We have developed a novel method that enables global subtelomere and haplotype-resolved analysis of telomere lengths at the single-molecule level. An in vitro CRISPR/Cas9 RNA-directed nickase system directs the specific labeling of human (TTAGGG) n DNA tracts in genomes that have also been barcoded using a separate nickase enzyme that recognizes a 7bp motif genome-wide. High-throughput imaging and analysis of large DNA single molecules from genomes labeled in this fashion using a nanochannel array system permits mapping through subtelomere repeat element (SRE) regions to unique chromosomal DNA while simultaneously measuring the (TTAGGG) n tract length at the end of each …

Purpose: The purpose of this study was to examine factors that might influence the treatment effectiveness of motor learning guided (MLG) treatment approach for apraxia of speech (AOS). Specifically, this study examined the effects home practice and the stimuli selection on speech production.

Method: This is a case study across two treatment cycles involving a 52 year-old male five months post left CVA (due to a carotid artery dissection). Each treatment cycle used three conditions of practice to investigate the influence of practice frequency on treatment outcomes. Nanosecond pulsed electric fields (nsPEF) are emerging as a novel modality for cell stimulation and tissue ablation. However, the downstream protein effectors responsible for nsPEF bioeffects remain to be established. Here we demonstrate that nsPEF activate TMEM16F (or Anoctamin 6), a protein functioning as a Ca2+-dependent phospholipid scramblase and Ca2+-activated chloride channel. Using confocal microscopy and patch clamp recordings, we investigated the relevance of TMEM16F activation for several bioeffects triggered by nsPEF, including phosphatidylserine (PS) externalization, nanopore-conducted currents, membrane blebbing, and cell death. An adhesive organ is a prominent, protruding mucus secreting gland that is used by newly hatched tadpoles and larvae of some fishes to attach to aquatic vegetation.  The objective of this research is to test the hypothesis that newly hatched cyprinid larvae of Hybognathus hankinsoni, Notemigonus crysoleucas, Cyprinus carpio and Gila atraria contain cephalic adhesive organs.  Newly hatched larvae of Semotilus atromaculatus, which do not attach to submerged aquatic vegetation, were used as the control.  SEM examination of newly hatched larvae indicate there were no adhesive organs on the control species (S. atromaculatus) or test species …

We monitored bat activity on the upper Coastal Plain of Virginia using mist nets and acoustic detection (ANABAT) during April-October 2000 and April-August 2001. We classified forty sites into three forest-cover classes (pine forest, mixed pine, and hardwood forest) and three landscape-feature classes (permanent water, riparian corridor, and upland). We captured 406 bats (8 species) in mist nets; red bats (Lasiurus borealis; n = 281), big brown bats (Eptesicus fuscus; n = 47), and eastern pipistrelles (Pipistrellus subflavus; n = 36) were the most commonly captured species. Because ground-nesting wild turkeys (Meleagris gallopavo) may sustain high incidences of nest predation in western Virginia, determining their predators is essential to understanding risk and managing the birds.  Our study investigated potential predators of wild turkey nests at the Radford Army Ammunition Plant, New River Unit (RFAAP; Pulaski Co., in western Virginia). Here, we established 8 artificial nests during the breeding season for wild turkey (March-April, 2017), and documented predators via game cameras.  Thirty-one species of mammals and birds visited the nests over the 31-day study. The distributions of the four most abundant isotopologues and isotopomers (N2O, 15NNO, N15NO, and NN18O of nitrous oxide have been measured in the Earth's stratosphere by infrared remote sensing with the Atmospheric Chemistry Experiment (ACE) Fourier transform spectrometer. These satellite observations have provided a near-global picture of N2O isotopic fractionation. The relative abundances of the heavier species increase with altitude and with latitude in the stratosphere as the air becomes older. The heavy isotopologues are enriched by 20-30% in the upper stratosphere and even more over the poles. These observations are …

The objectives of this research were twofold: test the infiltration rate of pervious concrete before and after vacuum cleaning and assess infiltration variability over time. Infiltration tests were performed on a 558.2 m2 area of pervious concrete, divided into sixteen 0.35 m2 test areas in a parking area at the Science Museum of Virginia in Richmond, Virginia on five dates from November 4-December 30, 2013. Average infiltration rates (avg.= 22.8 and 36.3 L/m2/min) immediately after vacuuming were lower than those (avg.=30.7 and 41.3 L/m2/min) before vacuuming. The juxtaposition of increasing suicide rates with continued calls for suicide prevention efforts begs for new approaches. Grounded in the Centers for Disease Control and Prevention (CDC) framework for tackling health issues, this personal views work integrates relevant suicide risk/protective factor, assessment, and intervention/prevention literatures. Based on these components of suicide risk, we articulate a Social-Ecological Suicide Prevention Model (SESPM) which provides an integration of general and population-specific risk and protective factors. Neonatal hypoglycemia is the most common metabolic abnormality in infants and is associated with neurological damage and death. The risk of developing hypoglycemia among infants born from diabetic mothers is even higher. Particle Swarm Optimization (PSO) has received increasing attention from the evolutionary optimization research community in the last twenty years. PSO is a metaheuristic approach based on collective intelligence obtained by emulating the swarming behavior of bees. A number of multi-objective variants of the original PSO algorithm that extend its applicability to optimization problems with conflicting objectives have also been developed; these multi-objective PSO (MOPSO) algorithms demonstrate comparable performance to other state-of-the-art metaheuristics. The black-legged tick Ixodes scapularis transmits the human anaplasmosis agent, Anaplasma phagocytophilum. In this study, we show that A. phagocytophilum specifically up-regulates I. scapularis organic anion transporting polypeptide, isoatp4056 and kynurenine amino transferase (kat), a gene involved in the production of tryptophan metabolite xanthurenic acid (XA), for its survival in ticks. RNAi analysis revealed that knockdown of isoatp4056 expression had no effect on A. phagocytophilum acquisition from the murine host but affected the bacterial survival in tick cells. Knockdown of the expression of kat mRNA alone or in combination with isoatp4056 mRNA significantly affected A. phagocytophilum survival …

Pulsed Electric Fields (PEF) have promised improved treatment results in a variety of cancer types including melanoma, pancreatic and lung squamous cancer. Recent studies show that PEF-based cancer therapy may be improved further with the assistance of moderate heating of the target. Experiments were performed to design, calibrate and implement a feedback-looped infrared laser irradiation system that could maintain specified temperatures during the treatment. Southeast Asia includes the Coral Triangle, a marine biodiversity hotspot that supports important fishery resources experiencing varied threats. Patterns of speciation and population structure in the Coral Triangle have been examined to test hypotheses relating to the historical geologic processes that may have influenced this biodiversity phenomenon. This thesis unlocks the lifespan story of nine Black participants as they reflected on the communicative practices that guided their career journey towards becoming a teacher. Through the lens of Bronfenbrenner’s (1979) socio-ecological development model, the microsystem, mesosystem, exosystem, macrosystem, and chronosystem were examined to understand the content of career-related memories and with whom or what the communicative experiences occurred with across the participants’ lifespan. Low molecular weight gelators (LMWGs) are an interesting class of compound that have gained considerable attention especially because of their potential applications for soft biomaterials. Carbohydrate based low molecular weight hydrogelators and organogelators are able to selfassemble and form ordered supramolecular structures, which are useful for exploring biomedical applications. The resulting gelators are responsive to external stimuli because of the weak noncovalent intermolecular forces and interaction that influences self-assembly. Through the years, various forms of media have become political institutions able to influence the public policy agenda. Analyzing policy helps to promote a better understanding of how the policy process works, how problems appear on the political agenda, and why governments choose to act or not to act on particular policy problems. This research utilizes Baumgartner and Jones’s (1993) Theory of Punctuated Equilibrium (PET) to examine the ways in which media attention influenced public policy on marijuana in the states of Kentucky, Massachusetts, and Vermont between 1996 and 2016.
