Determination Of Hydrogen Peroxide With An Enzymeless Amperometric Sensor Based On Poly(Vinylferrocene)-Supported Ag Nanoparticles, Mutlu Sönmez Çelebi̇, Kübra Öztürk, Mehmet Dumangöz, Fi̇li̇z Kuralay
Turkish Journal of Chemistry
An enzymeless electrochemical sensor for detection of low amounts of H$_{2}$O$_{2}$ with the aid of Ag nanoparticles supported on conducting poly(vinylferrocene) (PVF) film was developed. Experimental results revealed that contribution of Ag nanoparticles led to remarkable improvement by means of reduction potential and reduction current. Influence of experimental parameters (i.e. polymeric film thickness, concentration of Ag precursor, immersion time in precursor solution, reduction time, and reduction potential) were investigated. The Ag/PVF-modified electrode system was characterized physically by scanning electron microscopy. The results revealed that the sensor developed was easy-to-prepare, economic, selective, and sensitive, with a fast response time of 3 …

n plant development and abiotic stress response processes. In this study, we examined the role of a HSP, the Medicago sativa DnaJ-like protein (MsDJLP), in alfalfa and its potential application for the development of abiotic stress tolerance in plants. We found that expression of the MsDJLP gene was induced by chilling (4 °C) and heat (42 °C), but not by cadmium (500 µM) or arsenic (500 µM) stresses. We then cloned the MsDJLP gene downstream of the strong constitutive CaMV 35S promoter and transformed it into tobacco plants. Ectopic expression of MsDJLP conferred enhanced tolerance to both chilling and heat …

Turkish Journal of Biology
Moringa oleifera Lam. and Centella asiatica (L.) Urb. leaves have been previously reported to exhibit antioxidant activity. The objective of the present study is to determine the in vitro antioxidant activity of the combined extracts of M. oleifera and C. asiatica (TGT-PRIMAAGE) and its effect on hydrogen peroxide (H$_{2}$O$_{2}$)-induced oxidative stress in human dermal fibroblasts. TGT-PRIMAAGE acted on the mechanism of hydrogen transfer as it showed scavenging activity in the DPPH assay. This is due to the presence of phenolics and flavonoids in TGT-PRIMAAGE. TGT-PRIMAAGE effectively reduced cellular generation of reactive oxygen species induced by H$_{2}$O$_{2}$. The activities of superoxide …

Polyphosphate polymer (polyP) plays a very important role in every living cell. Synthesis of this linear polymer of phosphate (P$_{i}$) residues is catalyzed by the polyphosphate kinase (PPK) enzyme. It was shown that high levels of intracellular polyphosphate stimulated endotoxin production by Bacillus thuringiensis subsp. israelensis (Bti). In this study, proteomic analysis of the wild-type and two mutant strains, overexpressing the ppk gene (Bti pHTppk) and without the ppk gene (Bti Δppk), were used to clarify the relation between polyP and endotoxin production. Intracellular proteins were separated by two-dimensional gel electrophoresis; 41 spots of interest (proteins differentially expressed) were obtained …

Deep Sequencing Reveals Two Jurkat Subpopulations With Distinct Mirna Profiles During Camptothecin-Induced Apoptosis, İpek Erdoğan, Mehmet İlyas Coşacak, Ayten Nalbant Aldanmaz, Bünyami̇n Akgül
Turkish Journal of Biology
MicroRNAs (miRNAs) are small noncoding RNAs of about 19-25 nt that regulate gene expression posttranscriptionally under various cellular conditions, including apoptosis. The miRNAs involved in modulation of apoptotic events in T cells are partially known. However, heterogeneity associated with cell lines makes it difficult to interpret gene expression signatures, especially in cancer-related cell lines. Treatment of the Jurkat T-cell leukemia cell line with the universal apoptotic drug, camptothecin, resulted in identification of two Jurkat subpopulations: one that is sensitive to camptothecin and another that is rather intrinsically resistant. We sorted apoptotic Jurkat cells from nonapoptotic ones prior to profiling miRNAs …

Despite high vaccination coverage after introduction of whole cell (wP) and acellular pertussis (aP) vaccines, pertussis resurgence has been reported in many countries. aP vaccines are commonly preferred due to side effects of wP vaccines and formulated with aluminum hydroxide (Alum), which is not an effective adjuvant to eliminate Bordetella pertussis. Low efficiency of current aP vaccines is thought to be the main reason for the resurgence for which newer generation aP vaccines are needed. In the present study, immunogenicity and protective efficacy of outer membrane protein Q (OmpQ) and a putative lipoprotein (Lpp) from B. pertussis were investigated in …

Crm1 Knockdown By Specific Small Interfering Rna Reduces Cell Proliferation And Induces Apoptosis In Head And Neck Cancer Cell Lines, Si̇bel Özdaş, Tali̇h Özdaş
Turkish Journal of Biology
Head and neck squamous cell carcinoma (HNSCC) is the most common and most aggressive type of head and neck cancer. Current approaches for the treatment of HNSCC are not sufficient to increase the patient survival or to reduce the high recurrence rate. Consequently, there is a need to explore the molecular characteristics of this cancer in order to discover potential therapeutic target molecules. The overexpression of chromosome region maintenance 1 (Crm1), responsible for the transport of different classes of macromolecules from the nuclear membrane to the cytoplasm, in various cancer cells has made it an attractive target molecule in cancer …

Effects Of Gold Nanorods Modified With Antiepidermal Growth Factor Receptor Monoclonal Antibody On Laryngeal Cancer Cells, Fangzhou Liu, Yan Zhu, Yichuan Qian, Yanbin Zhao, Xiaotong Zhao, Jia Zhang, Yu Zhang, Yuan Zhang
Turkish Journal of Biology
We aimed to evaluate the photothermal effects of gold nanorods (GNRs) modified with antiepidermal growth factor receptor (EGFR) monoclonal antibody (mAb) on laryngeal cancer cells. EGFR protein expressions in HEP-2 cells, and normal laryngeal epithelial and laryngeal cancer tissues were detected by western blot. The cytotoxicity against HEP-2 and BEAS-2B cells was tested by MTT assay, and the photothermal effects were assessed by near-infrared (NIR) irradiation. The apoptosis of HEP-2 cells was detected by flow cytometry. EGFR expression in laryngeal cancer tissues was significantly higher than that in normal tissues (P < 0.05). The inhibitory effects of GNRs and anti-EGFR mAb/GNRs on the apoptosis of HEP-2 and BEAS-2B cells were enhanced with increasing dose. Anti- EGFR mAb/GNRs were more cytotoxic to HEP-2 cells and less cytotoxic to BEAS-2B cells than GNRs. NIR irradiation inhibited cell proliferation, which was enhanced with rising power, accompanied by continuously dropping survival rate. The apoptosis rate of the anti-EGFR mAb/GNRs group was significantly higher than that of the GNRs group, and the apoptosis rate of the irradiation + anti- EGFR mAb/GNRs group also significantly exceeded that of the irradiation + GNRs group (P < 0.05). Anti-EGFR mAb/GNRs promoted HEP-2 cell apoptosis more evidently than GNRs did. Functional modification of GNRs augmented the targeted specificity to cancer cells, biocompatibility, and stability. Anti-EGFR mAb/GNRs have great potential in biomedical fields.

In the present study we demonstrated that composite PPHMDS/DND coatings with elastic moduli close to those of mature bone tissue (0.2-2.8 GPa) stimulated growth and osteogenic differentiation of human adipose-derived mesenchymal stem cells (hAD- MSCs). Composite coatings were prepared by a method of plasma polymerization (PP) where detonation nanodiamond (DND) particles in different amounts (0.1, 0.5, and 1 mg/mL) were added to hexamethyldisiloxane (HMDS) before plasma deposition. This method allows variation only in the reduced elastic modulus (Er') with increase in the particle concentration, while the other surface properties, including surface wettability and topography, did not change. The response of …

In this study, transformation of BrCHS var 2 into B. rotunda cell suspension culture, followed by chalcone synthase enzymatic assay and HPLC analysis was conducted to investigate whether the substrate specificity for BrCHS var 2 is either cinnamoyl-CoA or p-coumaroyl-CoA. The HPLC profile showed an increase in the amount of pinocembrin chalcone when cinnamoyl-CoA and malonyl-CoA were added but not p-coumaroyl-CoA. Molecular docking was performed to explore the binding of cinnamoyl-CoA and p-coumaroyl-CoA to BrCHS var 2 receptor and the docking results showed that cinnamoyl-CoA formed numerous hydrogen bonds and more negative docked energy than p-coumaroyl-CoA. Cinnamoyl-CoA showed good interactions …

Biological Influence Of Cry1ab Gene Insertion On The Endophytic Bacteria Community In Transgenic Rice, Xu Wang, Mengyu Cai, Yu Zhou
Turkish Journal of Biology
The commercial release of genetically modified (GMO) rice for insect control in China is a subject of debate. Although a series of studies have focused on the safety evaluation of the agroecosystem, the endophytes of transgenic rice are rarely considered. Here, the influence of endophyte populations and communities was investigated and compared for transgenic and nontransgenic rice. Population-level investigation suggested that cry1Ab gene insertion influenced to a varying degree the rice endophytes at the seedling stage, but a significant difference was only observed in leaves of Bt22 (Zhejiang22 transgenic rice) between the GMO and wild-type rice. Community-level analysis using the …

Studies conducted in the past decade have reported nucleostemin (NS) as a nucleolar protein that has a role in self-renewal and cell cycle regulation in cancer/stem cells, but is absent in differentiated cells. The localization and expression patterns of NS have always been disputed, as reports indicate its varied levels among tissues and cells. This study evaluates the expression and localization pattern of NS in normal cells, cancer cell lines, and stem cells. Our findings revealed that the expression of NS was high in cancers originating from the skin and liver compared to the normal cell lines. NS knockdown affects …

Ikbke Inhibits Tsc1 To Activate The Mtor/S6k Pathway For Oncogenic Transformation, Serkan İsmai̇l Göktuna
Turkish Journal of Biology
IKBKE (IKK?) has emerged as a key modulator of multiple substrates, controlling oncogenic pathways in various malignancies. mTOR signaling, required for cellular growth, proliferation, and vascular angiogenesis in cancer, is potentially one of the pathways regulated by IKK?. Upon activation by various stimuli, PI3K/AKT or similar effectors can relieve the inhibitory effect of the TSC1/TSC2 complex through their phosphorylation to favor mTOR/S6K activation in the downstream. Therefore, any activity that interferes with PI3K/AKT or their downstream targets, such as TSC1/2 or GSK3?/ß, may activate the mTOR/S6K pathway for oncogenic transformation in normal cells. Previous studies have shown that PI3K/AKT can …
