Two-dimensional (2D) and 3D through-space 13C-13C homonuclear spin-diffusion techniques are powerful solid-state nuclear magnetic resonance (NMR) tools for extracting structural information from 13C-enriched biomolecules, but necessarily long acquisition times restrict their applications. In this work, we explore the broad utility and underutilized power of a chemical shift-selective one-dimensional (1D) version of a 2D 13C-13C spin-diffusion solid-state NMR technique. Obesity is a global epidemic characterized by an expansion of adipose tissue. Adipose tissue, composed of adipocytes and a stromal vascular fraction, is an endocrine organ that regulates whole body homeostasis. Obesity leads to the dysregulation of adipocytes and is often associated with increased susceptibility to metabolic diseases such as Type II diabetes mellitus (T2DM). Understanding the mechanisms by which adipocyte function and development are regulated is crucial. Mitochondrial pyruvate carriers (MPCs) are transmembrane proteins that transport pyruvate from the cytosol into the mitochondrial matrix to generate acetyl CoA. Objective Multiple clinical trials fail to identify clinically measurable health benefits of daily multivitamin and multimineral (MVM) consumption in the general adult population. Understanding the determinants of widespread use of MVMs may guide efforts to better educate the public about effective nutritional practices. The objective of this study was to compare self-reported and clinically measurable health outcomes among MVM users and non-users in a large, nationally representative adult civilian non-institutionalised population in the USA surveyed on the use of complementary health practices. Purpose: This study aims to investigate the process of service gifting (i.e. unexpected upgrades or benefits) and examine why service gifts do not always result in firm-beneficial reciprocal behaviors from consumers. Design/methodology/approach: Through a series of three studies including both scenario-based and game-theory-based experiments, this research proposes and empirically validates a conceptual model that examines the effect of service gifts on firm-beneficial reciprocal behaviors, and the role of collective social connection and norm of positive reciprocity (NPR) in this process. Solid phase extraction (SPE) is a widely used method to separate and concentrate the target molecules in liquid mixture. Traditional SPE has to be conducted in the laboratory with professional equipment and skilled operators. The microfluidic and 3D printing technology have opened up the opportunity in developing miniaturized automatic instruments. The main contribution of this research is to integrate the SPE process on a novel centrifugal platform. Various valves are applied on the platform to help control the aqueous sample and reagents in the cartridge.

Boxwood (Buxus spp. L) is one of the most common and widely planted perennial ornamentals in both home gardens and commercial landscapes. Grown for its evergreen, dark green foliage, boxwood cultivation dates back to 4000 BC in Egypt. Although considered hardy, boxwood is susceptible to several plant pathogens. Recently reported boxwood dieback, a fungal disease caused by Colletotrichum theobromicola, has been spreading at an alarming rate within the United States. The broad goal of the research described in this thesis was to better understand the potential for biologically mediated production of p-cresol in groundwater at a Superfund site located in south Louisiana. Previous literature reports that microorganisms known to carry out the transformation from L-tyrosine to p-cresol occur in diverse environments.

Although leak incidents continue, a pipeline remains the most reliable mode of transportation within the oil and gas industry. It becomes even more important today because the projection for new pipelines is expected to increase by 1 billion BOE through 2035. This dissertation examines how media attention and domestic opposition parties affect third-party intervention in foreign civil wars. Previous research defines some of the ways that political executives in democratic states are constrained in their choice of actions, while also pointing out the influences and limitations of outside actors. This dissertation builds on existing research pertaining to opposition parties and the news media by providing new theories on how these outside actors can push a leader in taking an action he otherwise might not take, such as intervening in a foreign civil war. In recent years, large scale deployments of electrical energy generation using renewable sources (RES) such as wind, solar and ocean wave power, along with more sustainable means of transformation have emerged in response to different initiatives oriented toward reducing greenhouse gas emissions. Strategies facilitating the integration of renewable generation into the grid and electric propulsion in transportation systems are proposed in this work.

P. ingrahamii is a halo-psychrophilic bacterium isolated from Arctic sea ice. We have cloned and purified the large fragment of the cold-active DNA polymerase I from P. ingrahamii, named Klenpin. The objective of this project is to directly compare the thermodynamic stability of Klenpin, and the salt dependence of that stability, with Klenow and Klentaq; two homologous polymerases from a mesophile (E. coli) and a thermophile (Thermus aquaticus).

In situ plant production is often assumed to be the major contributor to organic matter (OM) accumulation and vertical accretion in tidal marshes. Here, we evaluate the contribution of mud-associated OM in salt and brackish marshes in Louisiana. Based on 14 soil cores, the OM content of the mud fraction—i.e., any material smaller than 64 μm—was 17% ± 7% for the salt marshes and 28% ± 14% for the brackish marshes. This remains nearly uniform over the top 35 cm depth, suggesting that this material is deposited contemporaneously with the mud. The dry bulk density of the mud (300–450 kg …

Tidal flats form around the estuarine and coastal zone by continuous terrigenous sediment transport and deposition processes. Now a large body of published carbon research work frame within the vegetated area (mangrove forests, sea grass bed, and salt marshes). Nonvegetated tidal flats, which are characterized by predominantly silts and clays sediment, were generally impressed with low carbon stock due to their meager primary productivity. However, these regions may be a potentially important carbon sink, given their high burial rate, expanding areal coverage, and detrial organic carbon derived from watershed and adjacent vegetated area. Atmospheric cold front-generated waves play an important role in the air–sea interaction and coastal water and sediment transports. In-situ observations from two offshore stations are used to investigate variations of directional waves in the coastal Louisiana. Hourly time series of significant wave height and peak wave period are examined for data from 2004, except for the summer time between May and August, when cold fronts are infrequent and weak. The intra-seasonal scale variations in the wavefield are significantly affected by the atmospheric cold frontal events. Indoor environmental quality (IEQ) plays a key role in determining occupants' productivity at work; however, analyses of the interconnected factors among building physical, attitudinal, social and demographic components in one study are lacking. To fill this research gap, this study investigates these interconnected factors' influence on occupants’ IEQ-productivity belief, defined as a personal, subjective evaluation of the linkage between the impacts of five IEQ aspects (the quality of indoor temperature, air, natural and electric lighting, and acoustics) and productivity. Scc4 is an unusual bi-functional protein from Chlamydia trachomatis (CT) that functions as a type III secretion system (T3SS) chaperone and an RNA polymerase (RNAP)-binding protein. Both functions require interactions with protein partners during specific stages of the CT developmental cycle. As a T3SS chaperone, Scc4 binds Scc1 during the late stage of development to form a heterodimer complex, which chaperones the essential virulence effector, CopN. © 2020 British Ecological Society Dispersal is a key process in shaping species spatial distributions. Species interactions and variation in dispersal probabilities may jointly influence species spatial dynamics. However, many studies examine dispersal as a neutral process, independent of community context or intraspecific variation in dispersal behaviour. Here, we use controlled, replicated communities of two Tribolium species (T. castaneum and T. confusum) to examine how intraspecific variation in dispersal behaviour and community context influence dispersal dynamics in simple experimental landscapes composed of homogeneous habitat patches. We found considerable individual-level variation in dispersal probability that was unrelated to body size variation. …

The development of Edwardsiella-containing-vacuoles (ECV) and the ability of Edwardsiella ictaluri to survive and replicate within macrophages suggests a unique process relative to normal phagosomal/lysosomal maturation and programed cell death. Developing ECV showed that endosomal membrane markers Rab5, EEA1, and Rab7 were all detected in both the wild type (WT) and an E. ictaluri type-3 secretion system (T3SS) mutant, 65ST. Co-localization with Lamp1, however, was significantly lower in the WT. © 2020 John Wiley & Sons Ltd Aim: The processes that generate biodiversity occur at finer scales than are often studied, particularly in freshwater systems of the northern Neotropics. We investigate whether fine-scale biogeographic patterns are present within the larger Grijalva–Usumacinta Area of Endemism (AoE) —a region in Middle America where nearly 60% of freshwater fishes are endemic—and if present-day river basins are single historical units. Nut-based milks and yogurts are gaining popularity, but may not offer the same benefits as dairy yogurts to consumers. Cashew nuts often cause severe allergic reactions, and cashew nut allergens are stable to several types of processing. To compare its characteristics to dairy yogurt and characterize the effects of fermentation on the Ana o 1-3 cashew nut allergens, a commercial yogurt made from cashew nuts (Cashewgurt) was evaluated for microbiological, physiochemical, and immunological properties. Average counts for lactobacilli and Streptococcus thermophilus were greater than 10 million colony forming units per milliliter, indicating the capacity to provide a health benefit. Cashewgurt …

Plant architecture is critical for enhancing the adaptability and productivity of crop plants. Mutants with an altered plant architecture allow researchers to elucidate the genetic network and the underlying mechanisms. In this study, we characterized a novel nal1 rice mutant with short height, small panicle, and narrow and thick deep green leaves that was identified from a cross between a rice cultivar and a weedy rice accession. Bulked segregant analysis coupled with genome re-sequencing and cosegregation analysis revealed that the overall mutant phenotype was caused by a 1395-bp deletion spanning over the last two exons including the transcriptional end site …

The distributions of many sister species in the sea overlap geographically but are partitioned along depth gradients. The genetic changes leading to depth segregation may evolve in geographic isolation as a prerequisite to coexistence or may emerge during primary divergence leading to new species. These alternatives can now be distinguished via the power endowed by the thousands of scorable loci provided by second-generation sequence data. Here, we revisit the case of two depth-segregated, genetically isolated ecotypes of the nominal Caribbean candelabrum coral Eunicea flexuosa. Introduction A key component to analyzing wearable sensor data is identifying periods of non-wear. Traditionally, strings of consecutive zero counts (e.g. >60-minutes) are identified indicating periods of non-movement. The non-movement window length is then evaluated as wear or nonwear. Given that non-movement is not equivalent to non-wear, additional criteria should be evaluated to objectively identify periods of non-wear. Identifying non-wear is especially challenging in infants due to their sporadic movement, sleep frequency, and proportion of caregiver- generated movement. Objective: This study aimed to describe the energy expenditure (EE) and macronutrient oxidation response to an individualized nonshivering cold exposure in young healthy adults. Methods: Two different groups of 44 (study 1: 22.1 [SD 2.1] years old, 25.6 [SD 5.2] kg/m2, 34% men) and 13 young healthy adults (study 2: 25.6 [SD 3.0] years old, 23.6 [SD 2.4] kg/m2, 54% men) participated in this study. Resting metabolic rate (RMR) and macronutrient oxidation rates were measured by indirect calorimetry under fasting conditions in a warm environment (for 30 minutes) and in mild cold conditions (for 65 minutes, with the individual wearing …
