Our earlier studies found that in vitro estradiol modulates mitochondrial Ca2+ transport in discrete brain regions. The present study examined the role of estradiol receptors (ERs) in estradiol-induced inhibition of Ca^{2+} efflux from synaptosomal mitochondria isolated from rat caudate nuclei and brain stems. Radioactively labeled CaCl_2 (0.6?0.75 µCi ^45CaCl_{2}) was used for Ca^{2+} transport monitoring. The results revealed that in the presence of ER antagonist 7\alpha,17ß-[9[(4,4,5,5,5-pentafluoropentyl)sulfinyl]nonyl]estra-1,3,5(10)-triene-3,17-diol (ICI 182,780) (1 µmol/L), the inhibitory effect of estradiol on mitochondrial Ca^{2+} efflux was more than 60% decreased, suggesting the involvement of ER in this mode of estradiol neuromodulatory action. Sine oculis homeobox 1 (Six1), a member of the Six homeoproteins, plays an important role in skeletal myogenesis and the specification of myofiber diversity. In this study, in order to scale up the production of recombinant porcine Six1 (pSix1), a pET-30a(+)-pSix1 plasmid was constructed and transformed into Escherichia coli BL21 (DE3). The recombinant pSix1 could be induced for efficient expression with 2 mM IPTG for 2 h at 30 °C, yielding approximately 4.6 mg/L. Genetic Variability, Heritability, And Genetic Advance In Strawberry(Fragaria × Ananassa Duch.), Pranav Kumar Mishra, Ram Badan Ram, Neeraj Kumar

Turkish Journal of Agriculture and Forestry

Strawberry (Fragaria × ananassa Duch.) genotypes were evaluated to study genetic variation and the relationship between yield and its component using a randomized complete block design during 2013/14. The results showed significant variance among genotypes of all traits. The phenotypic coefficient of variation (PCV) for all the characters was slightly higher than genotypic coefficient of variation (GCV), which signified the presence of environmental influence to some degree in the phenotypic expression of characters. Lentil, which belongs to the family Leguminosae (Fabaceae), is a diploid (2n = 2x = 14 chromosomes) self-pollinating crop with a genome size of 4063 Mbp. Because of its nutritional importance and role in the fixation of nitrogen from the atmosphere, lentil is a widely used crop species in molecular genetic studies. The current study aimed to verify the preferred guava fruit ripening stage for egg laying by Anastrepha fraterculus (Diptera: Tephritidae) and to determine the influence of peel coloration of the fruits of 2 cultivars on infestation. The cultivars used were Paluma and Século XXI. The infestation level was evaluated in cages, considering 2 conditions: multiple-choice and nonchoice tests. The infestation levels of A. fraterculus differed between the green and the green-ripe stages in the multiple-choice test. Empirical standard weight equations and length-weight and length-length relationships were estimated for Barbus pergamonensis, Capoeta bergamae, and Ladigesocypris irideus, three endemic cyprinid species of Turkey that are still not widely studied. The distribution of crustaceans in 3 transects (İskenderun, Mersin, and Anamur bays, including depths of 10, 25, 50, 75, 100, and 200 m) located within Turkish coastal borders of the Cilician shelf was studied between 2005 and 2007 to show the association with habitat types, eutrophication level, and depth-dependent environmental parameters in representative months of each season (winter, spring, summer, and autumn). For the first time, morphological polymorphism among the anterior and posterior sets of primary teeth of the periphallic organ was observed among natural populations of Drosophila. Natural and laboratory populations of 3 different species (D. Ananassae, D. Bipectinata, and D. Malerkotliana) belonging to the ananassae subgroup were used to study the intra- and interspecific variations in the number of teeth of the anterior and posterior sets of the periphallic organ. Length-weight relationships, distribution of length and weight groups, sex ratio, and length compositions of the crayfish Astacus leptodactylus (Esch., 1823) population in Lake İznik (Bursa), along with some reproductive-period characteristics (spawning periods, pleopod egg numbers, and egg incubation periods), were investigated between February 2010 and May 2011. For this purpose, 1897 freshwater crayfish samples (896 female, 1001 male) were collected, using fyke nets of 34-mm nominal bar length. Roost characteristics of Indian flying fox Pteropus giganteus at the Jinnah (n = 1052) and Lalazar (n = 40) gardens in Lahore were observed for a period of 1 year to find roost preferences in these bats. The Jinnah garden harbors 4119 trees belonging to 46 families, 103 genera, and 132 species, of which 44 trees belonging to 17 families, 19 genera, and 21 species served as bat roosts. The seasonality of abundance and live/dead compositions of copepods was studied in the northeastern Mediterranean Sea. Zooplankton, chlorophyll-a, and PO4 sampling was performed on a monthly basis from March 2006 to February 2007 at both a coastal station and an open water station. At the coastal station, high phytoplankton biomass was driven by PO4 input from the Lamas River. On annual average, copepod abundance was 53,075 and 140,227 ind. m-2 at the coastal and open water stations, respectively. In total, 626 individuals of Alburnus mossulensis and 586 individuals of Acanthobrama marmid were collected monthly from Karakaya Dam Lake on the upper Euphrates River. The gonadosomatic index, somatic condition, oocyte size, and fecundity were calculated on a monthly basis. Reading of scales indicated that the maximum age was 4+ years for A. mossulensis and A. marmid. The estimated length-weight relationships were W = 0.206 × FL^{2.065} for females and W = 0.119 × FL^{2.138} for males in A. mossulensis, and W = 0.029 × FL^{2.678} for females and W = 0.030 × FL^{2.631} for males in A. marmid. Life history characteristics (e.g., longevity, age at maturity, body size, and body mass) of the green toad, Bufotes variabilis, were investigated from 2 populations inhabiting different altitudes (Çankırı, 960 m a.s.l., n = 57; Adana, 13 m a.s.l., n = 52) in Turkey. Age was determined using skeletochronology. The oldest individual was 11 years old (range: 4-11 years) in the highland population, while the oldest individual was 6 years old in the lowland population (range: 2-6 years). A porcine fat mass and obesity-associated gene (pFTO) was cloned into the expression vector pET30a(+) and heterologously expressed in Escherichia coli. The effects of isopropyl ß-D-thiogalactopyranoside (IPTG) concentration and induction time on the expression of recombinant pFTO were assessed. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that the molecular mass of the recombinant pFTO protein was about 56 kDa, and the recombinant protein could be induced efficiently in E. coli BL21 by the addition of 0.75 mM IPTG for 4 h at 30 °C. This work was conducted in the context of postmarketing biosafety assessment of genetically modified products. It presents a systematic approach based on a chronic toxicity study on Wistar albino rats, with a range of combined parameters including biochemical, histopathological, and cytogenetic to evaluate the negative impact of a genetically modified (GM) diet on animal health. Histopathological and biochemical analysis procedures were performed in the liver, kidney, and testis. Cytogenetic analysis was evaluated in germ cells and the liver. Our research sought to investigate whether host plant quality affects first-instar grub development, weight gain, or mortality. Ten plant taxa of the families Polygonaceae, Brassicaceae, Asteraceae, Fabaceae, and Pinaceae were tested. The quality of each plant species was estimated based on the root content of phenols, condensed tannins, soluble sugars, starch, nitrogen, and carbon. This study investigated the possibility of determining iron deficiency in orchard leaves using visible/near-infrared methods. Sampling was performed in 24 different healthy orchards (apple, cherry, and peach trees) grouped depending on the severity of iron deficiency. The study was conducted on a total of 120 plant samples including 40 apple, 40 cherry, and 40 peach trees. Spectral reflectance of leaves was measured with a FieldSpec HandHeld spectroradiometer (ASD Inc.) using a plant probe and a leaf clip. Land degradation and declining soil fertility challenge the productivity of forest plantations. Changes in soil properties could act as indicators for monitoring forest plantation management and productivity. This study highlights changes in soil properties and stand productivity over time (1, 3, and 5 years) in an Acacia mangium plantation in Bintulu, Sarawak, Malaysia. Soil pH, total N concentration, and available P were found to decline significantly with stand age.
