(First paragraph) The Virginia Department of Environmental Quality (DEQ) intends to unite elements of its National Coastal Assessment Program (NCA) with its Chesapeake Bay Probabilistic Benthic Monitoring (CBP-PBM) Program beginning in the summer of 2005. The primary motivations for this decision were two-fold. First, the probable reduction of federal grant support for the NCA Program after the summer of 2004 would require considerable restructuring of the of DEQ’s estuarine probabilistic monitoring to continue with a comprehensive program in the future. Coastal habitats that serve as nursery grounds for numerous marine species are badly degraded, yet the traditional means of modeling populations of exploited marine species handle spatiotemporal changes in habitat characteristics and life history dynamics poorly, if at all. To explore how nursery habitat degradation impacts recruitment of a mobile, benthic species, we developed a spatially explicit, individual-based model that describes the recruitment of Caribbean spiny lobster (Panulirus argus) in the Florida Keys, where a cascade of environmental disturbances has reconfigured nursery habitat structure. A molecular dynamics (MD) scheme is combined with a distributed circuit model for a self-consistent analysis of the transient membrane response for cells subjected to an ultrashort (nanosecond) high-intensity (approximately 0.01-V/nm spatially averaged field) voltage pulse. The dynamical, stochastic, many-body aspects are treated at the molecular level by resorting to a course-grained representation of the membrane lipid molecules. Coupling the Smoluchowski equation to the distributed electrical model for current flow provides the time-dependent transmembrane fields for the MD simulations. A good match between the simulation results and available experimental data is obtained. Predictions include pore formation times of about 5-6 …

Cyclic stratification was examined in the Upper Cretaceous (Santonian-Campanian) section (Eagle Formation) within the Bighorn Basin of Wyoming. Of particular concern was the 10³ to 10⁴ year band, which in marine settings has been attributed to orbital forcing (Milankovitch cyclicity). A series of 19 sections were measured through the Virgelle Member of the Eagle Formation. Most were measured on the “J”-shaped escarpment that constitutes the nose and north flank of the Thermopolis anticline. This dissertation presents the identification, molecular structure and expression of defensin, an antimicrobial peptide, in the ticks, Dermacentor variabilis and Ixodes scapularis. A 624 bp cDNA molecule for defensin was cloned and sequenced from hemocytes, fat body and midgut of unstimulated D. variabilis ticks. Defensin peptide was detected only in the hemocyte lysates. These results suggest that the defensin from D. variabilis is synthesized and stored in the hemocytes, although the role of other tissues is not excluded.

The ability of the juvenile stage ticks to express the defensin gene was confirmed. Detection of the defensin transcript in all …

Several species of dinoflagellates feed on a wide variety of food types including fish. However the actual feeding preferences of dinoflagellates have been much less studied. In the few studies that have been carried out on dinoflagellate feeding preference, none have looked at possible factors that may affect preference. This investigation identified the morphological defects caused by different concentrations of chromium, copper, and arsenic (CCA) on the developing zebrafish embryo; in addition to defects caused by the three combined components (complete CCA). A change in the DNA of the developing embryos in response to the three components of CCA and complete CCA was also determined. Early Dania rerio embryos were cultured for five days with various concentrations of CCA components and complete CCA. In response to microbial challenge, invertebrates rely on their innate immune response to recognize and destroy harmful pathogens. One of the primary proteins produced by the response and that has been observed in many invertebrates, including insects, mussels, and ticks, is defensin. These proteins are responsible for forming pores in the membranes of invading microbes thereby destroying them. This study examined the gene coding region for defensin in two important tick disease vectors, Dermacentor variabilis and Ixodes scapularis. In 1855, the town of Portsmouth, Virginia was devastated by an epidemic of yellow fever. Most citizens fled. Of those who remained, most became infected and a thousand died. The municipal government collapsed. In their place, a small organization known as the Portsmouth Relief Association assumed responsibility for ensuring the survival of the town. This organization managed the care of the sick, the burial of the dead, and the care of orphans. It was the sole agent receiving and allocating the funds and resources that poured into the community. Previous research on attitudes towards homosexuality has concentrated on characteristics of individuals, and neglected the capitalist structure of society that provides the contextual setting for the creation and maintenance of cultural attitudes.

This thesis uses responses collected in the general social survey of the United States between 1993 and 1998, to provide a more comprehensive explanation of differences in attitudes towards homosexuality, through an analysis of social position and the capitalist agenda. The analysis revealed significant relationships between sex, age, race, income, beliefs regarding the domestication of women, and attitudes towards homosexuality.

Preimplantation Genetic Diagnosis (PGD) allows for couples to obtain genetic information at the embryo stage, therefore preventing lethal genetic diseases in their offspring. A recent multi-center study determined a 17% amplification failure rate overall and a 3.4% misdiagnosis rate (ESHRE PGD consortium steering committee, 2002). Experiments presented here were conducted to decrease those rates by optimizing single cell sensitivity and specificity of PGD techniques. Primer extension preamplification (PEP) was used after initial amplification failure, followed by gene specific PCR. Reamplification experiments yielded 54% amplification rates using blastomeres that previously failed amplification. In the clinical trial, four of six blastomeres were …

Metastatic colon carcinoma is the second leading cause of death from malignancy in the United States, and development of more effective treatments is essential. Heterologous expression of Herpes Simplex Virus Thymidine Kinase (HSVtk) in combination with the prodrug, ganciclovir (GCV), has shown great promise for the genetic therapy of many cancers, but most patients have had only a partial or minimal response to the therapy. After screening a panel of two drug combinations, our laboratory has shown that the combination of GCV and the protein kinase inhibitor UCN-01 (7-hydroxystaurosporine) enhances tumor cell death more effectively than either drug alone. Synthesis and applications of two types of one-dimensional nanomaterials, carbon nanotubes (CNTs) and tungsten oxide nanorods, are investigated in this dissertation. Multi-walled CNTs have been successfully synthesized using two types of chemical vapor deposition (CVD) methods: microwave plasma enhanced CVD and atmospheric pressure thermal CVD. CNTs and their synthesis processes are characterized with various analysis techniques including scanning electron microscopy (SEM), transmission electron microscopy (TEM), Raman spectroscopy, and optical emission spectroscopy. The current project examined the associations between childhood psychological abuse, disclosure, and perpetrator relationship on sexual attitudes and satisfaction among adults. A total of 274 female graduate and undergraduate psychology students from Old Dominion University were recruited. Participants anonymously filled out the survey online. It was hypothesized that there would be a positive relationship between childhood psychological abuse and sexual communion and permissiveness. In terms of perpetrator relationship, it was hypothesized that as the objective relationship between the individual and perpetrator became closer, sexual attitudes and satisfaction would be more negatively affected. Arglabin-DMA, an analog of farnesyl pyrophosphate (FPP), reportedly inhibits farnesyltransferase (FTase) directly by competitively blocking the binding of Ras protein and its posttranslational modification, as suggested in previous studies. But, the mechanisms by which Arglabin-DMA inhibits tumor growth in vivo and in vitro are still relatively poorly characterized. Modeling and simulating such variables is difficult because the correlations are restricted by the marginal means via Fréchet bounds in a complicated way. In this dissertation we will first discuss partially specified models and methods for estimating the regression and correlation parameters. We derive the asymptotic distributions of these parameter estimates. The primary objective of this study is to document differences between image characteristics of two sources (illustrations in Naville, 1898; and images in the cast of the relief at Virginia Museum of Fine Arts (VMFA) of the Punt relief from the temple of Hatshepsut at Deir El-Bahri. Our second is to compare cast images to photographs of the original relief. Characteristics of 30 species in the illustrations were described and compared to descriptions of the corresponding 30 species photographed from the cast at VMFA.
