Phylogeny Of Korean Opuntia Spp. Based On Multiple Dna Regions, Krishnamoorthy Srikanth, Sung Soo Whang
Turkish Journal of Botany
Although Opuntia species are of high agronomic value in Korea, the taxonomic position of Korean Opuntia species has never been investigated. The taxonomic position of Korean Opuntia spp. Within the tribe Opuntieae was examined based on DNA sequence analysis of matK, trnL-F, atpB-rbcl, and ITS regions. The total amplified sequence length was 2977 bp; only 18 parsimonious informative sites were present, even though they belonged to different species. A phylogenetic tree using both the maximum likelihood method with 2000 bootstrap replications and Bayesian posterior probabilities was constructed. The new forma, Opuntia humifusa f. Jeollaensis, used in this study was placed …

Floral Function In Relation To Floral Structure In Two Periploca Species (Periplocoideae) Apocynaceae, Samia Heneidak, Yougasphree Naidoo
Turkish Journal of Botany
This study comprises an investigation into the floral function in relation to the floral structure of Periploca aphylla and P. Angustifolia, using light and scanning electron microscopy. Both species display the following floral structures: style-head bearing pollen translators, shelf-like nectaries, staminal feet, nectar-collecting troughs, trisegmented corolline corona lobes and corolla lobes, and anthers and nectaries covered by unicellular hairs. Lateral segments of adjacent corona lobes are fused with half of the adaxial base of the adjacent corolla lobes, creating openings through which pollinators can reach the nectar-collecting troughs in the corolla base. Flowers are rotate with spreading corolla lobes and …

Towards A New Classification Of Salvia S.L.: (Re)Establishing The Genus Pleudia Raf., Maria Will, Natalie Schmalz, Regine Classen-Bockhoff
Turkish Journal of Botany
Salvia L. İn its traditional circumscription is the largest genus within the mint family. To date, the magnitude of the task has rendered it difficult to provide a genus-wide revision based on morphological data. Current molecular investigations based on a dense taxon sampling representing the whole phenotypic diversity and distribution range of Salvia confirmed that the genus is polyphyletic. Salvia species fall in 4 distinct clades, although all of them, except Clade IV, also include non-Salvia genera. A taxonomic revision is thus urgently needed with two different approaches that have to be considered: (1) to include the 5 morphologically distinct …

Random Amplified Polymorphic Dna As A Method To Screen Metal-Tolerant Barley (Hordeum Vulgare L.) Genotypes, Süleyman Cenkci̇, Nevra Doğan
Turkish Journal of Botany
Differentially responding genotypes are comparatively used to understand mechanisms of metal tolerance in higher plants. The feasibility of the random amplified polymorphic DNA (RAPD) technique was evaluated to screen for metal tolerance within plants as an alternative to physiological growth parameters. Eight hydroponically grown barley (Hordeum vulgare L.) genotypes were exposed to different concentrations (75, 150, and 225 μM) of cadmium and copper for 7 days and subsequently harvested for determining metal tolerance indices by comparing dry biomass to genomic template stabilities based on their RAPD profiles. Dose- and genotype-dependent reductions in dry weights of shoot and root tissues were …

Generating Salt-Tolerant Nicotiana Tabacum And Identification Of Stress-Responsive Mirnas In Transgenics, Ferhunde Aysi̇n, Ayşe Eli̇f Erson Bensan, Füsun Eyi̇doğan, Hüseyi̇n Avni̇ Öktem
Turkish Journal of Botany
Identification of vacuolar Na+/H+ antiporters facilitates the basis of salt stress tolerance mechanisms. Na+ accumulation into the vacuole is crucial for the avoidance of cytoplasmic Na+ toxicity. In this study, we show that the introduction of AtNHX1 into tobacco generated more tolerant plants when compared to wild-type plants. Transgenic tobacco plants exhibited higher germination rates in the presence of increasing salt concentrations compared to wild-type plants. In addition, proline levels were higher under salt stress conditions in both the shoots and the roots of transgenics compared to wild-type plants. Increase in malondialdehyde production during lipid peroxidation by salinity was lower …

Effects Of Cobalt Nanoparticles On Artemisinin Production And Gene Expression In Artemisia Annua, Bita Ghasemi, Ramin Hosseini, Fatemeh Dehghan Nayeri
Turkish Journal of Botany
The low artemisinin content in Artemisia has caused this compound to be expensive among medicines. Several attempts have been made to increase its production by altering the expression of different genes. However, no approach has been cost-effective. In this study, the expression levels of SQS and DBR2 genes were quantified by qRT-PCR, and artemisinin content was measured by HPLC in Artemisia annua cell suspension culture under nano cobalt particles elicitation. For this purpose, nano cobalt particles were used in 0.25, 2.5, and 5 mg L-1 concentrations and samples were collected after 8, 24, 48, and 72 h. The highest artemisinin …

Relationships And Generic Delimitation Of Eurasian Genera Of The Subtribe Asterinae (Astereae, Asteraceae) Using Molecular Phylogeny Of Its, Elena Korolyuk, Alexey Makunin, Tatiana Matveeva
Turkish Journal of Botany
The subtribe Asterinae (Astereae, Asteraceae) includes highly variable, often polyploid species. Recent findings based on molecular methods led to revision of its volume. However, most of these studies lacked species from Eurasia, where a lot of previous taxonomic treatments of the subtribe exist. In this study we used molecular phylogenetics methods with internal transcribed spacer (ITS) as a marker to resolve evolutionary relations between representatives of the subtribe Asterinae from Siberia, Kazakhstan, and the European part of Russia. Our reconstruction revealed that a clade including all Asterinae species is paraphyletic. Inside this clade, there are species with unresolved basal positions, …

Development Of Aflp Markers Associated With Zucchini Yellow Mosaicvirus Resistance In Cucumber (Cucumis Sativus L.), Hasan Özgür Şiğva, Ahmet Fi̇kret Firat, Gülden Hazarhun, Ahmet İpek
Turkish Journal of Botany
Zucchini yellow mosaic virus (ZYMV) is one of the most important pathogens that cause significant yield losses in many cucurbit crops including cucumber (Cucumis sativus L). ZYMV resistance in cucumber is inherited by a single recessive gene. The purpose of this study was to identify molecular markers linked to the gene conferring ZYMV resistance in cucumber. We developed a population of 188 F2 plants derived from inbred cucumber lines. Individual F2 plants were self-pollinated to generate F3 populations. Ten randomly selected plants from each F3 population were tested for ZYMV resistance. We used a bulk segregant analysis method to identify …

Overexpression Of A Soybean Expansin Gene, Gmexp1, Improvesdrought Tolerance In Transgenic Tobacco, Thanh Son Lo, Hoang Duc Le, Vu Thanh Thanh Nguyen, Hoang Ha Chu, Van Son Le, Hoang Mau Chu
Turkish Journal of Botany
The EXP1 gene encodes expansin, which has the ability to loosen the plant cell wall. The soybean expansin gene GmEXP1 is activated specifically during the root elongation process, and thus it plays important roles in root development. During the drought period, changes in pressure within the cell and the fast development of the root allow plants to collect water from deep soil, which in turn helps plants grow and develop. In this study, we have successfully cloned and generated a GmEXP1 construct expressing recombinant expansin protein in tobacco plants. GmEXP1 is expressed in transgenic tobacco plants and passed on to …
Aquaporins As Targets For Stress Tolerance In Plants: Genomic Complexity And Perspectives, Wonkeun John Park, Benjamin Todd Campbell
Aquaporins As Targets For Stress Tolerance In Plants: Genomic Complexity And Perspectives, Wonkeun John Park, Benjamin Todd Campbell
Turkish Journal of Botany
As a multigene family, plant aquaporins function not only in water transport but also in transport of small elements that are important for vegetative and reproductive growth of plants. Increasing evidence exhibits the relevance of aquaporins to tolerance against abiotic and biotic stresses such as drought, nutrient deficiency, and herbivore attack. With the accumulation of crop genome sequencing, it is suggested that several aquaporin genes are conserved in subchromosomal locations as tandem duplicated members. In this review, we will discuss the compelling nature of aquaporins as multifunctional transport channels that are often encoded in clustered regions of genomes and relevant …

Systematic Placement Of The Turkish Endemic Genus Ekimia (Apiaceae) Based On Morphological And Molecular Data, Dmitry Lyskov, Galina Degtjareva, Tahir Samigullin, Michael Pimenov
Turkish Journal of Botany
The systematic position of the monotypic genus Ekimia H.Duman and M.F.Watson (Apiaceae), a narrow endemic to Turkey, was evaluated on the basis of morphological data and nrDNA ITS sequences. Ekimia bornmuelleri (Hub.-Mor. And Reese) H.Duman and M.F.Watson was initially described in Prangos Lindl. Due to the unique fruit morphology uncommon for this genus it was later shifted to an independent genus. In the Bayesian and most parsimonious trees, E. Bornmuelleri is sister to Laserpitium petrophilum Boiss. And Heldr and Laserpitium glaucum Post within the Daucinae clade. This result is consistent with its morphology: the presence of the primary and secondary …

Molecular Phylogeny And Biogeography Of The South American Genus Metrodorea (Rutaceae), Pedro Dias, Renata Giassi Udulutsch, Jose Rubens Pirani
Turkish Journal of Botany
The genus Metrodorea (Rutaceae), with 6 species, has representatives that vary from small shrubs to tall trees, and its leaf sheaths make it unique in the family; it is restricted to South America, with species diversity concentrated in Brazil. In this study, we applied Bayesian and parsimony methods to infer the molecular phylogeny of Metrodorea, and cladistic biogeographic methods to propose an area cladogram for the genus. All results supported Metrodorea as monophyletic, and the presence of leaf sheaths is reinforced as its key diagnostic feature. Bayesian and parsimony trees were identical for relationships within Metrodorea. The crown clade (M. …
