Provided is a separatome-based recombinant peptide, polypeptide, and protein expression and purification platform based on the juxtaposition of the binding properties of host cell genomic peptides, polypeptides, and proteins with the characteristics and location of the corresponding genes on the host cell chromosome, such as that of E. coli, yeast, Bacillus subtilis or other prokaryotes, insect cells, mammalian cells, etc. This platform quantitatively describes and identifies priority deletions, modifications, or inhibitions of certain gene products to increase chromatographic separation efficiency, defined as an increase in column capacity, column selectivity, or both, with emphasis on the former. Inquiry: The University of Arkansas Undergraduate Research Journal

This study investigated the behavior of vanadium redox flow batteries (VRFBs), which are batteries capable of easily switching between charging and discharging modes, making them a suitable option for storing intermittent sources of alternative energies (solar, wind, etc). Since different sizes of the battery provide varying voltages, optimal parameters for a particular home are key for implementation. These parameters, specifically the cell and tank volumes of the battery that are capable of providing consistent on-load voltage, were determined using data from a 13 kW solar array and a medium-sized house. Terahertz Imaging Platform To Characterize The Growth Of In-Vitro Breast Tumors, Scarlett-Marie Acklin

Inquiry: The University of Arkansas Undergraduate Research Journal

This study aimed at evaluating the ideal plating method and density for imaging with the terahertz (THz) spectrometer. In this study, different methods were used to grow in-vitro tumors using the 4T1 cell line. Here, attempts to grow breast tumors in-vitro were conducted. Results were produced in two environments, flat-bottomed plates and round-bottomed multiwell plates. The second method allowed for faster clumping and increased cell aggregation, producing tumors up to 7mm. Terahertz spectroscopy produced images that correlated well to photomicrographs taken of the in-vitro tumors. This methodology shows great promise for providing a reliable, parameter-controlled source of in-vitro breast tumors …

Interface Property Of Collagen And Hydroxyapatite In Bone And Developing Bioinspired Materials, Clint Paul

Inquiry: The University of Arkansas Undergraduate Research Journal

Bone at the nanoscale consists of type I collagen and hydroxyapatite (HAP). Type I collagen and HAP [Ca10(PO4)6(OH)2] are responsible for most of the structural integrity of bone. Collagen fibrils contain HAP platelets of varying size dispersed between the collagen. We investigate heterotrimeric collagen interaction with HAP using Steering Molecular Dynamics to obtain the force-displacement relation as the collagen is undergoing shearing and peeling on the surface of HAP. Results indicate that the collagen requires 40% less force to separate form the HAP surface under peeling, when compared to shear loading conditions. In both shearing and peeling, the number of …

Inquiry: The University of Arkansas Undergraduate Research Journal

Sepsis, or dysregulated inflammation caused by bacterial infection, places a disproportionately high burden on newborns in developing countries. This is due in part to a lack of diagnostic tools suitable for sustainable use in resource-limited nurseries. One potential vehicle for a new diagnostic assay is loop-mediated isothermal amplification (LAMP), a high-yield DNA amplification method. LAMP has previously been used to detect genes from single species of bacteria in blood serum samples to aid in sepsis diagnosis. LAMP could be adapted to detect a broad set of bacteria, while retaining a degree of specificity that allows clinicians to begin directed antimicrobial …

Investigating The Effect Of Stratospheric Radiation On Seed Germination And Growth, B. N. Fong, K. V. Newhouse, M. J. Huss, E. Roberts, J. T. Kennon, H. Ali

Three seed types: bean (Phaseolus vulgaris), corn (Zea mays) and radish (Raphanus sativus) were flown in a high altitude weather balloon into the mid-stratosphere to investigate the effects of high altitude radiation on germination success and seedling growth. After recovering and planting the seeds, the bean seeds showed lower germination success with exposure to high altitude radiation, and consequently stunted seedling growth. Cord and radish seeds experienced a statistically significant positive effect on germination success form radiation exposure compared to control seeds, but negative effect on seedling growth. Overall, the field experiments presented here support laboratory studies that show radiation …

Structure loading by vortices is a relevant phenomenon in numerous fields of engineering significance. Computer modelling is a powerful tool that can be used to study the loading produced on structures by impacting vortices. Realistic simulation of vortex-loading of structures necessitates the use of a realistic vortex tangential velocity profile (TVP). The present study compiles measured TVPs from various types of experimentally-produced vortices as well as real-world tornado and hurricane vortices. The measured TVPs are compared with commonly-used, analytical TVPs. Case Of Frugivory In A Green Treefrog (Hyla Cinerea) From Northeastern Arkansas, C. S. Thigpen, S. E. Trauth, T. D. Marsico

An adult green treefrog (Hyla cinerea) was collected in mid-September 2014 from Jonesboro, Craighead County, Arkansas. Contents included the remains of a beetle and two fruits. The contents were photographed, measured, and identified to the lowest achievable taxonomic level. The beetle was identified as a spotted cucumber beetle (Diabrotica undecimpuncta howardi Barber [Coleoptera: Chrysomelidae]). The fruits were identified as Persicaria sp. (likely P.punctata (Elliot) Small; Polygonaceae) and Oryza sativa L. (Poaceae). Fruits in the stomachs of frogs are rarely reported in the literature, but could represent possible mechanisms for seed dispersal in plants. It is unknown if frogs select to …

Low-Level Mercury Causes Inappropriate Activation In T And B Lymphocytes In The Absence Of Antigen Stimulation, K. L. Weigand, J. L. Reno, B. M. Rowley

The immune system primarily utilizes two cell types for adaptive immunity: T lymphocytes and B lymphocytes. T lymphocytes are activated when antigen presenting cells (APCs) present antigen to membrane-bound T cell receptors. B lymphocytes are activated when an antigen binds to receptors embedded in the plasma membrane. In both T and B cells this antigen binding crosslinks the receptor complexes and initiates the signal transduction cascade. These cascades frequently consist of a series of intracellular molecules becoming phosphorylated in a step-wise fashion. Foraging Behavior Of Swainson's Thrushes (Catharus Ustulatus) During Spring Migration Through Arkansas, S. Wiley, R. Kannan, D. A. James, A. Deshwal

Foraging behavior of Swainson’s Thrushes on spring migration was studied in western Arkansas in the spring of 2013 and 2014. Observations were made in two forested field sites, one of them urban and the other suburban. The former had a significantly higher woody stem area (cm2) than the latter. For each foraging observation, the following three parameters were noted: Foraging Stratum (Ground, Shrub, Sapling, Sub canopy, and Canopy); Foraging Substrate (Ground/Litter, Herb, Foliage, Bark, and Air); and Foraging Maneuver (Glean, Probe, Dive/Glean, Hover, Jump Hover, and Hawking).
