Polymerase Chain Reaction (PCR) is a powerful enzymatic reaction commonly used to amplify specific sequences of Deoxyribo Nucleic Acid (DNA). Since the introduction of the lab on a chip concept, numerous Continuous Flow PCR cyclers were realized with success at the micro scale. As reducing the reactor size and improving thermal management led to reduced sample volumes, results could be achieved much faster with these CF-PCR cyclers than with common commercial cycler. Static low density media (SLDM) filtration has been received with great admiration in the aquaculture industry and consequently spawned into the domestic wastewater setting on the pilot scale. This method of granular filtration uses plastic beads to facilitate the roles of both clarification and biofiltration into one compact unit. The fundamental operating principles utilized in this form of treatment can be greatly enhanced when they are coupled with self contained recirculation from airlift pumps. In November 2004, bluetongue virus (BTV) serotype 1 was detected for the first time in the U.S. from a hunter-killed deer in the marsh area of the Atchafalaya Delta in St. Mary Parish, LA. Subsequent serum surveys of three cattle farms further inland from the area where the deer was shot found bluetongue virus serotype 1 positive cattle on two of the three farms. Recently we have begun to understand the importance of inshore hard-bottom substrate, including oyster reefs, to estuarine fish communities in the Gulf of Mexico, especially in the context of identifying Essential Fish Habitat (EFH). However, problems such as habitat loss, disease, overharvest, and failure to replace shell have severely decreased the amount of high-relief oyster reef habitat available to finfish. The purpose of this research was to determine the influence of dietary Se on glutathione peroxidase (GPx)-1 and 3 activities and relative liver GPx mRNA levels in growing Holstein bull calves. Calves (n = 14) were started 28 d after birth on either a Se adequate (0.15 ppm Se) or deficient (0.01 ppm Se) diet consisting of 3 growth phases and maintained on the diet until 180 d of age. Forty-eight calves (heifers n=20, bulls n=28) were randomly assigned one of four dietary treatments which included no additive (C); the yeast culture Saccharomyces cerevisiae (YC); the probiotics Bacillus lichenformis and Bacillus subtilis (P); and both yeast culture and probiotics (YCP). Calves were administered treatments from day 2 to 56 in starter and from day 57 to 84 in grower diets. Body weights were measured at birth and weekly thereafter until day 112 of age. The biological profile is one of the most important things that forensic anthropologists accomplish in their work. This includes the determination of age, race, sex, and stature. These four components of the biological profile aid in the identification of an individual in the forensic context. Since the beginning of the field of physical anthropology, osteologists and anatomists have studied human remains in order to provide new and more accurate ways of building the biological profile. This research was conducted to determine the effects of pre-incubation warming on the hatchability of post-peak egg production broiler breeder eggs. An experiment with six trials was conducted with 7,920 freshly laid eggs from Ross 308 and 708 broiler breeders from 61-67-wks of age. For each trial, 1,320 eggs were used to determine if pre-incubation warming treatments of 0, 2, 4, and 6, or 0, 3, 6, and 9, or 0, 9, 12 and 15 hrs (at 37.6°C) could improve the hatchability of eggs stored (at 15.5°C) for three days. A conditional silencing system using anti-gB siRNAs was devised to investigate the structure and function of the KSHV gB. Transient co-transfection of plasmids constitutively expressing gB and anti-gB siRNAs in 293 cells substantially inhibited gB mRNA levels and protein production. Similarly, transient expression of siRNAs into the basal cavity-based lymphoma cells (BCBL-1) caused substantial reduction of gB transcription and protein synthesis. TaqMan real-time PCR and infectivity assays showed that gB was essential for virion egress and infectivity. Transfection of a codon-optimized gB not recognized by the anti-gB siRNAs, efficiently rescued virion egress and infectivity in BCBL-1 cells in the presence …

Adiponectin is a hormone secreted from adipocytes that plays an important role in insulin sensitivity and fatty acid oxidation. The secretion of adiponectin from adipose tissue has been shown to be attenuated by treatment with prolactin (PRL). PRL is a potent STAT5 activator, and studies have indicated that it can modulate the expression of several genes in adipocytes. In this study, we demonstrate that 3T3-L1 adipocytes treated with PRL exhibit a reduction in adiponectin levels. The purpose of this research is to identify key components of community wildfire risk reduction programs and potential influences on the adoption of these program elements. Community wildfire programs have been developed to educate and encourage property owners to adopt “vegetation management” practices such as creating defensible space around structures, landscaping with fire-resistant plants, and removing potential wildfire fuels such as trees and shrubs. This study assessed the efficacy of the ozonation process in degrading aflatoxin in corn, and investigated the chemical reaction between aflatoxin and gaseous ozone. Ozonation (12-13 wt%) totally degraded aflatoxin B1 in a model system. Conversion of aflatoxin into polar compounds was observed during ozonolysis of 100 µg aflatoxin B1 in an aqueous environment and in solid form. Seven intermediate reaction products were separated by two-dimensional thin layer chromatography. HPLC analysis of ozonated AFB1 revealed the presence of six major peaks. MALDI-MS analysis detected compounds that have higher molecular weights than AFB1. The dichloromethane fraction contained compounds with molecular ion …

The global conformations of DNA polymerase I from Thermus aquaticus (Taq/Klentaq) and E. coli (Pol I/Klenow) both in isolation and in complex with DNA have been examined in solution using hydrodynamic (analytical ultracentrifugation) and small/wide angle X-ray scattering techniques and then compared to their known crystal structures to assess the similarities and differences in the overall structure of these enzymes and enzyme complexes within a solution environment. This study examined the reinforcing value of caffeine and food in a sample of 14 normal-weight females who indicated some degree of dietary restraint, and consumed caffeine daily. Eligible individuals participated in two sessions, one of which required a 24-hour fast. During both sessions, participants completed measures assessing caffeine withdrawal symptoms, urge to drink caffeine, hunger, and a multiple-choice questionnaire (MCQ) requiring them to earn points towards either snack foods or caffeinated beverages. There were no significant differences between the fasting and non-fasting conditions on MCQ scores, though the means were in the predicted direction (i.e., participants appeared to work …

Heteroepitaxial nanostructures have a diverse array of applications and show novel phenomena that arise from the exotic physics exhibited in reduced dimensions. We have investigated two nano-structured systems in order to gain insights into the dynamics of their nucleation, growth and have observed striking differences, due in part to a competition between lattice strain, surface and interfacial free energies. When Ag is deposited on clean, single-crystal Cu(110), it initially wets the surface with a (111) monolayer, and spontaneously nucleates nanowires as the coverage is increased. The nanowires nucleate at defects and step edges and grow aligned along the [1–10] direction. …

Mitochondrial dysfunction has been linked to numerous chronic diseases. The antioxidant quercetin has been shown to protect against damage caused by exogenous and endogenous free radicals. Material properties of industrial polymer blends are of great importance. X-ray tomography has been used to obtain spatial chemical information about various polymer blends. The spatial images are acquired with synchrotron X-ray tomography because of its rapidity, good spatial resolution, large field-of-view, and elemental sensitivity. The spatial absorption data acquired from X-ray tomography experiments is converted to spatial chemical information via a linear least squares fit of multi-spectral X-ray absorption data. Because of their high preservation potential and uses in foraging and defense, decapod crustacean dactyls (movable fingers of claws) are potentially excellent test subjects for an ongoing debate concerning the relative importance of top-down (predators) and bottom-up (prey) controls on morphologic diversification and evolution. In this work, a new model relating temperature and pressure to dielectric relaxation or viscosity of a glassformer is developed. The model is based upon cluster kinetics, reaction-like mechanisms describing interactions between glassformer monomers and their clusters. Mathematical solutions of population balance equations for monomer and cluster lead to molar concentrations in terms of rate coefficients of the reaction-like mechanisms. These are then related to viscosity or dielectric relaxation time through free volume theory. The synthesis and sequential surface characterization of silica-polypeptide composite particles is described. Nearly monodisperse colloidal silica cores were obtained by the alkaline hydrolysis of tetraethyl orthosilicate (TEOS). The hydrodynamic radius can be easily controlled in the range of ƒî 20-140 nm by the water/TEOS ratio in the starting reaction mixture. The surface of the synthesized silica cores was further passivated/functionalized (P/F) by a mixture of (3-aminopropyl) trimethoxysilane (APS) and methyl-trimethoxysilane (MTMS).
