A model analysis of electroporation dynamics in biological cells has been carried out based on the Smoluchowski equation. Results of the cellular response to short, electric pulses are presented, taking account of the growth and resealing dynamics of transient aqueous pores. It is shown that the application of large voltages alone may not be sufficient to cause irreversible breakdown, if the time duration is too short. Failure to cause irreversible damage at small pulse widths could be attributed to the time inadequacy for pores to grow and expand beyond a critical threshold radius. We address the development of a new and innovative method, in vivo and possibly non-invasive, for tumor and healthy tissues control during and after the radiation treatment. The radiation treatment is delivered in an almost standardized manner for particular classes of tumors. Influenza infections result in activation of cellular and humoral immune responses, leading to stimulation of cytotoxic T cells (CTL) and helper T cells with subsequent viral clearance.

By using an assay of granzyme B activity, the in vivo cellular-mediated immune response of old adults was found to be significantly decreased in comparison to young adults. The effect of aging on the CTL response was further investigated via enzyme-linked immunospot technique. Intrusion of Upper Circumpolar Deep Water (UCDW), which was derived from the Antarctic Circumpolar Current (ACC), onto the western Antarctic Peninsula (WAP) shelf region in January 1993 provided a reservoir of nutrient-rich, warmer water below 150 m that subsequently upwelled into the upper water column. Four sites, at which topographically-induced upwelling of UCDW occurred, were identified in a 50 km by 400 km band along the outer WAP continental shelf. One additional site at which wind-driven upwelling occurred was also identified. Diatom-dominated phytoplankton assemblages were always associated with a topographically-induced upwelling site. The chemical composition of high molecular weight dissolved organic matter (DOM) and particulate organic matter (POM) was examined along the salinity gradient of the Delaware Estuary. DOM was collected and fractionated by tangential-flow ultrafiltration into 1-30 kDa (HDOM; high molecular weight) and 30 kDa to 0.2 μm (VHDOM; very high molecular weight) and compared to particles collected in parallel. Polysaccharides comprised 12-43% of particulate organic carbon (POC), 30-56% of VHDOM carbon, and 7.5-19% of HDOM carbon. Hydrolyzable amino acids comprised 17-38% of POC, 5.4-12% of VHDOM carbon, and 1.5-4.2% of HDOM carbon. Only 7-43% of dissolved organic nitrogen in VHDOM …

Nitrate reductase activity (NRA) and ¹⁵NO₃- uptake (NU) were determined in the East China Sea and the adjoining Kuroshio in May 1996, at six stations covering a range of hydrographic conditions: the nutrient-rich and fresher plume of Changjiang Diluted Water along the Chinese coast, the nutrient-rich upwelling Kuroshio Subsurface Water at the shelf edge northeast of Taiwan, the oligotrophic Kuroshio Surface Water and the mixing zones among these water masses on the shelf. The values of NRA in the surface mixed layer ranged between 16 and 0.1 nM-N h^-1, whereas those of NU ranged between …

The requirement for selenium (IV) was assessed in five strains of the toxic dinoflagellate Gymnodinium catenatum Graham, representing three populations from Tasmania (Australia), as well as one each from Japan and Spain. Strains were grown in nutrient-enriched sea water medium with 10^-9 M selenium added as selenite (H₂SeO₃), or with no added selenium, and monitored for growth and cell yield. Strains exhibited different selenium (Se) requirements, as evidenced by (i) a decrease in exponential growth rate (10-20%) and cell yield (up to 80%) (Japanese strain); (ii) a decrease in cell yield only (Tasmania Derwent Estuary …

Dissolved iron and manganese and total dissolvable iron were measured in water column samples from the Polynya Region of the southern Ross Sea in cruises in Nov.-Dec. 1994 (spring 1994) and Dec. 1995 to Jan. 1996 (summer 1995). Iron and manganese addition bottle incubation experiments were also performed on these cruises to assess the nutritional sufficiency of ambient iron and manganese concentrations for phytoplankton growth. Generally high dissolved iron concentrations (>0.5 nM) and relatively complex iron and manganese vertical profiles were obtained in spring 1994 vs. summer 1995. A doubly labelled tracer molecule of glucose, [6,6^-2HJglucose, has been used to measure the rate of glucose appearance in blood due to endogenous glucose production (by the liver and kidneysj.^l,2 Considered 'non-recirculating', this tracer generally provides a realistic estimate of glucose kinetics.^2,3 Used as a dual tracer in conjunction with [l-¹³C]glucose, the extent of glucose recycling can be quantitated.^2-4 but the extent of possible recirculation of a single deuterium atom on newly formed glucose must then be considered.

The Optical Plankton Counter (OPC) has been used in oceanic and fresh waters to estimate zooplankton abundance and biovolume. However, it is not clear whether the OPC can produce accurate estimates of zooplankton abundance and biovolume in waters with high detritus. In order to test the capability of the OPC to estimate zooplankton abundance and biovolume in Chesapeake Bay, two sets of laboratory experiments were conducted using water with high detritus concentrations collected from the upper Choptank estuary of Chesapeake Bay and laboratory cultured Artemia. Current velocity and water density profiles were obtained along two crossestuary transects with the purpose of determining the fortnightly variability of the transverse dynamics in a partially stratified coastal plain estuary. The profiles were measured with a towed acoustic Doppler current profiler and a conductivity-temperaturedepth recorder in the James River estuary, Virginia. The cross-estuary transects were sampled during the spring tides of October 26-27, 1996, and the ensuing neap tides of November 2-3, 1996. Salinity, a measure of the dissolved salts in seawater, is a fundamental property of seawater and basic to understanding biological and physical processes in coastal waters. In the open ocean long term salinity measurements are identified as necessary to understand global climate studies, hydrological cycle, and circulation. Bacterial hyaluronidases, enzymes capable of breaking down hyaluronate, are produced by a number of pathogenic Gram-positive bacteria that initiate infections at the skin or mucosal surfaces. Since reports of the hyaluronidases first appeared, there have been numerous suggestions as to the role of the enzyme in the disease process. Unlike some of the other more well studied virulence factors, much of the information on the role of hyaluronidase is speculative, with little or no data to substantiate proposed roles. Response Of The Tick Dermacentor Variabilis (Acari: Ixodidae) To Hemocoelic Inoculation Of Borrelia Burgdorferi (Spirochetales), Robert Johns, Daniel E. Sonenshine, Wayne L. Hynes

When Borrelia burgdorferi B31 low passage strain spirochetes were directly injected into the hemocoel of Dermacentor variabilis(Say) females, the bacteria were cleared from the hemocoel within < 24 h. Viable spirochetes were not found in hemolymph, salivary gland, or ovary tissues by subculturing or by IFA. The hemocyte population increased ≈6 times within the first 6 h after inoculation compared with the uninoculated controls. In contrast, the soluble total hemolymph protein content decreased inversely with the increase in hemocytes. Borreliacidal activity was demonstrated with cell-free hemolymph from D. variabilis. In vitro antimicrobial assays using hemolymph from borrelia-challenged and nonchallenged ticks resulted in 72% spirochete reductions compared with only 11.5%, respectively, within 1 h. Additional evidence of induced antimicrobial hemolymph protein activity was demonstrated by SDS-PAGE, which revealed upregulation of a lysozyme-like peptide (≈ 15 kDa) (22% increase) and the induction of a ≈ 5.8 kDa peptide in the B. burgdorferi-challenged ticks. We discuss the utility of wavelet transform methods in signal processing in general, and in particular, demonstrate the technique in optoacoustic applications. In several optoacoustic experiments with different samples, we have successfully enhanced the signal to noise ratios. Wavelet transforms optimize resolution by utilizing a tailored, variable time-window in different frequency regions. The technique's great advantage lies in the fact that the wavelet transform adds some redundancy to the original signal, and some desired features can be enhanced in the transformed space. Zooplankton and water quality parameters were investigated at eight mesohaline stations in the lower Chesapeake Bay and Elizabeth River from January through December 1994 to identify the changes of zooplankton community structure with increased eutrophication. The total micro- and mesozooplankton biomass decreased with the increase of eutrophication. However, the relative proportion of microzooplankton increased with increased eutrophication. Within highly eutrophied waters, the small oligotrichs (m) and rotifers dominated the total zooplankton biomass (as carbon). However, tintinnids, copepod nauplii and mesozooplankton significantly decreased with the increase of eutrophication. Using the human placental choriocarcinoma JEG-3 cell line as an in vitro human placental model, I studied the mechanisms of the tissue-specific expression and steroid hormone regulation of the hGnRH gene in the human placenta. The results showed that all of the previously identified four elements are required for the full activity of the hGnRH upstream promoter in JEG-3 cells, while the element 4 (FP4, −987/−968) is the most important. Studies performed with 5′ end deletion of this region confirmed these observations. This is the first work examining the utility of scale chemistry for determining natal origins and habitat associations. I quantified a relationship of scale chemistry to water chemistry, quantified geographic variation in scale chemistry, and evaluated stability of scale chemistry through maturation. Scale chemistry accurately reflected trace element composition of the water in which fish had lived. Juvenile spot (Leiostomus xanthurus) were held in four concentrations of Sr, Cd, and Ba maintained at either 20°C or 25°C, for 42 days. Strontium:Ca, Cd:Ca, and Ba:Ca levels in scales were linearly related to environmental concentrations while temperature had no effect. …

Recently, a one-dimensional model was developed which gives a reasonable explanation for the existence of a Critical Size Defect (CSD) in certain animals [1]. In this paper, we examine the more realistic two-dimensional model of a circular wound of uniform depth to see what modifications are to be found, as compared with the one-dimensional model, in studying the CSD phenomenon. It transpires that the range of CSD sizes for a reasonable estimate of parameter values is 1 mm-1 cm. More realistic estimates await the appropriate experimental data.

The present study examines complex relationships between fathering and children's social, behavioral, and cognitive development. The participants were 57 families with a child between the ages of 7 and 12. Fathers were interviewed about their disciplinary practices, work history, and day to day involvement with their children. Fathers also completed the Child Behavior Checklist (CBCL; Achenbach, 1991) which is designed to assess children's internalizing and externalizing behavior. Mothers were administered the Vineland Adaptive Behavior Scale - Survey Form (Sparrow, Balla, & Cicchetti, 1984), the CBCL, and a background questionnaire.
