Today, the eastern equatorial Pacific (EEP) plays a critical role in the global CO₂ budget as a major source of CO₂ to the atmosphere, but recent studies suggest the region may shift to a sink for atmospheric CO₂ under different climate states. Here, I focus on two transitional periods, the last deglaciation (25 kyr to present) and last glaciation (the Marine Isotope Stage (MIS) 5a-4 transition, 96 to 60 kyr), to investigate how the carbon system in the EEP responds to major climate changes. The Last of Us and The Walking Dead video games deploy father-daughter relationship pairings between their main characters in ways that disrupt the hegemonic patriarchal understandings of those very roles, though in different ways. The Last of Us and The Walking Dead utilize paternal mentorship in ways that subvert patriarchal ideology’s established patterns for gendered behavior through role-switching and alternative models of masculine care respectively. Where video games too often still cater to an audience that is heterosexual, white, and male, these games feature narratives that challenge the heteropatriarchal messaging common to this medium. The overarching goal is to predict three-dimensional structures from their amino acid sequences. Gene electrotransfer (GET) holds great promise for the delivery of therapeutic agents. The skin serves as an attractive target for GET due to its availability and unique cellular composition. Protein replacement therapy and DNA vaccination are potential applications for intradermal GET. The combination of moderate tissue preheating and GET has been shown to achieve elevated gene expression levels while reducing the necessary applied voltage. In the current work, we utilized a 16-pin multi-electrode array (MEA) and incorporated nine optical fibers, connected to an infrared laser, to pre-heat the tissue to 43°C before application of GET. More frequent storms due to climate change may impact estuarine species such as the mummichog (Fundulus hetereoclitus), an ecologically important salt marsh fish. This study investigated the effect of storm events and month on consumption of terrestrial insects by mummichogs in Hoffler Creek, Portsmouth, VA, as well as the effect of storms on consumption of major categories of benthic prey. Samples were taken monthly in the summers of 2017 and 2019. This study examined the role motivational dispositions had on completing a massive open online course (MOOC) using identifiable data from 10,726 students who enrolled in an iteration of the HarvardX MOOC, Super Earths and Life. As part of the course registration process, learners had the option to complete a pre-course survey and self-report information including their level of education, gender and registration motivations. Using these pre-course survey responses, latent profiles linked to learners’ course performance were created. Results showed education background, gender, and motivation were all significantly related to students’ performance. Prostate apoptosis response-4 (Par-4) is an apoptosis-inducing tumor suppressor protein. Full-length Par-4 has previously been shown to be a predominantly intrinsically disordered protein (IDP) under neutral conditions, with significant regular secondary structure evident only within the C-terminal coiled coil domain. However, IDPs can gain ordered structure through the process of induced folding, which often occurs under non-neutral conditions. Previous work has shown that the Par-4 leucine zipper, which is a subset of the C-terminal coiled coil domain, is disordered under neutral conditions, but forms a dimeric coiled coil at acidic pH. Latinx students are likely to enter postsecondary education at a community college. This phenomenon has led to the increase in community colleges being designated as Hispanic Serving Institutions. The designation of Hispanic Serving is not driven by mission, but rather by number of enrolled students who identify as Latinx. Dispersal is a critical life-stage with consequences not only for the individual, but for population dynamics and thus the fate of the whole species. The creation of dispersal coalitions can lead to complex outcomes and affect how changes in abundance occur on the landscape. I examined population dynamics and dispersal coalitions in a population of cooperatively breeding acorn woodpeckers in central California, USA. Using a 34-year dataset on occupancy and abundance, I determined that increases in abundance were determined by dispersal to new locations. Although African-American students start STEM majors with higher levels of interest compared to their racial majority peers, they drop out of these majors at higher rates. One often tested explanation for this racial disparity is stereotype threat–the anxiety related to being judged stereotypically or the fear of confirming such stereotypes. Stereotype threat negatively impacts academic outcomes through a variety of psychological mechanisms including declined motivation. Accordingly, in this study, I examined expectancy-value beliefs as motivational mechanisms for the effects of stereotype threat on STEM outcomes. Although this process is necessary for system security, users view authentication as a frequent disruption to their primary tasks. This thesis demonstrates the unique correlation between myth and the propagation of narrative across generational boundaries. It argues that myth occurs in the intersection of belief, semiotics, and context, and further enables a way of re-encoding a narrative with a dual contextuality. This dual context preserves a narrative’s literal context while endowing it with a new or modified myth context and affords the audience a selection of choices for how to receive a narrative experienced as myth. The human pathogen Clostridioides difficile is increasingly tolerant of multiple antibiotics and causes infections with a high rate of recurrence, creating an urgent need for new preventive and therapeutic strategies. The stringent response, a universal bacterial response to extracellular stresses, governs antibiotic survival and pathogenesis in diverse organisms but has not previously been characterized in C. difficile. This dissertation explores the ability of C. difficile to mount the stringent response. The bacteria encode a full-length, canonical bifunctional Rel/Spo Homolog or RSH enzyme. C. difficile RSH is incapable of utilizing GTP as a substrate but readily synthesizes putative 5’-pGpp-3’ alarmones. …

This research project investigated the impacts of urbanization on plant species biodiversity within Southeastern Virginia. The hypothesized results of this research were if there was an increase in urbanization, then plant diversity would decrease. Plant biodiversity can increase the decomposition rates, pollination success, and biomass production of the area. Decreasing in plant biodiversity can increase the spread of pathogens through plant populations and increase herbivore damage. There was data compiled from SERNEC herbarium records of four counties, Accomack, Northampton, Norfolk, and Virginia Beach. Exposure to metals poses potential health risks, including insulin resistance (IR), to those exposed to them in excess. Limited studies have examined such risks in occupational workers, including welders, and these have yielded inconsistent results. Thus, we examined the associations between exposure to welding metals and IR in welders. We recruited 78 welders and 75 administrative staff from a shipyard located in northern Taiwan. Past studies examining the health outcomes of diabetes mellitus (DM) patients found that social determinants of health disparities were associated with variabilities in health outcomes. However, improving access to healthcare, such as health insurance, should mitigate negative health outcomes. The aim of the study was to explore the association between four types of health insurance, namely, Medicare Fee-For-Service (FFS), Medicare Managed Care (MC), Private FFS, and Private MC plans, and the health outcomes of DM patients, controlling for patients’ social determinants of health.
