The mediator kinase module plays a critical role in the regulation of transcription during metabolic processes. Here we demonstrate that in human adipose-derived stem cells (hASCs), kinase module subunits have distinct mRNA and protein expression profiles during different stages of adipogenesis. In addition, siRNA-mediated loss of MED12 results in decreased adipogenesis as evident through decreased lipid accumulation and decreased expression of PPARγ, a master regulator of adipogenesis. Moreover, the decrease in adipogenesis and reduced PPARγexpression are observed only during the early stages of MED12 knockdown. At later stages, knockdown of MED12 did not have any significant effects on adipogenesis or …

Background & aims: The validity of most commercially available metabolic cart is mostly unknown. Thus, we aimed to determine the accuracy, precision, within-subject reproducibility, and concordance of RMR and RER measured by four commercially available metabolic carts [Cosmed Q-NRG, Vyaire Vyntus CPX, Maastricht Instruments Omnical, and Medgraphics Ultima CardiO2]. Further, we studied whether a previously proposed simulation-based post-calorimetric calibration of cart readouts [individual calibration control evaluation (ICcE)] modify the RMR and RER reproducibility and concordance. Methods: Three experiments simulating different RMR and RER by controlled pure gas (N2 and CO2) infusions were conducted on 5 non-consecutive days. Moreover, 30-min methanol …

The Counterbalancing Effects Of Energy Expenditure On Body Weight Regulation: Orexigenic Versus Energy-Consuming Mechanisms, Paolo Piaggi, Alessio Basolo, Corby K. Martin, Leanne M. Redman, Susanne B. Votruba, Jonathan Krakoff

Clinical Research Faculty Publications

Objective: Weight change is a dynamic function of whole-body energy balance resulting from the interplay between energy intake and energy expenditure (EE). Recent reports have provided evidence for the existence of a causal effect of EE on energy intake, suggesting that increased EE may drive overeating, thereby promoting future weight gain. This study investigated the relationships between ad libitum energy intake and 24-hour EE (24-h EE) in sedentary conditions versus long-term, free-living weight change using a mediation analysis framework. Methods: Native American individuals (n = 61, body fat by dual-energy x-ray absorptiometry: 39.7% [SD 9.5%]) were admitted to the clinical …

Faculty Publications

Sweetpotato [Ipomoea batatas (L.) Lam.], grown as an edible ornamental, is propagated in home gardens using locally purchased transplants. However, vigorous growth of sweetpotato limits the period of transplant salability due to root confinement. The objective of the experiment was to evaluate reductions in phosphorus (P) fertility to slow transplant root growth and extend the period of salability. Cuttings were planted into containers and fertilized at 0, 5 (0.0007), 10 (0.0012), 15 (0.0020), 20 (0.0024), and 31 mg P.L-1 (0.0040 oz P.gal-1) using a modified Hoagland solution. All transplants fertilized at 5 mg P.L-1 increased in shoot length, color, and …

Background: Although 10 000 steps per day is widely promoted to have health benefits, there is little evidence to support this recommendation. We aimed to determine the association between number of steps per day and stepping rate with all-cause mortality. Methods: In this meta-analysis, we identified studies investigating the effect of daily step count on all-cause mortality in adults (aged ≥18 years), via a previously published systematic review and expert knowledge of the field. We asked participating study investigators to process their participant-level data following a standardised protocol. The primary outcome was all-cause mortality collected from death certificates and country …

Post-Acute Sequelae Of Covid-19: A Metabolic Perspective, Philipp E. Scherer, John P. Kirwan, Clifford J. Rosen

Division of Basic Science Faculty Publications

The SARS-CoV-2 pandemic continues to rage around the world. At the same time, despite strong public health measures and high vaccination rates in some countries, a post-COVID-19 syndrome has emerged which lacks a clear definition, prevalence, or etiology. However, fatigue, dyspnea, brain fog, and lack of smell and/or taste are often characteristic of patients with this syndrome. These are evident more than a month after infection, and are labeled as Post-Acute Sequelae of CoV-2 (PASC) or commonly referred to as long-COVID. Metabolic dysfunction (i.e., obesity, insulin resistance, and diabetes mellitus) is a predisposing risk factor for severe acute COVID-19, and …

The Influence Of Whey Protein Heating Parameters On Their Susceptibility To Digestive Enzymes And The Antidiabetic Activity Of Hydrolysates, Kungnang Bunsroem, Witoon Prinyawiwatkul, Siwatt Thaiudom Mar 2022

The Influence Of Whey Protein Heating Parameters On Their Susceptibility To Digestive Enzymes And The Antidiabetic Activity Of Hydrolysates, Kungnang Bunsroem, Witoon Prinyawiwatkul, Siwatt Thaiudom

Faculty Publications

The inhibition of dipeptidyl peptidase-IV (DPP-IV) and the release of glucagon-like peptide-1 (GLP-1) could normalize blood glucose levels in diabetic patients. This study evaluated the susceptibility of whey proteins to enzyme hydrolysis and the antidiabetic properties of protein hydrolysates from β-lactoglobulin (β-LG) and α-lactalbumin (α-LA) solutions compared with whey protein isolate (WPI) solution treated at different heating temperatures (65, 75, and 85◦ C). α-LA hydrolysate provided the lowest degree of hydrolysis (DH). Those heating temperatures did not significantly affect the DH of all protein hydrolysates. α-LA hydrolysate significantly increased GLP-1 levels and DPP-IV inhibitory activity more than β-LG hydrolysate. WPI …

Based on the spectator expansion of the multiple scattering series we employ a chiral next-to-next-to-leading order (NNLO) nucleon-nucleon interaction on the same footing in the structure as well as in the reaction calculation to obtain an in leading-order consistent effective potential for nucleon-nucleus elastic scattering, which includes the spin of the struck target nucleon. As an example we present proton scattering off 12C.

Evaluating Stratified Substrates Effect On Containerized Crop Growth Under Varied Irrigation Strategies, Kristopher S. Criscione, Jeb S. Fields, Jim S. Owen, Lisa Fultz, Edward Bush

Faculty Publications

Growers rely on soilless substrates to provide sufficient water and nutrients to containerized crops. Traditional bark-based substrates are engineered to have relatively low water-holding capabilities, which can lead to nonuniform rewetting patterns and inefficient usage of water resources. Engineering substrates to redistribute water dynamics and maximize aeration within the container may improve water resource efficiencies. The goal of this study was to evaluate whether more efficient irrigation schedules can be used when stratifying unique substrates within a container for added crop water and nutrient efficiency. Loropetalum chinense ‘Ruby’ liners were planted and grown in a conventional pine bark substrate or …

The H3.3 Chaperone Hira Complex Orchestrates Oocyte Developmental Competence, Rowena Smith, Andrej Susor, Hao Ming, Janet Tait, Marco Conti, Zongliang Jiang, Chih Jen Lin

Faculty Publications

Successful reproduction requires an oocyte competent to sustain early embryo development. By the end of oogenesis, the oocyte has entered a transcriptionally silenced state, the mechanisms and significance of which remain poorly understood. Histone H3.3, a histone H3 variant, has unique cell cycle-independent functions in chromatin structure and gene expression. Here, we have characterised the H3.3 chaperone Hira/Cabin1/Ubn1 complex, showing that loss of function of any of these subunits causes early embryogenesis failure in mouse. Transcriptome and nascent RNA analyses revealed that transcription is aberrantly silenced in mutant oocytes. Histone marks, including H3K4me3 and H3K9me3, are reduced and chromatin accessibility …

The Potential Of Dietary Bioactive Compounds Against Sars-Cov-2 And Covid-19-Induced Endothelial Dysfunction, Jack N. Losso

Faculty Publications

COVID-19 is an endothelial disease. All the major comorbidities that increase the risk for severe SARS-CoV-2 infection and severe COVID-19 including old age, obesity, diabetes, hypertension, respiratory disease, compromised immune system, coronary artery disease or heart failure are associated with dysfunctional endothelium. Genetics and environmental factors (epigenetics) are major risk factors for endothelial dysfunction. Individuals with metabolic syndrome are at increased risk for severe SARS-CoV-2 infection and poor COVID-19 outcomes and higher risk of mortality. Old age is a non-modifiable risk factor. All other risk factors are modifiable. This review also identifies dietary risk factors for endothelial dysfunction. Potential dietary …

Topological Edge States At Singular Points In Non-Hermitian Plasmonic Systems, Yin Huang, Yuecheng Shen, Georgios Veronis Mar 2022

Topological Edge States At Singular Points In Non-Hermitian Plasmonic Systems, Yin Huang, Yuecheng Shen, Georgios Veronis

Faculty Publications

We introduce non-Hermitian plasmonic waveguide-cavity systems with topological edge states (TESs) at singular points. The compound unit cells of the structures consist of metal-dielectric-metal (MDM) stub resonators side-coupled to an MDM waveguide. We show that we can realize both a TES and an exceptional point at the same frequency when a proper amount of loss is introduced into a finite three-unit-cell structure. We also show that the finite structure can exhibit both a TES and a spectral singularity when a proper amount of gain is introduced into the structure. In addition, we show that we can simultaneously realize a unidirectional …

Event-Triggered Control For Linear Time-Varying Systems Using A Positive Systems Approach, Frederic Mazenc, Michael Malisoff, Corina Barbalata, Zhong Ping Jiang

Faculty Publications

This paper presents a new design for the event-triggered output feedback control of uncertain linear time-varying systems. Departing from the traditional treatments of event-triggered linear time-invariant systems, it is shown that factoring of fundamental solutions as products of solutions of matrix differential equations, together with small-gain arguments and the notions of interval observers and positive systems, lead to a new class of robust event-triggered output-feedback controllers. A key innovation is our use of new event triggers, which use vectors of absolute values instead of the usual Euclidean 2-norms. An illustrative example of a curve tracking system from marine robotics validates …

Nanoscale delivery systems have the potential to improve the effectiveness of agrochemicals while reducing their negative environmental impacts. Herein, we explore the use of lignin nanoparticles (LNPs) to enhance the translocation of methoxyfenozide (MFZ), a non-systemic pesticide, in soybean plants under hydroponic conditions. LNPs (113.8 ± 3.5 nm and zeta potential – 53.3 ± 6.9 mV) were synthesized by emulsion evaporation from lignin-graft-poly(lactic-co-glycolic) acid and MFZ was incorporated into the LNPs (2.7% w/w). Twenty eight day old soybeans were grown hydroponically and were treated with 0.01, 0.1 or 1 mg/ml of LNPs. Plants were harvested after 6, 12, and 24 …

One of the current pressing problem for all DOTs is the corrosion-oriented deterioration of the existing metal culverts. These metal culverts typically are designed for a life of 50 years. However, corrosion is making them last no longer than 30years. Here we propose use of Glass Fiber Reinforced Polymers (GFRP) pipe section as a fit-in GFRP profile liner for complete repair and rehabilitation of the corroded metal culvert with an expected life of 75 years. This is mainly because of the corrosion free nature of the GFRP material. In the current study, the design method for using glass fiber-reinforced polymer …

The viscosity of iron alloy liquids is the key for the core dynamo and core-mantle differentiation of terrestrial bodies. Here we measured the viscosity of Fe-Ni-C liquids up to 7 GPa using the floating sphere viscometry method and up to 330 GPa using first-principles calculations. We found a viscosity increase at ∼3–5 GPa, coincident with a structural transition in the liquids. After the transition, the viscosity reaches ∼14–27 mPa·s, a factor of 2–4 higher than that of Fe and Fe-S liquids. Our computational results from 5 to 330 GPa also indicate a high viscosity of the Fe-Ni-C liquids. For a …

My Favorite Murder As Discursive Performance, Taylor Dawson

LSU Doctoral Dissertations

My Favorite Murder as Discursive Performance uses performance-centered discourse analysis to explore the major narratives informing the popular true crime comedy podcast, My Favorite Murder (MFM). Hosts, Karen and Georgia draw from podcast, comedy, true crime, feminist, and mental health discourse to create a unique discursive space where “murderinos” (fans of the podcast) express and explore aspects of their own life experiences. I explore the performative strategies MFM uses and the effects of those choices, drawing on some of my own experiences as a murderino. Ultimately I argue that a performance lens reveals some of the imperfect but powerful ways …
