Ex situ conservation of Dianthus giganteus subsp. banaticus was obtained by in vitro culture on different culture media supplemented with phytohormones. Assessment of somaclonal variability over a period of preservation by in vitro culture is required prior to introduction and acclimatization in natural habitats. An efficient in vitro bulblet production procedure from immature zygotic embryos of endemic and endangered Muscari muscarimi Medik. was described in the current study. Zygotic embryos were first isolated from immature seeds and cultured on different nutrient media compositions supplemented with various combinations of \alpha-naphthalene acetic acid (NAA), picloram, dicamba, 6-benzylaminopurine (BAP), and thidiazuron (TDZ). The best bulblet regeneration (59 bulblets per explant) was achieved in Murashige and Skoog (MS) medium containing 4 mg/L BAP and 0.5 mg/L NAA after 1 year of culture initiation. The effect of various factors on dye decolorization activity of crude laccase enzyme from repeated-batch culture of Funalia trogii ATCC 200800 to obtain rapid and high decolorization activity against Reactive Black 5 and Reactive Blue 171 dyes was investigated. All conditions used were important for dye decolorizing activity of this crude laccase enzyme. The optimum pH of decolorization was tested at 30 °C and it was around 3.0. The cytidine-5'-triphosphate (CTP) synthase (EC 6.4.3.2) gene (pyrG) was cloned and sequenced from the thermophilic bacterium Anoxybacillus gonensis G2 (Ago). The gene is 1590 bp in length and encodes a protein of 530 amino acids, with a molecular mass of 59.5 kDa. The amino acid sequence of CTP synthase shares approximately 90%-94% similarity to Bacillus sp., and it belongs to the triad glutamine amidotransferases, which utilize a Cys-His-Glu triad for activity. Multiple sequence alignments revealed that the enzyme includes conserved amino acids responsible for catalytic activity and the binding of a divalent metal ion (Mg^{+2}). The combined effect of anther incubation time on CP induction medium (12, 14, and 16 days) and 2 concentrations of kinetin in R1 regeneration medium (0.1 and 0.3 mg/L) on the effectiveness of androgenesis was investigated in 17 genotypes of Capsicum grown in Poland. A high-quality and low-cost genomic DNA isolation method is needed for use in microbiology laboratories. In this study, we developed a new modified guanidinium isothiocyanate method to isolate DNA from Escherichia coli, Salmonella enterica subsp. enterica serotype Typhimurium, and Staphylococcus aureus and compared it with 4 other DNA isolation methods. The results show that the modified guanidinium isothiocyanate method developed in our laboratory is simple, fast, and inexpensive and yields DNA whose quality and quantity are similar to that of 2 commercial extraction kits but far better than 2 conventional DNA extraction methods used for comparison.

The release of neurosecretory material from A2 neurosecretory neurons (NSNs) was stimulated in Lymantria dispar fourth instar caterpillars exposed to a temperature of 35 °C for 1, 12, and 24 h, as well as those allowed recover after exposure (12 h at 35 °C, then 12 h at 23 °C). The levels of 2 protein forms with the same molecular mass as bombyxin (3-4 and 4-5 kDa) increased with prolonged exposure to 35 °C. Fourteen genotypes and 3 different gamma ray doses were tested to develop an efficient haploidization protocol in squash. For this purpose, male flowers collected 1 day before anthesis were irradiated with 150, 200, and 300 Gy gamma ray doses, and female flowers were pollinated with the irradiated pollens the next day. In the first year of the study, 1858 embryos were obtained from 219 fruits. The effects of physical and chemical sublethal treatments on the antibacterial activity of enterocin KP produced by Enterococcus faecalis KP against Escherichia coli O157:H7 and Salmonella Typhimurium were investigated. Enterocin KP was not active against intact cells of E. coli O157:H7 or S. Typhimurium. Various elicitors have already been reported to enhance the production of taxol and related taxanes. In this study, effects of a fungal metabolite, squalestatin, on production of taxol and baccatin III were studied. Expressions of 4 key involved genes, ts, dbat, bapt, and dbtnbt, in suspension cultures of Taxus baccata were also detected using qPCR. Results showed that application of squalestatin significantly increased taxol and baccatin III yields. Increased expressions of the genes were in accordance with measures of taxol and baccatin accumulations in cells and medium. The intention of this study was to investigate the toxic effect of cobalt on the bone metabolism of rats, including calcium, inorganic phosphorus, and bone-specific alkaline phosphatase of serum during short- and long-term experiments. Rats were injected intraperitoneally (IP) with either 16 mg kg^{-1} BW of cobalt for 5 and 10 days or 6 mg kg^{-1} BW for 20 and 40 days. It was found that IP administrations of cobalt could increase all serum parameters associated with bone metabolic processes. NKX3.1 is a prostate-specific transcription factor that is regulated by the androgen receptor in the presence of androgens. It functions as a tumor suppressor against the development of prostatic intraepithelial neoplasia and primary prostate tumors. Here, a recognized approach combining in silico analysis and chromatin immunoprecipitation (ChIP) was used to identify the genes directly regulated by NKX3.1 promoter binding in LNCaP cells. Quantitative PCR using ChIP-captured DNAs as templates verified a subset of NKX3.1 binding motifs. Licorice (Glycyrrhiza spp.) is an important medicinal plant and its distribution of natural habitats has been shrinking day by day due to extensive collection. Genetic diversity identification will likely assist in conserving the different Glycyrrhiza species. A total of 127 simple sequence repeat (SSR) markers from Medicago truncatula Gaertn., Phaseolus vulgaris L., and Cicer arietinum L. were used and 26 of them were amplified on Glycyrrhiza genomes. Xanthium strumarium L. is a major weed affecting flour corn in Khyber Pakhtunkhwa Province of Pakistan. Studies conducted in 2006 and 2007 evaluated corn yield and yield component responses to competition from X. strumarium over a range of corn populations (5, 7.5, 10, and 12.5 plants m^{-2}) and X. strumarium densities (0, 2, 4, 6, 8, 10, and 12 plants m^{-2}). This research was an attempt to study the effect of heavy metals lead and cadmium (0.05 mM and 0.3 mM) on growth and antioxidant enzymes of seedlings of 2 mung bean genotypes (NM 19-19 and Azri mung-2006). Results revealed that germination percentage and seedling length decreased when compared with the control for both genotypes. However, seedling length and germination percentage was better in NM 19-19 as compared to Azri mung-2006. Elevated levels of protein were observed under metal stress in both genotypes. A survey was conducted for entomopathogenic nematodes in various agricultural fields in the Eastern Black Sea region of Turkey. A total of 77 soil samples were collected from 15 distinct geographic areas during 2010-2011. Seven entomopathogenic nematode isolates (ZET02, ZET04, ZET09, ZET28, ZET31, ZET35, and ZET76) were detected from the soil samples (9.1% positive) using the Galleria baiting technique. Morphological and molecular characterizations of the isolates were performed for species identification. In this study, the effects of organic silicon (Si) on the toxicity of imidacloprid to a new invasive mealybug, Phenacoccus solenopsis (Hemiptera: Pseudococcidae), infesting Portulaca grandiflora (Caryophyllales: Portulacaceae) plants were evaluated using spotting and dipping methods under laboratory and field conditions to improve management strategies for solenopsis mealybug. Results showed significant synergistic effects of 0.06% organic Si with imidacloprid solution against third-instar P. solenopsis nymphs and adult females. The effects of Trichoderma harzianum, T. hamatum, T. viride, T. polysporum, and T. koningii on the wilt disease complex of chickpea caused by Fusarium oxysporum f. sp. ciceri and Rhizoctonia solani were investigated under field conditions during 2 consecutive years. Chickpea cultivar Avrodhi, grown in plots inoculated with F. oxysporum f. sp. ciceri and R. solani, showed chlorosis of leaves and wilting of foliage and exhibited a 22%-25% decrease in yield. A nucleopolyhedrovirus (NPV) was isolated from diseased larvae of the eastern tent caterpillar, Malacosoma americanum (Fabricius), collected from the northeastern part of Turkey. Light and electron microscopic observations showed that this virus is a multiple nucleopolyhedrovirus (MNPV). The dimensions of the polyhedral inclusion bodies varied from 1.0 to 2.1 µm, and the sizes of the virions were also measured to be approximately 194.5 nm. Restriction analysis of the new isolate and ManeNPV-T2 (previously isolated from M. neustria larvae) showed that the new virus is closely related to ManeNPV-T2. In the present study, response surface methodology was conducted for the determination of effects of some phenolics (gallic acid, ellagic acid, and quercetin) on some stability parameters of mixed oil prepared with sunflower and hazelnut oils (50:50, v/v). In this respect, peroxide value (PV), free fatty acids (FFAs), p-anisidine value, induction period, and refractive index of mixed oil were investigated. Predictive regression equations were constructed for the estimation of each studied parameter (R^2 > 0.861). This study reports the presence of the summer fruit tortrix moth, Adoxophyes orana, in pome and stone fruit orchards for the first time in northwestern Turkey. Based on growers complaints about cosmetic damage to peach and pear fruits, pheromone traps were set up in mid-April of 2009 and 2010 to monitor the occurrence of the summer fruit tortrix moth. We conducted studies in conventional apple, pear, peach, and sweet cherry orchards in Bursa, northwestern Turkey. Moths caught in pheromone traps showed a trimodal flight pattern in all orchards.
