This dissertation investigates the ruminal microbiome of American plains bison (Bison bison) to uncover the metabolic activities that enable efficient utilization of plant fiber, with a particular focus on bacterial composition and functional capabilities in response to forage quality. Using both 16S rRNA gene sequencing and metagenomic approaches, this research provides a comprehensive view of microbial diversity and metabolic potential in the rumen of bison, offering insights into their ecological resilience and efficiency in nutrient extraction. In the aquaculture industry, plant-based protein sources have emerged as an economically and environmentally sustainable replacement for traditional animal protein sources. Yet, due to reduced digestibility, the presence of anti-nutritional factors, as well as an increased risk of inflammation of the digestive system, the inclusion of plant-based protein ingredients must be limited, especially in the diets of carnivorous fishes. This study presents the development of a collagen-based bioink for 3D printing tendon-to-bone tissue grafts, specifically designed for rotator cuff repair applications. Three different bioink formulations were developed: (i) 5.1% (w/v) collagen I, (ii) 5% (w/v) collagen I and collagen II, and (iii) 4.5% (w/v) collagen I combined with β-TCP. The bioink containing collagen I and collagen II was prepared by mixing the two components in a 1:1 ratio, while the collagen I plus β-TCP formulation was made by incorporating 20 wt % β-TCP into the collagen I solution. The Prairie Pothole Region (PRR) is a landscape defined by a dynamic surface water system, with wetlands, lakes, and rivers playing critical roles in supporting biodiversity, regulating floods, and maintaining hydrological balance. This study conducts a time-series analysis of surface water change in the PPR from 1984 to 2024, using remote sensing tools including Google Earth Engine, ArcGIS Pro, and ENVI. 41 annual surface water presence rasters were compiled into a cumulative dataset, allowing for both year-to-year comparisons and broader trend analysis. Results indicate that the presence of surface water in the PPR has not followed a simple linear trend. …

Beginning with a literature review of the estrous cycle and the endocrinology driving it. Building on this foundation, the physiological basis for estrous synchronization is described, followed by the advancement of pre-synchronization protocols designed to increase estrus and conception. Roads and streams are synonymous with their connection to movement, the acquisition of resources, and as pathways to new opportunities. However, the factors that define good roads and ecologically connected streams are rarely without conflict. Road infrastructure at stream-road crossings has the potential to fragment stream networks. Tube culverts, where streams pass under the road through metal pipes, are particularly concerning for stream connectivity. Undersized, aging, or inappropriately installed culverts can develop vertical drops at the outflow due to high velocities and stream bed scouring. Thin stillage (TS) and whole stillage (WS) are major by-products of the corn ethanol industry. These by-products are generated in large quantities and contain nutrients that remain after yeast fermentation. Current industrial utilization of these by-products involves a series of downstream processing steps such as centrifugation and evaporation, which are energy-intensive and often result in nutrient losses. Traditionally, these by-products are dried to produce distillers dried grains with solubles. Recently, cattle feeders have extended days on feed (DOF) resulting in hot carcass weights (HCW) exceeding the limits of existing serial slaughter datasets. This study's goal was to determine the impact extended DOF exerts on growth performance, feed efficiency, and carcass characteristics of steers and heifers with varying proportions of Angus (AN) and Limousin (LM) genetics. In 2022, commercial AN cows from two MT ranches were artificially inseminated to 9 sires from 3 breeds (AN, LM, and Limflex (LF)) with parentage confirmed by DNA. Pre-weaning mortality (PWM) remains a significant challenge in modern swine production systems, particularly in the context of hyper prolific sows. Although genetic selection has successfully increased litter sizes, these advancements have also intensified physiological stress on sows and increased the vulnerability of neonatal piglets. The majority of PWM occurs within the first 72 hours postpartum, with crushing by the sow as the leading cause of death. Additionally, iron deficiency anemia remains one of the most widespread nutritional disorders in piglets, further compromising survival, immune function, and growth. The objective of this dissertation was to evaluate strategies to mitigate metabolic disorders in confinement fed steers. The first study was conducted on yearling beef steers to investigate the effect of a rumen buffer, calcium magnesium carbonate (CMC), when fed with or without tylosin phosphate (TYL), on growth performance, rumination activity and liver abscess prevalence. Supplementation of a rumen buffer was not an effective alternative to a feed-grade antimicrobial for the control of liver abscesses in beef steers. Supplementation of CMC reduced DMI, thus negatively influencing BW and ADG. No associative effects of CMC and TYL were observed for any …

Following a sharp moose (Alces alces; mooz, Anishinaabemowin, Ojibwe language) population decline in northeastern Minnesota during the early 2000s, the Grand Portage Band of Lake Superior Chippewa initiated a long-term moose collaring effort to assess survival and mortality causes. Moose are a culturally important species to the Band, with annual subsistence hunts held on the Reservation and adjacent 1854 Ceded Territory. Continued research guides culturally appropriate management decisions aimed at bolstering calf and adult survival, helping to preserve this moose in Minnesota today and for subsequent generations. Soil moisture (SM) plays a central role in climatic and environmental processes, influencing shear strength of soil, agricultural productivity, land–atmosphere interactions, and hydrologic functioning. However, accurately estimating SM across diverse climatic regions remains challenging due to spatial heterogeneity, limited in situ measurements, and inconsistencies in sensor resolution. The pinyon jay (Gymnorhinus cyanocephalus) is a highly social, colonially nesting corvid endemic to the interior western United States whose populations have declined by ~72% since 1966, prompting the species’ current review for listing under the Endangered Species Act. Pinyon jays inhabit pinyon pine (Pinus subsect. Cembroides) and juniper (Juniperus spp.) woodlands and other dry conifer woodlands, portions of which are treated through tree thinning and removal for fuels reduction, ecological restoration, and habitat improvement for wildlife. Walleye Sander vitreus are a popular sportfish among anglers in South Dakota with large economic impacts to local communities. A popular destination for Walleye anglers is Lake Sharpe, a 25,000-ha reservoir on the Missouri River, in central South Dakota. Gill net catches in Lake Sharpe show high annual variation, prompting concern that gill net catch rates may be poorly related to population abundance. This thesis presents the design, implementation, and validation of the Mid Scale Rover Gravity Offloader (MRGO), a terrestrial testbed developed to simulate partial-gravity conditions for planetary rover testing within a three-dimensional environment. The MRGO enables consistent, user-defined offloading force as a rover drives freely within a 20 by 18 ft test area, while a stepper motor rail gantry autonomously tracks the rover’s motion and maintains overhead alignment. Mounted to the moving gantry is a vertical carriage column that houses the offloading mechanism, which accommodates up to 5 ft of vertical displacement without loss of force accuracy. The Prairie Pothole Region of eastern South Dakota contains numerous small depressional wetlands, colloquially known as potholes, that provide important ecosystem services, including water storage, water quality improvement, and wildlife habitat. The Agricultural Conservation Easement Program (ACEP) was established by the U.S. Department of Agriculture to protect and restore wetlands and grasslands on private land through easements such as Wetland Reserve Easements. Although ACEP requires management practices intended to maintain or improve habitat quality, few studies have evaluated their effectiveness at supporting wildlife communities and vegetation integrity. Rangelands cover nearly half of the Earth’s surface, representing about 54% (79 million km2), and occur in different types and climates worldwide. Alfalfa (Medicago sativa L.) is a major component in rangelands and covers about (320 thousand km2), representing about 0.4% of total global rangeland area. As a critical perennial legume crop, alfalfa can improve soil health by fixing approximately 336 kg/ha/year of nitrogen and sequestering up to 614 kg/ha/year of carbon, while enhancing livestock production due to its high content level of vitamins, magnesium, minerals, fiber, and protein. Soybean cyst nematode (SCN), Heterodera glycines (HG) Ichinohe, is an endoparasitic nematode and one of the major yield-limiting pathogens of soybean (Glycine max L.) in the United States. Soybean cyst nematode alone accounts for more than $1.5 billion economic loss annually in North America. First reported in the U.S.A. in 1954 in New Hanover County, North Carolina and later in 1995 in Union County, South Dakota. Prior to this study, SCN has spread in 34 counties in the state. This dissertation presents the design, formulation, and immunogenicity evaluation of a novel mRNA vaccine targeting the L1 ORF1p protein, a tumor-associated antigen. The vaccine was synthesized using in vitro transcription and encapsulated in lipid nanoparticles (LNPs), with both unmodified uridine and N1-methylpseudouridine variants tested. Physicochemical characterization confirmed optimal particle size (160–220 nm), acceptable zeta potential, and encapsulation efficiency ranging from 74% to 92%, with preserved RNA integrity post-formulation. These results validate the robustness of the formulation workflow and its suitability for antigen delivery. Immunogenicity studies in C57BL/6 mice revealed modest but promising immune responses. The increasing demand for soybean production in the Midwest, driven by new soybean crushing plants, may reduce the cost of soybean meal (SBM), making it a more accessible and economically viable feed ingredient in swine diets. Replacing dried distillers grains with solubles (DDGS) with SBM and soy hulls at different inclusion rates may influence growth performance, carcass traits, nutrient digestibility, manure nutrient output, pelleting energy use, and economic returns in wean-to-finish pigs. Cover crops (CCs) play an essential role in sustainable agriculture by providing multiple ecosystem services. However, adoption remains low due to concerns about economic returns, management complexity, and potential yield reductions in cash crops. Applying CCs uniformly across fields without accounting for spatial variability can result in inconsistent outcomes. A precision cover cropping system (PCCS), which incorporates site-specific management zones (MZs), offers a more targeted approach to improving performance while minimizing costs and uncertainties. This study aimed to evaluate the effects of rumen-pro- tected Met on lactation performance, inflammation, and immune response, and liver glutathione of lactating dairy cows during a subclinical mastitis challenge (SMC). Thirty-two Holstein cows (145 ± 51 DIM) were enrolled in a randomized complete block design. At −21 d relative to the SMC, cows were assigned to dietary treatments, and data were collected before and during the SMC. Mozzarella cheese (MC) is widely used for pizza ap- plications, and it is generally shredded either in conver- sion plants or in pizzerias. The shreddability of MC is influenced by a variety of factors and it is critical to understand how different Mozzarella cheese types and storage conditions (temperature and age) affect this prop- erty. Three batches each of 3 types of MC (low-moisture Mozzarella (LMM), low-moisture part-skim Mozzarella (LMPS), and reduced fat Mozzarella (RFM)) representing 3 different fat levels were procured directly from a com- mercial manufacturer and stored at 2 different tempera- tures (1.67 and 4.44°C) and evaluated …
