Engineers need to develop professional skills, including the ability to work successfully in teams and to communicate within and outside of their discipline, in addition to required technical skills. A collaborative multi-disciplinary service learning project referred to as Ed+gineering was implemented in a 100-level mechanical engineering course. In this collaboration, mechanical engineering students, primarily in the second semester of their freshman year or first semester of their second year, worked over the course of a semester with education students taking a foundations course to develop and deliver engineering lessons to fourth or fifth graders. Gastrointestinal (GI) symptoms, such as nausea and gas, are common problems for athletes in endurance sport. There is considerable evidence that psychological factors influence GI function, but little research has evaluated this in the context of exercise-induced GI symptoms. The overall purpose of this dissertation was to explore the role of psychological factors in the incidence and management of GI symptoms during endurance running. Study 1 assessed associations between several psychological factors, GI symptoms, and nutrition intake before and during runs. The purpose of this study was to describe faculty member experiences related to identifying and addressing prohibited speech in the classroom. The researcher studied faculty members at University of North Carolina (UNC) system institutions using a multiple case study research approach based on the constructivist paradigm. For the purposes of this study, prohibited speech was defined as behaviors that fall into any category deemed not protected or prohibited in case law by the Supreme Court of the United States. These prohibited behaviors included Harassment, Obscenity, Defamation/Libel, Incitement, and True Threats.

Low molecular weight gelators (LMWGs) are attractive molecules that have been explored extensively due to their practical applications in many disciplines. These small molecules self-assemble forming solid-like gels via three-dimensional cross-linked networks with the solvent as the key component within the matrix. As the Arctic experiences consequences of climate change, a shift from thicker, multi-year ice to thinner, first-year ice has been observed. First-year ice is prone to extensive pools of meltwater (“melt ponds”) forming on its surface, which enhance light transmission to the ocean. Changes in the timing and distribution of melt pond formation and associated increases in under-ice light availability are the primary drivers for seasonal progression of water column primary production and warming. Current literature explains that events resulting from instances of white supremacy and racism are traumatizing for Black individuals (Phillips, 2020). Prior literature illuminates the impact of racism and resulting race-based trauma on Black individuals but is lacking regarding the lived experiences of race-based trauma in Black individuals in the U.S. and Black individuals from Jamaica. Young adults are vulnerable to a range of mental health concerns and tend to drink in high quantities and tend to not seek help for these concerns. Specifically, college students involved in athletics tend to have low help-seeking rates—though help-seeking research for this population is relatively limited. Athletic identity (i.e., identification with the athlete role) is a relevant construct for examining this population, however little is known about its association with help-seeking beliefs and ideas. Binge eating is a prominent concern with 2.8 million Americans meeting criteria for binge eating disorder and an additional 10-15% reporting loss of control and overeating behaviors that fail to meet diagnostic criteria. Despite the risk associated with binge eating in emerging adulthood, studies exploring differences in binge eating between Black and White college women have been limited. Black women may be more likely than White women to deny disordered eating behaviors, such as binge eating, due to pressure to reflect historical body positive ideals and heightened stigma regarding mental health issues in Black communities. Thyroid hormones (THs) in mammalian tissues are crucial for development and maintaining metabolic homeostasis. Iodothyronine deiodinases (Dios) remove iodines from THs by a selenocysteine (Sec) residue, which either activates or inactivates them. Halogen bonding (XB) has been proposed to describe the interaction between the Se and I atoms of the T4-Dio complex. Disruption of TH homeostasis by xenobiotics, such as polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) can cause deleterious effects on the endocrine system. Experimental studies have indicated that PBDEs and PCBs could disrupt TH homeostasis by inhibiting Dio through XB formation. However, no current quantitative study exists …

Stress is a known contributor to immune system suppression associated with higher illness susceptibility, including acute infectious respiratory illness or the common cold. Sleep quality is an additional mechanism that may underlie the association between stress and cold symptomatology. Although the associations between stress and sleep and cold symptomatology have been examined separately, little is known about the mechanistic role of sleep in these associations. The present study fills that void by examining archival data from the Common Cold Project (Pittsburgh Cold Study 3). Interface problems have many applications in fluid dynamics, molecular biology, electromagnetism, material science, heat distribution in engines, and hyperthermia treatment of cancer. Mathematically, interface problems commonly lead to partial differential equations (PDE) whose in- put data are discontinuous or singular across the interfaces in the solution domain. Alexandrium tamerense is a well-studied dinoflagellate known for its ability to produce the neurotoxin that causes paralytic shellfish poisoning. Until 1970 Alexandrium tamerense was only found in Europe, North America, and Japan but has been increasingly found all over the globe. Alexandrium is characteristically found in temperate and subtropical regions and as the Arctic warms, there is considerable concern that it may be expanding into the Arctic. Marine microbial dinitrogen (N2) fixation, the conversion of gaseous N2 to bioavailable species, is the primary source of new oceanic nitrogen (N). N is present in nucleic acids, amino acids, and proteins, and is essential to all life. Nano-Pulse Stimulation (NPS), a pulsed power-derived technology, stimulates structural and functional changes in plasma membranes and cellular organelles. NPS induces a Ca2+ influx and opening of the mitochondrial permeability transition pore (mPTP) that dissipates the mitochondrial membrane potential (ΔΨm) and, when sustained, induces regulated cell death. Here we show that in rat cardiomyoblasts (H9C2) cyclophilin D (CypD) is a mitochondrial sensor for NPS as defined by observations that loss of ΔΨm is Ca2+ and mitochondrial reactive oxygen species (mROS) dependent and cyclosporin A (CsA)-sensitive, which are diagnostic qualities for effects on CypD and the mPTP. …

Dinitrogen (N2) fixation is the process of taking widely abundant but mostly biologically inaccessible N2 gas and converting it into more biologically accessible forms of the essential macronutrient nitrogen. Only a small fraction of organisms known as diazotrophs can perform biological N2 fixation. Trichodesmium is one such genus of N2-fixing marine cyanobacteria that is commonly observed in waters along the West Florida Shelf (WFS). We hypothesize that the two main Trichodesmium clades (T. erythraeum and T. thiebautii) occupy distinct environmental niches, one being more coastal and one being more oceanic. To test …

Prostate apoptosis response-4 (Par-4) is a pro-apoptotic tumor suppressor protein. We have shown that this 38 kDa full-length Par-4 (Fl-Par-4) protein is predominantly intrinsically disordered in vitro. In vivo, Par-4 is cleaved by caspase-3 at Asp-131 to generate a 24 kDa functionally active cleaved Par-4 (cl-Par-4) fragment. The cl-Par-4 protein inhibits the NF-κB-mediated cell survival pathway and causes selective apoptosis in various tumor cells. Our laboratory is interested in how the disorder-order balance within Fl-Par-4 and cl-Par-4 may be related to the balance between cell survival and cell death. The Arctic Ocean is a rapidly changing environment, and a key observational system for monitoring climate change. The Arctic is going under a rapid transition from thicker, multi-year ice, to thinner first-year ice, that may have many potential consequences. The upper Roanoke River has 11 species of Catostomidae including Thoburnia rhothoeca, Torrent Sucker; Moxostoma cervinum, Blacktip Jumprock; and Moxostoma ariommum, Bigeye Jumprock. Resource partitioning appears to be a key component of maintaining diverse fish assemblages with habitat and food partitioning cited as especially important in communities containing members of the same family. The diets of these species have been documented in previous work revealing only modest differences among them. Snorkeling observations and subsequent quantification of microhabitat were conducted to illuminate habitat partitioning among these morphologically and ecologically similar species. LGBTQ students are ubiquitous on community college campuses across the United States. The exact number of LGBTQ students is unknown and often their needs are ignored. LGBTQ students face harassment and discrimination at higher rates than other minority groups. This study was conducted to gather the current narratives of LGBTQ students attending community colleges in the Appalachian Region. This study aimed to determine what LGBTQ students are experiencing on community college campuses across Appalachia. Concussion is the most common form of traumatic brain injury (TBI). However, there is a disproportionate level of understanding between the acute and chronic impairments associated with traumatic brain injury. Specifically, problems maintaining balance during standing and walking are cardinal signs of acute concussion, but the temporal extent to which postural control deficits remain following the initial injury are not well defined or understood. Underwater sound is a cue used by many marine larvae to orient to coastal habitats including backreef, sponge-dominated hardbottom habitat in the Florida Keys (Florida, USA) – a particularly “noisy” coastal habitat. However, the distance over which acoustic cues are attractive to settlement-stage larvae - is generally unknown. I examined this phenomenon in a region of the Florida Keys where mass sponge die-offs have diminished both underwater soundscapes and larval settlement. Ticks were removed from three species of Malagasy tortoises, Astrochelys yniphora, A. radiata, and Pyxis arachnoides (comprising two subspecies P. a. arachnoides and P. a. oblonga), between 2012 and 2015. The ticks were presumed to be from the genus Amblyomma. Ticks were morphologically identified and then checked molecularly to confirm their classification or identify any ticks that could not be morphologically identified. Molecular identification was done via end-point PCR that amplified tick cytochrome oxidase (CO1) and tick 12S rRNA genes. Ticks were screened via a real-time polymerase chain reaction assay for the presence of Rickettsia spp. …

Vector-borne diseases (VBDs) are illnesses transmitted to humans and other animals by arthropods such as ticks, mosquitoes, and fleas. These arthropod vectors transmit infectious pathogens such as viruses, bacteria, and protozoa, to humans during blood-feeding. We have very few control strategies to treat or control these diseases. Human anaplasmosis, caused by the bacterium Anaplasma phagocytophilum, is the second most common tick-borne disease in the United States. This work defines three studies elucidating Anaplasma phagocytophilum-mediated modulation of cell signaling in mammalian cells and arthropod vector Ixodes scapularis ticks.

Understanding the relationship between ecological characteristics and genetic change in natural populations in different time scales can reveal how anthropogenic stressors affect natural populations and can improve the success of conservation strategies. The purpose of the Philippines Partnerships for International Research and Education (PIRE) project is to examine levels of genetic change between historical fish samples collected by the USS Albatross expedition in the early 1900s in the Philippines and contemporary populations collected at the same localities. The selection and computation of meaningful features is critical for developing good deep learning methods. This dissertation demonstrates how focusing on this process can significantly improve the results of learning-based approaches. Specifically, this dissertation presents a series of different studies in which feature extraction and design was a significant factor for obtaining effective results. The first two studies are a content-based image retrieval system (CBIR) and a seagrass quantification study in which deep learning models were used to extract meaningful high-level features that significantly increased the performance of the approaches.
