistence of the animal population is determined by examining the effects of changing parameters on …

Electronic Theses and Dissertations, 2020-2023
This study examined graduate students' experiences with social support in relation to stress and uncertainty during the COVID-19 global pandemic. Graduate students as a population have significant levels of stress and uncertainty which may have negative effects on their lives and academic experiences. Graduate students often utilize social support as a way to cope with or mitigate the effects of stress and uncertainty. This qualitative study consisted of six focus groups, ranging from three to six participants, total of 22 participants, who were found using a purposeful snowball sampling method. The participants were masters and doctoral level students in communication …
Reassessing The Transcriptional Regulation Of Protective Cd4 T Cell Responses Against Influenza A Virus, Kunal Dhume
Reassessing The Transcriptional Regulation Of Protective Cd4 T Cell Responses Against Influenza A Virus, Kunal Dhume
Electronic Theses and Dissertations, 2020-2023
An attractive strategy to improve efficacy of current Influenza A Virus (IAV) vaccines is to promote protective CD4 T cell responses. Antiviral CD4 T cell responses are predominantly classified as strong IFN-gamma producing T helper type 1 (Th1) cells. Yet, the role of IFN-gamma in protection against IAV is still unclear, suggesting absolute Th1 polarization may be expendable for effective IAV immunity. Here we test this hypothesis using models that restrict the deficiency of known transcriptional regulators of Th1 immunity in only CD4 T cells, avoiding indirect impact on other immune cell responses. We find the 'master regulator' of Th1 …
Loss Of Control Eating Predicted By The Interaction Between Emotion Regulation Difficulties, Distress Tolerance, And The Expectancy That Eating Reduces Negative Affect, Emily Burr
Electronic Theses and Dissertations, 2020-2023
A prominent theory of binge eating is the affect regulation theory, which posits that individuals binge eat to alleviate negative affect, and subsequently reduced negative affect reinforces the behavior. Although it is well-supported that individuals experience elevated negative affect pre-binge, findings do not consistently evince reduced negative affect after binge eating. Therefore, the affect regulation theory does not fully account for binge eating. However, habitual binge eating without reliable improvement in affect may be accounted for by expectancy theory. Expectancies may be predictive of behavior whether the outcomes of a behavior are inconsistent. Additionally, there is an increasing scientific awareness …
Actin Cytoskeleton Dynamics And Organization Modulated By Macromolecular Crowding, Cation Interaction, And Nanomaterials, Jinho Park
Electronic Theses and Dissertations, 2020-2023
The assembly of actin, the essential cytoskeleton protein, into filaments and bundles/networks is important in various cellular processes including cell movement and morphogenesis. Actin bundle formation occurs in crowded intracellular environments with the aid of actin-crosslinking proteins. The role of actin-crosslinking proteins such as fascin and a-actinin in bundle formation has been investigated, however, how intracellular environments affect actin crosslinker-induced bundle formation is unknown. In the first two parts of this dissertation, we explore the effects of macromolecular crowding and cation interactions on the organization and mechanics of actin crosslinker-induced bundles. To determine how changing environmental conditions modulate actin bundling, …
Modulation Of Actin Filament Severing And Mechanics By Gelsolin In Varying Ph Conditions, Claire Toland
Modulation Of Actin Filament Severing And Mechanics By Gelsolin In Varying Ph Conditions, Claire Toland
Electronic Theses and Dissertations, 2020-2023
Actin is an essential cytoskeletal protein that plays a critical role in cell mechanics, structure and organization with the help of actin binding proteins (ABPs). Gelsolin is a calcium-dependent ABP that severs actin filaments and caps them at their barbed end, regulating cell motility and signaling through dynamic actin cytoskeleton remodeling. A recent study has indicated that low pH stabilizes the active conformations of gelsolin. Additionally, the binding of gelsolin to the barbed end of an actin filament induces a conformational change that propagates along the actin filament. However, it has not been well understood how the complex intracellular environments …
Impact Of Crowder Size On Actin Filament Assembly Kinetics, Bryan Demosthene
Impact Of Crowder Size On Actin Filament Assembly Kinetics, Bryan Demosthene
Electronic Theses and Dissertations, 2020-2023
Actin is an abundant and essential cytoskeletal protein that plays a central role in eukaryotic cell structure and motility. The intracellular environment where actin assembly occurs is crowded with various organelles, proteins, and macromolecules that limit the accessible volume for biomolecular reactions. Macromolecular crowding induces excluded volume effects influencing the activity of biological molecules as well as the shape and conformation of proteins. Crowding agents, such as polysaccharides or inert polymeric molecules, are used to mimic the conditions present in intracellular spaces and provide a better understanding of interactions inside the cell. Macromolecular crowding has been shown to affect actin …
Correlations In Microplastic Abundance Between Water, The Eastern Oyster, Crassostrea Virginica, And Their Biodeposits In A Dynamic Florida Estuary, Casey Craig
Electronic Theses and Dissertations, 2020-2023
Estuaries have been identified as hotspots of microplastic pollution because they are transitional zones where coastal freshwater and oceans converge. Microplastics (MP) are transported through estuaries by a dynamic series of forces such as surface flow and tides, which influence MP abundances and trends. The eastern oyster, Crassostrea virginica, is an estuarine bivalve known to ingest MP, resulting in negative impacts on organism physiology. I investigated MP pollution as a threat to C. virginica in a dynamic Florida estuary, the Indian River Lagoon (IRL), and determined there are both regional and small-scale spatial and temporal fluctuations in MP abundance. Tributaries …
Evaluating Carbon Uptake And Storage Potential Of Three Pine Species Across Environmental Gradients In Florida, Alicia Formanack
Evaluating Carbon Uptake And Storage Potential Of Three Pine Species Across Environmental Gradients In Florida, Alicia Formanack
Electronic Theses and Dissertations, 2020-2023
Natural climate solutions, such as reforestation, are increasingly called for to lower atmospheric CO2 concentrations and prevent further warming of the climate. Predictive modeling of forest stand dynamics provides a quantitative framework that can be used to select the tree species with the highest carbon (C) uptake potential for restoration efforts based upon site-specific and species-specific data. To simulate stand dynamics and compare C uptake and storage potential of three pine species across Florida, I developed individual-based models that combined environmentally-sensitive tree growth models with background mortality taken from the literature. Growth model parameters were estimated using space for time …
Dual Beam Frequency Comb Ftir Spectroscopy, Connor Kelly
Dual Beam Frequency Comb Ftir Spectroscopy, Connor Kelly
Electronic Theses and Dissertations, 2020-2023
A visible and Infrared (IR) range dual beam frequency comb Fourier transform spectrometer was developed. Using dual mode-locked Ti:Sapphire lasers a comb-interferogram was generated in the visible range. This spectrum was calculated and used to measure the transmittance of a Nd-doped crystal. The system was further developed to generate an IR interferogram by Difference Frequency Generation (DFG) using a Gallium Selenide (GaSe) crystal placed in the mode-locked pump beam. Numerical work was done to calculate the expected DFG spectrum confirming the necessary IR range can be reached. This has been conducted in support of an IR holographic spectroscopic microscopy spatial …
Understanding The Dietary Behavior Of A 7th - 8th Century Avar Community From Jagodnjak, Croatia Using Stable Isotope Analysis Of Bone Collagen, Marlon Koci
Electronic Theses and Dissertations, 2020-2023
The dietary behavior of past communities is influenced by several factors such as environmental sources, social and economic organization, individual preferences, and physiological needs. Every society has its characteristics and understanding the dietary choices of its individuals expands the knowledge about its lifestyle. Applying biochemical tracers, we can explore the food choices of an Early Medieval Avar community. The Jagodnjak cemetery (n=35) in Croatia is dated to the 7–8th century. Historical and archaeological records are currently limited for Avar populations. Jagodnjak is a transitory community from a period where different historical events shaped the community's way of life. The research …
The Intersection Between Multi-Voiced Narratives And First-Person Narratives Of War Fiction: Epistemology And The Meaning Of War, Prashant Patel
The Intersection Between Multi-Voiced Narratives And First-Person Narratives Of War Fiction: Epistemology And The Meaning Of War, Prashant Patel
Electronic Theses and Dissertations, 2020-2023
The topic for this project concerns a critical rhetorical analysis of the correlation between texts of first-person narratives and multi-voiced narratives of the Vietnam War (1965-1975) and the American invasion of Iraq (2003-2011). Moreover, this project establishes a correlation between the textual design in these novels and concepts of epistemology that reside in the Western cannon of rhetorical theory. The theoretical framework that is incorporated within this study extracts concepts of (t)ruth from philosophical works such as Michel Foucault's Birth of the Clinic and Friedrich Nietzsche's On Truth and In a Nonmoral Sense. In comparison, I examine the concepts and …

Long-Term Changes In Juvenile Green Turtle Abundance And Foraging Ecology In The Indian River Lagoon, Florida, Christopher Long
Electronic Theses and Dissertations, 2020-2023
Marine turtles are distributed in temperate, sub-tropical and tropical waters and beaches worldwide, often in areas heavily impacted by humans. Although there are many threats to marine turtle populations, the growing threats of nutrient pollution and harmful algal blooms are relatively understudied despite their widespread impacts on coastal marine ecosystems that marine turtles depend on. By studying juvenile green turtles (Chelonia mydas) in the Indian River Lagoon, Florida, where nutrient pollution and HABs are a widespread and longstanding issue, I aimed to conduct a case study of how these threats may affect this federally Threatened species. In Chapter 2, I …
Enhancement And Evaluation Of Proton Pencil Beam Spot Placement Algorithms, Mahboob Ur Rehman
Enhancement And Evaluation Of Proton Pencil Beam Spot Placement Algorithms, Mahboob Ur Rehman
Electronic Theses and Dissertations, 2020-2023
Intensity modulated proton therapy (IMPT) in the form of pencil beam scanning (PBS) has shown improvement in treatment plan quality as compared to conventional proton and photon-based radiotherapy techniques. However, in IMPT maintaining a sharp lateral dose falloff is crucial for sparing organs at risk (OARs), especially when they are in close proximity to the target volume. The most common approach to improve lateral dose falloff is through the use of physical beam shaping devices, such as brass apertures or collimator-based systems. This work has shown that IMPT can be further improved by implementation of advanced spot placement techniques by …

Electronic Theses and Dissertations, 2020-2023
Birds are often used as indicators for biodiversity and ecosystem health. While birds have been monitored in other parts of the Indian River Lagoon (IRL), there has been little research on birds in Mosquito Lagoon (ML, the northernmost portion of the IRL). This thesis first examines the abundance and diversity of birds in ML by using two years of photographic observations to assess seasonal variations and the use of various habitat features by specific bird taxa. Abundance and species richness were highest in winter, while evenness and Simpson's diversity were highest in summer. Moreover, natural and artificial habitat features were …
Forensic Implications For The Diagenetic Alteration Of The Stable Isotopic Composition Of Pig Ribs Following Exposure To Aqueous Conditions., Erin Martin
Electronic Theses and Dissertations, 2020-2023
This research involves the examination of the contribution of stable isotope analysis to the forensic anthropological objective of human identification. Oxygen isotopes, which vary in water in a predictable manner according to climatological and geographical factors, can be used to determine the region of origin of an unidentified individual following isotopic analysis of human skeletal remains. However, stable oxygen isotopes from human remains recovered from various aqueous environments may be altered by diagenetic processes that occur following deposition. The focus of this research is to determine potential diagenetic changes in the oxygen isotopic value (d18O) of Sus scrofa (domestic pig) …
Extending Open Mobility To Mobility Data Justice: A Localized Case Study Of Gtfs-Oriented (Infra)Activism In Defense Of The Mobile-Digital Commons, David Moran
Electronic Theses and Dissertations, 2020-2023
Through a humanist lens, this research aims to conceptualize a Mobility Data Justice (MDJ) framework that re-orients both GTFS open transit data (General Transit Feed Specification) and transit rider activists towards collaborative Mobility Data Justice tactics. Transit riders and activists, in the age of Uberfication, have to account for and navigate the dynamics of how Big Data and platform economies are not only augmenting but co-opting political decision-making processes about public transit policy, planning and investment. Building on mobility justice and data justice theorists, I examine how the GTFS open data ecosystem can be operationalized via justice ontologies to support …
Biophysical Analysis Of The Structure And Aggregation Of Amyloid Beta Peptide, Faisal Abedin
Biophysical Analysis Of The Structure And Aggregation Of Amyloid Beta Peptide, Faisal Abedin
Electronic Theses and Dissertations, 2020-2023
Alzheimer's disease (AD) is the major cause of dementia and is characterized by neuronal death and brain atrophy. The amyloid ß (Aß) peptide is tightly associated with neuronal dysfunction during AD, but the molecular mechanism underlying the neurotoxic effect of Aß is poorly understood. Extracellular fibrillar deposits (plaques) of Aß were initially believed to be the cause of AD, but currently there is overwhelming evidence that the prefibrillar Aß oligomers are the major toxic entities. Structural characterization of Aß oligomers and fibrils is important for understanding the structural features determining the toxic potency of the peptide. This project has studied …
Identifying Biomarkers Of Alzheimer's Disease From The Dakhleh Oasis Kellis 2 Cemetery Using Icp-Ms, Emily Barron
Identifying Biomarkers Of Alzheimer's Disease From The Dakhleh Oasis Kellis 2 Cemetery Using Icp-Ms, Emily Barron
Electronic Theses and Dissertations, 2020-2023
Alzheimer's disease is a prevalent disease that affects roughly 5.4 million individuals each year (Fisher et al., 2018). Understanding the prevalence of Alzheimer's disease within the bioarchaeological record can help anthropologists gain an understanding of a population's health while also understanding the potential social ramifications. Trace element analysis is a valuable tool that allows bioarchaeologists to gain insight into individuals' health and development within the context of the past. In bioarchaeological studies, hair analysis has been used to map the nutritional status and migration patterns of individuals of past peoples, with recent studies focusing on disease prevalence. Analysis of hair …
Non-Lethal Predictors Of Organ Level Metal Contaminants In Central Florida Red-Shouldered Hawks, Jennifer Bouchenot
Non-Lethal Predictors Of Organ Level Metal Contaminants In Central Florida Red-Shouldered Hawks, Jennifer Bouchenot
Electronic Theses and Dissertations, 2020-2023
Elemental contaminants aggregate in the environment due to human activity and can induce detrimental physiological effects within organisms. Birds exhibit exceptional susceptibility to many pollutants and are popular biomonitoring agents throughout the world. Birds of prey are of special interest due to their substantial biomagnification potential. Lethal sampling of internal organs yields accurate contaminant measurements, but recent sampling trends are endeavoring towards non-destructive sampling such as feathers. This methodological shift stems from researchers' preference for less-regulated samples, enhanced animal welfare, and broadening the possible collaborative audience with simpler protocols. Unfortunately, investigations of feathers have shown that their accuracy is capricious …
Modulation Of Fat Taste By Diet And Hormones, Ashley Calder
Modulation Of Fat Taste By Diet And Hormones, Ashley Calder
Electronic Theses and Dissertations, 2020-2023
The prevalence of obesity worldwide continues to rise despite efforts to reverse the trend. While many factors contribute to the onset and maintenance of obesity, caloric intake and dietary composition have been shown be primary contributors. The oral cavity is one of the first systems to encounter food and determine its hedonic value. As the gateway to ingestion, the taste system plays a unique role in the initial decisions surrounding the control of food intake. Nutrients like carbohydrates, protein, minerals, and fat all have dedicated systems to allow their recognition at this outermost site of the enteric nervous system. Recent …
Transcriptomic Analyses Reveal Distinct Responses Of Differentiated And Poorly-Differentiated Intestinal Epithelial Cells To Lipid Peroxides, Nisreen Faizo
Electronic Theses and Dissertations, 2020-2023
Lipid peroxides (LOOHs) abound in processed food and have been implicated in the pathology of diverse diseases including gut, cardiovascular, and cancer diseases. However, the molecular mechanisms by which LOOHs contribute to disease have not been fully characterized. Caco-2 cells have been widely used to model human intestinal epithelium in metabolic studies. As differentiated (Diff) and poorly-differentiated (PDiff) Caco-2 cells represent good models of human enterocytes and intestinal tumor cells, respectively, we investigated the cellular response of Diff and PDiff Caco-2 cells to the most common dietary LOOH, 13-hydroperoxyoctadecadienoic acid (13-HPODE), in terms of differential gene expression, gene ontology and …

Bioarchaeological And Mortuary Patterns At Holtun, Guatemala: Integrating A Comparative Osteobiographic Approach, Horvey Palacios
Electronic Theses and Dissertations, 2020-2023
Bioarchaeological analysis can help identify patterns of mortuary ritual and social experience of ancient Maya peoples. However, there is limited bioarchaeological and mortuary evidence for the relationship between the development of social complexity and social experience. Particularly, how is social organization reflected in patterns of burial practice and skeletal markers of stress. This thesis uses osteobiographies to contextualize the mortuary and biological profiles of 20 individuals interred at the Maya site of Holtun, Guatemala to examine bioarchaeological variation during the Preclassic (800 B.C. – 250 A.D.) and Late Classic (550 A.D. – 900 A.D.) periods. This work highlights the integration …

Electronic Theses and Dissertations, 2020-2023
Introduction: The development and regulation of depressive symptoms and the ability to regulate their development is a complex process. Both working memory and sleep disturbance relates to depressive symptom endorsement, though the mechanisms relating these variables have not been examined longitudinally. The current manuscript contains a series of three interrelated studies that aim to elucidate the relationship between potential emotion regulation resources longitudinally within the context of the Selection, Optimization, and Compensation of Emotion Regulation (SOC-ER) model. Study one examined the temporal relationship between working memory and depression, study two examined working memory and depression following loss of spouse, and …
Analysis Of Sexual Lubricants As A Form Of Trace Evidence For Sexual Assault Cases, Brooke Baumgarten
Analysis Of Sexual Lubricants As A Form Of Trace Evidence For Sexual Assault Cases, Brooke Baumgarten
Electronic Theses and Dissertations, 2020-2023
A major gap in sexual assault casework is demonstrated when DNA is not recovered. Oftentimes, if DNA evidence is not present on the collection swabs, the sexual assault kit (SAK) is not further analyzed. Due to the "CSI effect," DNA is commonly understood as highly identifiable evidence, potentially leading to increased condom usage to eliminate or reduce DNA transfer during a sexual assault. Therefore, the analysis of condoms and sexual lubricants is pertinent. The purpose of this research is to develop analytical protocols to potentially connect unknown substances recovered in a SAK to known lubricant reference samples. Sexual lubricants were …
