Genetic Integrity Assessment Of Cryopreserved Tomato (Lycopersicon Esculentum Mill.) Genotypes, Ana Coste, Dana Suteu, Ioan Bacila, Constantin Deliu, Sergiu Valimareanu, Adela Halmagyi

Turkish Journal of Biology

Shoot tips of five tomato (Lycopersicon esculentum Mill.) genotypes were successfully cryopreserved by droplet vitrification. Recovered plants were studied for genetic stability by two different assays: amplified fragment length polymorphism (AFLP) using 4 primer combinations and sequencing of lycopene $\beta$-cyclase gene (LCY-B) from leaves. The highest shoot regeneration after cryopreservation was 70% (Pontica) following 24 h osmoprotection in 0.5 M sucrose and 30 min dehydration in plant vitrification solution 2 (PVS2). The data inferred from AFLP showed no genetic dissimilarities between in vitro regenerants derived from cryopreserved tissues compared with the noncryopreserved controls. Although a single nucleotide polymorphism, a G→T …

A survey of both ectomycorrhizal (ECM) root tips and fruiting bodies of ECM fungi was carried out during a four-year period (2009-2012) in two similar forest stands in central Italy, previously investigated in detail being habitats of the Italian white truffle Tuber magnatum Pico. This research is one of the few conducted in mixed forest stands and, for the first time, it took into account the cryptic fungi (corticiaceous, hypogeous, and sclerotia). This survey led to an exhaustive description of the ECM community by integrating all the 197 taxa recorded (147 species as fruiting bodies, 65 as ectomycorrhizas, and 15 …

Random Amplified Polymorphic Dna As A Method To Screen Metal-Tolerant Barley (Hordeum Vulgare L.) Genotypes, Süleyman Cenkci̇, Nevra Doğan

Turkish Journal of Botany

Differentially responding genotypes are comparatively used to understand mechanisms of metal tolerance in higher plants. The feasibility of the random amplified polymorphic DNA (RAPD) technique was evaluated to screen for metal tolerance within plants as an alternative to physiological growth parameters. Eight hydroponically grown barley (Hordeum vulgare L.) genotypes were exposed to different concentrations (75, 150, and 225 μM) of cadmium and copper for 7 days and subsequently harvested for determining metal tolerance indices by comparing dry biomass to genomic template stabilities based on their RAPD profiles. Dose- and genotype-dependent reductions in dry weights of shoot and root tissues were …

Aquaporins As Targets For Stress Tolerance In Plants: Genomic Complexity And Perspectives, Wonkeun John Park, Benjamin Todd Campbell Jan 2015

Aquaporins As Targets For Stress Tolerance In Plants: Genomic Complexity And Perspectives, Wonkeun John Park, Benjamin Todd Campbell

Turkish Journal of Botany

As a multigene family, plant aquaporins function not only in water transport but also in transport of small elements that are important for vegetative and reproductive growth of plants. Increasing evidence exhibits the relevance of aquaporins to tolerance against abiotic and biotic stresses such as drought, nutrient deficiency, and herbivore attack. With the accumulation of crop genome sequencing, it is suggested that several aquaporin genes are conserved in subchromosomal locations as tandem duplicated members. In this review, we will discuss the compelling nature of aquaporins as multifunctional transport channels that are often encoded in clustered regions of genomes and relevant …
