Individuals with diabetes mellitus frequently develop severe skin and soft tissue infections (SSTIs) that are recalcitrant to antibiotic treatment. We examined how diabetes affects the emergence of antibiotic resistance in a Staphylococcus aureus SSTI. We determined that S. aureus evolves antibiotic resistance rapidly in diabetic mice, while resistance did not occur in nondiabetic mice over the course of infection. Diabetes-associated immune cell dysfunction plays a minor role in the emergence of resistance, while hyperglycemia plays a dominant role facilitating the expansion and takeover of resistant mutants in diabetic infections. In this study, a new hybrid nanoparticle composed of magnesium hydroxide and copper oxide (Mg(OH)2/CuO) with an optimized ratio of magnesium (Mg) to copper (Cu) was designed and incorporated into a 3D-printed scaffold made of polycaprolactone (PCL) and gelatin. These hybrid nanostructures (MCNs) were prepared using a green, solvent-free method. Their topography, surface morphology, and structural properties were characterized using scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS). Background
Photon-counting CT (PCCT) systems acquire multiple spectral measurements at high spatial resolution, providing numerous image quality benefits while also increasing the amount of data that must be transferred through the gantry slip ring.
Purpose
This study proposes a lossy method to compress photon-counting CT data using eigenvector analysis, with the goal of providing image quality sufficient for applications that require a rapid initial reconstruction, such as to confirm anatomical coverage, scan quality, and to support automated advanced applications. The eigenbin compression method was experimentally evaluated on a clinical silicon PCCT prototype system.

Aims. A new dataset of collisional rate coefficients for transitions between the rotational states of H2O collided with H2 background gas is developed. The goal is to expand over the other existing datasets in terms of the rotational states of water (200 states are included here) and the rotational states of hydrogen (10 states). All four symmetries of ortho- and para-water combined with ortho- and para-hydrogen are considered.

Rhythmic, daily fluctuations in minute ventilation are controlled by the endogenous circadian clock located in the suprachiasmatic nucleus (SCN). While light serves as a potent synchronizer for the SCN, it also influences physiology and behavior by activating Brn3b-expressing, intrinsically photosensitive retinal ganglion cells (ipRGCs). It is currently unclear the extent to which the external light environment shapes daily ventilatory patterns independent of the SCN. Quadriceps weakness after anterior cruciate ligament reconstruction (ACLR) is a well-known phenomenon, with more persistent quadriceps weakness observed after ACLR with a bone-patellar tendon-bone or quadriceps tendon autograft than with a hamstring tendon autograft. Longstanding quadriceps weakness after ACLR has been associated with suboptimal postoperative outcomes and the progression of radiographic knee osteoarthritis, making the recovery of quadriceps size and strength a key component of ACLR rehabilitation. The NIH All of Us Research Program aims to advance personalized medicine by not only linking patient records, surveys, and genomic data but also engaging with participants, particularly from groups traditionally underrepresented in biomedical research (UBR). Purpose:
Warfarin is one of the most widely used anticoagulants that functions by inhibiting vitamin K epoxide reductase. Warfarin overdose, whether intentional or unintentional, can cause life-threatening bleeding. Here, we present a novel warfarin adsorbent based on mesoporous silica that could serve as an antidote to warfarin toxicity.

Amino-functionalized mesoporous silica (MS-NH2) was synthesized based on the co-condensation method through a soft template technique followed by template removal. Background
Due to lower maximum strength, older adults purportedly need to exert greater muscular effort to stand up from a chair. This study investigated joint-specific muscular effort during a sit-to-stand by calculating relative-muscular-effort and investigating differences between healthy older and young adults.
Methods
Fifteen older (age: 69.9 ± 3.5 years) and 15 young (age: 22.2 ± 2.6 years) adults performed five self-paced sit-to-stand trials. Kinematic and kinetic data were collected and used to calculate inverse dynamics net joint moment. Introduction: Alzheimer’s disease (AD) prevalence and severity are associated with increased age, female sex, and apolipoprotein E4 (APOE4) genotype. Although estrogen therapy (ET) effectively reduces symptoms of menopause including hot flashes and anxiety, and can reduce dementia risk, it is associated with increased risks of breast and uterine cancer due to estrogen receptor alpha (ERα)-mediated increases in cancer cell proliferation. Because ERβ activation reduces this cell proliferation, selective targeting of ERβ may provide a safer method of improving memory and reducing hot flashes in menopausal women, including those with AD. Chatogekko amazonicus is a miniaturized gecko from northern South America and is among the smallest of toe pad bearing lizards. The toe pads of C. amazonicus are miniscule, between 18% and 27% of the plantar surface area. We aimed to investigate the relationship between adhesive toe pad morphology, body size, and adhesive capabilities. Using scanning electron microscopy, we determine that the adhesive pads of C. amazonicus exhibit branched setae similar to those of other geckos, but that are generally much smaller. Pathologies of the skull and teeth are well documented for many human populations, but there are fewer studies of other primates. We contrast lesion prevalence and patterning among cercopithecoid primates and map variation onto socioecological variables. We compare craniodental lesions in six species: Nasalis larvatus (n = 54), Colobus polykomos (n = 64), Cercopithecus mitis (n = 65), Macaca fascicularis (n = 109), Theropithecus gelada (n = 13), and Papio anubis (n = 76). Purpose: To examine whether time of year (relative to hibernation emergence) influences the retinal degenerative effects of intravitreal injection of adenosine triphosphate (ATP) in the 13-lined ground squirrel (13-LGS).
Methods: Eighteen (9 male, 9 female) 13-LGS in three experimental cohorts (early season, mid-season, late season) (n = 6 each) underwent baseline imaging using scanning light ophthalmoscopy (SLO) and optical coherence tomography (OCT). Animals then received a 10-µL intravitreal injection of 0.723 M ATP, followed by OCT and SLO imaging at 3, 10, and 21 days. Introduction Prenatal opioid exposure (POE) is a major public health consequence of the opioid epidemic. Long-term health outcomes associated with POE remain unclear, especially for children with POE without a diagnosis of neonatal abstinence syndrome (NAS). Germ cell development and gamete production in animals require small RNA pathways. While studies indicate that microRNAs (miRNAs) are necessary for normal sperm production and function, the specific roles for individual miRNAs are largely unknown. Here, we use small RNA sequencing (RNA-seq) of dissected gonads and functional analysis of new loss-of-function alleles to identify functions for miRNAs in the control of fecundity and sperm production in Caenorhabditis elegans males and hermaphrodites. By 2050, the projected 9.7 billion global population will increase food demand, especially for staple crops. Climate change, with extreme temperature fluctuations, heavily impacts cold-sensitive Asian rice (Oryza sativa L.) grown in tropical and subtropical areas. Therefore, understanding the response mechanisms of two Asian rice subspecies, japonica and indica, which have distinctive tolerance to cold stress, becomes important for improving crop cold tolerance. Millions of children in the United States struggle with reading acquisition, many of whom are identified with developmental dyslexia (DD). Although experimental research indicates that evidence-based reading interventions can bring many of these children to average-range reading skills, such promising outcomes are not commonly achieved within K-12 public schools. Molecular chaperones assist in maintaining protein homeostasis by limiting protein aggregation, including populations of protein aggregates that may be associated with the onset of debilitating and/or often fatal diseases. Chaperones are known to limit protein aggregate populations through disaggregase activity, in which chaperone complexes work together to disassemble multi-subunit aggregate species into smaller populations, or holdase activity, in which chaperones bind to proteins or smaller aggregates to prevent further aggregation. S-Nitrosothiols (RSNOs) are chemical compounds capable to transport nitric oxide (NO) and show promise in the treatment of cardiovascular diseases. RSNOs occur naturally in-vivo, and others have been synthesized in-vitro as potential drug candidates for NO transport and release. However, RSNOs are inherently unstable due to their weak S–N bond (D0(S-N)=25–30 kcal/mol), so developing a deeper understanding of the factors influencing the RSNOs stability is crucial. In Chapter 2, we investigated the substituent effect on the -SNO group properties. Osteogenesis imperfecta (OI) is a heritable bone fragility disorder characterized by skeletal deformities and frequent fractures. Impairment of the collagen network in bone reduces bone quality and quantity, increasing the risk of fracture. The corrective osteotomy required to insert a rod into a long bone during corrective orthopaedic surgeries in OI excises at least one small bone wedge. Research on these specimens is beginning to offer valuable insight into the material behavior and modeling potential for improved orthopaedic treatment. Pyruvate carboxylase (PC) is implicated in a variety of diseases, including type-2 diabetes, cancer, and bacterial/viral infections. However, there are currently no molecular tools capable of precisely manipulating PC activity both in vitro and in vivo. This dissertation describes the identification and characterization of 1,3-disubstituted imidazolidinetriones as a novel class of potent, selective, and permeable allosteric inhibitors of Staphylococcus aureus PC. Based on kinetic, structural, and biophysical data, this class of inhibitors is hypothesized to bind at the noncatalytic “exo binding” site on PC. Endotracheal tube (ETT) intubation is a medical procedure routinely used for achieving mechanical ventilation in critically ill patients. Appropriate ETT placement is crucial as undetected tube migration may cause multiple complications or even fatalities. Therefore, prompt detection of unplanned movement of the ETT and immediate action to restore proper placement are essential to ensure patient safety. Despite this necessity, there is not a widely adopted tool for real-time assessment of ETT displacement. Surface grinding plays a pivotal role in the machining industry, constituting roughly 25% of all machining operations worldwide. Its precision and efficiency are crucial, particularly in sectors requiring high-quality surface finishes, such as aerospace and semiconductor manufacturing. For thermal damage prevention, traditional approaches to parameter selection use thresholds to exclude burn-prone parameters. In this project, I argue that the trajectory of Western European civilization has resulted in increasingly tragic predicaments that are not to be taken as given features of being human. We describe a new species of miniaturized gecko (genus Pseudogonatodes) from the Peninsula de Paria in northeastern Venezuela. Externally, the new species resembles Pseudogonatodes furvus and Pseudogonatodes manessi, from the Sierra Nevada de Santa Marta in Colombia and the Central Coastal Range in Venezuela, respectively; however, it differs from these species in terms of molecular genetic data (12S rRNA, 16S rRNA, and c-mos), osteological characters, and scale counts. The new species is unique in skull osteology, and we adopt the term ‘telescoped’ from the literature to describe the overlap of bones in the snout, in particular the …
