Phthalocyanines (Pcs) are excellent candidates for use as fluorophores for near-infrared (near-IR) fluorescent tagging of biomolecules and as photodynamic therapy (PDT) agents. Synthesis of Pcs with asymmetrical substitution on the periphery is often difficult due to the problems during the purification of the Pc mixtures obtained. The objective of this project is to design and synthesize chemically robust near-IR fluorophores for bioanalytical applications and to develop new synthetic methods for rapid synthesis of the target compounds. In arid and semi-arid parts of the Old World, Phlebotomus paptasi is a significant biting pest of man and is the primary vector of Leishmania major, the causative agent of zoonotic cutaneous leishmaniasis (ZCL). Phlebotomus papatasi exhibits a close association with the burrowing rodents that serve as the reservoirs of L. major. Rodent burrows are considered to be the primary habitat of immature P. papatasi in ZCL foci, and sand fly larvae have been observed feeding on rodent feces. Heme oxygenase-1 (HO-1) catalyzes the initial and rate-limiting reaction in heme catabolism (yielding equimolar quantities of iron, carbon monoxide, and biliverdin), which is further metabolized to bilirubin by biliverdin reductase. HO-1, an important enzyme with antioxidant and cytoprotective properties, functions against oxidative stress generated in response to heme and to various stress stimuli, such as zinc. The cytoprotective function of HO-1 is dependent on both the activities of the reaction products and on the inducibility of the heme oxygenase gene (hmox-1), which is regulated primarily at the level of gene transcription by two distal enhancers, E1 and E2. Oncolytic herpes simplex virus type-1 (HSV-1) has shown great potential as an effective agent for cancer therapy ¨C oncolytic virotherapy. To enhance oncolytic capabilities of previously reported agents, new oncolytic and fusogenic HSV-1 OncSyn and OncdSyn viruses were constructed based on wild type HSV-1 (F) strain. To provide for safety and tumor selectivity, the viruses carried a large deletion including one of the two ¦Ã134.5 genes. The unique configuration of the human clavicle and mastoid process suggests a functional connection between the head and shoulders in humans. The hypothesis in this study is that the clavicle, scapula and head form a functional complex and are interconnected by the sternocleidomastoid and trapezius muscles. In this complex, the trapezius muscles attach to the skull and become active when loads are carried. The sternocleidomastoid muscles are anchored to the clavicles; when loads are being carried, they act as guy ropes for the head, thereby keeping the head from being extended by the force of the contracting trapezius muscles. This dissertation explores the development of analytical methods for studies of CVD biomarkers and related biomolecular indicators. Initially, spectroscopic studies were conducted to investigate the chemical reactivity of homocysteine (Hcy), an independent CVD risk factor and serological biomarker. Consequently, we proposed an alternate theory for in vivo Hcy clearance based on spontaneous pyridoxal tetrahydrothiazine (PT) formation from Hcy and pyridoxal. The validity of PT-assisted Hcy clearance was further evaluated by use of capillary electrophoretic methods, which allowed rapid monitoring of protein oligomerization in PT-protein reaction mixtures. Rapid rotation of guanine base derivatives about Pt–N7 bonds results in fluxional behavior of models of the key DNA intrastrand G–G cross-link leading to anticancer activity of Pt(II) drugs (G = deoxyguanosine). This behavior impedes the characterization of LPtG2 models (L = one bidentate or two cis-unidentate carrier ligands; G = guanine derivative). The objective of this study is to understand the types of conformers formed as L is systematically varied. I take a comparative approach to literatures of the Americas because of the dearth of research examining the literatures of both continents side by side, particularly literatures by and about Indigenous Americans. The overall objective of the research presented here is to understand the nanoscopic and microscopic structures that exist in dispersions of laponite and montmorillonite in poly(ethylene oxide), (PEO) and to correlate their shear responses with the final structure and properties of multilayered nanocomposite films prepared from these dispersions. The purpose of this qualitative study was to describe the lived experience of couples residing in Southeastern Louisiana who were diagnosed with infertility and underwent infertility treatments. The study was a phenomenological qualitative research design, with researcher-developed guiding questions to help direct interviews. The sample was purposeful and was drawn from the patient base of a local endocrinologist. The couples who participated were greater than 18 years of age and had discontinued treatment one year prior to implementation of the study. Investigations of hurricane disturbances on coastal vegetated communities are common, but relatively few are comprehensive across broad geographic regions. The 2005 hurricanes, Katrina and Rita, exposed Louisiana coastal landscapes to physical modifications and extensive and prolonged flooding, resulting in measurable physicochemical changes. This research used remote sensing and field investigations to regionally assess (1) porewater salinity and sulfide impacts to and recovery of coastal Louisiana vegetation communities, and (2) the importance of mineral sediment deposition on accretionary processes. Experiments were conducted in the state of Louisiana to evaluate methods to asses and potentially reduce water quality issues. Such events can lead to the collapse of a bridge, causing deaths, shutting down a primary route of transportation and costing millions of dollars in repairs. The overall goal of the work is to develop methods and instrumentation to minimize sample treatment and maximize sensitivity and selectivity for bioaerosol analysis. This work is divided into three parts: sample preparation, test bioaerosol analysis, and instrument development. The delay in early height growth (EHG) has been a limiting factor for artificial regeneration of longleaf pine (Pinus palustris Mill.). Simple Sequence Repeat (SSR) markers have been used to map the genome and quantitative trait loci controlling the EHG in a backcross family (longleaf pine x slash pine) x longleaf pine. A total of 228 locus specific SSR markers were screened against 6 longleaf pine recurrent parents and a sample of 7 longlef x slash pine hybrid parents. The dimensions of microfluidic devices closely parallel those of biological cells; thusly, they are excellent platforms for the speciation, transport, manipulation, and analysis of cells. Electrokinetic transport of Escherichia coli and Saccharomyces cerevisiae was evaluated in microfluidic devices fabricated in pristine and UV-modified poly(methylmethacrylate) and polycarbonate. The magnitude and direction of transport of the cells was dictated by the buffer composition, conduit surface chemistry, and intrinsic cellular electrical properties. Baker’s yeast in all devices migrated toward the cathode, because of their smaller electrophoretic mobility compared to the electroosmotic flow of the polymer. This research focused on four previously identified single nucleotide polymorphisms in a leptin gene on chromosome 4 of beef cows. The SNP were E2FB, T945M, UA1, and UA2. Speciation, geographic variation, and genetic differentiation are fundamental processes that generate diversity, and understanding these processes are major goals of evolutionary biology. Evolutionary phenomena may be more observable on islands as compared to continental landmasses as a result of small population sizes, unoccupied niches, and the relative simplicity of island systems and their populations: physical isolation, shorter (and often well documented) geologic time scale, reduced faunal diversity, and lack of outside faunal influence. A ~5000 yr history of modern and prehistoric hurricane landfall and environmental change has been reconstructed from coastal lake sediments of the northern West Indies. Hurricane overwash layers and environmental shifts were identified by changes in biological and sedimentary stratigraphies, core lithology, and loss-on-ignition techniques. 14C and 137Cs were used to establish a chronology of events. Many hurricane-induced overwash layers were identified within the lake sediments. Evidence indicates distinct periods of temporal shifts in hurricane activity for the Northern Caribbean islands of Anguilla, Barbuda, and Acklins Island, Bahamas, providing the first paleotempestological records for the region. The rice stink bug (Oebalus pugnax) is the major pest of late-season rice in Louisiana. Prior studies indicate that large populations of mosquitoes are often present in rice fields during the time that rice fields are infested with rice stink bugs. Lambda-cyhalothrin (Karate®) is widely used to control rice stink bugs. The produce of travels to the area of the Mississippi River delta, along with text-based research, formed the materials from which two evenings of participative performance were crafted. Personal narratives were used alongside evaluations of administrative programs, and the space in the Backyard Gallery was transformed using shovels, buckets, and teamwork. Found chairs, rope, and other objects, through ritualized interaction, became an aesthetic formation; everyone present worked together in the assembly of a boat, which contained the results of these interactions between myself, six volunteers, and an audience turned participants. Microcosm and mesocosm scale experiments were carried out to investigate the effects of vegetation and substrates on the performance of engineered wetland systems (EWSs) meant to treat mixtures of volatile organic compounds (cVOCs). The experiments were conducted with a view to optimizing contributions of these wetland components. Phragmites communis and Typha latifolia, the commonest wetland plant species, were compared in mesocosm scale EWSs. Eight common wetland plant species were compared using microcosm experiments based on the potential of their rootmatter to enhance treatment of cVOC mixtures.
