Characterization Of The Human Sialidase Neu4 Gene Promoter, Volkan Seyrantepe, Murat Delman

Turkish Journal of Biology

There are 4 different sialidases that have been described in humans: lysosomal (Neu1), cytoplasmic (Neu2), plasma membrane (Neu3), and lysosomal/mitochondrial (Neu4). Previously, we have shown that Neu4 has a broad substrate specificity and is active against glyco-conjugates, including G_{M2} ganglioside, at the acidic pH of 3.2. An overexpression of Neu4 in transfected neuroglia cells from a Tay-Sachs patient shows a clearance of accumulated G_{M2}, indicating the biological importance of Neu4. In this paper, we aimed to characterize a minimal promoter region of the human Neu4 gene in order to understand the molecular mechanism regulating its expression. We cloned 7 different …

Turkish Journal of Biology

Colicin N is a bacterial toxin that kills Escherichia coli and related cells. Its mode of action is of interest in protein import and toxicology. Colicin N translocates across the E. coli outer membrane and periplasm by interacting with several receptors. The translocation process involves the interaction of the colicin N with the outer membrane porin OmpF and subsequently with the integral membrane protein TolA. The N-terminal domain of colicin N is involved in the import process. TolA consists of 3 domains. The N-terminal domain of colicin N interacts with the C-terminal domain of TolA at later stages of the …

Turkish Journal of Biology

A novel gram-negative bacterium, OG2, was isolated from the body microflora of cockroaches (Blatta orientalis). Based on morphological, biochemical, and 16S ribosomal DNA sequence analysis, the isolated strain OG2 was identified as Stenotrophomonas maltophilia. This bacterial strain was screened for its alpha-cypermethrin-degrading potential with minimal salt medium (MSM). The alpha-cypermethrin degradation and utilization ability of the isolated organism was verified. Bacterial growth was measured by optical density in the presence of the pesticide at different concentrations (50-200 mg/L). S. maltophilia OG2 utilized alpha-cypermethrin as the sole carbon source for growth, and alpha-cypermethrin degradation increased when MSM was supplemented with glucose. …

Long-Term Water Balance Of A Bare Soil With Slope In Chuncheon, South Korea, Mehmet Aydin, Yeong-Sang Jung, Jae E. Yang, Hyun-Il Lee Jan 2014

Long-Term Water Balance Of A Bare Soil With Slope In Chuncheon, South Korea, Mehmet Aydin, Yeong-Sang Jung, Jae E. Yang, Hyun-Il Lee

Turkish Journal of Agriculture and Forestry

Water balance components of a bare soil with slope varying from 5% to 30% in Chuncheon, South Korea, were simulated using the E-DiGOR model, which proposed an interactive way to quantify runoff, drainage, soil water storage, and evaporation. Daily computations were carried out during the period of 1980 to 2009 for the identified soil-topography-climate combination. A strong correlation between measured pan evaporation and calculated potential soil evaporation was observed (R^2 = 63.8%, P < 0.01) based on the monthly data of the past 30 years (hereafter 'long-term'). When examining long-term dynamics of simulated soil evaporation, monthly mean potential and actual soil evaporations ranged from 9.1 and 9.0 mm in December to 110.5 and 75.2 mm in June and July, respectively. The ratio of actual to potential soil evaporation (E_a/E_p) had a close linkage with soil water content. The higher the soil water amount, the greater the E_a/E_p ratio was. A nonlinear relationship between rainfall and surface runoff was obtained at a given slope. Excess surface runoff and subsurface flow (percolation + interflow) occurred throughout the rainy months from July to September, with peaks in July. The ratio of direct surface runoff to rainfall increased with the natural logarithm of slope. The long-term mean annual direct surface runoff and subsurface flow at the maximum slope were 408.1 and 437.6 mm, respectively. Furthermore, mean annual surface runoff from the slope of 30% was approximately 2 times higher than that from the slope of 5%.

Autophagy And Cancer, Hacer Ezgi̇ Karakaş, Devri̇m Gözüaçik

Turkish Journal of Biology

Autophagy is an evolutionary conserved intracellular degradation and stress response mechanism that is mainly responsible for the breakdown and recycling of cytoplasmic materials, including long-lived proteins, protein aggregates, and damaged organelles. In this way, autophagy provides the cell with building blocks and allows the maintenance of homeostasis under stress conditions such as growth factor deficiency, nutrient deprivation, hypoxia, and toxins. Consequently, abnormalities of autophagy contribute to a number of pathologies ranging from neurodegenerative diseases to cancer. Autophagy was reported to have a dual role in cancer. Depending on cancer stage, autophagy seems to act as tumor suppressor or as a …

It Takes 2 Antioxidants To Tango: The Interaction Between Manganese Superoxide Dismutase And Glutathione Peroxidase-1, Dede N. Ekoue, Alan M. Diamond

Turkish Journal of Biology

Reactive oxygen species (ROS) are generated during mitochondrial oxidative metabolism. Accumulation of ROS without an effective antioxidant response can lead to oxidative stress, resulting in macromolecular damage that is implicated in the etiology of various diseases including cancer. ROS detoxification is regulated by various antioxidant proteins, specifically manganese superoxide dismutase (MnSOD), which catalyzes the conversion of superoxide into H_{2}O_{2}, and the subsequent conversion of H_{2}O_{2} into water is catalyzed by glutathione peroxidase 1 (GPx-1). In vitro and in vivo evidence supports a conflicting role of MnSOD in tumor biology and indicates that an interaction between MnSOD and GPx-1 can modulate …

Failure Of Immunological Cells To Eradicate Tumor And Cancer Cells: An Overview, Rohit Sharma, Daizee Talukdar, Parth Malik, Tapan K. Mukherjee

Turkish Journal of Biology

Inflammation can be broadly understood as a successive immune response of an organism's immune system towards nonnative or foreign antigens. This is a protective mechanism of the immune system, mediated by diverse immunological cells, to ensure homeostasis of an individual. Once activated, these immunological cells release a number of cytokines, chemokines, reactive oxygen species, reactive nitrogen species, histamines, prostaglandins, and other materials leading to inflammation. Tumor cells express altered proteins due to mutations of their genes, DNA modifications such as histone modification, DNA methylation, or other mechanisms of altered protein expression. The body's immunological cells actively recognize these altered proteins, …
