An Analysis Of Mitochondrial Dna In Rett Syndrome And Other Neurodegenerative Disorders, Catherine Erickson Burgess

Mitochondrial dysfunction resulting from mutations on mitochondrial DNA (mtDNA) is being recognized in a growing spectrum of diseases. These diseases, resulting from single base mutations, large deletions, or insertions, have been largely neuromuscular in origin. However, as an understanding of the effects of mtDNA mutations progresses, attention is now focusing on neurodegenerative diseases. Rett Syndrome (RS), a progressive neurodegenerative disease with predominantly female cases, demonstrates morphologic mitochondrial changes, mitochondrial enzyme deficiencies and maternal inheritance (characteristic of mtDNA diseases). No investigation of mtDNA involvement has been previously conducted and, to date, no biological marker exists for this disorder.

This study analyzes Iranian foreign policy since 1979 to determine if Iran is a pariah state in the international community. To determine Iran's isolation in the international community, various factors were examined, including diplomatic relations, political leverage, relations with the United Nations, and the ability to acquire military hardware. Iran's status was compared with three generally accepted pariah states, Taiwan, Israel, and South Africa to determine the degree of isolation faced by the Islamic Republic. This study determined that Iran remains a pariah state, but is less isolated than most pariah states because its objectionable behavior, its aggressive foreign policy, …

This thesis studies the radioactive contamination of Kazakhstan's environment, concentrating on the period between February 1989 and January 1993. For each type of nuclear degradation in Kazakhstan the thesis asks the questions: what is its cause and extent, what are its consequences for humans, and what are the human responses. The main causes of Kazakhstan's nuclear devastation are atomic weapons' tests, uranium mining, manufacturing of uranium fuel, and the operation of a fast breeder reactor power generator unit. At this time there is no NIOSH approved means for bag sampling 1,3-butadiene. An attempt was made to use the application of cold (0°F) to inhibit the reactivity of the gas prior to analysis. The State of California Air Resources Board has an approved method for the gas chromatographic analysis of 1,3-butadiene from stationary sources. The method was utilized in the application of industrial hygiene monitoring of affected workers. The application of cold was found to be ineffective at inhibiting the highly reactive nature of 1,3-butadiene. Analyte losses were too great to classify the method as an accurate measure of …

Soil microorganisms are important sources of the nitrogen trace gases NO and N₂O for the atmosphere. Present evidence suggests that autotrophic nitrifiers such as Nitrosomonas europaea are the primary producers of NO and N₂O in aerobic soils, whereas denitrifiers such as Pseudomonas spp. or Alcaligenes spp. are responsible for most of the NO and N₂O emissions from anaerobic soils. It has been shown that Alcaligenes faecalis, a bacterium common in both soil and water, is capable of concomitant heterotrophic nitrification and denitrification. Comparisons were made between 55 heterosexual males, 49 heterosexual females, 56 homosexual males, and 35 homosexual females on measures of attachment, relationship satisfaction and relationship success. Results indicated that adult homosexuals experience greater difficulty than adult heterosexuals in becoming close to others. Individuals endorsing the secure attachment style reported greater relationship satisfaction than individuals endorsing either of the two insecure attachment styles. In addition, females reported greater relationship satisfaction than males. Significantly more individuals endorsing the secure attachment style reported their mothers to have been warm/responsive; significantly more heterosexual males and females than homosexual females reported their mothers to have …

Identification And Quantitation Of Volatile Amines In Chesapeake Bay Sediments, Neeraja Y. Krishna

Sediments from different parts of the Chesapeake Bay were analyzed to determine the presence of volatile amines. A HF-HCl extraction method was used to extract both exchangeable and fixed amines from the sediments and a purge-and-trap technique was used to concentrate the amines. Following derivatization with heptafluorobutyric anhydride (HFBA), the HFBA-amines were analyzed by gas chromatography with electron capture detection and with mass spectrometric detection. One primary amine, i-amylamine with a concentration ranging from 0. 78 nmoles/g dry weight to < 0.05 nmoles/g dry weight was found in a total of twenty seven samples collected on three different dates in 1993. Another primary amine, 2-methylbutylamine with a concentration ranging from 0.18 nmoles/g dry weight to < 0.05 nmoles/g dry weight was found in a total of twenty three samples collected on three different dates in 1993. A secondary amine, diethylamine was detected but could not be quantified in a total of seven samples collected in September 1993. The concentrations of these amines varied with location in the Bay, depth in the sediment, and time of year the sediments were collected. The origin of i-amylamine and 2-methylbutylamine might be from the biological decarboxylation of amino acids leucine and isoleucine, respectively. The origin of diethylamine is unknown.

This study examined the influence of prior satisfaction, perceptions of problem severity, and attachment style in determining Rusbult's Exit, Voice, Loyalty, and Neglect responses to dissatisfying events depicted in a psychotherapy simulation. Constructive responses to therapy dissatisfaction (i.e., Voice and Loyalty) were positively related to prior satisfaction with the therapist and negatively related to perceptions of problem severity. Destructive responses (i.e., Exit and Neglect) were negatively related to prior satisfaction and positively related to problem severity. Attachment style was associated with individual differences in approaches to resolving conflict, but it was not consistently related to Exit, Voice, Loyalty, and Neglect …

The lower Chesapeake Bay spawning stock of the blue crab, Callinectes sapidus, (Rathbun), is examined for seven years (1986 - 1992) in an attempt to better understand stock behavior. Three methods of post-stratification are used to describe the distribution and movement of the population over time. The three methods, density strata, geographic zones, and depth strata, did well in explaining movements of the population, indicating a trend of increased concentration of blue crabs near the eastern Bay late in the spawning season - October. The data suggest a bimodal period of spawning and a trimodal period of abundance. The …

Atlantic croaker, Micropogonias undulatus, collected from commercial catches in Chesapeake Bay and in Virginia and North Carolina coastal waters during 1988-1991 (n=1,967) were aged from transverse otolith sections. Ages 1-8 were recorded, but eight-year-old fish were rare. Marginal increment analysis showed that for ages 1-7, annuli are formed once a year during the period April-May. Otolith age readings were precise: >99% agreement within and between readers. Observed lengths-at-age were highly variable and growth rate decreased after the first year. Despite the high variability in sizes-at-age, observed lengths for ages 1-7 fit the von Bertalanffy growth model (r2=0.99; n=753) well. …

The purpose of this study was to determine if variations in cam profiles cause differences in muscle contraction as measured by surface EMG, knee joint angular displacement, and torque. There were seven hypotheses tested. The ten subjects were adult male volunteers (18-24 years) who participated in a counterbalanced treatment design study testing three variously shaped cams over five lifting tests. The results indicated that for the EMG data, the cam and lifts effect had significant Wilk's values. For the cam effect the consistent cam had the highest millivolt value. H₂0₂ in seawater has complicated sources and sinks. The relative importance of biological regulation of H₂0₂ compared to other processes is not well understood. In addition, environmental factors affecting the biological regulation of H202 are largely unknown. Marine phytoplankton was examined for the kinetics of the production and decomposition of H₂0₂ in the dark. Effects of varying environmental factors such as light, temperature, salinity, nutrients, amino acids, trace metals and growth phase, were examined. H₂0₂ was determined with the scopoletin-fluorescence decay method.

Mammalian mitochondria contain a unique DNA topoisomerase I with characteristics of both nuclear and bacterial type I DNA topoisomerases. This enzyme, purified from bovine liver, requires cations as cofactors and possesses optimal activity at pH 7.0, 37°C and either 150 mM K+ or Na+ ions. Similar to nuclear enzymes and different from the bacterial enzyme, divalent ions such as Ca²⁺ or Mg²⁺ will stimulate but are not essential for the enzymatic activity.

A non-estrogenic hormone, 6-methoxybenzoxazolinone (6-MBOA), found in young growing grasses, has been shown to be a reproductive initiator in several herbivorous rodents. However, the mechanism by which 6-MBOA initiates reproduction has not been determined. Sagittal otoliths were used to determine the age and growth of 605 larval and juvenile Atlantic croaker, Micropogonias undulatus, collected in the Middle Atlantic Bight (MAB) and estuarine waters of Virginia. A Laird-Gompertz model (r2 = 0.95) was used to describe the growth of croaker up to 65 mm standard length (SL) and 142 days (t): S1_(t) = 2.657 Exp {4.656 [1-Exp (-0.008lt)]}; SL_(t) = standard length at day t. Generally, croaker collected inshore were larger and older than those collected offshore, indicating estuarine immigration from offshore spawning grounds. Iodate and iodide are the two major species of iodine in seawater. In comparison to iodate, iodide is thermodynamically unstable in oxic seawater. The conversion (or reduction) of iodate to iodide may be mediated via biological activities. Since biological activities vary with the season, the conversion of iodate to iodide may also vary seasonally. The conversion (or oxidation) of iodide to iodate is thermodynamically feasible, but the mechanism is poorly known. Hydrogen peroxide, which is ubiquitous in surface seawater, may oxidize iodide to iodate.

Grape seed extract is being marketed in France for cardiovascular use and skin care. The active constituents of both these drugs are certain polyphenols called procyanidins. A similar seed extract has now been obtained from Virginia Chardonnay grape seeds. Ten of the major components in this extract have been isolated in their pure form by the semi-preparative HPLC separation on a YMC ODS-AQ (Octadecylsilane-AQ) column. The perinea! gland in the hispid cotton rat, Sigmodon hispidus, is a seasonally cyclic, subcutaneously located organ which is associated with the reproductive system. On gross dissection, the gland possesses a strong attachment to the penis with only loose fascia! connections to the rectum and surrounding muscle and skin. Histologic exam confirms this connection to the reproductive tract and shows the organ to be a compound tubuloacinar gland with a projection leading into the penis. Florida Bay is a major nursery for the Caribbean spiny lobster, Panulirus argus. The Bay contains a series of hardbottom or seagrass-covered basins separated by mudbanks, which serve as barriers to water circulation and presumably to transport of postlarvae. Temperatures can fluctuate dramatically in the basins and salinities range from 35 ppt to > 50 ppt. Two species of bont ticks, Amblyomma hebraeum and Amblyomma variegatum, common to Africa have been studied to compare types and quantities of compounds known or believed to serve as components of the attraction-aggregation-attachment pheromone (AAAP). A complex of attraction, aggregation and attachment pheromones are used by these ticks to detect hosts, mates and perhaps minimize interspecific breeding. Solvent extraction of pheromone emitting ticks followed by Gas Chromatography and Mass Spectrometry revealed little difference in the composition of the AAAP in these two species. Masculinity ideology is the extent to which an individual endorses traditional male-role norms. The present study examined the discriminant and convergent validity of this construct relative to other gender-role constructs. Two-hundred sixty-nine male participants completed a questionnaire containing measures of masculinity ideology, gender-role conflict, sex-role orientation, and gender-relation attitudes. Participants also completed measures of constructs known to correlate with masculinity ideology. support for discriminant validity was indicated by masculinity ideology accounting for variance in outcome measures that was unaccounted for by sex-role orientation or gender-relation attitudes. A bloom of the dinoflagellate Ptychodiscus brevis occurred off the North Carolina coast in late 1987, and samples were forwarded by NOAA to the Old Dominion University Phytoplankton Laboratory for total phytoplankton analysis. Six sampling stations were chosen to represent different bloom scenarios and these six were clustered into three site groups based on the concentrations of statistically significant taxa. Abundances of phytoplankton taxonomic groups over time were examined in relation to P. brevis concentrations to identify any general tendencies. Petrographic and sedimentologic analyses of eleven shallow subsurface cores (3 to 19 meters in length) from the Lucayan Limestone where it occurs along northeastern Great Abaco Island (little Bahama Bank) are used to delineate five carbonate lithofacies. The most prevalent lithofacies is a mottled, nonskeletal, peloidal packstone to grainstone. Other lithofacies include a rudstone, a laminated nonskeletal grainstone, a crossbedded grainstone and a mudstone-wackestone.

Cementation varies from poor to very well cemented core intervals. Cements observed included equant granular, equant blocky, drusy, meniscus, and needle fiber "whisker" cements. These cements reflect a variety of diagenetic environments such as the vadose, …

An experiment was conducted to study the association of foods of different levels of palatability and environmental events present during eating. Rats were exposed to different. foods in a start box and then allowed to run to a goal box containing food of a lesser or equal palatability. All animals showed a decrease in latency to eat in the start box and an increase in the amount of food consumed in the start box, with no significant differences in treatment groups at the end of conditioning. Two hundred and forty-six species of benthic foraminifera have been recognized in fourteen short cores (≤ 24cm) taken from within the Wilmington and South Heyes submarine canyons along the slope and rise of the United States Atlantic Continental Margin. Of these one hundred and forty-five species were identified to the species level, thirty were closely related to known species, sixty-six could not be assigned specific names, and five species were of uncertain generic placement. The petrologic character of the sediments was determined for each of the seventy-eight micropaleontogical samples.

Previous studies have indicated that, in our culture, female genitalia are unlabeled, mis-labeled, and/or negatively labeled, while male genitalia are not.

Twenty-five mothers of young children were interviewed to determine what labels they were teaching their children for sexual body parts. The women later met in small groups to discuss the i r reactions to a soft-sculpture of female genitalia.

The study found that twice as many male children were taught common designations for their own genitalia than were female children. In addition, a greater proportion of female children were given a label for male genitalia, than male children were …
