DNA-based vaccines were employed to assess protective immunity against herpes simplex virus in experimental infections of hairless (strain SKH1) and BALB/c mice. Mice were vaccinated with plasmids containing the herpes simplex virus type-1 (HSV-1) glycoprotein B (gB) or D (gD) genes under the human cytomegalovirus immediate-early promoter control. Vaccines were injected intramuscularly (i.m.) or intraperitoneally (i.p.) as purified DNA alone or as formulations supplemented with different non-ionic block copolymers. Antibody responses were assessed by immunofluorescence and radioimmunoprecipitation assays. Mice inoculated with either gB or gD plasmid, alone or with non-ionic block copolymers CRL 1029 and CRL 1190, produced high levels …

The presence of the capacitative Ca2+ entry mechanism was investigated in porcine oocytes. In vitro-matured oocytes were treated with thapsigargin in Ca2+-free medium for 3 h to deplete intracellular calcium stores. After restoring extracellular calcium, a large calcium influx was measured by using the calcium indicator dye fura-2, indicating capacitative Ca2+ entry. A similar divalent cation influx could also be detected with the Mn2+-quench technique after inositol 1,4,5-triphosphate-induced Ca2+ release. In both cases, lanthanum, the Ca2+ permeable channel inhibitor, completely blocked the influx caused by store depletion. Heterologous expression of Drosophila trp in porcine oocytes enhanced the thapsigargin-induced Ca2+ influx. …

The mammalian sex determining gene, SRY, is the founding member of the new growing family of Sox (SRY-like HMG-box gene) genes. Sox genes encode transcription factors with diverse roles in development, and a few of them are involved in sex determination and differentiation. We report here the existence of Sox genes in the rice field eel, Monopterus albus, and DNA sequence information of the HMG box region of five Sox genes. The Sox1, Sox4 and Sox14 genes do not have introns in the HMG box region. The Sox9 gene and Sox17 gene, which each have an intron in the conserved …

Bovine brucellosis is a serious zoonotic disease affecting some populations of Rocky Mountain elk (Cervus elaphus nelsoni) and bison (Bison bison) in the Greater Yellowstone Area, USA. The fear that elk and/or bison may spread Brucella abortus to livestock has prompted efforts to reduce or eliminate the disease in wildlife. Brucella abortus strain RB51 (RB51) vaccine has recently been approved for use in cattle. Unlike strain 19 vaccine, RB51 does not cause false positive reactions on standard brucellosis serologic tests. If effective, it may become the vaccine of choice for wildlife. In February 1995, 45 serologically negative female elk calves …

An indirect enzyme-linked immunosorbent assay (ELISA) was developed to identify elk (Cervus elaphus nelsoni) with Brucella abortus strain RB51 (RB51)-specific antibodies using a mouse monoclonal antibody specific for bovine IgG1. This test was relatively easy to perform, accurate, and easily reproducible; therefore it could be standardized for use between laboratories. In addition, we attempted to compensate for inherent variabilities encountered when comparing ELISA readings from multiple samples taken from many animals over time. Optical density (OD) readings for each sample were converted into a percent positivity value for analysis. A negative cutoff value was determined above which a sample was …

Brucella abortus strain RB51 is used as a vaccine because it induces antibodies that do not react on standard serologic tests for brucellosis allowing differentiation between vaccination and infection. Strain RB51 was evaluated in captive elk (Cervus elaphus) to determine if vaccination protected against abortion following experimental challenge. Thirty elk were vaccinated intramuscularly with 1.0 × 1010 colony-forming units (CFU) of strain RB51 in March 1998. Fourteen of these were given a booster dose of 1.13 × 1010 CFU exactly 1 yr later. All vaccinated elk seroconverted via a modified dot blot assay to strain RB51 with the booster group …

The Ta (transcribed, subset a) subfamily of L1 LINEs (long interspersed elements) is characterized by a 3-bp ACA sequence in the 3′ untranslated region and contains ∼520 members in the human genome. Here, we have extracted 468 Ta L1Hs (L1 human specific) elements from the draft human genomic sequence and screened individual elements using polymerase-chain-reaction (PCR) assays to determine their phylogenetic origin and levels of human genomic diversity. One hundred twenty-four of the elements amenable to complete sequence analysis were full length (∼6 kb) and have apparently escaped any 5′ truncation. Forty-four of these full-length elements have two intact open …

The bilayered rhodium oxide (formula presented) was synthesized by high-pressure and high-temperature heating techniques. The single-phase polycrystalline sample of (formula presented) was characterized by measurements of magnetic susceptibility, electrical resistivity, specific heat, and thermopower. The structural characteristics were investigated by powder neutron diffraction study. The rhodium oxide (formula presented) (formula presented) is isostructural to the metamagnetic metal (formula presented) with five (formula presented) electrons per Rh, which is electronically equivalent to the hypothetical bilayered ruthenium oxide, where one electron per Ru is doped into the Ru-327 unit. The present data show the rhodium oxide (formula presented) to be metallic with …

Fåk, van Dijk, and Wills (FDW) question our analysis of elastic neutron-scattering experiments in the antiferromagnetic phase of (formula presented) They state that our analysis is incorrect because we average over magnetic structures that are disallowed by symmetry. We disagree with FDW and reply to their criticism below. FDW also point out that we have mistaken the magnetic-field direction in the experiment reported in N. H. van Dijk, B. Fåk, L. P. Regnault, A. Huxley, and M-T. Fernández-Díaz, Phys. Rev. B 58, 3186 (1998). We correct this error and note that our previous conclusion is also valid for the correct …

Density Of States In (Formula Presented)-Wave Superconductors Disordered By Extended Impurities, İnanç Adagideli, Daniel E. Sheehy, Paul M. Goldbart
Faculty Publications
The low-energy quasiparticle states of a disordered d-wave superconductor are investigated theoretically. A class of such states, formed via tunneling between the Andreev bound states that are localized around extended impurities (and result from scattering between pair-potential lobes that differ in sign), is identified. Its (divergent) contribution to the total density of states is determined by taking advantage of connections with certain one-dimensional random tight-binding models. The states under discussion should be distinguished from those associated with nodes in the pair potential. © 2002 The American Physical Society.

Magnetic Field Induced Charge And Spin Instabilities In Cuprate Superconductors, M. Franz, D. E. Sheehy, Z. Tešanović
Faculty Publications
A [Formula presented]-wave superconductor, subject to strong phase fluctuations, is known to suffer an antiferromagnetic instability closely related to the chiral symmetry breaking in ([Formula presented])-dimensional quantum electrodynamics ([Formula presented]). Based on this idea we formulate a “ [Formula presented] in a box” theory of local instabilities of a [Formula presented]-wave superconductor in the vicinity of a single pinned vortex undergoing quantum fluctuations. As a generic outcome we find an incommensurate 2D spin density wave forming in the neighborhood of a vortex with a concomitant “checkerboard” pattern in the local electronic density of states, in agreement with recent neutron scattering …

An Optimal Extraction Of Spatially Blended Spectra, R. I. Hynes
An Optimal Extraction Of Spatially Blended Spectra, R. I. Hynes
Faculty Publications
We describe an algorithm to optimally extract individual spectra of blended sources from a long slit spectrum. A semi-analytic model for the spatial profile is used: a Voigt profile for the undersampled core with a numerical correction applied to the wings. This is derived from an isolated source which must also be placed on the slit. χ2 fitting is used to separate the blended components with maximum weight given to the best data. We demonstrate the successful application of the algorithm to data on two X-ray binaries in crowded fields: XTE J2012+381 and V404 Cyg.

We compare ultraviolet (UV) spectra of the recent soft X-ray transients XTE J1118+480 and XTE J1859+226. The emission line strengths in XTE J1118+480 strongly suggest that the accreting material has been CNO processed. We show that this system must have come into contact with a secondary star of about 1.5 M⊙, and an orbital period ∼ 15 h, very close to the bifurcation value at which the nuclear and angular momentum loss time-scales are similar. Subsequent evolution to the current period of 4.1 h was driven by angular momentum loss. In passing through a period of 7.75 h the secondary …

We present a compilation of spectroscopic observations of the sgB[e] star CI Cam, the optical counterpart of XTE J0421+560. This includes data from before, during, and after its 1998 outburst, with quantitative results spanning 37 years. The object shows a rich emission line spectrum originating from circumstellar material, rendering it difficult to determine the nature of either star involved or the cause of the outburst. We collate all available pre-outburst data to determine the state of the system before this occurred and provide a baseline for comparison with outburst and post-outburst data. During the outburst all lines become stronger, and …

The Sudbury Neutrino Observatory (SNO) has measured day and night solar neutrino energy spectra and rates. For charged current events, assuming an undistorted [Formula presented] spectrum, the night minus day rate is [Formula presented] of the average rate. If the total flux of active neutrinos is additionally constrained to have no asymmetry, the [Formula presented] asymmetry is found to be [Formula presented]. A global solar neutrino analysis in terms of matter-enhanced oscillations of two active flavors strongly favors the large mixing angle solution. © 2002 The American Physical Society.

Limits on [Formula presented] and [Formula presented] oscillations are extracted using the NuTeV detector with sign-selected [Formula presented] and [Formula presented] beams. In [Formula presented] mode, for the case of [Formula presented], [Formula presented] is excluded, and for [Formula presented], [Formula presented]. The NuTeV data exclude the high [Formula presented] end of [Formula presented] oscillation parameters favored by the LSND experiment without the need to assume that the oscillation parameters for [Formula presented] and [Formula presented] are the same. We present the most stringent experimental limits for [Formula presented] oscillations in the large [Formula presented] region. © 2002 The American …
