Molecular mechanisms that drive metabolic acclimation to environmental shifts have been poorly characterized in phytoplankton. In this laboratory study. the response of light- and N-limited Skeletonema costatum cells to an increase in light and NO₃ availability was examined. C assimilation was depressed relative to N assimilation early in enrichment, and the photosynthetic quotient (O₂: CO₂) increased, consistent with the shunting of reducing equivalents from CO₂ fixation to NO₃- reduction. The concomitant increase in dark respiration was consistent with the increased energetic demand associated with macromolecular synthesis. The accelerations of N-specific rates of …

The seasonal and inter-annual concentrations of phytoplankton were studied over a 50 month period in the lower James, York and Rappahannock Rivers (USA). Differences in the onset, duration and magnitude of major seasonal growth periods varied from year to year. There was a tendency for spring, summer and fall maxima, with a winter period of reduced abundance. An additional study of picoplankton over a 12 month period indicated greatest abundance during summer and fall, with least development in winter.

Uniqueness and stability criteria are established for the steady states of a nonlinear model of growth factor production. A specific expression for the nonlinearity is chosen, containing three parameters which can be adjusted to fit a specific biological context, but much of the analysis applies to a general class of source terms that exhibit the same qualitative behavior.

Understanding recruitment and identifying factors critical to that process are imperative if adult spiny lobster Panulirus argus stocks are to be conserved and properly managed. The goal of our research has been to obtain ecological information linking inshore postlarval spiny lobster recruitment to later life stages, thereby providing the basic framework for assessing and predicting adult stock. Supernatant proteins in Vibrio alginolyticus batch cultures were analyzed by SDS-PAGE before copper was added, 24 and 48 hours after the addition of copper, and in 24 hour control (no Cu) cultures. Two proteins, one 21 kilodalton (kDa) and one 19 kDa, were found to be copper-induced, and were designated copper-binding protein 1 (CuBP1) and CuBP2. CuBP1 and CuBP2 became detectable in supernatants during the Cu-induced lag phase, and increased in concentration over the following 48 hours. Chloramphenicol inhibited production of these proteins. The eggs of the Japanese medaka (Oryzias latipes) were employed in a number of studies to develop a routine, standardized assay which can assess the acute and sublethal impacts of individual toxicants and complex mixtures. The eggs of this Cyprinodont minnow were topically treated with each toxicant or mixture dissolved in membrane permeable dimethylsulfoxide (DMSO) solvent. Nine chemicals were initially evaluated with two, pentachlorophenol (PCP) and tributyltin chloride (TBTCl), subsequently selected for extensive evaluations of salinity tolerance and the accuracy of the topical exposure method for representing a typical immersion exposure. Local gamma-amino butyric acid (GABA) neurons in the hypothalamus can modulate the luteinizing hormone releasing hormone (LHRH) pulse generating system.

Two animal models (intact and ovariectomized rats) were used to determine the nature of modulation of LHRH release by GABA. The experiment determined the release of LHRH, serotonin (5-HT), and 5-hydroxyindole acetic acid (5-HIAA) during two hormonal states with each model. In the intact rat the proestrus and estrus states were examined, and in the ovariectomized rat in-vitro release was determined with (OVX₂) and without (OVX) estradiol treatment.

The survival, revascularization, reinnervation, and recovery of function of transplanted newborn rat pineal glands were studied following grafting into four different locations in pinealectomized rats. These locations were the third cerebral ventricle, in situ (where the pineal gland is normally located), the renal capsule, and the anterior chamber of the eye.

Pinealocytes in the grafts maintained high metabolic activity as evidenced by the presence of euchromatic nuclei, prominent nucleoli, infolded nuclear envelopes, numerous mitochondria and Golgi apparati, and relatively abundant endoplasmic reticulum and ribosomes. Fisheries scientists use roving creel survey methods to estimate total catch and total effort of recreational fisheries. Bias in the roving creel estimators had been known to the scientific community, but it had not been quantified. I developed a roving-creel-survey computer simulation model to investigate the bias and precision of roving creel estimators, and estimated parameters for the model based on field observations from the blue crab (Callinectes sapidus) fishery of the Chesapeake Bay. Test populations of different-sized guppies, Poecilia reticulata, were exposed to individuals of two natural predatory species, the pike cichlid Crenicichla a/ta and the killifish Rivulus harti, under conditions of varying prey (guppy) density and habitat complexity in the laboratory. Rivulus fed most frequently on newborn and juvenile guppies < 14 mm. Crenicichla consumed more and larger guppies than did Rivulus. The mean guppy size eaten by Crenicichla was dependent on the length of the individual predator, but as a group, the 15 Crenicichla tested were non-selective with respect to guppy size and gender. The population dynamics of two species was studied for one year on Craney Island in Portsmouth, Virginia. This dredge disposal site provided good habitat for the house mouse, Mus musculus, whose opportunistic behaviors can withstand the sudden changes in a dynamic environment. Although the site was not optimal habitat, a population of Microtus pennsylvanicus, the meadow vole, was sustained there. Of the seven species of small mammals captured, 65% were house mice and 28% were meadow voles. Meadow voles were more successful at establishing short periods of residency than were house mice; on average, each meadow vole was …

A study of the subtidal macrobenthos in Back Bay, Virginia was conducted to examine community structure in relation to sedimentary and water quality characteristics. Samples were collected in August and November of 1987 and February and May of 1988 at ten stations.

Three site groups and three temporal groups were identified by cluster analyses of species abundance. Species composition between site groups was relatively homogeneous and eight species accounted for most of the variation between site groups. be identified that would aid in the identification and distribution of the threatened subspecies Sorex l._ fisheri, endemic to the Great Dismal Swamp, the more upland nominate subspecies Sorex l. longirostris, and possible intergrades between the two subspecies. Several processes can affect variations in the chemical composition of an estuary, including (1) biogeochemical reactions in the water column, (2) advective transport, and (3) diffusion from underlying sediments.

The purpose of the study was to establish whether estuarine redox reactions are controlled by a complex interaction of processes 1 to 3. Short-term and long-term studies were conducted in Chesapeake Bay. Redox species involved in redox cycling include oxygen, iron, manganese, hydrogen sulfide, nitrogen species, arsenic and antimony species, while phosphate is indirectly involved in redox cycles. Silicate was used as a non-redox tracer.

The objective of this research was to develop a method for the separation of different types of collagen using a large pore size (100 nm) reversed-phase C-8 column. Three different types of collagen (type I, III, and V) were sufficiently separated on the column using a mobile phase containing water-acetonitrile-trifluoroacetic acid. Collagens found in each HPLC peak were identified using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) to assay the column effluents.

Acid demineralized bone (DMB) implanted in extraosseous sites induces bone formation. In vivo studies have shown that DMB stimulates the differentiation of mesenchymal cells into osteogenic cells. In this study an in vitro system was developed to demonstrate the osteoinductivity of DMB quantitatively. This in vitro working system was used to assess the osteoinductivity of variously processed DMB products as a means to evaluate their processing protocols. Hydrothermal plumes associated with nearshore lava flows from Kilauea Volcano, Hawaii were studied on five occasions during 1989-1990 to address the current lack of data on direct lava-seawater interactions. The following enrichments were found in the sea-surface hydrothermal plumes above the active underwater lava flows: H₂, 15,000x ambient seawater concentrations; Mn, 250x; and Si, 20x. Water temperatures reached 46°C. Lower concentrations and temperatures were observed in the plumes with increasing distance from shore, with H₂, Si, and Mn concentrations linearly related to seawater temperature. The relationship between gender, classroom type, seating preference and locus of control with respect to how they predict attitudes about classrooms and seating preferences was investigated in 38 males and 44 females. A number of significant relationships were found between the perceived environments and various attitudes of students about the environments. The Classroom Environment Scale and Seating Preference Questionnaire were used to assess student attitudes about classroom environments and seating preferences. Students perceived small classrooms as enhancing friendships, affiliation and competition as compared to large and lecture classrooms. The study examined subjects' perceptions about disclosure of emotion by a stimulus person who was depicted as extraverted or introverted, and subjects' self-disclosure of emotion based on their own level of extraversion. In addition, the role of gender in disclosure of emotion was examined. The findings on differences in disclosure of emotion based on gender are considered tentative. Males self-disclosed anger more to same-sex stimulus persons than did females, but this was the only significant difference. An introverted compared to an extraverted stimulus person was found to elicit more disclosure about emotions associated with calmness and depression. Emotions which an …

This study investigated the impact of menopause stage and level of exercise on depression and perceived body image with 101 women aged 40-60 years. Instruments included a demographic information sheet, the Life Experience Survey (LES), the Beck Depression Inventory (BDI), and the Multidimensional Body-Self Relations Questionnaire (MBSRQ). The relationship between menopause stage and depression was not significant. The relationship between menopause stage and perceived body-image was also not significant. Menopausal women do not appear to be more depressed or to have lower body-image perceptions when compared to premenopausal women. The main effect for amount of exercise and perceived body image …

Model solutions of the amino acid phenylalanine were chlorinated to seven Cl/N mole ratios. The by-products were characterized using gas chromatography, high performance liquid chromatography, GC/MS, infrared spectroscopy, and nuclear magnetic resonance spectroscopy. At the lower Cl/N ratios N-chlorophenylalanine and phenylacetaldehyde were identified as the main products while phenylacetonitrile and N-chlorophenylacetaldimine were the major products at Cl/N ratios of 2.0 and beyond. The concentration of phenylalanine in a municipal wastewater was determined. Using a radiotracer the chlorination products of phenylalanine in the wastewater at the 7 Cl/N ratios were identified as those seen in the model solutions. The distribution of …

A murine monoclonal antibody (MAb), 7E11-C5, recognizes a prostate organ-specific antigen. Under reducing conditions, the 7E11-C5 antigen exhibited major 100-Kd and minor 70-Kd bands, suggesting that the antigen comprises two peptide chains. The antigen was detected in pooled normal human seminal plasma, xenograft tissue extract of a prostate adenocarcinoma cell line, LNCaP, and tissue extracts of benign prostatic hyperplasia (BPH) and prostate carcinomas, but was not identified in any of the non-prostate tissue extracts tested. This study investigated the effect of client gender on several aspects of clinical decision-making processes and clinical judgement when a client presented with a combination of depressive and alcohol-dependent symptoms. Forty-four male and 44 female clinicians, who came from various mental health disciplines (i.e., psychology, social work, counseling, nursing, and psychiatry), volunteered to participate as subjects.

The central purpose of this study was to evaluate the effectiveness of the Welcome Baby Project (WBP), a primary prevention program with the mission of promoting child development and a positive relationship between "at-risk" adolescent mothers and their infants and to prevent disorders of attachment, child abuse and neglect, and unwanted, repeat pregnancy. Intervention involved weekly home visits by trained parent volunteers for children from birth to two years.

This thesis examines Foreign Minister Edgardo Mercado Jarrin's policies between 1968 and 1971, investigating the degree to which he formulated an independent Peruvian foreign policy. It traces the ideological underpinnings behind Peruvian diplomacy. The study analyzes three basic dimensions of foreign policy: the seizure of La Brea and Parinas oil fields, the capture of unregistered fishing vessels, and the extension of relations to nonaligned and communist countries. New optimal algorithms for recognition, center, diameter, maximum matching, Hamiltonian path and domination in indifference graphs are presented. The recognition algorithm produces a linear order with properties which allow the solution of the other problems in linear time. The purpose of this study was to investigate whether practicing dental hygienists promote their profession as a desirable career. A self-designed, 48 item questionnaiire, Dental Hygienists' Participation in Career Recruitment, was mailed to a random sample of 600 dental hygienists residing in the Commonwealth of Virginia. Data were obtained from 393 dental hygienists for a 65.5 percent response rate. Responses from each item were analyzed by frequency distributions, percentages and chi-square test for independence at a 0.05 level of confidence. The purpose of this study was to identify the range of problems and coping strategies as perceived by antepartal patients experiencing medical treatment to decrease risk of preterm labor. In this qualitative, interpretive study problems and coping strategies were identified through semi-structured interviews with a convenience sample of seven antepartal patients. Using content analysis of interview data, identified problems were categorized, and for each problem, coping strategies were identified and grouped using the categories of the Jalowiec Coping Scale.
