Assesing The Degree Of Intereron Gamma Induced Stat1 Activation On Peripheral Lymphocytes In Patietns With Rheumatoid Arthritis, Vishal Sharma

All ETDs from UAB

Rheumatoid Arthritis (RA) is a systemic autoimmune disease with debilitating side effects, from joint swelling to bone erosion and bone deformation. Geographically, RA affect 0.5 – 1% of the worldwide populations, affecting women more than men. Studies have shown that CD4 T cells infiltrate RA synovium and are key to the pathogenesis of RA. Furthermore, drugs targeting co-stimulatory T cell molecules such as abatacept can be beneficial in controlling RA disease activity.Interferon gamma (IFN) is a pleotropic cytokine that contributes to inflammation by aiding in CD4 T cell differentiation into Th1 subtype, enhancing MHC class II presentation, supplementing lymphocyte trafficking, …

Mechanism Of Stat3 Mediated Drug Persistence In Chronic Myeloid Leukemia Stem Cells, Sweta B. Patel Jan 2021

Mechanism Of Stat3 Mediated Drug Persistence In Chronic Myeloid Leukemia Stem Cells, Sweta B. Patel

All ETDs from UAB

Drug persistence is one of the major impediments for cancer therapeutics. There are two known evolutionary models for tumor drug persistence. First, a pre-existing sub-population is inherently resistant to treatment; and second, a sub-population acquires resistance by drug-induced molecular reprograming. One ideal model to study the mechanism of drug persistence is chronic myeloid leukemia (CML), a hematopoietic stem cell (HSC) disorder. HSCs acquire a single translocation between chromosome 9 and 22, forming BCR-ABL, a constitutively active tyrosine kinase. Tyrosine kinase inhibitors (TKI), the standard regimen, have been able to efficaciously control CML and yet many patients relapse upon treatment discontinuation …

Examining Approaches For Peacebuilding In Post-Conflict, Post-Good Northern Ireland, Evan Wiley Smith Jan 2021

Examining Approaches For Peacebuilding In Post-Conflict, Post-Good Northern Ireland, Evan Wiley Smith

All ETDs from UAB

In this paper, I broadly describe the timeline of the conflict in Northern Ireland, examine negative and positive peace approaches to peacebuilding including political (or consociationalism-based), human rights-based, and reconciliation-based methods, and present my own conclusions based on the literature review I conducted as to the most efficacious strategy for ensuring long-lasting peace in Northern Ireland. To do this, I reviewed numerous articles, papers, and chapters on the conflict in Northern Ireland, approaches in post-conflict reconciliation, and conflict resolution theory. Because the conflict in Northern Ireland ended approximately twenty years ago, much of the research referenced in this paper is …

The Kinetic Effects Of Nanoconfinement, Aggregate Phase Type, And Pressure On Some Thermally Stimulated Processes, Victoria Lynn Stanford Jan 2021

The Kinetic Effects Of Nanoconfinement, Aggregate Phase Type, And Pressure On Some Thermally Stimulated Processes, Victoria Lynn Stanford

All ETDs from UAB

This research explores the effects of nanoconfinement, aggregate state type, and an inert gas pressure on the kinetics of thermally stimulated processes. The processes considered are solution crystallization and thermal decomposition. The kinetic effects are measured via differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and high pressure differential scanning calorimetry (HPDSC), combined with isoconversional kinetic analysis. The first chapter demonstrates that DSC can be applied to measure the kinetics of nonisothermal crystallization. The crystallization of potassium nitrate and ammonium perchlorate are from unsaturated and saturated aqueous solutions. An advanced isoconversional method demonstrates that the process is represented by negative values …

Waffle is a novel about a high school senior realizing she has an anxiety disorder and is pansexual. Aside from wanting to tell a version of my own story, I want this to be a young adult novel that has appropriate, diverse representation. Given that several characters have identities different than mine (Jay having a single parent, Ginger being Black and having Type One Diabetes, Ella being transgender with divorced parents, and Birdie being Taiwanese-Korean American), I will need a lot of research and help to write these characters well. I want the information and experiences regarding race, gender, sexuality, …

An Investigation Into The Effects Of Diet On Pain And Pain Disparities, Larissa Jane Strath Jan 2021

An Investigation Into The Effects Of Diet On Pain And Pain Disparities, Larissa Jane Strath

All ETDs from UAB

Chronic pain is one of the most common and disabling disorders in Westernsociety. Recently, it has been suggested that dietary quality may play a key role in the development and maintenance of pain. Additionally, diet differs between demographic groups, as does physiological differences in response to metabolites. We sought to explore 1) the central way in which diet influences pain sensitivity and 2) if dietary patterns affected various demographic groups differently. We first employed a rat model to observe diet-related changes in the dorsal horn of the spinal cord. In the absence and presence of an injury, rats fed a …

Characterization Of Color Centers In Diamond For Laser Applications, Shova Devi Subedi Jan 2021

Characterization Of Color Centers In Diamond For Laser Applications, Shova Devi Subedi

All ETDs from UAB

Diamond has been gaining a lot of interest in the field of photonics such as quantum optics, nano-photonics, quantum computation, magnetometry, sensing, laser development, and color center physics as it provides a remarkable platform to combine its non-linear optical properties in conjunction with the unique optical and spin properties of its color centers. Besides that, energetic pulsed lasers operating in the visible to near IR spectral range are mostly obtained from non-linear frequency conversion which is less efficient, expensive, and requires nonlinear crystal with excellent optical quality, a special cut to meet the perfect phase-matching condition. Another alternative is dye …

From Deviance To Disease: How Congress Frames Opioid Use, 1994-2019, Stephanie Kirkland

All ETDs from UAB

Over the past thirty years, the use of opioids like heroin, fentanyl, oxycodone, and hydrocodone led to tens of thousands of deaths from overdoses and caused a public health crisis. Researchers have noted the roles of various social organizations and groups in creating a public discourse around this topic and have studied the changing public views of opioid use in light of new scientific research. And yet, there remains a gap in research on the role of the members of Congress in this public discourse. These legislators pass policies that directly affect people who live with or who know someone …

Embodied Fat Stigma And Health Beliefs About Exercise And Diet, Brie Scrivner Jan 2021

Embodied Fat Stigma And Health Beliefs About Exercise And Diet, Brie Scrivner

All ETDs from UAB

The purpose of this dissertation is to examine how individuals navigate the cultural mandate for thinness, which pressured individuals to pursue weight loss through exercise and restrictive diets. While sustainable weight loss is not probable for the majority of people, there are still significant physical and mental health benefits to be had from engaging in regular, enjoyable movement. For many, physical activity is an important component of a healthy lifestyle. It can improve quality of life for individuals regardless of weight, body size, chronic illness, or disability. However, many fitness facilities focus on weight loss as the most essential goal …

Characterizing The Role Of Necroptosis Of Airway Epithelial Cells In The Immune Response To Respiratory Pathogens, Ashleigh Nichole Riegler Jan 2021

Characterizing The Role Of Necroptosis Of Airway Epithelial Cells In The Immune Response To Respiratory Pathogens, Ashleigh Nichole Riegler

All ETDs from UAB

Necroptosis, a programmed form of lytic cell death, is initiated by various viral and bacterial pathogens through irreparable ion dysregulation and energy depletion. This cellular damage results in the activation of the cellular kinases RIPK1 and RIPK3, consecutively, and the activation and membrane targeting of MLKL, the latter responsible for lysis. Here we have demonstrated that necroptosis of airway epithelial cells is key in the development of the adaptive immune response to asymptomatic colonization by Streptococcus pneumoniae (Spn). Briefly, necroptotic deficient animals or wildtype animals colonized with Spn lacking the necroptosis-triggering pneumolysin toxin, failed to recruit CD11c+ leukocytes to the …

Cns Antigen Presentation And Immune Cell Infiltration In An Α-Syn Model Of Parkinson Disease, Aubrey Michelle Schonhoff Jan 2021

Cns Antigen Presentation And Immune Cell Infiltration In An Α-Syn Model Of Parkinson Disease, Aubrey Michelle Schonhoff

All ETDs from UAB

Parkinson disease (PD) is a neurodegenerative movement disorder that is characterized by aggregated alpha-synuclein (a-syn), loss of dopaminergic neurons, and neuroinflammation. This neuroinflammation includes the activation of immune cells that reside in the CNS, such as microglia and border associated macrophages (BAMs), in addition to the infiltration of peripheral leukocytes and increased cytokine and chemokine production. Because of the involvement of both the innate and adaptive immune systems, we sought to investigate their interaction through antigen presentation and determine what role this played in disease progression. Using an a-syn based mouse model of PD, our studies have provided evidence via …

The Cellular Role Of Tyrosyl-Dna Phosphodiesterase 1 N-Terminal Domain In Topoisomerase 2-Dna Covalent Complex Repair In Yeast, Isaac Andres Segura Jan 2021

The Cellular Role Of Tyrosyl-Dna Phosphodiesterase 1 N-Terminal Domain In Topoisomerase 2-Dna Covalent Complex Repair In Yeast, Isaac Andres Segura

All ETDs from UAB

Tyrosyl-DNA phosphodiesterase 1 (Tdp1) removes adducts from DNA-ends, including damaged nucleotides and protein-DNA crosslinks such as DNA topoisomerases (Topo1 and Topo2) and Tdp1 itself. The N-terminal domain (NTD) of Tdp1 has been understudied, structurally unresolved, and seemingly dispensable for in vitro catalytic activity. However, our lab made the novel observation that the NTD is critical for Tdp1 cellular activity in removing Topo1-DNA covalent complexes (Topo1cc). Removal of the first 80 residues (yeast Tdp1 NTD) results in the loss of the toxic phenotype induced by full-length Tdp1 catalytic mutants. This suggests that in cells the NTD has a critical function in …

“Pertinent And Plain”: Robert Barclay And The Transformation Of Quaker Silence, Samuel Phillips Jan 2021

“Pertinent And Plain”: Robert Barclay And The Transformation Of Quaker Silence, Samuel Phillips

All ETDs from UAB

Scholarship on Quaker rhetoric has frequently neglected the subversive possibilities of Quaker pamphleteering, particularly in the period following the publication of Robert Barclay’s Apology. This article develops an epistemological framework based on the Apology and demonstrates that although Quaker pamphleteers during this “Quietist” period asserted the honesty and transparency of their use of language, the Apology works to license certain circumlocutionary moves which Quaker authors used to obscure potentially seditious or heretical ideas. It focuses on two classical rhetorical figures, paralipsis and aposiopesis. Through a close reading of pamphlets written before and after the publication of the Apology, it demonstrates …

Development And Evaluation Of A Functional Human Cardiac Tissue Equivalent Fabricated From Multi-Lineage Human Induced Pluripotent Stem Cell Derived Cells, Danielle Pretorius Jan 2021

Development And Evaluation Of A Functional Human Cardiac Tissue Equivalent Fabricated From Multi-Lineage Human Induced Pluripotent Stem Cell Derived Cells, Danielle Pretorius

All ETDs from UAB

The human heart is an exceptionally complex muscular organ that is vital for survival. Electrical impulses signal activation of mechanical contractions, pumping blood through the entire body and allowing for oxygen-exchange to occur. Unfortunately, certain disease states or traumatic injuries such as myocardial infarctions hamper the heart’s efficiency and function by damaging its structure. Ideal treatment would allow for the replacement or regeneration of the damaged tissue with cells and material harvested from the patient, thus avoiding the potential for immune rejection. Engineered cardiac tissues fabricated from human induced pluripotent stem cells have shown great promise for restoring function in …

Inflammation, Executive Function, And Adiposity In Children With Or At Risk For Obesity, Kathryn L. Prendergast Jan 2021

Inflammation, Executive Function, And Adiposity In Children With Or At Risk For Obesity, Kathryn L. Prendergast

All ETDs from UAB

Obesity is associated with executive function (EF) deficits across the lifespan. Higher body mass index (BMI), greater severity of obesity, and poorer adherence and weight outcomes in obesity treatment have all been associated with EF deficits. Adult literature has begun to emphasize neuroinflammation in obesity as a possible pathway to later cognitive impairment. However, the pediatric obesity literature has yet to even establish associations between peripheral inflammation and EF. Thus, the present study aimed to examine associations and variability in inflammatory biomarkers, EF, and adiposity in children with or at risk for obesity. Additionally, inflammation was examined as a mechanism …

Biomarker And Target Discovery In Cancer, Alyncia Dominique Robinson Jan 2021

Biomarker And Target Discovery In Cancer, Alyncia Dominique Robinson

All ETDs from UAB

Cancer is a complex disease characterized by uncontrolled growth of transformed cells that can arise in many tissue types throughout the body (e.g., breast, lung, prostate, pancreas, lymph nodes) and is a major cause of death worldwide. Cancer progression resulting in aggressive or metastatic disease accounts for one of the leading causes of death worldwide, second only to heart disease in the US. Incidentally, cancer-related mortality has been on a steady decline, dropping 25% over the last 25 years [1]. This could be attributed to improved and earlier diagnoses and better treatment options being developed over the past few decades. …
