In this study, lipid peroxidation, intracellular, membrane and extracellular antioxidant activities in Turkish Angora goats (Genotype I) and American x Turkish crossbred Angora goats (Genotype II) were investigated, in order to show the effects of genotype on the antioxidative metabolism. Plasma malonlydialdehyde (MDA) levels of Angora goats in different genotype were analyzed to show the effects of free radicals. Glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and catalase (CAT) activities in hemolysates of erythrocytes were analyzed as intracellular antioxidants, \beta-carotene in serum as membrane antioxidants and ascorbic acid in plasma, albumin, total bilirubin, glucose, uric acid and ceruloplasmine in serum as …

Detection Of Antibodies Against Salmonella Enteritidis In Chicken Serum And Egg Yolk With Elisa, Gülay Altay, Hakan Yardimci

A total of 473 sera from hens of different age groups and different breeds and a total of 430 eggs were collected from 11 breeders and egg producers in the districts of Adapazarı, Afyon, Ankara and Konya. Lipopolysaccharide (LPS), heat-extract (HE) and flagella antigens were tested in the standardizing ELISA technique. Flagella antigen was found useful in the screening of field samples. Of the 48 (10.2%) rapid plate agglutination positive sera, 7 were negative in ELISA. One hundred and seventy-three (36.6%) of the 473 sera and 76 (17.7%) of the 430 egg yolks were found to be positive with ELISA. …

Photoinactivation Of Buffalo Liver And Kidney Tissue Arginases And Its Kinetic Properties, Necmi̇ Özdemi̇r, Mehtap Özçeli̇k

Exposure of buffalo liver and kidney tissue arginases to a 150 w light source from a distance of 8-10 cm, in the presence of 0.05mM methylene blue, caused significant inhibition of arginase activity. It was determined that this inhibition occurred differently in liver and kidney tissues. Arginase activity was spectrophotometrically measured using the Thiosemicarbazide diacetyl-monoxime urea (TDMU) method. Two studies were conducted to evaluate if Fertirelin acetate (an analogue of GnRH) and Gentamycin sulphate would improve pregnancy rate in healthy cows or in nonpregnant cows although they were inseminated three times or more. The first investigation was carried out in sixty-eight healthy cows. Cows were alloted randomly to one of two treatment groups or one of two control groups when found to be in oestrus. The first treatment group was treated with 100 micrograms of Fertirelin acetate, while the first control grup was given saline (2 ml) intramuscularly immediately after artificial insemination. This investigation was carried out to demonstrate some production traits and egg quality characteristics of Japanese quails (Coturnix coturnix japonica). One hundred female quails were used in the study. The experiment was carried out for six months. Egg production, egg weight, feed consumption and some egg quality characteristics were taken into consideration. Egg quality characteristics were measured at 8-24 weeks of age. Average egg production (% HD) at 8, 12, 16, 20 and 24 weeks of age was 20.5, 33.8, 63.7, 73.8 and 72.5 respectively. In this study, how to obtain high quality sugar beet pulp silage was investigated. Sugar beet pulp was mixed with wheat straw or grass hay in order to increase the level of dry matter to 20, 25 and 30%, and 5% molasses and different levels of urea were added (0, 1, 1.5 and 2% for wheat straw containing groups; 0, 0.5 and 1% for grass hay containing groups). Enzyme based biosensors are highly selective devices which rely on the specific binding of the target analyte (the substrate) to the active-site regions of the enzyme. The enzymatic reaction between mushroom tissue polyphenol oxidase and its substrate phenol is coupled with the use of potassium ferrocyanide (K_{4}Fe(CN)_{6}) as the mediator in ammonia buffer solution (pH 8.80). New Photochemical And Electrochemical Methods For The Degradation Of Pesticides In Aqueous Media. Environmental Applications, J. J. Aaron, Mehmet A. Oturan

The development of new electrochemical and photochemical methods for the decontamination of natural water containing significant concentrations of aromatic pesticides is described. The electrochemical method is based on the electro-Fenton process, i.e. the simultaneous reduction of O_{2} and Fe^{3+} ions. Hydroxyl (OH^{.}) radicals are electrosynthesized in aqueous solutions, followed by complete mineralization of the initial pollutants. The photochemical methods involve either a direct photodegradation reaction of pesticides by UV light, or indirect photodegradation processes including H_{2}O_{2} photolysis or photo-Fenton reagents (H_{2}O_{2}/ Fe^{3+} ). Considering the whole landscape of ecosystems is the focus of landscape management. This paper examines the evolutionary process of forest management and compares and contrasts the landscape management planning concept to its predecessor, the conventional forest resource management approach. The study was conducted at Alata Horticultural Research institute between 1994 and 1996. The aim of the study was to obtain a strong colony by way of supersedure and empty frames, feeding with saccharose syrup and encouraging egg laying of queen bees at the end of summer. According to the results, there were significant differences among treatments in terms of the development of colony population, brood rearing activity and honey yield. Treatments had no significant effect on wax building activity and swarming tendency.

This investigation was carried out to determine the effect of amitraz poisoning on the the formation of methemoglobin and osmotic fragility of erytrocytes. in this study, 12 guinea-pigs, weighing 250-300 gr were used. They were equally divided into two groups, control and experimental. The animals in the experimental group were intraperitoneally injected with amitraz at a dose of 1 mg/kg body weight. Trichloroethylene (TE) is an environmental toxic solvent hazardous to human and domestic animals and well known in the industrial sector. The purpose of this study was to determine whether oral TE plays a role in lipid oxidation and tissue damage. Fourteen dogs were treated with an oral toxic dose of 0.5 ml/kg T.E. This study was carried out to investigate the prevalance of leptospirosis in the urine of cattle slaughtered in three major abattoirs in the east of Turkey. A polymerase chain reaction (PCR) based on a pair of genus-specific primers was used to detect leptospiral DNA in the urine samples of 473 cattle, 284 of which were from Elazig, 112 from Malatya and 77 from Diyarbakir. The detection limit of the method was determined to be approximately five bacteria per ml of urine. Development Of An Enzyme Immunoassay For The Determination Of Ovine Growth Hormone In Plasma, Behi̇ç Serpek, Seyfullah Hali̇loğlu

A competitive enzyme immunoassay for the determination of ovine growth hormone (oGH) was developed. The assay is based on (anti-rabbit IgG) goat IgG-coated microtitreplates, anti-oGH from rabbits, biotinyl-oGH and streptavidin-horseradish peroxidase. The assay is done directly with 40 ml of plasma and the calibration graph (90% relative binding at 0.38 ± 0.02 ng ml -1 and 50% relative binding at 3.80 ± 0.18 ng ml -1 ) is prepared in plasma without measurable endogenous oGH. Recovery in different plasma samples with added oGH amounted to 96.78 - 100.03 %, and all variabilities were < 10.44. The assay shows no significant binding of oFSH, oLH, oTSH and oPRL. The assay detects high and low plasma oGH levels within the physiological variation as well as changes in plasma oGH after stimulation with growth hormone releasing factor. Hence, it offers a reliable alternative radioimmunoassay. However, a reference method providing evidence of the identity of exogenous oGH will be required for proving illegal treatment with oGH and for forensic purposes.

One hundred pneumonic lungs from calves were examined by an immunoper oxidase technique using avidin-biotin- per oxidase complex method in formalin-fixed, paraffin-embedded sections for Haemophilus somnus. In the histological examination of tissue sections, strong positive reactions against H. somnus were detected in 14% of the lungs. H. somnus antigens were observed at the surface and/or within the epithelial cells, macrophages, necrotic alveolar walls, alveolar -bronchial/br onchiolar exudates, fibrins, distended interlobular septa and perivascular areas. In addition, dense antigen accumulations were detected in the necrotic macrophages and neutrophils.

This research was carried out on mirror carps ( Cyprinus carpio, L.,1758) taken from Seyhan Dam Lake and Seyhan River between January and October 1996 to determine some blood parameters. Erythrocyte, leucocyte count, leucocyte cell formulas, total serum protein and serum glucose values of mirror Carps ( Cyprinus carpio, L.,1758 ) caught from Seyhan Dam Lake and Seyhan River were also compared statistically. Different values were recorded in erythrocyte and serum protein amounts for both environments (P0.05).

This study was conducted in pregnant and aborted sheep to determine Toxoplasma gondiiantibodies. Blood samples were collected from a total of 154 sheep from 22 different herds during the period December 1996-January 1997 in Elazig and vicinity. Of these 154 sheep, 56 were pregnant, 57 aborted in the previous year and the remaining 41 sheep aborted within the period of the study. Sera obtained properly from these blood samples were examined by Sabine-Feldman (SF) test.
