In a glasshouse experiment, the effectiveness of the entomopathogenic nematode (EPN) Steinernema feltiae (Filipjev) (Rhabditida: Steinernematidae) was compared with thiamethoxam for the control of adults of the greenhouse whitefly (GWF), Trialeurodes vaporariorum (Westwood) (Homoptera: Aleyrodidae), on slicer cucumbers. In a period from mid June to the end of August (2007 and 2008), cucumbers were grown in 4 different growth substrates: expanded perlite, expanded vermiculite, light expanded clay, and peat. Molecular Cloning, Characterization, And Homologous Expression Of An Endochitinase Gene From Bacillus Thuringiensis Serovar Morrisoni, Sezer Okay, Gülay Özcengi̇z

The endochitinase gene (chi3023) of Bacillus thuringiensis (Bt) serovar morrisoni strain 3023 was amplified via polymerase chain reaction (PCR) and cloned in Escherichia coli. The ORF of chi3023 (GenBank Accession Number: DQ993175) consists of 2031 nucleotides encoding a 676-residue protein with a calculated molecular mass of 74.5 kDa and a pI value of 6.0. The amino acid sequence of Chi3023 was compared with previously sequenced Bt chitinases and the phylogenetic relationships among them were determined. The objective of this study was to evaluate the effects on the structure and ultrastructure of pepper plants (Capsicum longum A.DC.) exposed to UV radiation under greenhouse conditions. The plants were grown in a growth chamber under uniform environmental conditions and after 35 days were exposed to UV-A and UV-C radiation for 15 and 8 days, respectively. Changes in root growth were not significant, but shoot growth decreased in UV-R-exposed plants and decreased significantly in UV-C-exposed plants. The role of mineral elements (ppm), protein and oil content (%), and saponification and free fatty acid values in contributing to resistance against insect infestation was investigated in 4 fish species: Clarias gariepinus, Parachuma obscura, Hydrocynus sp., and Malapterus electricus. Smoked fish samples were purchased from a local market in Ilorin, Kwara State, Nigeria, packaged, and maintained at 4 ^°C for 7 days. In vitro antioxidant properties of 3 different extracts of black mulberry (Morus nigra L.) were investigated. Acidified methanol, acidified water, and non-acidified methanol/water solutions were used to prepare extracts. Different solvents caused different protonation on black mulberry anthocyanin structures, which were predicted according to their UV-VIS spectrum. Extracts indicated 2 main peaks at about ~280 and ~520 nm with different peak areas in their UV-VIS spectrum. The rank of extracts' total phenolic content and reducing power values were both found to be in correlation with their absorbance at ~280 nm. We have examined the consequences of Ultraviolet-B (UV-B) irradiation in the thallus of the lichen Physcia semipinnata (J.F.Gmel) in terms of cell viability and apoptotic-like formation. UV-B induced oligonucleosomal DNA fragmentation was detected by TUNEL assay and this is the first study showing DNA fragmentation in thalli. The intensity of TUNEL-positive cells after exposure to UV-B at doses up to 95.9 J/cm2 was higher in the photobiont layer than the mycobiont layer.

Detergent compatible alkaline protease production by a newly isolated Bacillus sp.Y from laundry soil was studied under shake flask conditions in growth medium comprised (g/L): peptone, 1; NaCl, 5; skim milk, 100; Na_2CO_3, 4. Different environmental process parameters such as fermentation period, initial pH, incubation temperature, and nutritional parameters such as supplementation of different starch sources and nitrogen sources were standardized for the maximum yield of alkaline protease. Possible seasonal variations of the fatty acid composition (neutral lipid, phospholipid, and total lipid content) of the freshwater snail Melanopsis praemorsa were investigated over the course of a year. Snail body lipids were fractionated by thin layer chromatography. Fatty acid composition of total body lipid, phospholipid, and neutral lipid of whole snail and total lipid of their algal food were analyzed by capillary gas chromatography and gas chromatography-mass spectrometry. The present study was conducted to determine the phenotypic and genotypic characterization of thermophilic bacteria isolated from Pasinler hot spring, Erzurum, Turkey. Fatty acid profiles, BOX PCR fingerprints, and 16S rDNA sequence data were used for the phenotypic and genotypic characterization of thermophilic bacteria. Totally 9 different bacterial strains were selected based on morphological, physiological, and biochemical tests. A total of 45 lactic acid bacteria were isolated from 10 different boza samples consumed in Turkey. The isolates were identified with respect to colonial, cellular morphology, and biochemical characters based on API CHL50 (Biomérieux) kit and also automated riboprinting was used to identify of the isolates. These isolates were tested for inhibitory activity against food-borne bacterial pathogens. DNA was extracted from 2 cyprinid fishes kept under formalin preservation in 2 different time durations, and the quality and quantity were analyzed. The tissue samples from specimens preserved for shorter duration yielded higher quantity of DNA compared to long-term preserved tissues. ANOVA (one way) on the quantity of the DNA from the samples of 2 periods showed that the genetic material obtained from the recently preserved samples was highly significant (P > 0.001). The purpose of the current study was to investigate the possible antimicrobial and anticancer properties of the aquatic moss Fontinalis antipyretica Hedw. (Fontinalaceae). We first obtained 8 different extracts (methanol; chloroform; acetone; ethyl acetate A, B, C, and D) by two different extraction processes. Ran binding proteins (RanBPs) are highly conserved members of the GTP-binding protein family that are involved in nuclear protein export between the nucleus and the cytoplasm. In this study, a CmRanBP gene from a melon was isolated (Cucumis melo L.) using the RACE (rapid amplification of cDNA ends) method. The 778 basepair long melon, with a RanBP cDNA encoding consisting of 197 amino acids (22.2 kDa protein), was characterized (GenBank accession no: EU853459). The sensitivity of the effective refractive index of slab waveguide sensors to variations in the refractive index of the cladding is compared to that of the ellipsometric parameters. The changes of the effective refractive index of a waveguide and the ellipsometric parameter \Delta , due to the index change of the cladding, were derived and plotted as a function of the guiding layer thickness and with the index of the cladding. Optical Characterization Of Fe^{3+} Doped Cds Nanoparticles Synthesized By Wet-Chemical Route, Seyed Mohammad Taheri Otaqsara, Mohammad Hassan Yousefi, Ali Azam Khosravi

Fe^{3+} doped CdS nanoparticles using Mercaptoethanol as a capping agent were prepared by colloidal wet-chemical method. UV-VIS spectra show that Fe-doping can effectively tune energy band structure, i.e. a large blue shift (\sim 30 nm) with increasing Fe concentration. PL measurement of doped CdS nanoparticles shows an additional emission band at \sim 532 nm, which can be due to radiative transition of dopant level to ground state. X-ray diffraction patterns studies revealed cubic crystal phase. TEM micrographs show a spherical shape and nearly well distribution with an average particle size of \sim 4 nm.

The aim of this study was to observe the microstructure and ultrastructure of the testis, demonstrate testicular cell development and apoptosis, and elucidate regularity of development and apoptosis in ostrich chick testes. By employing light microscopy 3 obvious development characteristics were detected with ostrich age increasing: first, many primordial germ cells and a few spermatogonia were found while seminiferous tubule integrity was not evident in testes of 1-day-old ostrich chicks nor were primordial germ cells. To investigate the global response of microvascular endothelial cells (MVECs) to foot-and-mouth (FMDV)disease vaccine (FMDV) antigen and to explore the role of MVECs in FMD vaccine immunity, the gene expression alterations of rat myocardium microvascular endothelial cells (RMMVECs) after stimulation with FMD virus (FMDV) 146S antigen were assayed using cDNA microarrays. We observed that 18 genes and 67 genes in RMMVECs were respectively altered by FMDV 146S antigen at 6 h and 24 h, respectively. Ovine scrapie is considered a neurodegenerative disease characterized by the accumulation of an abnormal, protease-resistant isoform of PrP. The polymorphisms at codons 136, 154, and 171 are associated with susceptibility to scrapie. In order to detect the existing polymorphisms at the 3 codons we used the real-time PCR technique. The obtained genotypes were confirmed by PCR amplification and sequencing of a 402 bp fragment of exon 3 of the prion gene. The aim of the present study was to determine the effects of replacing fish oil (FO) with cottonseed oil (CSO), as an alternative plant lipid source in diets for rainbow trout (89.3 ± 1.1 g mean individual weight), on the growth and fatty acid composition of fish. Regulatory T lymphocytes (T-regs) may be involved in suppressing the host's immunity, which is important for viral clearance, and may therefore contribute to persistent viral infection. However, nothing is known regarding the situation of porcine T-regs during persistent infection with porcine reproductive and respiratory syndrome virus (PRRSV). This study, which used flow cytometry and real-time PCR, determined that the frequencies of CD4+CD25+ T-cells and porcine Forkhead/winged helix transcription factor (FoxP3) mRNA levels in peripheral blood significantly increased during PRRSV persistence, indicating that CD4+CD25+ T-regs might play an important role in PRRSV persistent infection.
