The inhibitor CO was used to investigate the mechanism of Mo and V nitrogenase. While low pressures of CO enhanced, high pressures inhibited V nitrogenase-catalyzed C$\rm\sb2H\sb6$ formation. This is not the case for $\rm C\sb2H\sb4$ formation, where the rate of electron flux determined if CO acted as an enhancer or inhibitor. A two-site model is proposed, in which CO binding to one site enhances, and CO binding to both sites inhibits product formation. Replacement of CO(g) by Ar or $\rm C\sb2H\sb2$ resulted in correlated decay of hi-CO and development of lo-CO EPR signals, suggesting that the two signals arise from …

At low CO$\sb2$ conditions, C. reinhardtii, like many other green algae, induces a CO$\sb2$ Concentrating Mechanism (CCM) to raise the internal Ci concentration. During the induction, at least five new polypeptides and six different genes are upregulated and some of the corresponding mRNAs or proteins are absent in high CO$\sb2$ requiring mutants. However the identities and functions of these genes are unknown. In this dissertation, I partially characterized four low CO$\sb2$ inducible genes and one low CO$\sb2$ inducible protein LIP-36. The factors that control plant species diversity have been examined by many researchers, but with little consensus as to the causes of diversity patterns. Plant diversity has been correlated with disturbance, competition, soil resources, and other variables. In the research presented here, I investigated the relationship between plant species richness and biomass, competition, nutrient enrichment, flooding, salinity, and herbivory in field studies in Louisiana coastal marshes. Biomass alone was a poor predictor of richness in two Louisiana marshes. When environmental variables (flooding, salinity, and soil fertility) were included with biomass in a multiple regression model, 82% of the variance in …

In the dissertation we have proposed the first systematic and formal approach to reduce connectivity of general purpose multiple bus systems. The approach is based on a probabilistic technique. The hypothesis on which this dissertation is based stipulates that bus connectivity in multiple bus systems can be much reduced by removing connections which are only needed for highly improbable request patterns. With this approach, performance comparable to that of the original multiple bus system could be achieved while significantly reducing memory bus connectivity. Johannes de Garlandia (fl. 1250, Paris), author of the seminal De mensurabili musica, has been linked to two plainchant treatises, De plana musica and Introductio musice. This dissertation presents the first critical edition and translation, with commentary, of these two treatises. It includes an introduction, Latin texts of the two treatises with English translations in parallel columns, critical apparatus, commentary, and appendices. De plana musica is transmitted in three sources, Introductio musice in four. The text of Introductio musice established here is shorter than that printed by Coussemaker (CS, 1:157-75), who included in the final portion of his text several …

Sequence specific protein-DNA interaction involves changes in protein conformation and dynamics. Research work involved in this thesis is the study of structure and dynamics of EcoRI restriction endonuclease N-termini. The N-termini of EcoRI endonuclease has been shown to be essential for DNA cleavage and also stabilize EcoRI-DNA complex. But they are not resolved in the X-ray crystal structure. An Asn3Cys mutation was made at the N-termini by site-directed mutagenesis and the mutant was subjected to cysteine crosslinking, pyrene labeling and fluorescence studies. Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous contaminants in aquatic environments. Because of their hydrophobicity, PAHs accumulate in bed sediments and pose a risk to the benthos. Information on the toxicity of sediment-associated PAHs is, however, limited and more information is needed to improve sediment quality-criteria and ecological-risk assessments. My research focused on improving our current knowledge of the acute and, especially, the sublethal effects of sediment-associated PAHs to aquatic invertebrates. Representatives of two phylogenetically distinct benthic invertebrate groups, freshwater tubificid oligochaetes and estuarine meiobenthic harpacticoid copepods, were used. This dissertation attempts to solve two problems related to an ATM network. First, we consider packetized voice and video sources as the incoming traffic to an ATM multiplexer and propose modeling methods for both individual and aggregated traffic sources. These methods are, then, used to analyze performance parameters such as buffer occupancy, cell loss probability, and cell delay. Results, thus obtained, for different buffer sizes and number of voice and video sources are analyzed and compared with those generated from existing techniques. The difficulty in debugging distributed programs motivates the development of formal methods for designing distributed programs that are easier to debug and maintain. We address state identification problem for distributed systems using the finite state I/O automaton model. A state S is identified based on the unique event sequences starting at S, called distinguishing sequences. An automaton is diagnosable if every state has a distinguishing sequence. A distributed program may not be diagnosable even if its components are diagnosable. The present research was performed to characterize immunoreactive (ir) pituitary cells in the equine species. Two experiments were performed to measure the prevalence of single- and double-hormone labeled gonadotropes, lactotropes, somatotropes, and mammosomatotropes. In Experiments 1 and 2, pituitaries from three intact pony mares were processed for ultrastructural evaluation of cells stained for luteinizing hormone (LH), follicle-stimulating hormone (FSH), prolactin (PRL), and growth hormone (GH). Primary antibodies and gold-conjugated antibodies were used to double-stain ir-cells. The effects of silvicultural treatments on mechanical and physical properties of loblolly pine (Pinus taeda L.) wood composites, veneer tensile and bending properties, and basic chemical properties were investigated. Stands managed in a plantation setting showed higher extractive contents. Holocellulose and alpha-cellulose were minimally affected by the silvicultural strategies. Klason lignin showed an inverse relationship with holocellulose. The effect of wood type was significant for the extractives because the innerwood region yielded much greater values than the outerwood. Holocellulose and alpha-cellulose increased in concentration from innerwood to outerwood. Veneer tensile strength was significantly affected by silvicultural practice, but other mechanical …

The objective of this study was to find whether DHI data could be used to estimate sire non-return rates to replace current technician data estimates. Bull weighted least squares means for non-return rates were calculated separately for five overlapping 60-90 day service periods from each data source. Models included stud, sire, service number, and linear and quadratic form of breeding month for both data sources, service unit for technician and lactation for DHI data. Sire and lactation were not significant $(P > .10).$ Technician differences $(P < .05)$ were in service unit, stud, service number, and linear and quadratic service month in all but one service period. DHI differed $(P < .05)$ for service number, month (linear and quadratic), and stud in two service periods. Technician R-square values were 0.23 to 0.28 versus 0.94 to 0.96 for DHI. Sire estimated non-return rates were weighted using the inverted estimator standard error squared and compared. Sire, stud, data source, service period, and appropriate interactions were modeled. Weighted bull non-returns differed $(P < .01)$ in magnitude across data sources. Stud, data source by stud, and sire within stud by data source were significant $(P .10).$ Four fertility categories based on mean and standard deviation of the weighted estimates were formed within the data sources across service periods. These categories were correlated (0.5 $>$ r $>$ 0.9) and 52.9 to 87.4% congruent within data source for adjacent …

Aliphatic polycarbonates were synthesized by condensation of the bischloroformate of 2,2,4,4-tetramethyl-1,3-cyclobutanediol with cyclic aliphatic diols. Polycarbonates that lacked flexible methylene units in the backbone processed T$\sb{\rm g}\ >$ 139$\sp\circ$C. Copolycarbonates acted as resists when combined with cobalt(III)-am(m)ine tetraphenylborate complexes which act as photointiators, with tetraphenylborate serving as counter anion. The photoinitiator liberates either ammonia or ethylene diamine base upon UV exposure and the polycarbonate backbone is subject to nucleophilic attack at the carbonyl linkage by the base. Image tone was dependent on the base used. Amine complexes, Co(NH$\sb3)\sb6\sp{3+}$ effected a positive tone due to ammonia induced main chain degradation at …

Geographic Information System (GIS) technology was used to develop a local scale screening procedure for assessing shallow ground water's vulnerability to nitrate contamination. The procedure was based on the spatial distribution of ten key physical and chemical properties of soils. Both flow characteristics and attenuation capabilities of soils were incorporated into the assessment. Land surface areas were classified by the GIS to show the level of vulnerability present at a site as a result of existing natural conditions. The impacts of south Louisiana crude oil on three types of marsh, salt, brackish, and freshwater, dominated by Spartina alterniflora, S. patens, and Sagittaria lancifolia, respectively, were studied in the greenhouse. The influence of a number of factors such as marsh type, plant species, oil dosage, oil coverage, soil composition, leaf surface structure, season and meteorological conditions, on the impact of crude oil to marsh vegetation and revegetation were investigated. The structures of a number of novel poly(aryl ether) sulfones (nitro, aminomethyl, methyl and ethyl derivatives), which have applications as separation membranes, were determined unequivocally for the first time by applying sophisticated two-dimensional nuclear magnetic resonance (NMR) methods. The degree of substitution (DS) was found to vary among the products from 0.1 percent to about 2 percent (DS: nitro, 2.0; amino, 0.11; aminomethyl, 0.76; methyl, 2.12; ethyl, 2.05). Poly($\gamma$-stearyl-$\alpha$,L-glutamate), PSLG, is a "fuzzy rod" polymer composed of a central backbone that can exist as an $\alpha$-helix. This backbone, a polypeptide, is hydrophilic. The primary purpose of this study was to develop a simple, low-cost, easy to administer and score post-course participant survey that examined perceptions of the influence of certain factors identified as affecting the transfer of learning to the workplace. These factors were in the areas of course design, delivery and relevance as well as personal, organizational and workplace influences on learning transfer. The purpose of this research is to develop and automatically extract an abstract representation model of object-oriented (abbreviated as OO) software systems that captures the structure of the system and code dependencies, in order to aid maintenance. The research resulted in the development of two abstract representation models--the low-level design pattern (LLDP) abstract model and the low-level software architecture (LLSA) abstract model. Deinococcus radiodurans is extremely resistant to ionizing radiation and can withstand 5 kGy $\gamma$ radiation without loss of viability or evidence of mutation. Available evidence argues that an efficient DNA repair mechanism is responsible for the resistance these organisms possess to ionizing radiation. The enzymes which coordinate DNA repair in D. radiodurans are poorly characterized. Two new loci have been detected in D. radiodurans, namely, irrB and irrI which provide resistance. Mutational inactivation of either locus resulted in a partial loss of resistance to ionizing radiation. A monoclonal antibody (MAb)-based western blot analysis (WB) was developed for detection of a 26-28 kD coproantigen in Fasciola hepatica infected cattle. Using WB the coproantigen was detected in feces from experimentally infected calves with 22 flukes or more. Using the biotin-streptavidin modification of WB, antigen was detected in the feces from experimentally-infected calves with 10 flukes or more as early as 6 weeks post-infection. The MAbs did not react with the ES of Paramphistomum sp. or Moniezia sp, but showed slight cross-reactivity with ES from F. gigantica, and there was moderate cross-reactivity with Fascioloides magna ES. The nucleotide sequence of the gene encoding the cytoplasmic $N,N\sp\prime$-diacetylchitobiase from Vibrio parahaemolyticus has been determined. The deduced peptide sequence surprisingly has minimum evolutionary relationship to two other reported $N,N\sp\prime$-diacetylchitobiases from vibrios, except for highly conserved regions which are also homologous with lysosomal beta-hexosaminidases from eukaryotes including humans. In contrast, the other two sequenced chitobiases from vibrios are much more closely related to each other. This 85 kDa cytoplasmic protein, as revealed from the sequence, appears to be a unique protein, lacking a signal sequence and genetically distant from other known enzymes of similar function. Community structure and relative habitat preferences of intertidal fishes from the eastern Canary Islands of Fuerteventura, Gran Canaria, and Lanzarote were examined in a study of 354 tidepools at 33 localities. Habitat use and behavior of the most abundant fish Mauligobius maderensis were examined on Gran Canaria and Lanzarote. Of the 27 species encountered, 11 were common to each of the islands. The most abundant species were: M. maderensis, Parablennius parvicornis, Gobius paganellus, Coryphoblennius galerita, and Chelon labrosus. The purpose of the present study was to determine the effects of two levels of instructional treatment on sixth graders' ability to solve mathematical story problems. The two levels of instructional treatment were instruction in the use of graphic organizers in conjunction with specific analytic reading skills and instruction in specific analytic reading instruction alone. I studied the nesting ecology of the White-faced Ibis (Plegadis chihi) during the summers of 1994 and 1995 in three colonies in Lacassine National Wildlife Refuge, southwestern Louisiana. Data were collected on colony-site characteristics, chronology (with abundance) of colony formation, reproductive success, and growth and development of ibis nestlings. Small colonies nested in dense buttonbush (Cephalanthus occidentalis) and water willow (Decodon verticillatus) "islands." The largest colony nested in black willows (Salix nigra) (3.2 ha)--a habitat unique to ibis populations in Louisiana--with nests heights up to 7.5 m. Cattle egrets (Bubulcus ibis) (62%) and ibises (36%) were the most abundant species. …

The present work is an examination of the problem and the responses to it in early modern England, and is set in the local context of the city of Exeter. This study analyzes the personal narrative of Mary Rhyne, an elderly African American domestic worker. The study describes the ethnographic process of collecting, transcribing and interpreting the informant's oral history. Interpretive and theoretical perspectives concerned with African American culture, domestic labor, and feminist practice are used to analyze the narrative. Further, the ethnographic process, the informant's oral "telling," and the events that she recalls and recounts are viewed as, and in terms of, performance. Performance foregrounds the collaborative and "fictional" aspects of the ethnographer-informant relationship. What immediately comes to mind when looking at the First String Quartet (1960) of Kryzstof Penderecki is his creative method of notation and captivating visual aspect of the score. The composer introduces effective and unusual sounds which require new performance techniques. In addition to these devices he places the composition in a non-metered notation which gives the work an indeterminate aspect. All of these mechanisms sometimes overshadow the more detailed information of the work.
