Mechanisms Of At101 [(-)-Gossypol] Induced Cytotoxicity In Malignant Peripheral Nerve Sheath Tumors, Niroop Kaza
Mechanisms Of At101 [(-)-Gossypol] Induced Cytotoxicity In Malignant Peripheral Nerve Sheath Tumors, Niroop Kaza
All ETDs from UAB
Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive Schwann cell-derived sarcomas and are the leading cause of mortality in patients with neurofibromatosis type 1 (NF1). Current treatment modalities have been largely ineffective resulting in a high rate of MPNST recurrence and poor five year patient survival. This necessitates the exploration of alternative chemotherapeutic options for MPNST patients. By evading apoptosis and utilizing protective mechanisms such as autophagy, cancer cells develop resistance to chemo- and radio-therapy. The overall goal of my thesis studies is to evaluate chemotherapeutic agents that can modulate both apoptosis and autophagy, thus target both cell death pathways …
The Direct Effects And Vertical Transfer Of Carbonaceous And Metal-Loaded Hydrochars In Aquatic Organisms, Katie Lauren Kohlenberg
The Direct Effects And Vertical Transfer Of Carbonaceous And Metal-Loaded Hydrochars In Aquatic Organisms, Katie Lauren Kohlenberg
All ETDs from UAB
Industrial use of reactive micro- and nano-sized carbonaceous hydrochar (CH) is expanding rapidly. These carbonaceous particulates are synthesized by the pyrolysis of biomass in an industrial process called hydrothermal carbonization (HTC) and have gained attention as a soil conditioner. Additionally, the HTC process is being expanded to coat heavy-metal nanoparticles, such as nano-copper and nano-iron, to prevent surface oxidation of these metals. These carbonaceous particles are likely to undergo secondary reactions with metals, toxicants, or other compounds. There is potential for these carbonaceous particulates to enter watersheds and have deleterious effects on aquatic food web dynamics. In this study, we …
The Role Of Il-7 And C-Kit Signaling Pathways In Specification Of B Lymphocytes, Oleg Kolupaev
The Role Of Il-7 And C-Kit Signaling Pathways In Specification Of B Lymphocytes, Oleg Kolupaev
All ETDs from UAB
B-lymphocyte commitment from common lymphoid progenitors (CLP) is orchestrated by a number of transcription factors and extracellular developmental cues. Most prominently, IL-7 signaling acts as a critical factor for promotion of B cell differentiation through its role in upregulation of the B-cell commitment factor, Ebf1, by activation of Jak-Stat5 signaling. Other cytokines and growth factors secreted by bone marrow stromal cells, such as Flt3 ligand, are known to modulate developmental outcomes by regulating the IL-7R-Jak-Stat5 pathway. The role of stem cell factor (SCF), the ligand of the receptor tyrosine kinase (RTK) c-Kit, and the mechanism by which it affects cell …

Purpose: The influence of the vertical abutment positions on the retention of a three-unit implant supported cantilevered FPD restoration was measured and compared. Methods: The study included four groups (n=10) of cantilevered FPDs restorations, fabricated of monolithic zirconia. Prefabricated titanium implant abutments were used and connected to their analogs. The analogs were fixated on a split mold and their vertical position was adjusted on a 2 mm bases depending on the group, resulting also to an alteration of the occlusal thickness of the restorative material. The prosthesis were cemented to the abutments with Rely X Luting Plus resin modified glass …
Osteomimmunology Of Bone Fracture Healing And Cell And Gene Therapy Approaches For Nonunion Bone Defects, Seth G. Levy
Osteomimmunology Of Bone Fracture Healing And Cell And Gene Therapy Approaches For Nonunion Bone Defects, Seth G. Levy
All ETDs from UAB
Despite the body's ability to repair bone fractures under normal circumstances, up to 10% of the 7.9 million fractures suffered in the United States each year do not achieve bony union. Bone fractures heal with overlapping phases of inflammation, cell proliferation, and bone remodeling. Osteogenesis and angiogenesis are known to work in concert to control many stages of this process, but when one is impaired it leads to failure of bone healing, referred to as a nonunion. Such nonunion fractures often result from critical-size defects that will not completely heal over the natural lifetime of the animal. Based on this …
Type I Interferons-Induced Follicular Translocation Of Lymphotoxin-Expressing Marginal Zone B Cells Initiates Lupus, Hao Li
All ETDs from UAB
Systemic lupus erythematosus (SLE) is characterized by elevation of type I interferon (IFN) signature genes and production of autoantibodies against apoptotic self-antigens. The complex nature of SLE, however, suggests that multiple immune dysregulations must integrate to initiate the disease. Here, we provided evidence that lupus can be initiated via type I IFNs-induced follicular translocation of membrane lymphotoxin (mLT)-expressing marginal zone (MZ) B cells in the spleen of lupus prone BXD2 mice. The mislocation of mLT+ MZ B cells induces two important pathogenic effects including deteriorations of marginal zone macrophages (MZMs) in the MZ, leading to defective clearance of apoptotic debris, …
Evaluation Of Tau As A Target For Alzheimer Disease And Other Conditions With Epileptiform Activity, Zhiyong Li
Evaluation Of Tau As A Target For Alzheimer Disease And Other Conditions With Epileptiform Activity, Zhiyong Li
All ETDs from UAB
Alzheimer disease (AD) is a devastating neurological condition that slowly destroys memory and thinking skills of the affected patients. No disease-modifying medications are available. While all the treatment against Aß (major component of one hallmark pathology of AD) failed in clinical trials, tau (major component of the other hallmark pathology of AD) is emerging as a better alternative. Germline knockout of tau does not cause overt abnormalities in young mice and prevents AD-like deficits in mouse models of AD. Germline knockout of tau also confers resistance to epileptiform activity in models of both AD and epilepsy. Therefore tau is proposed …
The Small Sample Inferences Of Cluster-Randomized Trials, Peng Li
The Small Sample Inferences Of Cluster-Randomized Trials, Peng Li
All ETDs from UAB
Generalized estimating equation (GEE) and generalized linear mixed model (GLMM) are two statistical modeling methods commonly used in the analyses of correlated outcomes from cluster-randomized trials (CRTs). The fact that most CRTs involve only a small number of clusters makes the small sample adjustment critical to preserve the type I error rates in testing the null hypothesis of intervention effects. To improve the small sample performance of GEE, some bias-corrected sandwich estimators have been proposed. To improve the small sample performance of GLMM, the approximated Wald F test is used to replace the asymptotic ÷^2 test and the denominator degrees …
A Low-Power Multichannel Telemetry System For High-Speed Wireless Neural Recordings, Yang-Guo Li
A Low-Power Multichannel Telemetry System For High-Speed Wireless Neural Recordings, Yang-Guo Li
All ETDs from UAB
The advances in microelectronics and wireless communication are going to change our lifestyles and benefit our healthcare system in the foreseeable future. New conception named "Ubiquitous Healthcare" has been proposed. It envisions a continuous, real-time, remote monitoring on the health conditions of people through the wireless communication network. Traditional biosignal monitoring devices need wire-connections for the recording, which may cause skin rupture and body infection. The physical connections also limited the number of the integrated recording channels. A low-power implantable multichannel telemetry system which can be implanted in the body and transmit recordings wirelessly can solve the problem of the …
Ksrp Ablation Enhances Lipolysis In White Adipose Tissue Through Reduced Mir-145 Expression, Yi-Yu Lin
Ksrp Ablation Enhances Lipolysis In White Adipose Tissue Through Reduced Mir-145 Expression, Yi-Yu Lin
All ETDs from UAB
White adipose tissue (WAT) releases fatty acids from stored triacylglycerol (TAG) for energy source. Here, we report that null mice for KH-type splicing regulatory protein (KSRP), an RNA-binding protein, resisted high fat diet (HFD) induced obesity. Target of deletion of KSRP regulates gene expression at multiple levels and enhances lipolysis in epididymal WAT (eWAT) due to up-regulation of genes promoting lipolytic activity. Expression of miR-145 is decreased due to impaired pri-miR-145 processing in Ksrp-/- eWAT. We show that miR-145 directly targets and represses Foxo1 and Cgi58, activators of lipolytic activity, and forced expression of miR-145 attenuates lipolysis. This study reveals …

Streptococcus mutans (Sm) and Streptococcus sobrinus (Ss) , are considered the major bacterium associated with dental caries. However, recent findings identify Scardovia species (Sc) is associated with early childhood caries, with or without the presence of Sm or Ss. This IRB approved study quantified the presence of Sm and Sc in saliva by real-time quantitative PCR (qPCR) in relation to caries. Saliva samples were collected from 72 preschool children in a high-caries risk community in rural Alabama. SYBR Green-based qPCR using extracted DNA and primers specific for Sm, Ss, Sc and total bacteria determined the copy number (CN/ml). Twenty two …
Expression Of The Influenza Protein M2 During Viral Infection Inhibits Cftr Activity, James David Londino
Expression Of The Influenza Protein M2 During Viral Infection Inhibits Cftr Activity, James David Londino
All ETDs from UAB
The purpose of these dissertation studies was to 1) determine whether influenza infection alters CFTR activity in polarized epithelium; 2) measure CFTR expression and activity in cells co-expressing influenza M2 protein; and 3) examine the role of M2 on the alteration of CFTR during viral infection. We determined that infection of polarized primary epithelial cells with influenza decreases CFTR expression and activity. In addition, individual cells infected with influenza had decreased CFTR conductance as measured by whole-cell patch clamp. We also found that the influenza ion channel, matrix protein 2 (M2), alone reduced CFTR expression and activity. M2 is expressed …

Background: Obesity and overweight contribute to worksite absenteeism and decreased productivity. Encouraging employees to frequent the stairs is one approach towards increasing worksite physical activity. Methods: An explanatory, mixed methods research design was used to explore the impact of a health communications campaign entitled "Stepping Up" on worksite stair use in health department employees. The campaign included octagonal shaped posters similar to traffic signs, email prompts and printed materials. A pretest and posttest were administered before and after the intervention to assess stair use frequencies and identify predictors. A Wilcoxon signed-rank test for related samples, Spearman correlations and multiple rank …
Sociocultural Disposition And Psychosocial Adjustment: An Examination Of The Relationship Of Self-Efficacy, Optimism, And Hope To Social And Psychological Adjustment Among Urban Minority Youth, Latoya J. O'Neal
All ETDs from UAB
This dissertation examines the relationship between sociocultural disposition, as protective factors, and social and psychological adjustment among urban minority youth. Pierre Bourdieu's concept of the habitus provides the theoretical foundation for sociocultural disposition as a factor influencing the reproduction of poor psychosocial adjustment. Sociocultural disposition is operationalized as self-efficacy, optimism, and hopelessness. Data from this study are from the Birmingham Youth and Technology Study (BYTS) which was conducted during the 2008-2009 school year by researchers at the University of Alabama at Birmingham (UAB). Ordinary Least Squares (OLS) Regression was used to test six hypotheses regarding the relationship between sociocultural disposition …

3-Dimensional Comparison Of Hard And Soft Tissue Asymmetry In, Philip H. Pan
All ETDs from UAB
Class III malocclusions are particularly prevalent in the Asian population. Threedimensional imaging has improved the ability to portray the three-dimensional characteristics of soft tissue face and underlying skeletal hard tissue. While soft tissue reflects the underlying hard tissue structures, soft tissue may either provide a compensation for hard tissue asymmetry or potentially exacerbate the asymmetry. The current study is novel in that the hard tissue and soft tissue asymmetries are both analyzed relative to the same reference point, and using 3D imaging, we have attempted to elucidate a compensation tendency between the hard and soft tissues. The sample was comprised …
Altered Dna Methylation Contributes To Temporal Lobe Epilepsy And Associated Memory Deficits, Robert Ryley Parrish
Altered Dna Methylation Contributes To Temporal Lobe Epilepsy And Associated Memory Deficits, Robert Ryley Parrish
All ETDs from UAB
Status epilepticus (SE) triggers molecular mechanisms that underlie the cellular and network changes that occur during the development of temporal lobe epilepsy (TLE). Additionally, gene expression alterations occur during epileptogenesis and in the epileptic brain that contribute to the persistence of the phenotype. However, the underlying epigenetic mechanisms responsible for aberrant gene expression that occur with epilepsy require further investigation. This dissertation investigates how DNA methylation in the hippocampus affects epilepsy and contributes to the memory impairments associated with the disorder. We found that DNA methylation was altered in a hippocampal subregion specific manner in the epileptic brain, displaying altered …

Biology, Ecology, And Conservation Of Hatchling And Post-Hatchling Diamondback Terrapin (Malaclemys Terrapin Pileata), Tandy Dolin Petrov
All ETDs from UAB
The diamondback terrapin has been considered a keystone species along its range in the coastal, brackish estuaries bordering the Atlantic Coast and Gulf of Mexico. Historically, diamondback terrapin populations have been numerous; however, in Alabama; the population has dramatically declined over the past century due to a variety of threats. The threats in Alabama include predation by raccoons, incidental drowning in crab traps, as well as road mitigated mortality. Because of these threats, collaboration efforts between UAB, USA, DISL, and the Alabama Department of Conservation and Natural Resources, a head-start program has been initiated to mitigate the predation of hatchlings. …
The Impact Of Interleukin-21 On Intestinal Inflammation During Health And Disease, Catherine Poholek
The Impact Of Interleukin-21 On Intestinal Inflammation During Health And Disease, Catherine Poholek
All ETDs from UAB
The intestinal environment is extremely complex due to the dynamic interactions between host cells and luminal contents. The immune cells located in the intestine are responsible for sensing and responding to intestinal pathogens as well as maintaining tolerance to non-pathogenic organisms and environmental antigens. Disruption in this delicate balance results in chronic intestinal inflammation. Particularly important to the maintenance of intestinal homeostasis are CD4 T cells and innate lymphoid cells (ILCs). These cells produce cytokines in an antigen-specific and antigen-independent manner, respectively, and have analogous subsets with similar cytokine profiles. CD4 T cells and ILCs expressing ROR gamma t (Th17 …
Power Issues And Internal Pilot Designs For Cluster-Randomized Trials With Unequal Cluster Sizes, Ashutosh Ranjan
Power Issues And Internal Pilot Designs For Cluster-Randomized Trials With Unequal Cluster Sizes, Ashutosh Ranjan
All ETDs from UAB
Cluster-randomized trials that randomize groups of individuals, instead of individuals themselves, run the risk of being underpowered when they are designed assuming equal cluster sizes but end up recruiting unequal clusters. The loss in power of the trial is directly related to the amount of variation present among the cluster sizes. To overcome the loss in power, researchers often employ a weighted analysis of the cluster means. Different weighting procedures have been developed that appear to maintain the power of the trial but there have been fewer attempts to describe the impact of different weights on the type I error …
Molecular And Functional Interaction Of Runx2 And Sp7 For Development Of The Osteoblast Phenotype, Harunur Rashid
Molecular And Functional Interaction Of Runx2 And Sp7 For Development Of The Osteoblast Phenotype, Harunur Rashid
All ETDs from UAB
Runx2 and Sp7 transcription factors are essential for skeletogenesis. Deletion of either gene in mice results in failure of bone tissue development. However, underlying mechanisms responsible for a surprisingly similar phenotype by two distinctly unrelated proteins remain unknown. Sp7 is a Runx2 downstream target gene and is not expressed in Runx2 null mice. Thus, the Runx2 null model represents a compound phenotype of loss of both proteins. In contrast, normal levels of Runx2 mRNA are noted in Sp7 null mice. The failure of Runx2 to promote bone formation in Sp7 null mice suggests that Sp7 is required for Runx2 function …

Epithelial Sodium Channel Purification And X-Ray Crystallographic Studies, Bharat G. Reddy
All ETDs from UAB
Epithelial sodium channels (ENaC) play a critical role in maintaining Na+ homeostasis in various tissues throughout our body. Understanding of ENaC structure is mediated from studies of the homologous acid sensing ion channel 1 (ASIC1). However, ENaC has sev-eral notable functional differences compared to ASIC1, thereby providing justification for determination of its three-dimensional structure. Unfortunately, this goal remains elu-sive due to several experimental challenges. Of the subunits that comprise a physiological hetero-trimeric ENaC (α, ß, and γ), the α-subunit alone is of significant interest. αENaC is unique in that it is capable of forming a homo-trimeric structure capable of conducting …

Finding Bands In Optical Coherence Tomography Images Using Curve And Function Fitting, Douglas Howard Ross
All ETDs from UAB
Optical Coherence Tomography (OCT) is a noninvasive method to create a volume of data that is a measurement of the reflectance properties of tissue. The retina of the human eye has a layered structure that presents as bands in the OCT images. Evaluation of OCT images can be used to diagnose abnormal structure due to disease or damage. The principal task in evaluating the OCT images is segmentation. While the images can be evaluated by trained personnel, the quantity of OCT image data is increasing rapidly, therefore, the creation of methods for automatic segmentation is vital. Prior work in segmenting …

Mycobacterium tuberculosis, the causative agent on tuberculosis in humans, is a globally important pathogen. In 2013, nearly 1.2 million people died from tuberculosis. Drug treatments and effective vaccines are lacking for this pathogen. A better understanding of the basic physiology of M. tuberculosis is required to improve disease outcomes. Upon inhalation into the lungs, M. tuberculosis is taken up by macrophages, in a process called phagocytosis, which normally destroys invading bacteria. As the phagosome inside the macrophage matures, bactericidal mechanisms are activated including: lowering pH, and introducing reactive oxygen and nitrogen species and hydrolytic enzymes to degrade bacterial cell walls. …

Coexisting lesions can reduce genomic heterogeneity in precancer progression analysis by reducing variation and bias that can mask gene effects. Our goal was to use novel methodologies to depict the neoplastic stage effect (Normal v. LSIL v. HSIL) in cervical precancer. We analyzed the neoplastic stage effect via in silico and in vivo methodologies. For in silico analyses, we calculated differential expression (DE) estimates from a systematic review of DNA methylation and gene expression literature. Significant genes (FC≤2.0 or p-value≥0.05) were grouped by histology for pathway analysis. For in vivo analyses, we performed RNA-seq on microdissected FFPE coexisting cervical tissue …

Vitally important to HIV-1 vaccine and prevention studies is a well-characterized animal model that recapitulates the salient features of HIV infection in humans. SIVmac and SIVsmm infection of rhesus macaques share key virological and pathological parallels with HIV-1 infection of humans including target cell entry via CD4 and CCR5 (Edinger et al., 1997), progressive T cell loss (Swanstrom & Coffin, 2012), generalized immune activation and preferential loss of mucosal Th17 cells (Brenchley et al., 2008; Klatt & Brenchley, 2010), and a vigorous but ultimately ineffective adaptive immune response. Though commonly used as a vaccine challenge strain in the macaque model, …
