Exercise is a first-line treatment for type 2 diabetes and preserves b-cell function by hitherto unknown mechanisms. We postulated that proteins fromcontracting skeletalmuscle may act as cellular signals to regulate pancreatic b-cell function. We used electric pulse stimulation (EPS) to induce contraction in C2C12myotubes and found that treatment of b-cells with EPS-conditioned medium enhanced glucose-stimulated insulin secretion (GSIS). Transcriptomics and subsequent targeted validation revealed growth differentiation factor 15 (GDF15) as a central component of the skeletal muscle secretome. Exposure to recombinant GDF15 enhanced GSIS in cells, islets, andmice. GDF15 enhanced GSIS by upregulating the insulin secretion pathway in b-cells,whichwas abrogated …

Quality Of Farmed Shrimp (Penaeus Monodon, Fabricius, 1798 And Macrobrachium Rosenbergii, Deman, 1879) As Affected By Melanosis Inhibiting Compounds, M. Khan, J. A. Lively Aug 2023

Quality Of Farmed Shrimp (Penaeus Monodon, Fabricius, 1798 And Macrobrachium Rosenbergii, Deman, 1879) As Affected By Melanosis Inhibiting Compounds, M. Khan, J. A. Lively

Faculty Publications

A problem associated with shrimp is blackspot or melanosis and to prevent blackspot, sulfites have been used for years but come with additional health risks. Newer 4-hexylresorcinol formulations exist that allow sulfite-free shrimp, but effects on the quality of Penaeus monodon Fabricius, 1798, and Macrobrachium rosenbergii, de Man, 1879, beyond melanosis prevention are unknown. Adverse effects on the quality of the shrimp from melanosis prevention treatments would negatively offset the benefits of a sulfite-free product. In order to determine if prevention compounds (sulfite powder or 4-hexylresorcinol based compounds, Everfresh® and Xyrex® Prawnfresh™) affect quality, proximate composition and total plate count …

When Can Local Bird Detection Radars Best Complement Broad-Scale Early-Warning Forecasts Of Risk Potential For Bird–Aircraft Strikes As Part Of An Integrated Approach To Strike Mitigation?, Melanie R. Colón, Ashley M. Long

Faculty Publications

Worldwide, wildlife–aircraft strikes cost more than US$1.2 billion in aircraft damage and downtime and jeopardize the safety of aircrews, passengers, and animals. Radar has long been used to monitor flying animal movements and can be a useful tool for strike mitigation. In the USA, the Avian Hazard Advisory System (AHAS) is an early-warning system that integrates data from next-generation weather radar (NEXRAD) weather surveillance radars (WSRs) with historic bird occurrence data to quantify avian activity and forecast the relative bird risk within a ~9.3-km radius of military and civilian airfields. Bird detection radars (BDRs) with both horizontal-surveillance and vertical-scanning components …

Context: Carnitine palmitoyltransferase-1A, encoded by the CPT1A gene, plays a key role in the oxidation of long-chain fatty acids in the mitochondria and may be important in triglyceride metabolism. Previous work has shown that high fat intake was negatively associated with CPT1A methylation and positively associated with CPT1A expression. Objective: We aim to investigate the association of DNA methylation (DNAm) at the CPT1A gene with reductions in triglycerides and triglyceride-rich lipoproteins (TRLs) in response to weight-loss diet interventions. Methods: The current study included 538 White participants, who were randomly assigned to 1 of 4 diets varying in macronutrient components. We …

Faculty Publications

The clapper rail (Rallus crepitans), of the family Rallidae, is a secretive marsh bird species that is adapted for high salinity habitats. They are very similar in appearance to the closely related king rail (R. elegans), but while king rails are limited primarily to freshwater marshes, clapper rails are highly adapted to tolerate salt marshes. Both species can be found in brackish marshes where they freely hybridize, but the distribution of their respective habitats precludes the formation of a continuous hybrid zone and secondary contact can occur repeatedly. This system, thus, provides unique opportunities to investigate the underlying mechanisms driving …

Science Diplomacy From The Global South: The Case Of Intergovernmental Science Organizations, Anna Lena Rüland, Nicolas Rüffin, Katharina Cramer, Prosper Ngabonziza, Manoj Saxena, Stefan Skupien

Faculty Publications

Intergovernmental science organizations (IGSOs) address many challenges of the 21st century. Several countries of the Global South have joined established IGSOs or have created new ones. Yet we know little about their interests in IGSOs. Our study addresses this blind spot by investigating which objectives Southern actors pursue in IGSOs and under which conditions they are likely to achieve their objectives. Using insights from three strands of literature, we compare four IGSOs with Southern participation: the European Organization for Nuclear Research, the International Thermonuclear Experimental Reactor, the Square Kilometer Array, and the African Lightsource. We show that countries of the …

The DELLA genes, also known as ‘Green Revolution’ genes, encode conserved master growth regulators that control plant development in response to internal and environmental cues. Functioning as nuclear-localized transcription regulators, DELLAs modulate expression of target genes via direct protein–protein interaction of their carboxy-terminal GRAS domain with hundreds of transcription factors (TFs) and epigenetic regulators. However, the molecular mechanism of DELLA-mediated transcription reprogramming remains unclear. Here by characterizing new missense alleles of an Arabidopsis DELLA, repressor of ga1-3 (RGA), and co-immunoprecipitation assays, we show that RGA binds histone H2A via the PFYRE subdomain within its GRAS domain to form a TF–RGA–H2A …

Deregulated protein kinases are crucial in promoting cancer cell proliferation and driving malignant cell signaling. Although these kinases are essential targets for cancer therapy due to their involvement in cell development and proliferation, only a small part of the human kinome has been targeted by drugs. A comprehensive scoring system is needed to evaluate and prioritize clinically relevant kinases. We recently developed CancerOmicsNet, an artificial intelligence model employing graph-based algorithms to predict the cancer cell response to treatment with kinase inhibitors. The performance of this approach has been evaluated in large-scale benchmarking calculations, followed by the experimental validation of selected …

The national K-12 science standards were reformed in April 2013. On March 8, 2017, the state of Louisiana’s Board of Elementary and Secondary Education approved the rewrite of the science standards to align with these national standards, and they were implemented in the middle school science classes. In March 2020, the COVID-19 pandemic began and thrust all students into the virtual learning world. The purpose of this study was twofold: to investigate certain changes made to the science standards in Louisiana and to examine how the pandemic interrupted NGSS implementation in the classroom both with teachers who were familiar with …

Modulating Water Distribution And The Intramicellar Assembly Of Sequence-Defined Ionic Peptoid Block Copolymers By The Ionic Monomer Position, Bailee N. Barrett, Chi Huan Tung, Guan Rong Huang, Istiak Hossain, Chang Woo Do, Vijay T. John, Wei Ren Chen, Donghui Zhang Jul 2023

Modulating Water Distribution And The Intramicellar Assembly Of Sequence-Defined Ionic Peptoid Block Copolymers By The Ionic Monomer Position, Bailee N. Barrett, Chi Huan Tung, Guan Rong Huang, Istiak Hossain, Chang Woo Do, Vijay T. John, Wei Ren Chen, Donghui Zhang

Faculty Publications

The intramicellar mass heterogeneity of a series of sequence-defined ionic peptoid block copolymers carrying a single charged monomer has been determined through contrast variation small-angle neutron scattering analysis. We observe that the internal micellar structure, namely, the number density radial distributions of invasive water and peptoid polymer, is significantly impacted by the location of the ionic monomer. By positioning the ionic monomer progressively closer to the hydrophilic/hydrophobic block junction, the micelles become less compact with increasing levels of chain folding and invasive water to accommodate electrostatic repulsion among the ionic monomers via solvation. This results in increasingly smaller micellar aggregates …

Physiologically Relevant Fluid Shear Stress Induced Receptor Conversion In Hormone Receptor-Positive (Hr+) Breast Cancer, Jonathan James Cuccia Jul 2023

Physiologically Relevant Fluid Shear Stress Induced Receptor Conversion In Hormone Receptor-Positive (Hr+) Breast Cancer, Jonathan James Cuccia

LSU Doctoral Dissertations

Breast cancer is the most common type of cancer among all people in the United States as of 2023 and often has devastating impacts on afflicted patients. Although some treatments show great efficacy against primary breast tumors, none have been able to significantly reduce the mortality rate of breast cancer once it has metastasized to distant sites. In primary, hormone receptor-positive (HR+) breast cancers, endocrine therapies are often used as first-line treatments and are generally well-tolerated. However, as the disease progresses, these previously sensitive cancers can become resistant to endocrine therapies and continue proliferating despite them. Therefore, there is a …

Defeat, Entrapment, And Suicide Desire: Analyzing The Role Of Mindfulness Using Ecological Momentary Assessment., Emma H. Moscardini Jul 2023

Defeat, Entrapment, And Suicide Desire: Analyzing The Role Of Mindfulness Using Ecological Momentary Assessment., Emma H. Moscardini

LSU Doctoral Dissertations

Extant literature has indicated that the experience of defeat confers risk for experiencing conceptions of entrapment, which in turns increases one’s risk of experiencing suicidal ideation. In addition, certain cognitive processes are hypothesized to increase or decrease the likelihood of feelings of entrapment emerging when one feels defeated. One such cognitive process is mindfulness, defined as present moment awareness with an orientation of openness and acceptance. Better understanding the interplay of defeat, entrapment, mindfulness, and suicidal thinking will contribute to intervention strategies aimed at reducing the severity of suicidal thinking; however, said research should be conducted with consideration that one’s …

Glucose is the basic fuel essential for maintenance of viability and functionality of all cells. However, some neurons —namely, glucose-inhibited (GI) neurons — paradoxically increase their firing activity in low-glucose conditions and decrease that activity in high-glucose conditions. The ionic mechanisms mediating electric responses of GI neurons to glucose fluctuations remain unclear. Here, we showed that currents mediated by the anoctamin 4 (Ano4) channel are only detected in GI neurons in the ventromedial hypothalamic nucleus (VMH) and are functionally required for their activation in response to low glucose. Genetic disruption of the Ano4 gene in VMH neurons reduced blood glucose …

Genetic Dissection Of Alkalinity Tolerance In Rice (Oryza Sativa L.), Lovepreet Singh Jul 2023

Genetic Dissection Of Alkalinity Tolerance In Rice (Oryza Sativa L.), Lovepreet Singh

LSU Doctoral Dissertations

Alkaline stress is a major threat to rice production globally. The resources for genetic improvement of alkalinity tolerance in rice are limited. We performed genome-wide association study (GWAS) and QTL mapping integrated with whole genome sequencing and expression analysis to identify and confirm QTLs and candidate genes. We developed a near-isogenic line (NIL) population to fine map the two important QTLs. In GWAS experiment, we genotyped and phenotyped a panel of indica and japonica genotypes for alkalinity tolerance and identified genomic regions and candidate genes conferring alkalinity tolerance. Principal component analysis revealed that alkalinity tolerance score, shoot dry weight, and …

An Observed Increase In Cardiovascular And Respiratory-Disease Related Hospitalizations And Mortality In An Area Surrounding A Hazardous Waste Thermal Treatment Plant, Liana Isabelle Dela Torre Baconguis Jul 2023

An Observed Increase In Cardiovascular And Respiratory-Disease Related Hospitalizations And Mortality In An Area Surrounding A Hazardous Waste Thermal Treatment Plant, Liana Isabelle Dela Torre Baconguis

LSU Master's Theses

Particulate matter (PM) pollution is highly correlated with respiratory and cardiovascular disease incidence. The generation of oxidative stress is a major way by which PM can damage the body. Environmentally persistent free radical-containing PM are generated by combustion processes and are capable of exerting increased damage on the respiratory and cardiovascular systems due to their ability to remain stable and regenerate reactive oxygen species for up to five years. Clean Harbors, LLC operates a thermal treatment plant near the city of Colfax, LA in Central Louisiana. Its regular open burning of explosives causes disturbances to the community, such as loud …

Wave Attenuation Through Mangrove Forest Using Lidar Representation, Elizabeth Bogan Jul 2023

Wave Attenuation Through Mangrove Forest Using Lidar Representation, Elizabeth Bogan

LSU Master's Theses

Mangrove forests can be found along the world’s intertidal tropical and sub-tropical coastlines. These forested wetlands provide many ecosystem services, including supporting coastal food chains, trapping sediment, and shoreline stabilization. Mangroves are also known for flood damage mitigation by dissipating waves and surge during flooding events. Thus, coastal communities declare a vested interest in the performance of the forest to mitigate flood damage. Physical models using artificial mangrove structures have shown that red mangrove-dominated fringe forests can protect built infrastructure by reducing wave height due to increased drag. One way to estimate the contribution of red mangrove (Rhizophora mangle) prop …

Women's Interracial Friendships: Cultivating Coalitional Possibilities Across Difference Through Dialogue, Akie Fukushige Wenk Jul 2023

Women's Interracial Friendships: Cultivating Coalitional Possibilities Across Difference Through Dialogue, Akie Fukushige Wenk

LSU Doctoral Dissertations

In my dissertation, I thread through the macro-structure as well as micro-operations of hegemonic ideologies, namely white-supremacist capitalist cis-hetero patriarchy (hooks, 1995, Valdes, 1996), that has historically plagued the United States, eventually gaining a global reach over time. I center interracial friendship between Black and White cis-women, where I locate coalitional possibilities that begin at the grassroots level. I situate this interracial relationship within the socio-ecological system where the personal meets the social (including racialized backgrounds). I use a series of analogies to equate these ideologies as toxins in the air, water, and soil. Soil is where we can work …

Morphological And Molecular Characterization Of The Olfactory System In The Formosan Subterranean Termite, Paula Castillo Jul 2023

Morphological And Molecular Characterization Of The Olfactory System In The Formosan Subterranean Termite, Paula Castillo

LSU Doctoral Dissertations

The Formosan subterranean termite, Coptotermes formosanus, is a eusocial insect species. Its members live in colonies consisting of reproductive (queen and king) and non-reproductive individuals (workers and soldiers). Olfaction is an essential sense for the social life of many insects. However, in termites, little is known about how olfactory cues are perceived in the peripheral sensory system (antennae) and processed in the central nervous system (brain) to elicit different behavioral responses across castes. In this study, we investigated the olfactory system in all castes (soldiers, workers, and reproductive alates), from the antennal and brain morphology to the antennal gene …

Tissue Resident Regulatory T Cells And Immunopathology During Influenza Infections, Michael Mcgee Jul 2023

Tissue Resident Regulatory T Cells And Immunopathology During Influenza Infections, Michael Mcgee

LSU Doctoral Dissertations

Influenza (flu) virus infections are a leading cause of respiratory illness. Globally there are approximately 290,000-650,000 deaths and 3-5 million cases of severe illness during the average flu season. Severe flu infection is associated with the combination of a strong pro-inflammatory and weak anti-inflammatory response. Regulatory T cell subsets suppress pro-inflammatory responses during flu infection, with production of the immunomodulatory cytokine IL-10 playing a critical role in this process. Tissue resident regulatory T cell subsets are maintained within nonlymphoid tissues and are optimally positioned to suppress localized pro-inflammatory responses. However, knowledge of the signaling pathways regulating the development of tissue …

Predicted Structure And Functional Roles Of Alphaherpesvirus Tegument Protein Ul37 In Virion Morphogenesis And Innate Immunity, Therese Marie Collantes

LSU Doctoral Dissertations

Several members of the Family Herpesviridae are clinically significant pathogens, affecting humans and a wide range of animals. Alphaherpesviruses infect epithelial cells at the primary site of infection and subsequently invade adjacent sensory neurons. Infectious bovine rhinotracheitis (IBR) is an economically important disease caused by bovine herpesvirus 1 (BoHV-1), which belongs to the subfamily Alphaherpesvirinae, genus Varicellovirus. To date, vaccination is the only preventive measure against IBR. Understanding determinants of herpesvirus replication and innate immune regulation is important to help further develop vaccines that are efficacious and incapable of establishing latency in the host nervous system. The alphaherpesvirus …

Neural Mechanisms Of Resiliency And Susceptibility After Repeated Social Defeat, Christy R. Wayne Jul 2023

Neural Mechanisms Of Resiliency And Susceptibility After Repeated Social Defeat, Christy R. Wayne

LSU Doctoral Dissertations

Social defeat is a powerful experience leading to drastic changes in physiology and behavior, many of which are negative. For example, repeated social defeat in vertebrates results in reduced reproductive success, sickness, and behavioral abnormalities that threaten individual survival and species persistence. However, little is known about what neural mechanisms are involved in determining whether an individual is resilient or susceptible to repeated social defeat stress. I used a resident-intruder experiment in the African cichlid fish, Astatotilapia burtoni, to create and investigate the behavior and neural correlates of these two opposing groups. Resilient fish used both searching and freezing behaviors …

Sportsman's Paradox: Conservationism And Social Progress In Modern Louisiana, Jacob T. Gautreaux Jul 2023

Sportsman's Paradox: Conservationism And Social Progress In Modern Louisiana, Jacob T. Gautreaux

LSU Doctoral Dissertations

Over the nineteenth and early twentieth centuries, sportsmen increasingly identified Louisiana as a destined paradise due to the abundant flora and fauna. Confirmed in the legendary visits of Theodore Roosevelt in the early 1900s, the conception soon served a dual purpose as individuals like the Tabasco Sauce patriarch, E. A. McIlhenny, coopted the visualization as a lure for business investment into the nascent industrial interests within the coastal region of the state. However, it should be noted that in the 1930s and beyond, cultural conservationists like McIlhenny and Caroline Dormon preserved elements of under-documented cultures throughout the state, although usually …

Reef Fish Movement Dynamics And Recruitment To Artificial Habitat, Jade Carver Jul 2023

Reef Fish Movement Dynamics And Recruitment To Artificial Habitat, Jade Carver

LSU Master's Theses

Artificial reefs provide critical habitat for fish in areas lacking benthic structure, yet our understanding of how artificial reefs function and develop is limited. Here, changes in reef fish community assemblages were monitored using baited remote underwater video (BRUV) surveys (n=6) before and after a new artificial reef was deployed. Movement of red snapper (Lutjanus campechanus, n=20) and gray snapper (Lutjanus griseus, n=20) between nearby platforms (n=3) and the new artificial reef was examined using acoustic telemetry, and residency was calculated for fish associated with platforms and the artificial reef …

The Role Of Metallothioneins In Copper Homeostasis In Arabidopsis, Matthew Vandal Jul 2023

The Role Of Metallothioneins In Copper Homeostasis In Arabidopsis, Matthew Vandal

LSU Doctoral Dissertations

Copper (Cu) is a transition metal that is an essential nutrient for plants and is required for a number of critical processes. Therefore, proper homeostasis of copper is vital for plant life and must be tightly regulated. Metallothioneins are small molecular weight proteins, rich in cysteine residues and have the capability of binding to several different transition metals such as copper. Cu induces the expression of MTs, therefore they are believed to be involved in Cu transport, however their true functions have yet to be resolved. T-DNA insertions were identified in four Arabidopsis MT genes expressed in vegetative tissues, and …

Evaluation Of Salvinia Weevil Establishment, Dispersal, And Environmental Pressures Impacting The Giant Salvinia Biological Control Program, Korey Danielle Pham

LSU Master's Theses

Giant salvinia (Salvinia molesta Mitchell), an invasive free-floating aquatic fern, invaded the Toledo-Bend reservoir along the Texas-Louisiana border in the 1990s. The rapid spread threatens critical ecosystems, economies, and cultural services by altering water quality, and impacting fishing, hunting, and navigation. To combat this issue, the LSU AgCenter has been rearing and distributing the salvinia weevil (Cyrtobagous salviniae Calder and Sands), which has demonstrated effectiveness in reducing giant salvinia populations within a matter of months to a few years. The first study assessed the impact of propagule pressure on salvinia weevil effectiveness in outdoor mesocosms and revealed that …

Learning Dynamic Information Of High-Dimensional Signal Time-Series Using Advanced Machine Learning/Artificial Intelligence, Guannan Liu

LSU Doctoral Dissertations

In this dissertation, we propose a novel simulation-based device-free indoor localization and tracking system using the received signal strength indicators (RSSIs) of WiFi signals as the input features. The Feko channel-propagation simulation software is used to process the RSSI maps of the given arbitrary indoor geometry. In order to learn the dynamic information of high-dimensional RSSI time-series, we propose three procedures for the localization and dynamic tracking system.

First, The indoor geometry is partitioned into several equi-size zones and the localization problem is treated as the typical \multi-classification" problem. The advanced machine-learning techniques such as decision tree (DT) classifier, random …

Integrating Cultivar Resistance And Entomopathogens For Pest Management Of Sweetpotato Weevil, Cylas Formicarius Elegantulus (Summers), Schyler M. Lee Jul 2023

Integrating Cultivar Resistance And Entomopathogens For Pest Management Of Sweetpotato Weevil, Cylas Formicarius Elegantulus (Summers), Schyler M. Lee

LSU Master's Theses

Sweetpotato, Ipomoea batatas (L.) Lam, is the number one vegetable crop in Louisiana, producing 21,178 kg/ha in the year 2021. The sweetpotato weevil, Cylas formicarius elegantulus (Summers), is an economically important pest of sweetpotato. C. formicarius elegantulus females lay eggs into feeding holes on the storage root surface. Larvae then hatch and tunnel through the sweetpotato flesh where they complete their development. This cryptic-natured brentid can cause up to 100% yield loss when left unmanaged. The Louisiana Department of Agriculture and Forestry mandates protocols to limit C. formicarius elegantulus spread (e.g., clean seed, pheromone trapping, and quarantines); however, scheduled chemical …
