Effects Of Assembly And Mutations Outside The Active Site On The Functional Ph Dependence Of Escherichia Coli Aspartate Transcarbamylase, Xiaoling Yuan, Vince J. Licata, Norma M. Allewell

Faculty Publications

Electrostatics are central to the function and regulation of Escherichia coli aspartate transcarbamylase, and modeling has suggested that long range electrostatic effects are likely to be important (Glackin, M. P., McCarthy, M. P., Mallikarachchi, D., Matthew, J. B., and Allewell, N. M. (1989) Proteins Struct. Funct. Genet. 5, 66-77; Oberoi, H., Trikha, J., Yuan, X., and Allewell, N. M. (1995) Proteins Struct. Funct. Genet., in press). To investigate this possibility from an experimental standpoint, we have examined the effects both of assembly and of removing ionizable and polar side chains outside the active site (Glu-50, Tyr-165, and Tyr-240) on the …

Spectrum Generating Operators And Intertwining Operators For Representations Induced From A Maximal Parabolic Subgroup, Thomas Branson, Gestur Ólafsson, Bent Ørsted

Faculty Publications

We present a new method of calculating intertwining operators between principal series representations of semisimple Lie groups G. Working in the compact realization we find the eigenvalues of the operators on the K-types, and give several examples. Among the advantages of our method is its applicability to bundle-valued cases. © 1996 Academic Press, Inc.

Effects Of Climate Change On Hypoxia In Coastal Waters: A Doubled Co2 Scenario For The Northern Gulf Of Mexico, D Justic, N N. Rabalais, R. Eugene Turner Jan 1996

Effects Of Climate Change On Hypoxia In Coastal Waters: A Doubled Co2 Scenario For The Northern Gulf Of Mexico, D Justic, N N. Rabalais, R. Eugene Turner

Faculty Publications

Projections of general circulation models suggest that freshwater discharge from the Mississippi River to the coastal ocean will increase 20% if atmospheric CO2 concentration doubles. This result is likely to affect water column stability, surface productivity, and global oxygen cycling in the northern Gulf of Mexico, which is the site of the largest (up to 16,500 km(2)) and most severe hypoxic zone (liter(-1)) in the western Atlantic Ocean. We use a coupled physical-biological two-box model to investigate potential effects of climate change on seasonal oxygen cycling and hypoxia in river-dominated coastal waters. The model was developed and calibrated using comprehensive …

Hydrologic Restoration Of Coastal Wetlands, R. Eugene Turner, R. R. Lewiss Iii Jan 1996

Hydrologic Restoration Of Coastal Wetlands, R. Eugene Turner, R. R. Lewiss Iii

Faculty Publications

Hydrologic modification of coastal wetlands is pervasive, continuing and longstanding in the US. Appreciation for the subtleties of the direct and indirect effects of hydrologic changes on emergent vegetation, soils and co-dependent flora and fauna is contributing to restoration efforts. However the results of wetland restoration/rehabilitation are mostly empirical, rather than scientific in understanding. Science is contributing to the management interest by providing documentation of the past and present failures and successes, and, unveiling the fundamental understanding necessary to move from one wetland to another in an informed and adaptable manner. The papers in this volume are introduced within this …

Security And Rational Choice: Household, Community, And Public Provision., John Collins Kilburn Jr Jan 1996

Security And Rational Choice: Household, Community, And Public Provision., John Collins Kilburn Jr

LSU Historical Dissertations and Theses

This dissertation examines the problem of order through an analysis of individual practices of security. Because security is a multi-dimensional issue, no unified perspective currently exists to explain security practices. Based on my analysis of data from a detailed survey of 137 residents of a Baton Rouge neighborhood, I argue that there are three main dimensions of security and each has a unique set of determinants, since individuals are contextually situated in their households, neighborhoods, and municipalities. This dissertation examines variables related to psychological, stratification, social network, and rational choice perspectives. The analysis demonstrates that although factors suggested by each …

Gender And Spousal Violence: A Test Of Social Control And Resource Theories., Deborah Marie Hanrahan Jan 1996

Gender And Spousal Violence: A Test Of Social Control And Resource Theories., Deborah Marie Hanrahan

LSU Historical Dissertations and Theses

This research provides an explanation of spousal violence that has previously been lacking in the family violence literature--a social control approach. This research tests the hypothesis that those with stronger bonds to society will be less likely to engage in spousal violence. Furthermore, this research will incorporate the role that family structures have in stratifying social control for males and females by testing power-control theory, which suggests that women from egalitarian households of origin will be more likely to commit wife-to-husband violence than those from patriarchal households. Conversely, men from egalitarian households should be less likely to commit husband-to-wife violence …

Black Women's Listening: An Ethnography Of Listening In A Southern Inner-City Adult Learning Center., Jennifer Ann Falls Jan 1996

Black Women's Listening: An Ethnography Of Listening In A Southern Inner-City Adult Learning Center., Jennifer Ann Falls

LSU Historical Dissertations and Theses

The research investigated the phenomenon of listening in the context of adult literacy education. The purpose of this study was to explore and describe listening from the points of view of African American women who were teachers and adult learners in a welfare reform program. Using ethnography as a research methodology, this two-year study explored listening as it occurred in the Writing Class, a classroom culture that was situated in a Southern, inner-city adult learning center (Spindler & Spindler, 1982). The study investigated women's assumptions about listening, their own listening behaviors, and the behaviors they expected of listeners. An aim …

The Effects Of Performance Feedback On Treatment Integrity In School-Based Consultation., Kevin Michael Jones Jan 1996

The Effects Of Performance Feedback On Treatment Integrity In School-Based Consultation., Kevin Michael Jones

LSU Historical Dissertations and Theses

This study examined the relative effectiveness of traditional consultation, with and without performance feedback, on treatment integrity. Six teachers employed in a residential treatment community were responsible for treatment implementation. Treatment integrity was defined as the percentage of 2 min intervals during which contingent teacher attention for student on-task behavior was directly observed. Teacher and child behavior were monitored across baseline, traditional consultation, and consultation with performance feedback conditions in a multiple baseline design. In only one of six cases did mean levels of contingent teacher attention, following a Problem Identification Interview and Problem Analysis Interview (Bergan, 1977), exceed 35%. …

Restructuring In Louisiana Schools., Sharon Lee Pol Jan 1996

Restructuring In Louisiana Schools., Sharon Lee Pol

LSU Historical Dissertations and Theses

This study employs both quantitative and qualitative techniques to study the restructuring processes at the district, school, and teacher levels. The quantitative procedures include surveys used to distinguish between teachers's perceptions of highly and moderately restructured schools. Qualitative research techniques included interviews, observations, and document analysis to describe the restructuring. A new instrument, the Attributes of School Restructuring Scale (ASRS), was developed to measure teachers' knowledge about restructuring efforts and their involvement in the restructuring projects. The final version of the ASRS, included 48-items spread across three subscales: Budget/Finance, Governance/Decision-Making, and Curriculum/Instruction. The quantitative results from the statewide study indicated …

Diagnosing Adhd In Adolescence: An Analysis Of Symptom Presentation And Comorbidity., Larissa Kay Kern Jan 1996

Diagnosing Adhd In Adolescence: An Analysis Of Symptom Presentation And Comorbidity., Larissa Kay Kern

LSU Historical Dissertations and Theses

The diagnosis of Attention Deficit Hyperactivity Disorder (ADHD) in adolescents poses a challenge for clinicians and researchers. Given the decline in hyperactivity and increased behavioral inhibition in adolescence, the differential diagnosis between ADHD and internalizing disorders becomes difficult. In addition, the high comorbidity rates found in adolescents with ADHD further complicate diagnostic decisions. The present study examines the diagnostic efficiency of the DSM-IV criteria for ADHD in a large sample of adolescents aged 11 to 17 years. The results suggest that Inattentive symptoms are more useful for classifying ADHD in adolescents than Hyperactive/Impulsive symptoms. However, Inattentive symptoms also misclassify a …

Pilot-Scale Solvent Sublation For The Removal Of Hydrophobic Compounds From Wastewaters., Jeffrey Scott Smith Jan 1996

Pilot-Scale Solvent Sublation For The Removal Of Hydrophobic Compounds From Wastewaters., Jeffrey Scott Smith

LSU Historical Dissertations and Theses

Solvent sublation is a nonfoaming adsorptive bubble process which exploits the surface active nature of organic compounds in their removal from aqueous systems. Gas bubbles are introduced into an aqueous media and are buoyed upward to an immiscible organic solvent. As the bubbles rise, organic materials are selectively adsorbed from the aqueous phase and transported to the overlying, quiescent solvent layer. This dissertation studied three-phase, continuous solvent sublation of pyrene and pentachlorophenol from simulated wastewater at the pilot-scale. Steady-state removal efficiencies as high as 96% for pyrene and 94% for pentachlorophenol were demonstrated and were significantly greater than the corresponding …

Genetics Of Oviposition Success Of A Thelytokous Fairyfly Parasitoid, Anagrus Delicatus, James T. Cronin, Donald R. Strong Jan 1996

Genetics Of Oviposition Success Of A Thelytokous Fairyfly Parasitoid, Anagrus Delicatus, James T. Cronin, Donald R. Strong

Faculty Publications

The foraging behaviour of the salt-marsh parasitoid, Anagrus delicatus (Hymenoptera: Mymaridae), is distinguished by few eggs laid per patch of hosts and frequent dispersal among patches. We investigated the within-population genetic variability in six quantitative traits associated with this unusual behaviour: fecundity (lifetime number of eggs), time on a patch, number of ovipositions per patch, oviposition rate, ovipositor length and tibia length (a measure of body size). Forty-one wasp isolines were initiated from single parthenogenetic females from three isolated salt marshes, and were maintained for up to eight generations in the greenhouse. We estimated the genetic variance and broad-sense heritability …

The Phytochrome Gene Family In Grasses (Poaceae): A Phylogeny And Evidence That Grasses Have A Subset Of The Loci Found In Dicot Angiosperms, S. Mathews, Robert A. Sharrock Jan 1996

The Phytochrome Gene Family In Grasses (Poaceae): A Phylogeny And Evidence That Grasses Have A Subset Of The Loci Found In Dicot Angiosperms, S. Mathews, Robert A. Sharrock

Faculty Publications

The phytochrome nuclear gene family encodes photoreceptor proteins that mediate developmental responses to red and far red light throughout the life of the plant. From studies of the dicot flowering plant Arabidopsis, the family has been modeled as comprising five loci, PHYA-PHYE. However, it has been shown recently that the Arabidopsis model may not completely represent some flowering plant groups because additional PHY loci related to PHYA and PHYB of Arabidopsis apparently have evolved independently several times in dicots, and monocot flowering plants may lack orthologs of PHYD and PHYE of Arabidopsis. Nonetheless, the phytochrome nucleotide data were informative in …

In Situ Analyses Of Methane Oxidation Associated With The Roots And Rhizomes Of A Bur Reed, Sparganium Eurycarpum, In A Maine Wetland, G. M. King Jan 1996

In Situ Analyses Of Methane Oxidation Associated With The Roots And Rhizomes Of A Bur Reed, Sparganium Eurycarpum, In A Maine Wetland, G. M. King

Faculty Publications

Methane oxidation associated with the belowground tissues of a common aquatic macrophyte, the burweed Sparganium eurycarpum, was assayed in situ by a chamber technique with acetylene or methyl fluoride as a methanotrophic inhibitor at a headspace concentration of 3 to 4%. Acetylene and methyl fluoride inhibited both methane oxidation find peat methanogenesis. However, inhibition of methanogenesis resulted in no obvious short-term effect on methane fluxes. Since neither inhibitor adversely affected plant metabolism and both inhibited methanotrophy equally well, acetylene was employed for routine assays because of its low cost and ease of use. Root-associated methanotrophy consumed a variable but significant …

Characterization Of The Denaturation And Renaturation Of Human Plasma Vitronectin I. Biophysical Characterization Of Protein Unfolding And Multimerization, Ping Zhuang, Michael N. Blackburn, Cynthia B. Peterson Jan 1996

Characterization Of The Denaturation And Renaturation Of Human Plasma Vitronectin I. Biophysical Characterization Of Protein Unfolding And Multimerization, Ping Zhuang, Michael N. Blackburn, Cynthia B. Peterson

Faculty Publications

Upon treatment with denaturing agents, vitronectin has been observed to exhibit conformational alterations which are similar to the structural changes detected when vitronectin binds the thrombin-antithrombin complex or associates with the terminal attack complex of complement. Denaturation and renaturation of vitronectin isolated from human plasma were characterized by changes in intrinsic fluorescence. Unfolding by chemical denaturants was irreversible and accompanied by self-association of the protein to form vitronectin multimers. Self-association was evaluated by equilibrium analytical ultracentrifugation which demonstrated that multimers form only during the refolding process after removal of denaturant, that multimeric vitronectin dissociates to constituent subunits readily upon treatment …

The Ecological Origins And Consequences Of Cattle Ranching In Sixteenth-Century New Spain, Andrew Sluyter Jan 1996

The Ecological Origins And Consequences Of Cattle Ranching In Sixteenth-Century New Spain, Andrew Sluyter

Faculty Publications

Despite the dramatic landscape changes Mexico has undergone due to the introduction of cattle ranching by the Spaniards in the early sixteenth century, the ecological origins of that pastroecosystem have remained obscure. About 1521, Gregorio de Villalobos implanted an Andalusian-derived herding ecology in the Gulf Coast lowlands that involved seasonal movement of cattle between wetlands and hill lands. Land-grant records demonstrate that as the colonial economy expanded, the transhumant model proliferated and came into conflict with those native settlements that had survived the pandemics of the early sixteenth century. In circumventing viceregal laws meant to protect native cultures and ecologies, …

Efficient Long-Pcr Site-Specific Mutagenesis Of A High Gc Template, Vladimir Chouljenko, Sukhanya Jayachandra, Galina Rybachuk, Konstantin G. Kousoulas Jan 1996

Efficient Long-Pcr Site-Specific Mutagenesis Of A High Gc Template, Vladimir Chouljenko, Sukhanya Jayachandra, Galina Rybachuk, Konstantin G. Kousoulas

Faculty Publications

A long-PCR method was developed for the efficient site-specific mutagenesis of herpes simplex virus (HSV-1) DNA fragments with high GC content. In this protocol, a PCR product was partially extended first using a cloned DNA fragment. The final mutagenized fragment was produced after a second extension using another PCR product and final amplification using external primers. The sequential use of two extension reduction increased the predicted frequency of the engineered mutation in the final product to 100%. This method was used to generate a mutated glycoprotein K (gK) gene specified by HSV-1. A recombinant virus that carried the mutated gK …

Optimal Culture Conditions For The Propagation Of The Oyster Pathogen Perkinsus Marinus (Apicomplexa) In Protein Deficient Medium, J. F. La Peyre, M. Faisal Jan 1996

Optimal Culture Conditions For The Propagation Of The Oyster Pathogen Perkinsus Marinus (Apicomplexa) In Protein Deficient Medium, J. F. La Peyre, M. Faisal

Faculty Publications

The protozoan, Perkinsus marinus, acclimated and proliferated in the culture medium JL-ODRP-1 without bovine serum albumin. The principal culture conditions for the optima propagation of P. marinus in this protein deficient culture medium, were determined. The greatest growth rates of the parasite occurred at 28 °C, 661 mOsm/kg and pH 7.5. Decreasing seeding densities from 16 × 105 to 1 × 105 also increased growth rate. After several passages in the absence of 5 % CO2 tension, the growth rote of P. marinus was similar to its original value in the presence of 5 % CO2 tension. This protein deficient …

The Htra Stress Response Protease Contributes To Resistance Of Brucella Abortus To Killing By Murine Phagocytes, Philip H. Elzer, Robert W. Phillips, Gregory T. Robertson, R. Martin Roop Jan 1996

The Htra Stress Response Protease Contributes To Resistance Of Brucella Abortus To Killing By Murine Phagocytes, Philip H. Elzer, Robert W. Phillips, Gregory T. Robertson, R. Martin Roop

Faculty Publications

Compared with virulent Brucella abortus 2308, the isogenic htrA mutant PHE1 shows decreased resistance to killing by cultured murine neutrophils and macrophages and significant attenuation during the early stages of infection in the BALB/c mouse model. These findings further define the contributions of the htrA gene product to the pathogenesis of B. abortus infections.
