Gram positive (G(+)) infections make up similar to 50% of all acute lung injury cases which are characterized by extensive permeability edema secondary to disruption of endothelial cell (EC) barrier integrity. A primary cause of increased permeability are cholesterol-dependent cytolysins (CDCs) of G(+)-bacteria, such as pneumolysin (PLY) and listeriolysin-O (LLO) which create plasma membrane pores, promoting Ca2+-influx and activation of PKC alpha. In human lung microvascular endothelial cells (HLMVEC), pretreatment with the nitric oxide synthase (NOS) inhibitor, ETU reduced the ability of LLO to increase microvascular cell permeability suggesting an endothelial nitric oxide synthase (eNOS)-dependent mechanism. Sea turtles reside in different acoustic environments with each life history stage and may have different hearing capacity throughout ontogeny. For this study, two independent yet complementary techniques for hearing assessment, i.e. behavioral and electrophysiological audiometry, were employed to (1) measure hearing in post-hatchling and juvenile loggerhead sea turtles Caretta caretta (19-62 cm straight carapace length) to determine whether these migratory turtles exhibit an ontogenetic shift in underwater auditory detection and (2) evaluate whether hearing frequency range and threshold sensitivity are consistent in behavioral and electrophysiological tests. Cephalopods have visual and mechanoreception systems that may be employed to sense and respond to an approaching predator. While vision presumably plays the dominant role, the importance of the lateral line analogue for predator evasion has not been examined in cephalopods. To test the respective roles of vision and the lateral line analogue, brief squid, Lolliguncula brevis, were observed in the presence of summer flounder, Paralichthys dentatus, under light and dark conditions with their lateral line analogue intact and ablated. Transcriptome Of The Female Synganglion Of The Black-Legged Tick Ixodes Scapularis (Acari: Ixodidae) With Comparison Between Illumina And 454 Systems, Noble Egekwu, Daniel E. Sonenshine, Brooke W. Bissinger, R. Michael Roe

Illumina and 454 pyrosequencing were used to characterize genes from the synganglion of female Ixodes scapularis. GO term searching success for biological processes was similar for samples sequenced by both methods. However, for molecular processes, it was more successful for the Illumina samples than for 454 samples. Functional assignments of transcripts predicting neuropeptides, neuropeptide receptors, neurotransmitter receptors and other genes of interest was done, supported by strong e-values (<-6), and high consensus sequence alignments. Transcripts predicting 15 putative neuropeptide prepropeptides ((allatostatin, allatotropin, bursicon α, corticotropin releasing factor (CRF), CRF-binding protein, eclosion hormone, FMRFamide, glycoprotein A, insulin-like peptide, ion transport peptide, myoinhibitory peptide, inotocin ( = neurophysin-oxytocin), Neuropeptide F, sulfakinin and SIFamide)) and transcripts predicting receptors for 14 neuropeptides (allatostatin, calcitonin, cardioacceleratory peptide, corazonin, CRF, eclosion hormone, gonadotropin-releasing hormone/AKH-like, insulin-like peptide, neuropeptide F, proctolin, pyrokinin, SIFamide, sulfakinin and tachykinin) are reported. Similar to Dermacentor variabilis, we found transcripts matching pro-protein convertase, essential for converting neuropeptide hormones to their mature form. Beginnings in Toni Morrison's novels enact an uncanny moment of disorientation. They are beginnings in medias res, and, more importantly, beginnings of spatial deictic uncertainties that leave a reader with the absence of a stable system of reference. They enact the predicament of a beginning that precludes the fantasy of an absolute point of origin. Morrison's beginnings self-consciously advocate an imperative to engage in a continual process of re-reading, of revisiting the initial disorientation so as to avoid a "conclusion to living." BACKGROUNDS: Electronic medical records (EMR) form a rich repository of information that could benefit public health. We asked how structured and free-text narrative EMR data should be combined to improve epidemic surveillance for acute respiratory infections (ARI).

METHODS: Eight previously characterized ARI case detection algorithms (CDA) were applied to historical EMR entries to create authentic time series of daily ARI case counts (background). An epidemic model simulated influenza cases (injection). From the time of the injection, cluster-detection statistics were applied daily on paired background+injection (combined) and background-only time series. Once every two days in the United States, a child aged 5 to 14 years old dies by suicide (World Health Organization, 2010). When viewed on a spectrum that includes suicidal ideation, verbalizations, behaviors, and attempts, the problem of youth suicidality is actually far greater than the numbers of completed suicides suggest (Cheng, Tao, Riley, Kann, Ye, Tian...Hu, 2009). This study examined suicidality in children ages 10 and younger, and included the characteristics of suicidal children, factors that influence childhood suicidality, and treatment implications. This dissertation focuses on the dynamics and bioeffects of electroporation of biological cell and ionic conduction in nanopores under high-intensity, nanosecond pulses. The electroporation model utilized the current continuity equation and the asymptotic Smoluchowski equation to explore the transmembrane potential and pore density of the plasma and intracellular membranes; the ionic conduction model employed the Poisson-Nernst-Planck equations and the Navier-Stokes equations to analyze the ionic current and ion concentration profile.

The high rates of mobility in the U.S. can produce negative consequences for children's academic achievement. The purpose of this study was to determine relationships among math and reading academic achievement, mobility characteristics, student characteristics, and school characteristics in order to develop a model to predict achievement using these variables. Using such a model, educational stakeholders could identify students that are at risk for academic failure. The study included 523 third grade students from a high poverty, predominantly Latino, suburban district. This paper examines the relationship between conscription laws (mandatory military or social service) and crime rates across countries. This cross-national study focuses on three major crimes: burglary, robbery, and homicide. In addition to conscription laws several control variables have been included in the analyses are: percent of the labor force that is military, level of civil liberties/freedom, level of industrialization, illiteracy rates, percent of urban population, unemployment rates, percent under the international poverty line, income disparity (measured using the Gini index), and population.

Haemulids are one of the most ecologically and commercially important groups of near-shore fishes. They are very diverse, with 145 putative species belonging to 18 genera. The phylogenetic relationships of the genera within the haemulids, however, are uncertain and the limits and relationships with other percomorphs are undefined.

Here, I present the first comprehensive phylogenetic hypothesis for the family based on a combined dataset of five genes (4731 bp; 16 genera, 56 species). Results show strong support for a monophyletic Haemulidae with the inclusion of the former inermiids. However, results of the analyses call into question the monophyly of a …

Several viruses, including viruses that cause cancer, contain conserved zinc finger (ZF) proteins that are essential for viral reproduction, making them attractive drug targets for cancer and viral treatment. ZFs are small protein domains that have Zn2+ tetrahedrally coordinated to at least 2 Cys and His. They form three classes of ZFs depending on the amino acid ligands, CCHH, CCCH, and CCCC. Zn2+ is stable towards redox reactions; however, the Cys thiolates are redox active. Building on the extant research of on-again/off-again (on-off) romantic relationships, the current thesis focused on building upon past findings by utilizing a unique theoretical methodology in an emerging demographic. A sample of 22 emergent adult (ages 18-29) participants who were currently in or had recently experienced an on-off relationship completed face-to-face interviews discussing communicative processes during romantic reconciliation. Psychopathic and sensation seeking traits are often correlated; however, sensation seeking alone is not inherently pathological. This study seeks to investigate possible moderating variables between individuals who are high on sensation seeking but low on measures of psychopathic or antisocial traits. Specifically, a positive family environment is hypothesized to be a moderating variable in the development of psychopathic traits among high sensation seekers. A college student sample assessed for psychopathy, sensation seeking, and family functioning is used to test this hypothesis. Significant relationships between all three constructs were found. Casitas are artificial structures placed on the seabed by fishermen to aggregate Caribbean spiny lobsters (Panulirus argus) for ease of capture. Some researchers suggest that they may also enhance lobster populations in shelter-limited environments. Conversely, aggregation of lobsters within casitas may be detrimental to the population if the nutritional condition or mortality of lobsters is poorer in casitas than in natural shelters. Small juvenile lobsters may be at particular risk because their foraging range is smaller and they are more readily preyed upon than larger lobsters. Net community metabolism (NCM), the balance between primary production and community respiration, was measured during two cruises, June 2011 and August 2012, in the Middle Atlantic Bight, Georges Bank and Gulf of Maine. Net heterotrophy was observed in Georges Bank and Gulf of Maine during the June 2011 cruise. During the August 2012 cruise, net heterotrophy was observed in the Middle Atlantic Bight and net autotrophy was observed in Georges Bank and the Gulf of Maine. Concentrations of atmospheric CO2 are expected to double by year 2100 as a result of anthropogenic activities. Under elevated CO2 conditions, cyanobacteria may reallocate energy from active accumulation and transport of dissolved inorganic carbon (C) required for photosynthesis to other growth processes. Organic Copper Binding Ligands And Thiol Compounds Produced By Bacteria And In The Elizabeth River, Virginia, Christina Louise Dryden

This dissertation presents work focusing on copper and organic copper binding ligands in laboratory cultures and the Elizabeth River, Virginia. Laboratory cultures of the marine bacterium Vibrio parahaemolyticus were used to demonstrate the influence of elevated copper concentrations on copper-complexing ligand and thiol production. Copper-complexing ligands similar in binding strength to the strongest natural ligands were detected in V. parahaemolyticus cultures (log K′CuL = 11.8–13.2). A strong correlation (r2 = 0.973) was found between total thiol and copper-complexing ligand concentrations at all copper concentrations examined.

Short-cut nitrogen removal avoids nitrite oxidation to nitrate by nitrite oxidizing bacteria (NOB) and allows a) reduction of formed nitrite to nitrogen gas via heterotrophic denitrification and/or b) oxidation of remaining ammonia with formed nitrite to nitrogen gas via anaerobic ammonia oxidation (anammox). The precondition for achieving shortcut nitrogen removal is suppression of NOB, which is favored by warm and high ammonia strength conditions found in internally generated ammonia-rich waste streams through anaerobic digestion of waste solids referred to as sidestreams or reject water. In the second half of the nineteenth century, a retired Victorian undertaker named William Banting (1796-1878) dramatically altered attitudes toward fat by initiating the profoundly consequential idea of the diet as a saleable commodity capable of marking identity within particular social and racial contexts and connecting obesity with degeneracy, illness, and evil. His work Letter on Corpulence Addressed to the General Public self-published in 1863 describes how, with physician William Harvey, Banting reduced his weight by nearly fifty pounds by following a high-protein, low-carbohydrate diet. Banting and his dieting phenomenon transformed the English cultural consciousness of fatness, and created a …
