© 2018 The horn fly, Haematobia irritans irritans (Linnaeus, 1758; Diptera: Muscidae), a hematophagous external parasite of cattle, causes considerable economic losses to the livestock industry worldwide. This pest is mainly controlled with insecticides; however, horn fly populations from several countries have developed resistance to many of the products available for their control. In an attempt to better understand the adult horn fly and the development of resistance in natural populations, we used an Illumina paired-end read HiSeq and GAII approach to determine the transcriptomes of untreated control adult females, untreated control adult males, permethrin-treated surviving adult males and permethrin …

Caseinolytic peptidase P mediates degradation of unfolded mitochondrial proteins and activates mitochondrial unfolded protein response (mtUPR) to maintain protein homeostasis. Clpp −/− female mice generate a lower number of mature oocytes and two-cell embryos, and no blastocysts. Clpp −/− oocytes have smaller mitochondria, with lower aspect ratio (length/width), and decreased expression of genes that promote fusion. A 4-fold increase in atretic follicles at 3 months, and reduced number of primordial follicles at 6–12 months are observed in Clpp −/− ovaries. This is associated …

Beta-delayed neutron emission is being studied by detecting the β particles and recoiling ions emerging from the Beta-decay Paul Trap. For β decays to the ground state or γ-emitting states of the daughter nucleus, the fraction of recoiling ions which reach the ion detector in coincidence with a β particle has been determined for 134, 135Sb, 137, 138, 140I, and 144, 145Cs. This value is needed for the determination of the β-delayed neutron emission branching ratio solely from the recoil-ion time-of-flight (TOF) spectrum. The β-particle energy and recoil-ion TOF spectra were used to constrain a simple decay model, which can …

We demonstrate that when a waveguide beam splitter (BS) is excited by N indistinguishable photons, the arising multiphoton states evolve in a way as if they were coupled to each other with coupling strengths that are identical to the ones exhibited by a discrete fractional Fourier system. Based on the properties of the fractional Fourier transform, we then derive a multiphoton suppression law for 50/50 BSs, thereby generalizing the Hong–Ou–Mandel effect. Furthermore, we examine the possibility of performing simultaneous multiphoton quantum random walks by using a single waveguide BS in combination with photon-number-resolving detectors. We anticipate that the multiphoton lattice-like …

This paper reports measurements of final-state proton multiplicity, muon and proton kinematics, and their correlations in charged-current pionless neutrino interactions, measured by the T2K ND280 near detector in its plastic scintillator (C8H8) target. The data were taken between years 2010 and 2013, corresponding to approximately 6×1020 protons on target. Thanks to their exploration of the proton kinematics and of imbalances between the proton and muon kinematics, the results offer a novel probe of the nuclear-medium effects most pertinent to the (sub-)GeV neutrino-nucleus interactions that are used in accelerator-based long-baseline neutrino oscillation measurements. These results are compared to many neutrino-nucleus interaction …

The Fermi Gamma-ray Burst Monitor (GBM) is currently the most prolific detector of gamma-ray bursts (GRBs). Recently the detection rate of short GRBs (SGRBs) has been dramatically increased through the use of ground-based searches that analyze GBM continuous time-tagged event (CTTE) data. Here, we examine the efficiency of a method developed to search CTTE data for sub-threshold transient events in temporal coincidence with LIGO/Virgo compact binary coalescence triggers. This targeted search operates by coherently combining data from all 14 GBM detectors by taking into account the complex spatial and energy dependent response of each detector. We use the method to …

© 2018 American Society of Plant Biologists. All rights reserved. The water stress-associated hormone abscisic acid (ABA) acts through a well-defined signal transduction cascade to mediate downstream transcriptional events important for acclimation to stress. Although ABA signaling is known to function in specific tissues to regulate root growth, little is understood regarding the spatial pattern of ABA-mediated transcriptional regulation. Here, we describe the construction and evaluation of an ABSCISIC ACID RESPONSIVE ELEMENT (ABRE)-based synthetic promoter reporter that reveals the transcriptional response of tissues to different levels of exogenous ABA and stresses. Genome-scale yeast one-hybrid screens complemented these approaches and revealed …

All three epithelial Na+ channel (ENaC) subunits (α, β, and γ) and the mineralocorticoid receptor (MR), a known regulator of ENaC, are located in vasopressin (VP) synthesizing magnocellular neurons in the hypothalamic supraoptic (SON) and paraventricular (PVN) nuclei. Our previous study showed that ENaC mediates a Na+ leak current that affects the steady-state membrane potential of VP neurons. This study was conducted in Dahl salt-sensitive (Dahl-SS) rats to determine if any abnormal responses in the expression of ENaC …

© 2018 The Authors Objective: Pancreatic tissue, and islets in particular, are enriched in expression of the interleukin-1 receptor type I (IL-1R). Because of this enrichment, islet β-cells are exquisitely sensitive to the IL-1R ligands IL-1α and IL-1β, suggesting that signaling through this pathway regulates health and function of islet β-cells. Methods: Herein, we report a targeted deletion of IL-1R in pancreatic tissue (IL-1RPdx1−/−) in C57BL/6J mice and in db/db mice on the C57 genetic background. Islet morphology, β-cell transcription factor abundance, and expression of the de-differentiation marker Aldh1a3 were analyzed by immunofluorescent staining. Glucose and insulin tolerance tests were …

Background: The structural information on proteins in their ligand-bound conformational state is invaluable for protein function studies and rational drug design. Compared to the number of available sequences, not only is the repertoire of the experimentally determined structures of holo-proteins limited, these structures do not always include pharmacologically relevant compounds at their binding sites. In addition, binding affinity databases provide vast quantities of information on interactions between drug-like molecules and their targets, however, often lacking structural data. On that account, there is a need for computational methods to complement existing repositories by constructing the atomic-level models of drug-protein assemblies that …

Copyright © 2018 The Authors. The Amazon River and its major tributaries delimit the distributions of hundreds of terrestrial taxa. It remains unclear whether river-bounded distributions and taxon replacements reflect the historical role of rivers in generating species diversity as vicariant forces, or are the result of their role as secondary barriers, maintaining current levels of species diversity by inhibiting gene flow and population introgression. We use a community-wide comparative phylogeographic and phylogenetic approach to address the roles that the Rio Negro and the Rio Branco play in the avian speciation process in the Guiana Shield. The Stochastic Order Of Probability Measures On Ordered Metric Spaces, Fumio Hiai, Jimmie Lawson, Yongdo Lim

The general notion of a stochastic ordering is that one probability distribution is smaller than a second one if the second attaches more probability to higher values than the first. Motivated by recent work on barycentric maps on spaces of probability measures on ordered Banach spaces, we introduce and study a stochastic order on the space of probability measures P(X), where X is a metric space equipped with a closed partial order, and derive several useful equivalent versions of the definition. We establish the antisymmetry and closedness of the stochastic order (and hence that it is a closed partial order) …

Representation Stability For The Cohomology Of Arrangements Associated To Root Systems, Christin Bibby

From a root system, one may consider the arrangement of reflecting hyperplanes, as well as its toric and elliptic analogues. The corresponding Weyl group acts on the complement of the arrangement and hence on its cohomology. We consider a sequence of linear, toric, or elliptic arrangements which arise from a family of root systems of type A, B, C, or D, and we show that the rational cohomology stabilizes as a sequence of Weyl group representations. Our techniques combine a Leray spectral sequence argument similar to that of Church in the type A case along with FI -module theory which …

Dissolved carbon in lakes play a vital role in the global carbon cycling. The concentration and dynamics of lake dissolved carbon can be influenced by both the surrounding landscape and a combination of physical, chemical and biological processes within the lakes themselves. From 2009 to 2016, we conducted a large-scale assessment of dissolved organic carbon (DOC) and dissolved inorganic carbon (DIC) in 249 lakes across a diverse range of climatic, geopedologic, topographical and hydrological conditions in five Chinese limnetic regions: the East Limnetic Region (ELR), the Northeast Limnetic Region (NLR), the Inner Mongolia-Xinjiang Limnetic Region (MXR), the Yungui Limnetic Region …

Discharging 680 km3 of freshwater annually to the Northern Gulf of Mexico (NGOM), the Mississippi-Atchafalaya River System (MARS) plays a significant role in transporting major and trace elements to the ocean. In this study, we analyzed total recoverable concentrations of thirty-one metals from water samples collected at five locations along the MARS during 2013-2016 to quantify their seasonal mass exports. The Atchafalaya River flows through a large swamp floodplain, allowing us to also test the hypothesis that floodplains function as a sink for metals. We found that the seven major elements (Ca, Na, Mg, Si, K, Al, and Fe) constituted …

Crappie, Pomoxis spp., are popular game fish throughout North America and are produced by public and private hatcheries. However, production is limited by a lack of information on tank culture and induced spawning methods. Development of techniques for storage of sperm and in vitro fertilization would increase flexibility in spawning. Therefore, techniques for sperm cryopreservation were examined in white crappie, Pomoxis annularis. Sperm from adult wild white crappie were used to evaluate sperm extender, cryoprotectant agent and concentration, and cooling technique based on post-thaw sperm motility. Percent egg fertilization was also compared between sperm stored in the two best cryopreservation …

Addressing Reproducibility In Cryopreservation, And Considerations Necessary For Commercialization And Community Development In Support Of Genetic Resources Of Aquatic Species, Leticia Torres, Terrence R. Tiersch
Faculty Publications
For the past six decades a repeated cycle of developing new cryopreservation protocols or simply reinventing them to counteract a lack of reproducibility has led to hundreds of published studies that have offered little to the establishment of a genetic resources community for aquatic species. This has hampered repository development and inhibited industrial application. Most protocols were developed without standardized approaches, leading to irreproducible studies and questionable or meaningless comparisons. Thus cryopreservation of germplasm in aquatic species would greatly benefit from strategies to facilitate reproducibility. Our objectives were to: (1) identify major sources of irreproducibility across research, small-scale, repository, and …

In this study, nutrient properties and carbon sequestration potential of biochars derived from chicken manure (CM) impregnated with mineral salts (calcium chloride, magnesium chloride, ferric chloride) were evaluated. Pretreatment with mineral salts reduced phosphorus (P) availability via the formation of insoluble metal phosphate minerals. Less carbon was lost during the pyrolysis of pretreated CM, and the produced biochars (BCCa, BCMg, and BCFe) were more stable (i.e., reduced C loss during chemical oxidation and less CO2 release during incubation) than pristine biochars. Spectroscopic evidence indicated that enhanced biochar stability via metal salt pretreatment before pyrolysis was related to increased aromatization and …
