Type 1 diabetes (T1D) is an autoimmune disease where the immune system targets the pancreatic islet beta-cells, which are responsible for insulin production and secretion, leading to a state of insulin insufficiency. Many of the molecular details of disease onset and progression remain poorly understood, including key aspects of cytokine signaling. Therefore, we have examined some of the consequences of inflammatory cytokine signaling as a critical module relevant to T1D. Using interleukin-1 (IL-1) signaling as a well-defined paradigm for inflammation, we investigated key components of this signaling pathway in pancreatic beta-cells. Emily Somebody is a chamber opera in three acts that tells the story of the friendship between the American poet Emily Dickinson and the man that would become her first editor, Mr. Thomas Higginson. This story, for me, is ultimately about a poet (or an artist) in the process of finding not only her own voice but also discovering what poetry and art mean to her, in the most genuine way, and standing up for these beliefs and values. All the lyrics used in this work are from Emily Dickinson’s poems and letters exchanged between her and Mr. Thomas Higginson.

Increasing environmental concerns are encouraging the adoption of sustainable landscapes that have environmental, social, and financial benefits. Ornamental groundcover systems are touted as sustainable landscape features due to the associated decreased demand of water, fertilizer, pesticide, and maintenance; however, limited research exists on soil property effects, planting density, weed density, or consumer preferences of groundcovers. Psychomotor retardation (PMR) is a diagnostic criterion that reflects a gross slowing of movement, speech, and thought. More so, PMR is a transdiagnostic, pervasive source of impairment and distress that can be observed across disorders, such as Major Depressive Disorder (MDD), Schizophrenia (SZ), and Bipolar Disorder (BD). In clinical settings, PMR is associated with more severe psychopathology and is used to both inform treatment and predict long-term treatment outcomes. Mud settling velocity is controlled by flocculation, which in turn strongly depends on turbulence and on the chemistry and biology of the water-sediment mixture. As a result, mud settling velocity can be poorly constrained in coastal areas and vary in space and time by orders of magnitude. Here we quantified mud settling velocity in Barataria Basin, a deltaic estuary in Louisiana (USA), using three independent methods: eddy covariance (one station for 200 days), floc cameras (4 stations at one time), and Rouse profile inversion (14 stations, replicated 10-30 times each). A series of amorphous hydrogenated chromium containing diamond-like carbon films were deposited on 316 stainless steel substrates with a chromium interlayer through inductively coupled plasma assisted direct current magnetron sputtering. Details on the deposition process of these films are discussed. The film is characterized using scanning electron microscopy, X-ray photoelectron spectroscopy, X-ray diffraction, energy dispersive spectroscopy, and transmission electron microscopy. The coating hardness and indentation modulus is measured by instrumented nanoindentation. The coefficient of friction and the upper bound of the coating wear coefficient is measured using the pin-on-disk method. The fracture toughness of the diamond-like carbon film and a …

The development of children’s pragmatic language, which can be defined as conversational competence in social contexts, has been attributed to variation in the language they hear in their environment. Russia launched a military invasion of Ukraine on 24 February 2022, setting off Europe’s largest war since World War II. The war captured the attention of American news organizations, which gave the conflict unprecedented levels of coverage. This thesis tested the coverage of the war in Ukraine by two major American news outlets: The New York Times and CNN. This study used qualitative content analysis to examine the top four front-page stories of the Times and CNN Newsroom show. The lack of involvement of the autistic community and its stakeholders in autism research has led to a call to action. At present, autism researchers have limited knowledge about stakeholders’ priorities for research. The current study aimed to bridge the disconnect between the extant autism research and the recent neurodiversity movement by surveying key stakeholders – parents of autistic children – on their perspectives on autism research that focuses on language. The McMurdo Dry Valleys is the largest ice-free area in Antarctica, but seasonal snow covers the valley floors sporadically throughout the year. In this study, a model to estimate areal snow coverage from satellite imagery was created. An area-volume model was created to estimate the amount of snow water equivalent (SWE) from the snow area extracted from the imagery. Snow cover influences the total albedo, the hydrologic budget, and the soil moisture and soil temperature in Taylor Valley (TV). Renal disease is a common ailment of captive reptiles that is often closely linked to chronic, subclinical states of dehydration. Currently, the diagnosis of renal disease in reptiles is poorly characterized and often relies on invasive diagnostic techniques (e.g., renal biopsy) for definitive diagnosis. A potential outlet for the further characterization of renal dysfunction in reptiles is the induction of dehydration by furosemide therapy. Levinas offers a rich philosophical insight into the kind of responsibility and ethics that we must have for the Other. This involves a certain conception of what it means to be hospitable which turns out to be impossible. In order to talk about how this impossible relationship can occur, I use Heidegger’s description of the existential Being-with structure and Derrida’s conception of the event in order to make sense of how this Levinasian relationship can possibly exist in spite of—or thanks to—its impossibility.

Passionate fandoms centered on popular film and television series can elicit large-scale positive fan activity. At the same time, however, negative consequences can arise as factions or subsets of members with varying opinions arise within fandoms. One possible explanation for this phenomenon is varied levels among fandom members of how prototypical they view themselves within the context of that fandom. There have been over 350 K-12 school shootings since the shootings at Columbine High School in 1999 (Cox et al., 2023). Thousands of journalists cover these attacks each in their own way with unique motives. This information goes to the public, and it is up to that person how they want to consume the story and view the shooting. National and local journalists tend to cover school shootings quite differently throughout time with unique frames. After spring migrations to natal rivers Gulf Sturgeon Acipenser oxyrinchus desotoi reside in freshwater holding areas prior to fall emigrations to nearby estuaries. Limited published information on the physical habitat characteristics of these holding areas is available, especially for juvenile fish in the western portion of the Gulf Sturgeon’s distribution. To further investigate freshwater occupancy and movement patterns, 32 acoustic receivers were deployed from late April to October of 2021 and 2022 in 1600-m reaches distributed throughout the lower Pearl River, Louisiana-Mississippi, the westernmost river within their distribution. The edges of a graph have natural cyclic orderings. We investigate the matroids for which a similar cyclic ordering of the circuits is possible. A full characterization of the non-binary matroids with this property is given. Evidence of the difficulty of this problem for binary matroids is presented, along with a partial result for binary orderable matroids.

Floods represent one of the most severe natural disasters worldwide. It is crucial to accurately assess flood hazards to mitigate the impact and enhance community resilience. This study focuses on three key components of flood hazard management, including (1) flood frequency estimation, (2) inundation delineation, and (3) flood susceptibility assessment. The first project presents a Bayesian Log-Pearson Type III model with Spatial Priors (BLP3-SP) that utilizes spatial regression priors to enhance the accuracy of flood frequency estimation for sites with limited data. This dissertation concerns a linear-quadratic elliptic distributed optimal control problem with pointwise state constraints in two spatial dimensions, where the cost function tracks the state at points, curves and regions of a domain.
First we explore the elliptic optimal control problem subject to pointwise control constraints. This problem is reduced into a problem that only involves the control. Seasonal energy demand requires relatively cheap, impermeable, non-reactive materials for storing vast amounts of natural gas and hydrogen. The present work examines the natural law jurisprudence of John Finnis. It argues that Finnis’s teaching is a genuinely new natural law theory. Finnis’s jurisprudence is not a re- presentation of the jurisprudence of St. Thomas Aquinas because its central element—a doctrine of natural rights—is a departure from Aquinas’s natural law teaching. In support of these claims, the present work relies upon the scholarship of Ernest L. Fortin, A.A. Following Fr. Fortin, it presents an understanding of the natural law that endorses a clear distinction between natural right and natural rights—between premodern political philosophy and modern political philosophy.

Though these two thinkers are not often examined from this angle, and though there is controversy surrounding just how phenomenological each thinker might actually be, an examination of the two thinkers in this regard serves to identify interesting connections between Hegel and Heidegger while also raising questions about phenomenology in general. Most existing autograders used for grading programming assignments are based on unit testing, which is tedious to implement for programs with graphical output and does not allow testing for other code aspects, such as programming style or structure. We present a novel autograding approach based on machine learning that can successfully check the quality of coding assignments from a high school-level CS-for-all computational thinking course. For evaluating our autograder, we graded 2,675 samples from five different assignments from the past three years, including open-ended problems from different units of the course curriculum. Changes to societal norms, such as the educational, marital, and child-bearing expectations have coincided with significant increases in infertility worldwide. Surprisingly, male infertility is responsible for approximately half of all infertility cases worldwide. Thus, a better understanding of sperm development, and how it is affected by age, may permit the design and application of therapeutics to treat various cases male infertility. Here, I have found that lysosomes acidify as germ cells enter the spermatocyte stage. Once active, lysosomes turn over E-cadherin, and likely other proteins, to support plasma membrane stability. We address the open question of which representations of the modular group SL(2,Z) can be realized by a modular category. In order to investigate this problem, we introduce the concept of a symmetrizable representation of SL(2,Z) and show that this property is necessary for the representation to be realized. We then prove that all congruence representations of SL(2,Z) are symmetrizable. The proof involves constructing a symmetric basis, which greatly aids in further calculation. We apply this result to the reconstruction of modular category data from representations, as well as to the classification of semiregular categories, which are defined via an …

Svip Dependent Tubular Lysosomal Restoration: A New Direction In Vcp Diseases, Ankita Basu

Multisystem Proteinopathy-1 (MSP-1) is a rare, autosomal dominant, degenerative disease resulting from single amino acid mis-sense mutations in the AAA+ ATPase, Valosin containing protein (VCP). Patients with VCP mutations vary widely in their pathology and clinical penetrance, making it difficult to devise effective treatment strategies. To investigate the pathology of VCP- related disease, we engineered CRISPR knock-in Drosophila models for nine MSP-1 associated VCP mutations. These mutant animals mimic human MSP-1 disease phenotypes, including progressive mobility decline and accumulation of protein aggregates, making them excellent models to study the pathology of VCP associated mutations and test potential treatment strategies.
