They are sensitive to changes in abiotic conditions, and shifts in community composition can provide insight into estuarine ecosystem functioning in the face of various physiochemical and anthropogenic stressors. Therefore, monitoring these communities is essential for understanding and maintaining estuarine health. Automated infrared (IR) imagery analysis is essential for persistent surveillance, defense, and security, yet it remains difficult when sensors move and when unknown objects appear. This dissertation addresses two fundamental problems: (1) detecting small moving targets amid platform-motion induced parallax and (2) distinguishing between known stationary targets and out-of-distribution (OOD) objects.   For detecting moving targets while the platform itself is in motion, auxiliary Global Positioning System and Inertial Navigation System data are combined with image analysis. Direction Cosine Matrices from calibrated inertial measurements enable sub-pixel frame alignment unattainable with purely image-based registration. Dextran sodium sulphate (DSS) is widely used in vivo to induce epithelial injury; however, the molecular effects of DSS on intestinal epithelial cells, that investigate inflammatory and metabolic repair mechanisms remain unclear. Here, we establish an in vitro DSS-induced epithelial damage model using differentiated Caco-2 cell monolayer. Exposure to 2.5%(w/v) DSS for 24 hours resulted in significant upregulation of pore forming protein Claudin-2(2.677-fold) and oxidative marker, NOX-1(1.866-fold), accompanied by increased ROS production confirmed via DHE staining. Increased epithelial permeability was observed by TEER analysis, without significant cytotoxicity. Surprisingly, ELISA analysis revealed significant downregulation of IL-6 (p-value …

Careless responding (CR) has been identified as a threat to the psychometric integrity of cognitive and non-cognitive tests, with much of the current research focusing on the identification and removal of carelessness from dataset. While this research has proved fruitful in improving data quality, there has been a recent push to move towards preventing carelessness as opposed to post-hoc removal, which harms statistical power. However, the most common prevention strategy to prevent carelessness (pre-survey warnings) has shown equivocal effects. Gastrointestinal (GI) cancers remain among the leading causes of cancer-related mortality worldwide, with therapeutic resistance continuing to limit the effectiveness of current treatments. Three complementary studies were conducted to identify therapeutic vulnerabilities and evaluate novel combination strategies in pancreatic ductal adenocarcinoma (PDAC) and colorectal cancer (CRC).  First, dual inhibition of fibroblast growth factor receptor 4 (FGFR4) and phosphoinositide 3-kinase (PI3K)/mammalian target of rapamycin (mTOR) signaling was investigated in PDAC models. A combination of targeted drug treatments reduced viability, clonogenic survival, migration, and suppressed signaling pathways involved in translational control. The introduction of invasive species can disrupt ecosystems not only through competition and predation but also by introducing novel pathogens to naïve host populations. In the southeastern United States, the invasive Burmese python (Python bivittatus) has been implicated in the introduction of the pentastome parasite Raillietiella orientalis, which has been associated with increased parasite burdens and mortality in native snakes. We developed a behaviorally explicit framework to separate true mortality from changes in availability caused by movement and redistribution. We evaluated this problem for juvenile green turtles (Chelonia mydas) and loggerhead turtles (Caretta caretta) captured at the St. Lucie Nuclear Power Plant intake canal on Florida’s Atlantic coast using a multi-decade capture–mark–recapture record spanning 1977 - 2023. Hidden Markov Models classified individuals into latent habitat-use strategies (Nomadic, Resident, and Transient) based on capture timing and body size. Despite clear behavioral and physiological evidence that fat can evoke distinct taste signals, the cellular and molecular mechanisms underlying its transduction remain poorly understood. A critical gap exists in our understanding of the cell-type-specific molecular pathways and how these signals are modulated by hormonal status. This research examines the regulatory role of HES4 and its isoforms in NK cell function during retroviral infection, with particular focus on how Notch signaling–dependent regulation of HES4 shapes NK effector responses. These forest islands have been shown to have been integral aspects of the pre-Contact indigenous cultures which existed there for thousands of years. The purpose of this thesis is to examine how structured narrative representation and reasoning strategies jointly affect the performance of Video Question Answering (VideoQA). Although recent advances in large multimodal and language models have improved video description and retrieval, causal and counterfactual reasoning remain challenging. This thesis studies an inference-only pipeline in which videos are converted into natural-language narratives, and those narratives may optionally be transformed into a structured NarrativeML format before multiple-choice questions are answered without task-specific fine-tuning. These garments are typically passive storytellers in theme parks, even if the performers interact directly with guests. The fibroblast growth factor 19 (FGF19)-fibroblast growth factor receptor 4 (FGFR4) signaling pathway regulates hepatic metabolism, bile acid (BA) synthesis, and hepatocyte proliferation. This signaling axis also is implicated in promoting tumor progression, therapeutic resistance, and metastasis in certain cancers.

In colorectal cancer (CRC) there is a critical need for non-invasive biomarkers for disease detection and monitoring. We identified FGF19 as a candidate CRC serum marker through meta-transcriptomics and found FGF19 to be expressed and secreted by CRC cell lines. Crohn’s disease (CD) is a chronic inflammatory bowel disease characterized by intestinal inflammation, epithelial barrier dysfunction, and abnormal immune signaling. CD pathophysiology is increasingly linked to Mycobacterium avium subspecies paratuberculosis (MAP) infection, which increases chronic macrophage activation and the production of inflammatory mediators that lead to epithelial damage. Among these mediators, CXCL10, TNF-α, and IL-6 contribute in inflammation, while NOX-1 and SERPINE-1 indicate epithelial oxidative stress and damage. This dissertation examines how modality-specific scaffolding shapes performance, presence, and transfer in immersive virtual reality (I-VR). Framed through theories of extended cognition, real-time guidance is conceptualized as cognitive scaffolding that structures perception and action by embedding task-relevant information within the environment. In this dissertation, I examine how Latinx and Latina audiences engaged with and interpreted Disney’s 2021 Encanto, an animated film celebrated for its representation of Colombian and broader Latinx culture. While scholarship on Latinx representation in media has expanded, less attention has been given to how Latinx audiences themselves respond to narratives that are meant to represent them, particularly within fan studies.
