Climate change is expected to intensify salinity and temperature fluctuations in estuarine and coastal habitats. However, the impact of salinity variation on ectotherms’ thermal tolerance remains underexplored, limiting our understanding of the interplay between these stressors. This study investigated salinity-dependent changes in life history traits (survival, growth rate, fecundity, and egg size) over a 33-day exposure to a salinity gradient (12, 17, 22, 27, 32, 35, 40, 45, 50) in the marine annelid Ophryotrocha labronica. Climate change has had devastating effects on tropical coral reefs; however, much less is known regarding how heat stress affects temperate coral. This research focuses on Astrangia poculata (Ellis and Solander, 1786) collected from Narragansett Bay, RI, during the summer and winter seasons and understanding the effect of experimental thermal extremes (i.e., 26 °C) on seasonally different populations. Photosynthetic efficiency (Fv/Fm), symbiont density (via an inverse relationship with pixel intensity), and oxidative stress via reactive oxygen species (ROS) concentrations were measured on symbiotic and aposymbiotic A. poculata. Climate change has caused drastic declines in corals. As sessile organisms, corals acclimate to environmental shifts through genome-wide changes in gene expression, epigenetic modifications, and alterations in microbiome composition. However, alternative splicing (AS), a conserved mechanism of stress response in many organisms, has been under-explored in corals. Using short-term acute thermal stress assays, we investigated patterns of AS in the scleractinian coral Acropora cervicornis during response to low (33°C), medium (35°C), and high (37°C) heat stress and subsequent overnight recovery. Coral reefs are vital marine ecosystems that support many different species and ecosystem services through protecting coastlines from storms and erosion, providing raw materials for medication, and facilitating diversity amongst marine species. Anthropogenic climate change is causing ocean temperatures to exceed the range of coral thermal tolerance resulting in more coral bleaching events globally. Amblyomma maculatum Koch (Acari: Ixodidae), Ixodes scapularis Say, and Dermacentor variabilis Say are three hard-bodied ticks responsible for vectoring pathogens that cause most human tick-borne diseases in the United States of America (USA). Tick surveillance is critical to elucidate high-risk areas for targeted vector control and public health interventions. Despite tick-borne diseases having a higher annual incidence compared to mosquito-borne diseases in the USA, tick traps used for surveillance are grossly under-developed compared to the suite of sophisticated mosquito traps available on the commercial market. The future of coral reefs remains uncertain as intensifying ocean warming continues to drive coral decline and demise worldwide. Corals occurring in extreme environments such as the Persian/Arabian Gulf (PAG), the world’s warmest sea, represent a promising resource to study the genetic architecture of thermal tolerance. However, their thermal capacities remain unresolved. Mixotrophic reef-building corals acquire nutrition via photosynthate translocation from endosymbiotic microalgae and by heterotrophic prey/particle capture. Heterotrophy promotes resistance to, and recovery from, environmental stress, but quantifying coral heterotrophy remains difficult due to complex resource exchanges within the coral holobiont. We interrogated the response of multiple biomarkers to coral heterotrophy using fatty acid profiling and stable isotope analysis of Stylophora pistillata grown along a controlled feeding gradient from pure autotrophy to pure heterotrophy. Motivation: Here, we make available a second version of the BioTIME database, which compiles records of abundance estimates for species in sample events of ecological assemblages through time. The updated version expands version 1.0 of the database by doubling the number of studies and includes substantial additional curation to the taxonomic accuracy of the records, as well as the metadata. Moreover, we now provide an R package (BioTIMEr) to facilitate use of the database.

Urine is an attractive biomarker analyte for non-invasive longitudinal monitoring of health and disease, particularly for diseases of the genitourinary tract, like prostate and bladder cancer. The composition of an individual's urine reflects both genetic and lifestyle characteristics that differ across geographies and populations, like diet, hydration and other socio-economic factors. While men of African ancestry have elevated prostate cancer risk, it is unclear to what extent this influences urinary biomarkers. We therefore quantified the urinary proteomes of 329 localized prostate cancer patients: 135 self-identifying as White and 194 self-identifying as Black. Introduction: Neonatal hypoxic-ischemic encephalopathy (HIE) is neurological disease caused by the deprivation of oxygen and blood flow to the brain during the developmentally-critical perinatal period. Therapeutic hypothermia (TH) is the standard of care treatment for HIE, though cognitive deficits can persistent throughout life despite treatment. The nature of these deficits, and the impacts of TH and sex are not well understood, and this presents a key barrier in the development of novel therapeutics.

One form of corporal punishment commonly used in the United States is spanking. Spanking is a well-known risk factor for adverse child development, although its influence on children's approaches to learning (ATL) has been largely overlooked. Existing research is particularly limited in inadequately considering multiple confounds and selection biases in children's exposure to spanking. As artificial intelligence (AI) becomes increasingly embedded in healthcare applications, concerns have emerged around the trustworthiness, interpretability, and context-awareness of these systems. Knowledge Organization Systems (KOS) hold considerable potential to address these challenges by supporting semantic standardization, explainability, and domain alignment. This study presents a bibliometric analysis of scholarly publications referencing both AI and healthcare concepts to examine how KOS are positioned within this evolving discourse. The findings indicate that while early literature frequently and explicitly referenced KOS—such as ontologies, controlled vocabularies, and classification systems—their visibility has declined relative to newer paradigms such as machine learning and large language models. …

Limited evidence exists regarding the long-term oncologic safety of biologic therapies, particularly interleukin (IL)-17 inhibitors (IL-17i) and IL-23i, in the management of psoriasis. This propensity-score-matched retrospective-cohort study assessed the risk of developing a primary malignancy within 10-years following exposure to an FDA-approved biologic among psoriasis patients compared to biologic-naïve controls. After propensity-score-matching, 32,230 biologic-exposed patients were further grouped into cohorts of tumor necrosis factor inhibitors (TNFi, n=16,011), IL-23i (n=5,604), IL-12/23i (n=3,856), and IL-17i (n=5,467). During a 10-year period, TNFi-treated patients had a reduced risk of developing any primary malignancy compared to biologic-naïve psoriasis patients [HR 0.80(0.73-0.87)]. Diabetes remains a critical global health challenge, with early detection is crucial for effective management. This study presents a comprehensive benchmarking analysis of 14 diverse machine learning and Bayesian models for early-stage diabetes risk prediction using clinical data [2] from Sylhet, Bangladesh. This research evaluated traditional methods (Logistic Regression, Decision Trees), ensemble techniques (Random Forest, XGBoost, LightGBM), Bayesian approaches (BART, Bayesian Logistic Regression), and advanced neural architectures (Deep Belief Networks) using both 70-30 train-test splits and 10-fold cross-validation. The results demonstrate that ensemble methods consistently outperformed other approaches, with Random Forest(RF) achieving the highest cross-validated AUC (0.9951) and accuracy (0.9699). …

Optimal decision-making requires consideration of internal and external contexts. Biased decision-making is a transdiagnostic symptom of neuropsychiatric disorders. We created a computational model demonstrating how the striosome compartment of the striatum constructs a context-dependent mathematical space for decision-making computations, and how the matrix compartment uses this space to define action value. The model explains multiple experimental results and unifies other theories like reward prediction error, roles of the direct versus indirect pathways, and roles of the striosome versus matrix, under one framework. Reef-building corals are being impacted by increasing seawater temperatures, with marine heatwaves leading to mass bleaching events and extensive mortality. However, corals can display variation in thermal tolerance, both within and among species. In this study, we sampled four coral species (Acropora hyacinthus, Montipora grisea, Pocillopora verrucosa, and Porites lobata) from two sites on Tutuila, American Samoa, with contrasting environmental regimes but similar Maximum Monthly Means (MMM). Host microbiota may impact disease vector behavior and pathogen transmission, but little is known about associations between ectoparasites and microbial communities in wildlife reservoir species. We used Illumina metagenomic sequencing to explore the impacts of tick parasitism on the rodent fecal microbiome in both a field and laboratory setting. We found that tick parasitism on wild hosts was associated with variation in the fecal microbiota of both the white-footed deermouse, Peromyscus leucopus, and the southern cotton rat, Sigmodon hispidus. Mobulas (manta and devil rays) are large-scale ram filter feeders that separate planktonic food particles from large volumes of water with minimal clogging. This contrasts with most human-made filters that can suffer from problematic clogging requiring additional mechanisms for clearing blocked surfaces and maintaining performance. Prior studies have shown that mobulas employ a unique mechanism referred to as ricochet separation to filter feed, whereby captive vortices in filter pores cause particles to bounce off the filter surfaces and away from the filter pores. Soybean (Glycine max) peptide lunasin exhibits significant cancer-preventive, antioxidant, and hypocholesterolemic effects. This study aimed to identify quantitative trait nucleotides (QTNs) associated with lunasin content and to annotate the candidate genes in the soybean genome. The mapping panel of 144 accessions was gathered from the USDA Soybean Germplasm Collection, encompassing diverse geographical origins and genetic backgrounds, and was genotyped using SoySNP50K iSelect Beadchips. Habitat suitability commonly differs between the core and periphery of ecosystems. The longleaf pine ecosystem is recognized for its value in providing habitat to a variety of specialist and endemic species, especially herpetofauna. However, at the northernmost extent of longleaf pine (in southeastern Virginia) little is known about the denizen reptiles and amphibians. Moreover, the remaining longleaf pine ecosystem at the latitudinal ecotone is fragmented amongst a mosaic of other ecosystems. We examined herpetofauna diversity, richness, and capture rates between four focal habitat types—mature longleaf pine, restored longleaf pine, maple-gum swamp, and pocosin-bog—in the Zuni Pine Barrens of southeastern Virginia. …

The activity of nucleoside and nucleotide analogs as antiviral agents requires phosphorylation by intracellular enzymes. Phosphate-substituted analogs have low bioavailability due to the presence of ionizable negatively-charged groups. To circumvent this limitation, several prodrug approaches have been proposed and developed. Herein, we hypothesized that the conjugation of anti-HIV fatty acids with the nucleotide tenofovir alafenamide (TAF) (1) could improve the anti-HIV activity of the nucleotide. Background/Objectives: Neuroendocrine tumors (NETs) are heterogeneous malignancies arising from enterochromaffin cells that can arise from the gastrointestinal (GI) tract and pancreas. Surgical management is the cornerstone of treatment, with the optimal approach tailored by tumor grade, size, location, and presence of metastasis. This review discusses the current strategies for the surgical management of NETs of the gastroenteropancreatic tract. Methods: A review of the available literature was conducted to evaluate surgical approaches to NETs. Consensus guidelines were incorporated to synthesize evidence-based recommendations. Marine sediments are a large reservoir of recalcitrant organic matter and host microbes at subsurface depths exceeding 2.4 kilometers and temperatures up to 120°C, yet the mechanisms supplying bioavailable substrates remain unclear. Here, we investigated 7.8-million-year-old sediment from IODP Site C0012 off the Nankai Trough, Japan, through incubations at 20°, 35°, 55°, and 85°C to simulate burial temperatures. Using 3D fluorescence spectroscopy and ultrahigh-resolution mass spectrometry, we tracked changes in dissolved organic matter (DOM). At 35°C, humic-like DOM was released alongside metal ions, exhibiting low bioavailability. At 55°C, abiotic decomposition of humic compounds generated smaller, more bioavailable DOM, promoting fermentation. …

The “transmembrane-electrostatically localized proton(s)/cation(s) charge(s) (TELC(s), also known as TELP(s)) model” may serve as a theoretical framework to explain protonic cell energetics including both delocalized and localized protonic couplings. TELCs are held by their corresponding transmembrane-electrostatically localized hydroxides anions (TELAs) across the membrane through mutual transmembrane-electrostatic attractive force, which is now calculated to be in the range from 1.96 × 10⁻¹¹ to 2.28 × 10⁻¹¹ newtons (N) across a 2.5 nm thick membrane in a range of transmembrane potential from 10 to 200 mV. This study successfully implemented microcrystal electron diffraction (microED) and X-ray powder diffraction (XRPD) for the crystal structure determination of a new phase, TAF-CNU-1, Ni(C₈H₄O₄)·3H₂O, solved by microED from single microcrystals in the powder and refined at the kinematic and dynamic electron diffraction theory levels. Dissolved organic matter (DOM) reference materials are critical to ensuring reliable comparability of measurements across experiments and between labs. TRM-0522, isolated in 2022 from 45 m deep seawater off of Sweden’s west coast, fills the niche of a previously unavailable coastal marine DOM reference material. After its isolation, we detailed a limited number of metrics for TRM-0522, initially disclosing Orbitrap high-resolution mass spectrometry, nuclear magnetic resonance, carbon proportion, absorbance, and fluorescence data. With TRM-0522 becoming more widely used, a variety of labs have generated different metrics that help to characterize and define the chemical properties of this coastal reference material. …
